1.Detection of Urinary Podocyte Marker Protein and Its Value on Diagnosis of Kidney Disease

ObjectiveTo explore the clinical value on the detection of urinary podocyte marker protein podocalyxin(PCX) in children with nephritic syndrome(NS) and Schonlein-Henoch purpura nephritis(HSPN).MethodsUrinary samples voided in the morning were obtained from 14 healthy children and 75 children with NS or HSPN or Schonlein-Henoch purpura(HSP),including 21 children with NS in the acute phrase,14 children with NS in the catabasis,16 children with HSPN in the acute phrase,14 children with HSPN in the catabasis,10 children with common HSP,and 14 healthy children for control group.And urinary PCX content of the first morning urine was quantified by enzyme-linked immunosorbent assay(ELISA).SPSS 13.0 software was used to analyze the data.Results1.The levels of PCX content were significantly higher in the urine from children with any case of NS and HSPN compared with those in the control group(P≤0.009),but there was no obvious difference between common HSP children and children in the control group(P=0.754).2.The level of urinary PCX content in acute phrase of NS was(0.593?0.271) ?g/L,in the catabasis of NS was(0.162?0.093) ?g/L,there were significant difference(P=0).The level of urinary PCX content in acute phrase of HSPN was(1.822?1.342) ?g/L,in the catabasis of HSPN it was(0.236?0.141) ?g/L,which was significantly different(P=0.004).The level of urinary PCX content in common HSP was(0.089?0.061) ?g/L,there were significant difference in any case of HSPN(Pa
2.Effect of catgut implantation at acupoint on the content of tumor necrosis factor alpha of patients with ankylosing spondylitis.

OBJECTIVETo observe the effect of catgut implantation at acupoint on the content of tumor necrosis factor alpha (TNF-alpha), and to explore its mechanism for treatment of ankylosing spondylitis (AS).
METHODSSeventy-four AS patients were randomly assigned to the observation group and the control group by digit random table, 37 in each group. Patients in the control group took Sulfasalazine Enteric-coated Tablet (SECT), 0.5 g each time, three times daily for 3 months. Those in the observation group additionally received catgut implantation at acupoint for 3 months. Changes of visual analogue scale (VAS), Schober test, pillow wall distance, thoracic activity, erythrocyte sedimentation rate (ESR), C reactive protein (CRP), and TNF-alpha were compared after 3 months of treatment.
RESULTSThere was obvious change in VAS, Schober test, pillow wall distance, thoracic activity, ESR, CRP, and TNF-alpha after treatment in the both groups, showing statistical difference when compared with before treatment (P < 0.05, P < 0.01). The aforesaid indices were obviously improved after treatment in the observation group (P < 0.05, P < 0.01).
CONCLUSIONCatgut implantation at acupoint, as an assistant therapy, could significantly improve clinical symptoms and signs, decrease the level of TNF-alpha, and its effect was superior to that of oral SECT administration alone.

3.Study on the Effect of Residual Uracil DNA Glycosylase upon HCV cDNA

To explore the destructive effect of residual uracil DNA glycosylase on dUTP incorporation to the cDNA PCR products,PAGE and DNA-EIA hybridization technique were applied to detect the efficiency of anti-contamination and the rate of DNA hybridization.In this study,it was found that:⑴Good effect could be achieved with the reaction of 2u of uracil DNA glycosylase(UDG)for 20min at 37℃.⑵UDG could be inactivated at 94℃ for 10min.The hybridization rate of control group of PCR products was 94 18% at room temperature for 65 hours.The hybridization rates of test groups at-20℃ for 65 hours were 87 69%,77 24%,76 83%,respectively.And the inactivation times of anti-contamination groups at 94℃ were“0”min,“10”min and “20”min,respectively;at room temperature for 65 hours,these hybridization rates were 74 77%,72 50%,70 29%,respectively.There were significant differences between control and rest groups(P0 05).Our study suggested that the hybridization rate of test groups kept at-20℃ decreased 19 41%,after 65 hours,the rate decreased 27 45%,but in control group only decreased 5 82% for 65h at room temperature.So that,we conclude that the residual UDP can cause above results.
4.Analysis of the relative factors of the onset of repeated ectopic pregnancy

Objective To determine the relative factors of the onset of repeated ectopic pregnancy. Methods The clinical data of the first time ectopic pregnancy of 28 cases with repeated ectopic pregnancy were analysed, and compared with those of 56 cases of non-repeated ectopic pregnancy onsetting at the same period. The factors measured included: age at onset of disease, age at first coitus, gravidity, parity, methods of contraception, duration of amenorrhea,duration of vaginal bleeding, serum ?-human chorionic gonadotropin level, volume of intraperitoneal bleeding, types of ectopic pregnancy, methods of therapy and inflammation evidence of fallopian tube. Logistic regression analysis was performed to determine the relative factors for onset of repeated pregnancy. Results The risk factors and its odds ratio (OR) from the multivariate analysis were as follow: anastomosis of the tube(62.74, P=0.043), positive evidence of inflammation of the tube (54.85, P=0.000), no contraception (11.29, P=0.002), contraception by condom occasionally (4.75, P=0.046); the protective factors and its OR were as follow: therapy being salpingectomy and sterilization of the opposite tube(0.06, P=0.049), oral contraception (0.10, P=0.050) and pharmacotherapy (0.33, P=0.002). Conclusions The risk factors of onset of repeated ectopic pregnancy include: anastomosis of the tube, positive evidence of inflammation of the tube, no contraception and contraception by condom occasionally; the protective factors include: therapy being salpingectomy and sterilization of the opposite tube, oral contraception and pharmacotherapy.

6.Retinervus luffae fructus protects against the myocardial ischemic injury in mice

AIM: To observe the effect of Retinervus luffae fructus (RLF) on the myocardial ischemia in mice. METHODS: Mice were supplied with RLF (166.7,333.3, 666.7 g/L) twice daily (0.01 mL/g, ig). One week later, 30 U/kg pituitrin was injected intraperitoneally. The electrocardiogram was recorded, and the content of lactose dehydrogenase (LDH) in the serum and the activities of superoxide dismutase (SOD), maleicdialdehyde (MDA) in the myocardium were examined. RESULTS: RLF reduced the height of T wave in electrocardigram in myocardial ischemia mice and inhibited the decrease in heart rate. RLF also reduced the content of LDH in serum and the content of MDA in myocardium. The increase in the activity of SOD in myocardium was also observed. CONCLUSION: RLF may protect myocardium against ischemia injury. The mechanism may be related to the increase in activity of SOD and the suppression of the lipid peroxidation.

Triterpenoids are active constituents of many medicinal plants. The biosynthetic pathways of triterpenes have been fully studied and significant progress has been made. CYP450s are mainly involved in the post-modification process of triterpenoids biosynthesis, which plays a key role in the diversity of triterpenoids. Currently, a lot of CYP450s, which related to the biosynthesis of triterpenoids, have been cloned from many plants. In this paper, CYP450s involved in the biosynthesis of triterpenoids were reviewed, which provided a reference for the analysis of the biosynthetic pathway of triterpenoids and the functional study of CYP450s.

OBJECTIVETo observe preventive and therapeutic effect of Yifei Jianpi Recipe (YJR) on chronic obstructive pulmonary disease (COPD) model rats and to explore its mechanism from the way of airway inflammation and airway mucus hypersecretion.
METHODSThe COPD rat model was established by using cigarette smoking combined with intratracheal injection of lipopolysaccharide (LPS). Male SD rats were randomly divided into the blank control group (control group), the model group, the YJR group, 6 in each group. Forced vital capacity (FVC), forced expiratory volume in 0. 1 second (FEV0. 1), FEVO. 1/FVC, peak expiratory flow (PEF) was tested by lung function device. Pathological changes of bronchi and lung tissues were observed by HE staining. Airway Goblet cells were observed using AB-PAS staining. Contents of IL-8, IL-17, and TNF-α in bronchoalveolar lavage fluid (BALF) were measured by enzyme-linked immunosorbent assay (ELISA). Protein expressions of intercellular cell adhesion molecule-1 (ICAM-1), nuclear factor KB (NF-KB), mucin 5AC (Muc5AC), and Toll-like receptor 4 (TLR4) in rat airway were detected by immunohistochemical assay. mRNA expressions of TLR4 and Muc5AC in bronchi and lung tissues were detected by real-time quantitative PCR (RT qPCR).
RESULTSChanges of bronchi and lung tissues in the model group rats were consistent with typical pathological manifestations of COPD. Compared with the model group, the degree of lung injury was significantly alleviated in the YJR group. Compared with the control group, FVC, FEV0. 1, FEVO. I/FVC, and PEF were decreased (P <0. 01), contents of IL-8, IL-17, and TNF-α in BALF were significantly increased (P <0. 01), protein expressions of ICAM-1, NF-KB, Muc5AC, and TLR4, mRNA expression levels of Muc5AC and TLR4 in bronchi and lung tissues were also significantly increased in the model group (P <0. 01). Compared with the model group, FVC, FEV0. 1, FEV0. 1/FVC, and PEF were significantly increased in the YJR group (P <0. 01, P <0. 05), but the rest indices were significantly lowered (P <0. 01, P <0. 05).
CONCLUSIONYJR could decrease contents of IL-8, IL-17, and TNF-α in BALF of COPD model rats, inhibit protein expression levels of ICAM-1, NF-κB, Muc5AC, and TLR4.in airway and lung tissues, thus playing preventive and therapeutic roles by reducing airway inflammation and airway mucus hypersecretion.
