1.Effect of Cluster Scalp Acupuncture on Pathological Changes and Expression of Nerve Growth Factor and Transforming Growth Factor in Rats after Acute Cerebral Infarction

Objective To explore the effect of cluster scalp acupuncture on pathological changes and the expression of nerve growth factor (NGF) and transforming growth factor (TGF) in rats after acute cerebral infarction.Methods30 male Wistar rats were randomly divided into the sham operation group (group A), model group (group B) and cluster scalp acupuncture group (group C) with 10 animals in each group. Animal model was made by the middle cerebral artery occlusion. The pathological changes and the expression of NGF and TGF in the brain tissues of rats were tested.ResultsOn the 7th day after operation, brain tissue of infarction area in the group B indicated significantly edema, neural cells decreased. Compared with the group B, the edema in the group C decreased and neural cells increased. At the same time, there were a few NGF and TGF expressions in hippocampus andcortex areas of the group A and group B, and there was no difference between the two groups ( P>0.05); but compared with the group A and group B, NGF and TGF in the group C increased significantly ( P<0.05, P<0.01).ConclusionCluster scalp acupuncture therapy can decrease edema, increase neural cells in infarction areas, as well as induce the expression of NGF and TGF in cortex and hippocampus.
2.Cerebral functions and immunohistochemical changes caused by rehabilitation in rats after infarction

ObjectiveTo study the rehabilitation mechanism of acute cerebral infarction.Methods30 male Wistar rats were randomly divided into 3 groups:group A(sham group),group B(model group) and group C(rehabilitation group).Animal model was made by the middle cerebral artery occlusion(MCAO).For each group,Bederson Neural Function was scored and balancing wood,rotating bar,and net screen were tested24 hours,3 days and 7 days after operation respectively.The nerve growth factor(NGF),brain derived neurotrophic factor(BDNF),transforming growth factor(TGF),basic fibroblast growth factor(bFGF)in brain tissues were tested.ResultsBederson neural function scores of group A had no changes,while those of the other two groups increased after 24 hours and had a tendency to restore gradually with time continuing,but indicated significant differences at each time,compared with group A(P<0.01).The abilities of grasping ,walking and coordinating of all the groups after 24 hours by operation handicapped,and there was no difference among the groups(P>0.05);with time continuing,every function of group A restored soon and that of group B or C improved,to some degree,but showed significant difference to group A(P<0.01).After 3 days by operation,balancing wood test of group C was better than that of group B(P<0.05);after 7 days,except Bederson neural function score,there was difference between the group C and B,that is,the former was superior to the latter. After 7 days by operation,there were a few NGF,BDNF,TGF and bFGF expressions in hippocampus and cortex areas of group A and B,and there was no difference between this two groups(P>0.05),while those of group C greatly increased,compared with group A and B(P<0.05-0.01).ConclusionsRehabilitation therapy can improve the MCAO rats' ability of equilibrating,grasping and walking and strengthen the myodynamia,as well as induce the expression of NGF,BDNF,bFGF and TGF in cortex and hippocampus.
3.Immunofecal occult blood test predicts the prognosis of patients with chronic kidney disease

Objective To evaluate the value of immunofecal occult blood test (IFOBT) as a prognostic indicator in CKD patients with colorectal impairment.Methods A total of 176CKD patients and 180 healthy adults as control were enrolled.Serum biochemistry was measured at baseline and gastrointestinal bleeding was determined by IFOBT.All the CKD patients were followed up for 4.5 years.Renal replacement therapy or death was defined as end-point event.The Logistic regression analysis was used for risk factors.Kaplan-Meier analysis and COX regression model were used for survival analysis.Results The positive rate of IFOBT in CKD patients was significantly higher than healthy control (17% vs 5.3%,χ2=13.236,P<0.01).When comparing with IFOBT negitive patients,IFOBT positive patients were older [(62.030±15.544) years old vs (48.660±19.018)years old,P<0.01],had higher ESR [(71.800±31.657) mu/h vs (57.210±32.712) mm/h,P<0.05],C-reactive protein [6.230 (3.000~14.148) mg/L vs 3.000 (3.000~6.833)mg/L,P<0.05],serum creatinine [419.100 (103.200~546.625) μmol/L vs 175.100 (68.150~462.950) μmol/L,P<0.05],and had lower hemoglobin level [(97.970±20.590) g/L vs (107.170±27.988) g/L,P<0.05] and eGFR [11.400 (8.671~53.544) ml·min1·(1.73 m2)1 vs 35.274(10.961~82.145) ml·min-1·(1.73 m2)-1,P<0.01].There was a negative correlation between IFOBT value and eGFR in CKD patients (r=-0.20,P<0.01).Positive correlations of IFOBT value with age (r=0.175,P<0.05) and serum creatinine (r=0.171,P<0.05) were found.Logistic regression and COX regression analysis showed that IFOBT value,eGFR and ESR were important factors that influenced the prognosis of CKD patients.Kaplan-Meier analysis revealed that IFOBT value >100μg/L predicted progression of renal function.Conclusions The prevalence of gastrointestinal bleeding disorder is high in patients with CKD.Value of IFOBT independently predicts decline in renal function of CKD patients.
4.Effects of rehabilitation training on behavior and sensory evoked potential of rats with acute cerebral infarction

ObjectiveTo observe effects of rehabilitation training on behavior and sensory evoked potential of rats with acute cerebral infarction.Methods30 male Wistar rats were randomly divided into sham operation groups (group A), model group (group B), and rehabilitation group (group C). Animal model was made by blocking the middle cerebral artery (MCA). For each group, the Bederson Neural Function was scored and balancing wood, rotating bar, and net screen were tested at the 24 hours, 3rd day and 7th day after operation respectively. The sensory evoked potential (SEP) latency of each group was also tested right after operation and on the 7th day after operation.ResultsComparing with group A, Bederson Neural Function scores of groups B and C indicated significant differences at each time (P<0.01). The ability of grasping, walking and coordinating of all groups handicapped 24 hours after operation, however, with time continuing, every function of group A restored and that of group B or C improved partly, but showed significant difference compared with group A (P<0.01). On the 3rd day after operation, balancing wood test of group C was better than that of group B (P<0.05), on the 7th day after operation, except Bederson Neural Function scores, there were significant difference between groups C and B (P<0.01 or P<0.05). Right after operation, compared with group A, the SEP latency of group B and C showed significant difference (P<0.01), on the 7th day after operation, the SEP latency of group C improved significantly compared with itself right after operation (P<0.01).ConclusionThe rehabilitation training can improve the MCAO rats' ability of equilibrating,grasping and walking and adjust the abnormal latency of SEP after MCAO, and prevent the nerve cells from damage caused by ischemic trauma.
5.Clinical observation on two-way quintuple puncture in the treatment of ganglion.

OBJECTIVETo explore the better therapy in the treatment of ganglion.
METHODSNinety cases of ganglion were randomized into a two-way quintuple puncture group, a common quintuple puncture group and a fire needling group, 30 cases in each one. In the two-way quintuple puncture group, the "9-in-1" multiple penetrating needling technique was used. In the common quintuple puncture group, the traditional "5-in-1" multiple penetrating needling technique was applied. In the fire needling group, the traditional multiple fire needling technique was adopted. The treatment was given once a day, 3 treatments made one session and the efficacy was analyzed statistically after 1 session treatment in the three groups.
RESULTSAll of the three therapeutic methods achieved the efficacy on ganglion. The curative rate was 96. 7% (29/30) in the two-way quintuple puncture group, which was better obviously than 66.7% (20/30) in the common quintuple puncture group and 60. 0% (18/30) in the fire needling group (both P<0. 01).
CONCLUSIONThe two-way quintuple puncture technique achieves the remarkably superior efficacy on ganglion as compared with the common quintuple puncture technique and fire needling technique.

6.Comparative study on schizontocidal activity of recrystallized or crude daphnetin against malaria parasites.

OBJECTIVETo compare the schizontocidal activity of recrystallized or crude daphnetin against malaria parasites in vivo.
METHODSSchizontocidal activity of recrystallized or crude daphnetin at various dosages was assessed in mice infected with Plasmodium berghei ANKA using a "4-day suppress assay".
RESULTSThe comparison of the reduction rate of parasitemia caused by either recrystallized or crude dephnetin showed that ED(50) of crude daphnetin was 18.36 mg/kg, with 95% confidence limit of 5.96-56.54 mg/kg while ED50 of recrystallized daphnetin was 11.46 mg/kg, with 95% confidence limit of 8.63-15.22 mg/kg.
CONCLUSIONThe results indicate that the efficacy of recrystallized daphnetin is 37.6% higher than that of crude daphnetin.

7.Application of PCR-based tRNAval Pol III-shRNA expression cassettes in screening of efficient siRNA for inhibiting hepatitis B virus.

OBJECTIVETo screen efficient siRNA for inhibiting hepatitis B virus using the technique of PCR-based tRNA(val) Pol III-shRNA expression cassettes (SECs).
METHODSBased on core gene sequence of HBV, five target sites of siRNA were designed. tRNAval Pol III-shRNA expression cassettes produced by one-step overlapping extension PCR strategy were co-transfected with HBV C gene and pC-EGFP plasmid into AD293 cells respectively. Forty-eight hours after transfection, fluorescence of HBVC-GFP protein was detected by fluorescence-activated cell sorting (FACS); HBV C mRNA was detected by semi-quantitative RT-PCR. HBV-producing HepG2. 2. 15 cells were transfected with selected SECs for 72 h, HBsAg and HBeAg in the cell culture medium were detected by radioimmunoassay assay (RIA). HBV pgRNA from cell total RNA was detected by semi-quantitative PCR.
RESULTCo-transfection with pC-GFP plasmid and SECs into AD293 cells resulted in inhibition expression of HBV C gene and decrease of EGFP fluorescence intensity. SEC-492i showed most significant inhibition effect on HBV C-EGFP expression compared with other SECs. Selected SEC-492i or SEC-282i targeting core gene could efficiently decrease expression of HBeAg and the level of HBV pgRNA in a dose-dependent manner. SEC-492i inhibited HBV replication and antigen expression in a more efficient way than SEC-282i at the same final concentration.
CONCLUSIONThe expressed shRNA, which targets sites on HBV C mRNA in 492i, is to have having most efficient RNAi effect. tRNAval Pol III-shRNA expression cassettes produced by one-step overlapping extension PCR strategy should be useful for identification of optimal siRNA.

8.Primary non-Hodgkin's lymphoma of the testis and penis: clinical analysis of 5 cases.

OBJECTIVETo improve the clinical diagnosis and treatment of primary non-Hodgkin's lymphoma of male genitalia.
METHODSWe retrospectively reviewed the clinical data of 5 cases of primary non-Hodgkin's lymphoma of male genitalia, 4 in the testis and 1 in the penis, we also analyzed the relevant literature and clinical significance of the disease.
RESULTSAll the 5 cases were treated by surgery and pathologically confirmed to be non-Hodgkin's lymphoma. Three of them received chemotherapy, and the other 2 (1 in the testis and 1 in the penis) underwent both chemotherapy and radiotherapy after the operation. Follow-up averaged 25 months, during which 1 of the patients died and the other 4 survived.
CONCLUSIONPrimary non-Hodgkin's lymphoma of male genitalia is an uncommon disease with atypical clinical presentations and poor prognosis, which occurs mostly in elderly males. Definite diagnosis of the disease mainly depends on histopathology and immunohistochemistry. Surgery with multiagent chemotherapy and radiotherapy is advisable for its treatment.

9.Inhibition of HBs-GFP fusion gene expression by RNA interference.

OBJECTIVETo develop an effective report gene system to test the effect of small interfering RNA (siRNA).
METHODSHBV S gene was fused with enhanced green fluorescent protein (EGFP) gene to form HBs-GFP and the plasmid containing HBs-GFP was constructed. A vector expressing small hairpin RNA (shRNA) pAVU6 + 4sh357 was also constructed. Two plasmids were co-transfected into HepG2 cells transiently. The fluorescence of HBs-GFP was detected by fluorescence-activated cell sorting (FACS). The mRNA expression in HepG2 cells was detected by conventional RT-PCR and real-time PCR.
RESULTSsiRNA inhibited the expression of HBs-GFP 72 hours post transfection. The fluorescence of HBs-GFP in HepG2 cells treated with pAVU6+4sh357 was reduced by 55.4% compared with that of controls. The HBs-GFP expression in HepG2 cells treated with pAVU6+4sh357 was reduced by 76.3% and 90% as measured with conventional RT-PCR and real-time PCR, respectively.
CONCLUSIONThis investigation demonstrated siRNA derived from shRNA expression vectors can inhibit the expression of HBs-GFP in HepG2 cells.

10.Establish a screening system for selection of mRNA target sites for HBsAg to construct siRNA with shRNA.

OBJECTIVETo find some effective short interfering RNA's sites targeting HBV surface gene sequence using shRNA expression vectors.
METHODSFour shRNA expression vectors targeting HBV surface gene sequence were constructed based on pAVU6 + 27 vector, and cotransfected into AD293 cells with HBs-EGFP fusion gene plasmid. The changes of HBs-EGFP image were detected by FACS and microscopy. The HBs-EGFP mRNA expression was evaluated by RT-PCR.
RESULTSFour shRNA expression vectors and HBs-EGFP fusion gene plasmid were successfully constructed. pAVU6 + 4sh579 vector inhibited the HBs-EGFP expression by 69.8% in AD293 and suppressed the HBs-EGFP mRNA expression by 74.6%.
CONCLUSIONSThe results showed that the 579 site of HBV surface gene sequence was an effective target and pAVU6 + 4sh579 vector could suppress the HBs-EGFP expression in AD293 cells
