1.Clinical application of magnetic attachment in the treatment for dentition defect

38 patients with dentition defect in our hospital for magnetic attachment dental treatment were analyzed. At the same time,34 cases of patients with dentition defects were ,being treated with conventional prosthodontics, selected to compare the stability of overdentures and masticatory function with magnetic attachment. The stability of overdentures and masticatory function after restored with magnetic attachment was significantly higher than before(P< 0. 01),while there was no difference after 4 weeks. Cementation language,comfort and appearance satisfaction after 4 weeks were significantly higher than before(P < 0. 05). The stability of overdentures,masticatory function, cementation language,comfort and appearance satisfaction after restored with magnetic attachment was significantly higher than conventional prosthodontics(P < 0. 05). It finally indicated that magnetic attachment had the advanta-ges of comfort,convenience and good effect. The stability of overdentures,masticatory function and application sat-isfaction was superior to conventional prosthodontics,and it had an application prospect.
2.Differentiation of adult adipose tissue-derived Flk1~+CD31~-CD34~- cells into pancreatic islet-like endocrine cells in vitro

Objective To promote the differentiation of Flk1+CD31-CD34-cells isolated from adult adipose tissues into pancreatic islet endocrine cells in vitro.Methods Flk1+CD31-CD34-cells were first cultured and plated in medium supplemented with B27,epidermal growth factor(EGF),and basic fibroblast growth factor(bFGF).Next,the culture medium was changed.The glucose concentration in the serum-free medium was increased.At the same time,betacellulin and nicotinamide were added.Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of nestin,ngn3,insulin promoter factor-1(IPF-1),insulin,and glucagon before and after differentiation induction;immunofluorescent staining for nestin,insulin and glucagon and radioimmunoassay(RIA) for insulin.Results Initially,a nestin positive precursor cell population was found,then small round cells increased in number after 6 days.Later on,they were differentiated into islet-like clusters.The induced cells resulted in the formation of clusters which exhibited higher insulin secretion and other pancreatic endocrine hormones.RT-PCR detected an enhanced expression of pancreatic genes in the differentiated cells.Immunofluorescence revealed a high percentage of insulin-expressing cells in the clusters.Furthermore,the intra-cellular insulin content was detected by RIA after the induction culture.Conclusion These cells represent a previously unidentified adult intrinsic pancreatic precursor population and are a promising candidate for cell-based therapeutic strategies.
3.Effects of Tripterygium Hypoglaucum (Levl) Hutch alkaloids on cell cycle and time-dependent apoptosis of Jurkat T lymphoma cells

Objective To study the effects of the cytotoxicity, cell cycle and time dependent apoptosis of Jurkat T lymphoma cells induced by Tripterygium Hypoglaucum (Levl) Hutch (THH) alkaloids so ad to explore the mechanisms of the apoptosis induced by the THH alkaloids. Methods Cell vitality and cell proliferation were measured by Typan blue staining. Cell cycle and the time dependent apoptosis were determined by DNA staining, TUNEL labeling and flow cytometry. Results THH alkaloids could effectively inhibit cell proliferation of Jurkat cells and induce the G 1 arrest and could induce apoptosis in G 2/S phase first and then G 1 phase. Conclusion THH alkaloids can inhibit DNA synthesis, cell proliferation and can also induce apoptosis of Jurkat T lymphoma cells in all phases.
4.Clinical Observation on Effect of Pingai Mixture in the Treatment of 41 HIV/AIDS Patients

This study was aimed to observe the clinical effect of Pingai Mixture intervention on HIV/AIDS patients in order to clarify the characteristics of Pingai Mixture for AIDS treatment. A total of 41 HIV/AIDS patients who met the inclusion criteria were selected. Pingai Mixture was applied in the treatment. Three months was one treatment course. The treatment was given for two courses. Clinical symptoms, physical signs and changes in CD4+ lymphocytes of HIV/AIDS patients before and after treatment were observed. The SPSS software was used for statistical analysis. The results showed that the posttreatment total score (primary, secondary symptoms and the main physical sign cred-its) was significantly reduced compared with pretreatment (P < 0.01). The posttreatment spontaneous perspiration, night sweats, hair loss and other symptoms of a separate score was significantly reduced compared with pretreatment (P< 0.05). There were no significant changes on other symptoms or physical sign indicators. There was no significant difference in the Karnovsky score after 6-month treatment compared with pretreatment. It was concluded that Pingai Mixture is effective in AIDS treatment. It can improve the clinical symptoms and the administration is safe.
5.High-throughput screening of telomerase inhibitors in actinomycetes fermentation broth

Aim To discover specific telomerase inhibitors by high-throughput screening in the more than 2 000 strains of actinomycetes fermentation broth.Methods After verifying the three kinds of screening models for telomerase inhibitor by yeast PCR,the three yeast strains were inoculated overnight into yeast liquid culture which was lacking in histidine and contained 3-AT.Then concentration of yeast strains in culture liquid and dosage of screening sample were regulated for better adjustment to the high-throughput screening.At last the adjusted liquid culture containing yeast strain piped to 96-well plates,drug samples and control article were also added,then the yeast was incubated in the thermostat oscillator.At the same time OD_(595nm) of the yeast liquid was measured every 12 h by using Multifunctional Microplate Analyzer,and the samples whose inhibitory efficiency was over 0.45 were selected for secondary screening.Results The high-throughput screening method related to yeast three-hybrid system was effective in screening samples that had potential telomerase inhibitory ability.The initial concentration of yeast liquid OD_(595nm) was 0.04 and the best dosage of samples was 5 ml.14 active samples,whose inhibitory efficiency was over 0.45,were founded after primary and secondary screening.Conclusions High-throughput screening method related to yeast three-hybrid system is simple and stable in discovering telomerase inhibitors,and 14 new antitumor compounds are identified,which lays the foundation in the development of new anti-tumor agents.
6.Relationship between MRAS gene polymorphism and young patients with ischemic stroke

Objective ToinvestigatetherelationshipbetweenmuscleRASoncogenehomolog (MRAS)genepolymorphismandyoungpatientswithischemicstroke.Methods Atotalof243young patients with ischemic stroke from Nanjing Stroke Registry Program from December 2009 to October 2012 were enrolled retrospectively. At the same time,512 age-and sex-matched healthy controls were selected. The polymorphisms of MRAS genes rs3755751 and rs9289559 loci were analyzed by the modified multiplex PCR-ligase detection reaction assay. The genotype of each locus and the allele frequencies were analyzed and compared. Results (1)The frequencies of AA,AG and GG genotypes for rs3755751 in the stroke group (n=243)were 7. 4%(n=18),37. 0%(n=90),and 55. 6%(n=135),respectively. There were no significant differences compared with those (6. 4%[n=18],36. 9%[n=189]and 56. 6%[n=190]) in the control group (n=512)(P>0. 05). The frequencies of AA,AG and GG genotypes for rs9289559 in stroke group were 7. 0 (n=17)%,42. 0%(n=102),and 51. 0%(n=124),respectively. There were no significant differences compared with 6. 1%,(n=31)37. 9%(n=194),and 56. 1%(n=287)in the control group (P>0. 05). (2)Further construction of an effect model (AA vs. AG+GG and GG vs. AG+AA),there was no significant difference between the stroke group and the control group (P >0. 05 ). Analyzing the effects of different genotypes on plasma lipid levels showed that the high-density lipoprotein cholesterol level of the GG genotype subgroup in the young ischemic stroke group was significantly higher than that rs3755751 of the AG +AA genotype subgroup (OR,6. 80,95%CI 2.18-21.27, P=0. 001 ). Conclusions MRAS gene polymorphism may have no significant correlation with the genetic susceptibility in young patients with ischemic stroke. Polymorphism of rs3755751 may be correlated with the level of serum high-density lipoprotein cholesterol.
7.Effect of Pioglitazone on Impaired Glucose Regulation

ObjectiveTo observe the effect of pioglitazone on the metabolism of glucose and lipid in patients with Impaired Glucose Regulation(IGR).Methods60 cases of IGR received conventional education on diabetes prevention,while the patients in intervention group(30 cases) accepted pioglitazone 15 mg once a day for 12 weeks.Blood pressure(BP),body weight,waist circumference,hip circumference were measured to calculate body mass index(BMI) and waist-hip ratio(WHR).The Fasting blood glucose(FBG),2 h postprandial blood glucose,glcosylated hemoglobin(GHb,HbA1C),fasting serum insulin(FINS),postprandial serum insulin(2hINS),total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),hepatic function,renal function were determined before and 12 weeks after intervention.ResultsAfter intervention,BP,BMI,FBG,2hBG,FINS,2hINS,HbA1C,TC,TG,LDL-C had been decreased significantly in intervention group(P<0.05 or P<0.01).There were significant differences between intervention group and control group(P<0.05 or P<0.01) in all indexes except body weight,WHR,hepatic function,renal function.ConclusionPioglitazone can obviously improve the metabolism of glucose and lipid in patients with IGR.
8.Role of nitric oxide in iron-induced toxicity in rat hearts.

The aim of the present study was to explore the effect of nitric oxide (NO) on iron-induced toxicity in rat hearts. Langendorff perfused rat heart and enzymatically isolated cardiomyocytes were used. It was shown that lipophilic Fe-HQ reduced the contractile amplitude, velocity and end-diastolic cell length in the cardiomyocyte, while the left ventricular developed pressure (LVDP), +/-dp/dt(max), heart rate and coronary flow showed biphasic alterations, which increased in the first 2 min and then was followed by a decline in isolated perfused rat heart; the contents of lactate dehydrogenase (LDH) and creatine kinase (CK) in the coronary effluent and the malondialdehyde (MDA) in the myocardium were increased. L-arginine (L-Arg), an NO precursor, reduced the contractile amplitude and end-diastolic cell length in the cardiomyocyte; but reversibly increased LVDP, +/-dp/dt(max), and coronary flow in isolated perfused rat heart. Pretreatment with L-Arg aggravated the Fe-HQ-induced decrease in contractile amplitude, velocity and end-diastolic cell length in the cardiomyocyte; LVDP, +/-dp/dt(max), heart rate and coronary flow were significantly reduced in the perfused heart, and the levels of LDH and CK increased in the coronary effluent. In contrast, the NOS inhibitor N(omega)-nitro-L-arginine methyl ester (L-NAME) blocked the Fe-HQ induced change in contractile amplitude, velocity and end-diastolic cell length in the cardio- myocyte; it inhibited the decrease in LVDP, LVEDP and +/-dp/dt(max), and reduced the LDH and CK. Removing endothelial cells in coronary vessels attenuated the increase in LVDP and +/-dp/dt(max) at the beginning of Fe-HQ perfusion. It is suggested that L-Arg aggravates the iron-induced cardiac dysfunction, NO can mediate the iron-induced toxicity in heart, and endothelial cells in coronary vessels play an important role in the early stage of the effect of iron.

10.Effect of octacostyl alcohol preparation on blood free radical metabolism and cardiac endocrine function of rats.

OBJECTIVETo evaluate the effect of octacostyl alcohol preparation on the motor function, blood free radical metabolism and cardiac endocrine function of rats.
METHODSA rat model of exercise fatigue was used for this study. The rats were made to perform continuous exhaustive exercise, and the total exercise time of the rats till exhaustion was recorded followed by examination of the serum superoxide dismutase (SOD), malonaldehyde (MDA), plasma endothelin (ET), calcitonin gene-related peptide (CGRP), and atrial natriuretic peptide (ANP).
RESULTSCompared with rats without interventions for exercise fatigue, the rats receiving octacostyl alcohol preparation had significant prolonged exercise time till exhaustion (P<0.05), with also significantly enhanced serum SOD activity (P<0.05), significantly decreased MDA level and plasma ET (P<0.05) and increased CGRP and ANP levels (P<0.05). Octacostyl alcohol preparation produced more favorable effect on all the indices measured than pyruvic acid-creatine (P<0.05).
CONCLUSIONThe octacostyl alcohol preparation can promote the motor function, increases blood antioxidase activity, suppressed lipid peroxidation in rats engaged in exhaustive exercise. The preparation also produces favorable effect on cardiac endocrine function for heart protection during exercise.
