1.Culture and transplantation of autologous corneal limbal stem cells

OBJECTIVE: Autotransplantation with cultured autologous limbal stem cell is viewed as the best treatment. Therefore, it is necessary to further recognize the molecular biological character of limbal stem cell and histological anatomy and to study the progression of transplantation of autologous limbal stem cell.DATA SOURCES: The papers of Medline January 1995 to December 2002 were looked up in computer by retrieved words "limbus cornea, stem cells,autotransplant" of English version. Simultaneously, the papers of January 1995 to January 2002 of Chinese version were looked up by the same retrieved words in Chinese on http:∥www.wanfangdata.com.cn.STUDY SELECTION: It is to examine initially the papers relevant with studies on histology and physiopathology of limbal stem cells and relevant with DATA EXTRACTION: More than 120 papers of relevant literatures were collected in total and 15 papers of those met the criteria and selected for synthetic analysis.cells: Stem cells of corneal epithelium present similar characters as other committed stem cells, with very strong capacity of proliferation and cell division, in poor differentiation state, long cellular cycle and unparallel divipapillary structure of limbal basal palisades of Vogt are limbal stem cells.sion of stem cells are controlled by serum-source factors and various kinds of cellular factors, the changes in factors in microenvironment affect directly the proliferation and division of limbal stem cells and result in various limbal stem cells of the healthy eye from the patient with several ocular surface diseases cultured are grafted to amniotic membrane carrier for ocular surface autotransplantation on affected eye. It is observed after operation that corneal epithelium is regenerated in 2 to 5 days and visual sensibility is remarkably increased in l month. It is visible in 4-month observation after operation that corneal epithelium is integrative and neogenetic vessels are removed persistently.CONCLUSION: Autotransplantation of cultured limbal stem cells is still at the phase of experimental research, in which, initial success has been achieved in treatment of ocular surface diseases; but the remote therapeutic effect of such transplantation is required a further clinical observation. A further study is expected on proliferation, differentiation, physiology and biochemistry of cultured limbal epithelial stem cells, on screening of unique monoclonal antibody of stem cells, on purification and culture in vitro in stem cells, as well as on how to control the differentiation of stem cells and the influence of cellular factors on stem cells. Whether is it possible to apply allogeneic transplantation of cultured limbal stem cells for disorders of both eyes with deficiency of any source of autologous limbal stem cells? Whether is it possible to reduce incidence of rejection by the possible alternations on antigenicity of cultured limbal stem cells? There would be more prospects in the study on those topics.
2.The relationship study of essential hypertension arteriosclerosis and early renal damage

Objective To discuss the value of essential hypertension arteriosclerosis as a predictor of early renal damage.Methods 90 cases of essential hypertension with early renal damage were selected and statistical analysis to result of AASI,Scr,ACR and eGFR with SPSS17.0、Excel statistical analysis software were carried out.Group A was defined as AASI≥0.55 and group B was defined as AASI<0.55.Compared the two groups with relevant analysis,and then built multivariate linear regression equation.Results ① The group A showed higher age(P<0.01),24 h DBP(P<0.05),ACR(P<0.01),ACR(+) rate(P<0.01)and lower eGFR (P< 0.01),compared with the group B.②AASI showed a positive correlation with age (r= 0.483,P= 0.000),24 h SBP (r= 0.350,P =0.001),ACR (r=0.474,P= 0.000),and a negative correlation with eGFR (r=-0.383,P= 0.000).Two multiple linear regressions showed that AASI was associate with eGFR (B =-0.001,P= 0.038) and ACR(B = 0.025,P = 0.000) independent of other factors.Conclusion ①Arteriosclerosis is closely related with age and blood pressure mutation rate day and night.②Arteriosclerosis has a closely relationship with essential hypertension early renal damage,and can predict early renal damage in hypertension patients.
4.The Toxic Effect of Bilirubin on Different Types of Neurons of Rat in Primary Culture

Objective To investigate cytotoxicity and its mechanism of bilirubin on different types of neurons of rat in primary culture. Methods Toxic effect of bilirubin on primary cultured neurons (at cerebellar oranule hippocampal and cortical neurons) was observed. Ncuron survival was detected by fluorescei diacetate (FDA) staining. Measurement of fluorescent intensity of intracellular dilirudin were performed by microspectroflurimetry imaging system. Results Biliruin induced cytotoxicity to different type neurons in a dose-dependent manner. Exposed at 8d to different concentrations of bilirubin (4.3 ?mol/L、8.6 ?mol/L and 17.1 ?mol/L); cerebellar gtanule neuron survival werc (50.1?3.1)%,(18.5?2.3)%, (2.0?2.4)%, cortical neuron survival were (90.7?7.8)%, (85.3?5.3)%,(74.5?2.4)%, and hippocampal neuron survival were (75?6)%, (62?5)%, (49?5)% respectively. The toxic effect of bilirubin on ccrebellar granule neuron was significantly higher than that on hippocampal and cortical neurons. The fluorescent intensity of bilirubin in gra- nule, hippocampal and cortical neurons were 33.4?2.3、10.7?3.0、21.9?4.3 respectively three hours after addition of bilirubin to the medium. 5.Clinical application of magnetic attachment in the treatment for dentition defect

38 patients with dentition defect in our hospital for magnetic attachment dental treatment were analyzed. At the same time,34 cases of patients with dentition defects were ,being treated with conventional prosthodontics, selected to compare the stability of overdentures and masticatory function with magnetic attachment. The stability of overdentures and masticatory function after restored with magnetic attachment was significantly higher than before(P< 0. 01),while there was no difference after 4 weeks. Cementation language,comfort and appearance satisfaction after 4 weeks were significantly higher than before(P < 0. 05). The stability of overdentures,masticatory function, cementation language,comfort and appearance satisfaction after restored with magnetic attachment was significantly higher than conventional prosthodontics(P < 0. 05). It finally indicated that magnetic attachment had the advanta-ges of comfort,convenience and good effect. The stability of overdentures,masticatory function and application sat-isfaction was superior to conventional prosthodontics,and it had an application prospect.
7.EFFECT OF MILK BASIC PROTEIN ON BONE METABOLISM IN NORMAL AND OVARIECTOMIZED RATS

Objective To investigate the effect of milk basic protein (MBP) on bone metabolism in normal and ovariectomized (Ovx) rats. Method Forty-eight female Sprague-Dawley rats were ovariectomized and another 12 rats received sham operation (Sham). After 10 d recovery period, the Ovx rats were randomly divided into 4 groups: control, low-dose, medium-dose, and high-dose MBP group. Another 44 normal female rats without ovariectomy were also divided into 4 groups as above. The MBP dosages for each group were respectively 0, 10, 20, 30 mg/kg bw. All rats were i.g. administered for 90 d. Bone mineral density (BMD) of the femur (at proximal end, middle of diaphysis, and distal end) was measured by dual-energy X-ray absorptiometry in vivo. The amounts of calcium, magnesium and phosphorus were analyzed by ICP-AES. Results BMD at distal end of femur was significantly higher in normal low-dose group than in normal control group while no significant effect was observed in Ovx MBP groups. As for the amounts of calcium, magnesium and phosphorus, there were no significant differences among normal experimental groups and also among Ovx experimental groups. However, some variations in the level of those minerals were observed. Conclusion MBP at 10 mg/kg bw significantly elevated BMD at femoral distal end in normal rats, while no similar effect was observed in Ovx rats. Besides its influence on bone minerals, there may be another mechanism involved in its effect on bone metabolism.
8.Inhibitory effect of VEGF antisense oligonucleotides on synthesis of VEGF by Lewis lung cancer cells

Objective: To study the inhibitory effect of VEGF antisense oligonucleotides (ASODN) on growth of Lewis lung cancer in C57BL/6 mice.Methods: Lewis lung cancer cells were cultured and implanted subcutaneously into 40 C57BL/6 mice, which were then divided into 4 groups: VEGF-ASODN treatment group, VEGF-SODN treatment group, BEGF-MODN treatment,and control group (normal saline). Mice in different groups were treated 24 hours after cell inoculation. The weight and volume of subcutaneous tumors was measured and the morphological changes of tumor cells was observed under microscope. VEGF protein and microvessel density were examined by immunohistochemistry.Partial tumor tissues were kept in liguid nitrogon.Results: The average tumor weights of the control, VEGF-ASODN, VEGF-SODN and VEGF-MODN groups were (7.33?0.71)g, (4.56?0.38) g, (7.59?0.32) g, and (7.62?0.39) g, respectively. The inhibition rates of tumor growth in VEGF-ASODN, VEGF-SODN and VEGF-MODN group were 43.8%, 5.5% and 3.1%, respectively. VEGF-ASPODN obviously inhibited the tumor cell growth and decelerated the tumor cell proliferation. Alzheimer’ s disease ( AD) , a central nervous system degenerative disease, is characterized by progressive cognitive impairment and memory damage. Although intensive research leads to a better understanding of AD pathology, no new medi-cines are found to prevent, delay or stop the progression of AD. Three cholinesterase inhibitors ( donepezil, rivastigmine and ga-lantamine) and an N-methyl D-aspartase NMDA receptor antag-onist, memantine have been approved in clinic for AD treat-ment, but these treatments only have modest symptomatic effects for relatively short time periods. Therefore, to find effective medicines to prevent, improve and treat AD is urgent. It is well known, the traditional Chinese medicine resources are abundant in China. Chinese medicines, including the active ingredients and compounds, have been used in dementia treatment for a long history. The review is aimed to summarize the application and related mechanisms of natural products such as huperzine A, ginsenosides, L-3-n-butylphathlide, TSG and so on in AD treat-ment.
10.Progress of positive airway pressure and upper airway multilevel surgeries in the treatment of obstructive sleep apnea-hypopnea syndrome.

This review focus on the progress of the treatment of obstructive sleep apnea-hypopnea syndrome by using positive airway pressure and upper airway multilevel surgeries. OSAHS is a disease caused by multiple etiologies. We should consider upper airway anatomical abnormalities and the severity of OSAHS before reasonable treatment plans were formulated. "PAP-surgery-PAP" is the classic procedure for treating moderate OSAHS and severe OSAHS. Preoperative use of PAP could reduce the hypoxic condition of patients and decrease surgical risk. Surgery can effectively reduce the optimal PAP pressure value, and increase the tolerance of patient. Postoperative use of PAP can improve surgical efficacy, shorten patient recovery time. However, the influence of different surgical methods on optimal PAP pressure value is lacking in large samples. More related studies need to be per formed in the future.
