1.Review of the expressed emotion application in the guardian of patients with schizophrenia

Expressed emotion was the describing term about the guardian of patients' emotiveness tendency to patients,which reflected attitude,tolerance degree and the coping way in crisis.It was a robust predictor of relapse of schizophrenia.Studies showed that the family intervention for the high expressed emotion guardian of patients could effectively reduce the disease relapse.By summarizing the expressed emotion's concept,assessment tools,influence factors,family interventions,the paper aimed to innovate perfect interventions and promote the patients recovery.
2.Analysis of bioactive components of novel jujube beverage from Laoling Zizyphus Jujuba cv.Jinsixiaozao

Objective To detect the main nutritional components of novel jujube beverage from Laoling Zizyphus Jujuba cv.Jinsixiaozao.Methods Free monosaccharides in novel jujube juice were determined by the HPLC-RID method while water-soluble polysaccharides were analyzed by UV spectrophotometric method with the phenol-sulfuric acid derivatization.Amino acid and mineral contents were obtained by the automatic amino acid analyzer and atomic absorption photometric methods, respectively.The quantification of total phenolic and flavonoids was accomplished by the UV spectrophotometric method after the respective complexation of the Folin-Ciocalteu reagent and aluminum nitrate.The contents of rutin and cyclic nucleotides were determined by the HPLC-UV method.Results The average contents of sucrose, glucose, fructose and polysaccharides in novel jujube juice were 3.85, 32.5,27.3 and 3.6 mg/mL, respectively.The total amino acid average content was 269.7 μg/mL.The mineral average contents of Ca, Fe, Mg, Zn, Mn, Cr, and Cu were 35.1,0.85, 55.8, 0.85, 0.44, 0.41, 0.21μg/mL, respectively.The novel jujube juice contained average 1.34μg/mL rutin equivalents of total phenolics and average 157.3 μg/mL gallic acid equivalents of total flavonoids.Finally, the average amounts of rutin, cAMP and cGMP were 7.15, 19.17 , and 8.11 μg/mL, respectively.Conclusion The newly developed jujube juice is worthwhile further marketing development attributed to its rich nutritional components and healthy values .

Objective To investigate whether CD4~+CD25~(high)regulatory T cells(tregs)were increased in the tumor tissue and peripheral blood of prostate cancer patients.Methods The prevalence of CD4~+CD25~(high)regulatory T cells inside the prostate tumor was detected compared with benign tissue from the same prostate.Furthermore,the frequency of CD4~+CD25~(high)regulatory T cells in peripheral blood was detected in prostate cancer patients compared with normal donors.Results Moreover,CD4~+CD25~(high)regulatory T cells from blood and supernatants from cultured prostate tumor tissue samples exhibited immunosuppressive function in vitro.CD4~+CD25~(high)T normal peripheral blood cells were found for the ratio was(0.5?0.1)%.Lower than the proportion of T cells in peripheral blood of patients with CD4~+CD25~(high)was(2.3?0.7)%,P

5.Preventive effect of early bowel intervention on the gastrointestinal dysfunction in patients with invasive mechanical ventilation

Objective To explore the preventive effect of early bowel intervention on the gastrointestinal dysfunction in patients with invasive mechanical ventilation.Methods Sixty patients with invasive mechanical ventilation were equally randomly divided into the observation group and the control group.patlents in the control group were treated by conventional nursing care,while Patients in the observation group were given the early bowel intervention including abdominal massage and stimulation of rectum.The two groups were compared in terms of gastrointestinal dysfunction.Result The rate of gastrointestinal dysfunction in the observation group was significantly lower than that of the control group(P<0.05).Conclusion The early bowel intervention may effectively promote intestinal peristalsis,protect the intestinal digestion, absorption of nutrients and mucosal barrier function and prevent gastrointestinal dysfunction in patients with invasive mechanical ventilation during enteral nutrition.

To develop a method to estimate the anti-Aspergilli activity of active ingredients of Chinese herbs. With the broth microdilution testing procedure proposed by NCCLS, anti-Aspergilli activity of active ingredients ( cinnamalde-hyde, cinn. and citral) of Chinese herbs was determined.The MIC values of cinn. to Aspergillus flavus, A.fumigatus were 0.100?g /mL , 0.050?g/mL.The MIC values of citral to A. flaws, A.fumigatus were 2.600?g/mL, 0.650?g/ mL.These results demonstrate that citral and cinn. have high potency activity against Aspergillus spp. .The research may provide references to establishing a standard for evaluating the effect of anti-Aspergilli Chinese herbs.
7.The Role of CNQX in the Different Types of Synaptic Release in Mice

Objective:To explore the role of 6-CYANO-2,3-DIHYDROXY-7-NITROQUIN OXALINE (CNQX) in different types of synapse secretion.Methods:The spontaneous mEPSCs and eEPSCs at different extracellular concentrations of CNQX in cultured cortical or hippocampal neurons were recorded respectively.Results:The half inhibitory concentration (IC50) of CNQX in evoked neurotransmitter release was significantly higher than that of spontaneous release,indicating that the spontaneous neurotransmitter release was more sensitive to CNQX.No apparent difference was observed between cortical and hippocampal cells,suggesting that the blocking effect of CNQX was similar in different brain regions.Conclusion:CNQX might have differential regulating mechanisms between excitatory spontaneous and evoked neurotransmitter release,but without brain regions specificity.

Type 2 diabetes is a hypermetabolic disease characterized with disorders of glucose/lipid metabolism, absolute or relative lack of insulin, and can induce skeletal muscle atrophy. Hyperglycemia, hyperlipidemia, insulin resistance, and abnormal release of inflammatory factors can lead to abnormal signal transduction in skeletal muscle, thus make protein synthesis and degradation imbalance and eventually causing muscle atrophy. Under normal conditions, insulin-like growth factor 1 (IGF-1)/insulin can activate phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT). AKT not only increases protein synthesis through mammalian target protein of rapamycin (mTOR), but also phosphorylates forkhead box O (FoxO) transcription factor and then inhibits the transcription of several ubiquitin ligases (such as MAFbx/atrogin-1 and MuRF1), or autophagy related genes. The weakened IGF-1/PI3K/AKT pathway in type 2 diabetes is an important factor leading to skeletal muscle atrophy. Studies have shown that the commonly used anti-type 2 diabetic drugs have different effects in regulating the synthesis and degradation of skeletal muscle protein. Studies reported that drugs with effect of anti-diabetic muscle atrophy include thiazolidinediones, glucagon-like peptide analogs, glucose-sodium cotransporter 2 inhibitors, etc.; drugs that are still in controversial or even promote skeletal muscle atrophy include metformin, and some sulfonylurea or non-sulfonylurea insulin secretagogues. This article overviewed and analyzed the currently commonly used drugs for type 2 diabetes and summarized the related mechanisms, with the aim to provide references for the rational applications of drugs for type 2 diabetes.
9.The evaluation of efflux transporter model based on RNA interference technology in vitro.

In the present study, the specifically knockdown models of P-gp or MRP2 were constructed by using a series of chemically synthesized small interfering RNA (siRNA) in vitro. The expression of P-gp and MRP2 was measured by real-time PCR and Western blot, and the function was evaluated by applying P-gp and MRP2 substrate, rhodamine and methotrexate. The results showed that MRP2 siRNA-3 or P-gp siRNA-2 significantly decreased the mRNA expression of MRP2 or P-gp, the inhibition ratio was 68% or 84%; MRP2 siRNA-3 or P-gp siRNA-2 at a dose of 80 nmol x L(-1) significantly reduced the protein expression of MRP2 or P-gp at 48 h after treatment, the inhibition ratio was 62% or 70%. Meanwhile, other transporters were not influenced by siRNA. When pretreatment with MRP2 siRNA-3 or P-gp siRNA-2, the efflux of methotrexate or rhodamine decreased significantly and the intra-cellular concentration increased. The results suggested that chemically synthesized siRNA could significantly inhibit the expression and function of MRP2 and P-gp, and the model of RNAi in vitro could be used to evaluate the role of efflux transporters in transportation of drugs.

10.Bioinformatics analysis and construction of eukaryotic expression plasmid of Cx50 V64G mutation

AIM : To construct and analyze eukaryotic expression plasmid inserted by Cx50 with V64G mutation through bioinformatics software.METHODS: The full coding domain sequence of Cx50 with V64G mutation was acquired from the blood of patients with cataract and was cloned into pcDNA3.1 /Amp (+).The constructed plasmid was identified with PCR , enzyme digestion and sequencing. The analysis of Cx50 with V64G mutation was performed with bioinformatics software.RESULTS : Cx50 with V64G mutation was successfully amplified and its eukaryotic expression plasmid was constructed. Valine-64 is well conserved in the first extracellular loop of connexin 50 in different species and also in different human α -type gap junctional proteins.CONCLUSION : The successive reconstruction and verification of eukaryotic expression plasmid containing Cx50 with V64G mutation established the foundation for further studying the mechanism of cataract.
