Objective:To evaluate the toxic effect of 3 different gingival retraction cords.Methods:DMEMextraction of DL-adren-aline HCl,aluminium sulphate and non-drug retraction cords with the extraction time of 5,10,15 and 30 min were respectively pre-pared and were used to culture human gingival fibroblasts(HGFs)in vitro respectively.Cell proliferation was tested by MTT assay. Cell apoptosis was examined by Annexin/PI method.Results:The 3 gingival retraction cord extractions inhibited the roliferation,pro-moted the apoptosis of HGFs(P <0.05),the effects were related to the extraction time.Conclusion:The 3 retraction cords have time-dependant cytotoxity.

Objective To investigate the effects of hippocampal neurogenesis in spatial learning and memory,especially in spatial long-term memory.Methods Totally 24 wistar rats were randomly divided into two groups:normal control group (NC group,n =12) and irradiation group (IRR group,n =12).We use low-dose irradiation at subgranular zone to inhibit adult hippocampal neurogenesis and new neurons were investigated by 5-Bromo-2-deoxyUridine/neuron specific nuclear protein double-labeling.Four weeks after irradiation,Morris water maze,including navigation test and space exploration assay,was used to detect spatial leaming and memory.Then,in the day 8 and day 15 after the navigation test,space exploration assay was repeated to detect spatial long-term memory.The expression levels of postsynaptic density protein 95 and synaptophysin were evaluated using western blot and real-time quantitative PCR.Results Hippocampal neurogenesis was inhibited by low dose irradiation(2.80 ± 0.44 vs 23.50 ± 1.12,t =20.21,P ＜ 0.01).After the inhibition,the escape latency did not change,while the time spent in target quadrant was significantly declined in the day 2((14.76 ±.2.04) s vs (20.47 ± 1.29) s),day 8((11.95 ± 1.34) s vs (19.52±1.43) s)and day 15((11.79 ±1.35) s vs (21.58 ±1.07) s) after the navigation test (t=2.45,P＜0.05; t =3.76,P＜0.01; t =5.39,P＜0.01).The postsynaptic density protein 95 and synaptophysin expressions were significantly decreased in IRR group.As to the RNA levels,there was also a significant difference between the two groups.Conclusion Hippocampal neurogenesis in the dentate gyrus plays an important role for the formation of spatial long-term memory.

Objective To establish a recombinant eukaryotic expression vector containing human interferon-?(IFN-?) cDNA,and investigate the expression of IFN-? gene in transfected hepatocellular carcinoma (HCC) cell lines. Methods pcDNA3-IFN-?,established by subcloning IFN-? cDNA into a mammalian expression vector pcDNA3,was mediated into HCC cell lines SMMC-7721 and QGY-7701 by lipofectamine. The transfected HCC cells were selected in RPMI1640 containing G418(400～700?g).RT-PCR analysis and ELISA assay were used for mRNA transcription and protein expression of IFN-? gene. Results IFN-? mRNA was detected only in pcDNA3-IFN-? transfected cell lines, and high levels of IFN-? protein were detected only in the supernatants of IFN-? gene-modified cell lines. Conclusions pcDNA3-IFN-? can stably expressIFN-? cDNA in the transfected HCC cell strains

This paper investigated contamination situation of Listeria monocytogenes(Lm). To compare dif- ferent selectivity enrichment broth for detectable effect of Lm and compare detectable effect in different samples by using different methods, furthermore, choose the best enrichment broth for specific food. One hundred and thirty five random samples from raw meat, aquatic product, fruit and vegetable, quick-frozen food in Baoding. Applied LB enrichment broth, EB enrichment broth, new modification FDA enrichment broth and Fraser enrichment broth before separated by PALCAM selective agar, then identified by interna- tional standard method after PCR. Results: Four methods showed that there were 23 Lm positive, detected 5 Lm by LB method, 6 Lm by Fraser method, 5 Lm by EB method and 7 Lm by new modification FDA method. The total detectable rate of four methods had no large specificity, but to specific kind of food was different.
