1.Reserch on the Tlyroid Function Reference Range of Different Pregnancy Period of Normal Pregnant Women

Objective To establish the reference range of thyroid function from the normal pregnant women in different pregnant period in Hanzhong region.Methods According to the NACB inclusion criteria,collected local resident pregnant women 4 031 cases,from July 2012 to December 2015.With the combination of random sampling,stratified sampling and cluster sampling,thyroid hormone levels were measured by a fully automated chemiluminescence analyzer and its accompanying reagents.All patients were divided into three groups:998 cases in early pregnancy (T1),1 543 cases in mid-pregnancy (T2),1 490 cases in late pregnancy (T3),and 105 cases of non-pregnant women in childbearing age (T0) were selected as control group.Results The levels of thyroid hormones were different among three periods of pregnant women.TSH were 0.25~5.32,0.42~6.26 and 0.61~7.68 mIU/L respectively in the early,middle and late stages.FT3 were 3.54~6.04,3.57~5.94 and 2.93~5.40 pmol/L,respectively.FT4 was 7.11~16.88,6.78~16.94 and 6.03~16.87 pmol/L.Thyroid hormone levels in pregnant women compared with non-pregnant women,there were significant differences (TSH:x2=233.183,P<0.05,FT4:x2 =388.12,P<0.05 and FT3:x2 =558.795,P<0.05).TSH were lower in early pregnant women comparing to non-pregnancy women,and higher in middle-late pregnant women.The change of FT3 and FT4 were consistent,and reduced with the extension of pregnancy comparing to non-pregnancy women.Conclusion The level of thyroid function in pregnant women were different from non-pregnant women.and the normal reference range of local pregnant specific thyroid hormone should be established.
2.Discrimination of Descurainiae Semen and Pantagirus Semen by HPLC fingerprints.

OBJECTIVETo discriminate Descurainiae Semen and Pantagirus Semen.
METHODA high-performance liquid chromatographic method was developed to establish the fingerprint of Descurainiae Semen, and hierarchical cluster analysis (HCA) and partial least squares discriminant analysis (PLS-DA) were applied to study HPLC fingerprinting and chemical recognition.
RESULTThere exists large difference of chromatographic peaks and its relative peak area of HPLC fingerprints between Descurainiae Semen and Pantagirus Semen, and after conducting statistical analysis, the result demonstrated that all samples were classified into three categories: Descurainiae Semen, Pantagirus Semen and their mixtures.
CONCLUSIONThe developed HPLC fingerprint combined with chemometrics can be utilized to discriminate between Descurainiae Semen and Pantagirus Semen, which was quick, simple, accurate and reliable an can provide the basis for the characterization and quality assess of Descurainiae Semen and Pantagirus Semen.

"Tinglizi", the ripe seed of Descurainia sophia and Lepidium apetalum, is a member of Brassicaceae (Cruciferae). Traditionally, the former is called "Nantinglizi" (Descurainiae Semen) while the latter is called "Beitinglizi" (Lepidii Semen). In the theory of traditional Chinese medicine, it has the power to purge lung-fire, relieve dyspnea, promote diuresis and reduce edema, and it is mainly indicated in a case with phlegm-fluid accumulation, cough with excessive sputum, dyspnea with being unable to lie, and general swelling. In view of its wide-spread application in clinic, a comprehensive review of Lepidii Semen and Descurainiae Semen was conducted from the following aspects: herbalogical study, variety identification, historical evolution of processing, chemical constituents, pharmacological effects, quantitative determination and toxicity which could provide reference for further research and development of "Tinglizi".

5.Effects of the ultra-filtration extract mixture from Hedysarum Polybotrys on human liver cells HepG2 radiosensitivity.

OBJECTIVETo investigate the effects of the ultra-filtration extract mixture from Hedysarum Polybotrys (UEMHP) on the radiosensitivity of HepG2 cells, and to explore its possible mechanisms.
METHODSThe proliferation inhibition effects of UEMHP on HepG2 cells was detected by CCK-8 assay. The colony formation assay was used for the survival fraction (SF) analysis. The distribution of the cell cycle and the apoptosis rate were detected using flow cytometry (FCM). The survivin mRNA expression level was detected using reverse transcription-PCR assay.
RESULTSThe inhibition of UEMHP on HepG2 cells was time-and dose-dependent at the concentration ranging between 5 -50 mg/L (P < 0.05). The parameters of the two curve for SF (P < 0.05) showed statistical difference between the irradiation group and the UEMHP irradiation group. UEMHP could inhibit the clone formation of HepG2 cells and enhance the radiosensitivity of HepG2 cells. The results of FCM showed that UEMHP could induce G2/M phase arrest. The apoptosis rate in the UEMHP irradiation group (21.42% +/- 3.74%) was higher than that in the control group (5.35% +/- 0.41%), the only UEMHP group (10.36% +/- 1.75%), or the irradiation group (10.58% +/- 2.01%) (P < 0.01). RT-PCR showed that the survivin mRNA expression level was lower in the UEMHP irradiation group (0.31 +/- 0.02) than in the control group (0.82 +/- 0.06) and the irradiation group (0.58 +/- 0.04) respectively, showing statistical difference (P < 0.01).
CONCLUSIONUEMHP can enhance the radiosensitivity of HepG2 cells, and its possible mechanisms might be correlated to down-regulating the survivin mRNA expression and promoting the apoptosis.

6.Advancements on the zebrafish glioma model.

Glioma derived from the neural ectoderm is the most common brain tumor and is of great damage to human health among all lethal tumors. Scientists have been trying their best to find new methods and develop new drugs to treat glioma in recent years. The animal glioma model is of great importance to the research. Researchers have developed many animal glioma models, like the rat and mouse model. Now we are trying to develop a new zebrafish glioma model, which has much more advantages and fewer disadvantages than the traditional models in regard to gene mutation, chemical induction, and xenografts. Establishing a glioma model in zebrafish is feasible and would be of great use to patients with this common brain tumor.

7.Association of matrix metalloproteinase-9 and platelet membrane glycoprotein VI polymorphisms with acute coronary syndrome.

OBJECTIVETo investigate serum level and gene polymorphisms of matrix metalloproteinase 9 (MMP-9), and platelet glycoprotein VI (GPVI) in patients with acute coronary syndrome (ACS).
METHODSIn a prospective study of 179 patients with documented ACS and 164 controls, we measured baseline serum MMP-9 levels using ELISA and determined the MMP-9/C-1562T and MMP-9/G5564A genotypes using PCR-restriction fragment length polymorphism. Fib serum level was measured by Clauss assay. We also analyzed the Fib/Bbeta-148C/T and GPVI/T13254C polymorphisms.
RESULTSSerum levels of MMP-9 and Fib in ACS patients were significantly higher than in controls (P < 0.001), and serum level of Fib in the acute myocardial infarction group was higher than in patients with unstable angina (P < 0.05). No significant difference between ACS patients and controls was found in frequencies of MMP-9/C-1562T, MMP-9/G5564A, Fib/Bbeta-148C/T, and GPVI/T13254C genotypes and alleles (P > 0.05). The T allele of the Fib/Bbeta-148T polymorphism was associated with increased plasma Fib level (P < 0.05). There was a strong positive correlation between serum level of MMP-9 and Fib (r = 0.289, P < 0.01).
CONCLUSIONSerum levels of MMP-9 and Fib were independent risk factors of ACS. There was an obvious relationship between the Bbeta-148C/T mutation and high Fib level. No significant difference between controls and ACS patients was found in the frequencies of MMP-9 C-1562T and G5564A, Fib Bbeta-148C/T and GPVI T13254C genotypes and alleles (P > 0.05).

8.Effect of the “problem solving for better health” method on improving the compliance of raising the head of bed of invasive mechanical ventilation patients in ICU

Objective To explore the strategies that can improve the nurses ’ compliance of raising the head of a bed for 30 -45 °on mechanical ventilation patients and evaluate the clinical effect .Methods Before implementing “problem solving for better health ( PSBH )”, 78 patients of invasive mechanical ventilation in ICU were included as the control group from January 2012 to July 2012 .After implementing PSBH , 119 patients were included as the experimental group , from October 2012 to May 2013 .By training the nurses , improving the working conditions and putting up posters , the compliance of nurses and the incidence of ventilator associated pneumonia from each group were compared .Results Nurses’ compliance of raising the head of bed for 30-45°was 55.2%in experimental group, which was higher than that of control group (22.0%).The difference was statistically significant (χ2 =237.4, P <0.01).The incidence of ventilator associated pneumonia was 10.1‰ in experimental group, which was lower than that of control group (26.8‰).The difference was statistically significant (χ2 =5.11, P <0.05).Conclusions After the PSBH intervention, the nurses’ compliance of raising the head of bed was increased , and the incidence of ventilator associated pneumonia was reduced.PSBH plays an important role in promoting the patients ’ health.
9.Application effect of time-sensitive incentive nursing in patients with multiple myeloma

Objective:To explore the application effect of time-sensitive incentive nursing in patients with multiple myeloma (MM) .Methods:From January 2019 to May 2021, the convenient sampling method was used to select 86 MM patients admitted to Henan Provincial People's Hospital as the research objects. According to the time of admission, 43 patients admitted from January 2019 to January 2020 were divided into the control group and 43 patients admitted from February 2020 to May 2021 were divided into the study group. The control group was given routine nursing, while the study group was given time-sensitive incentive nursing based on the control group. The psychological resilience and quality of life of the two groups before and after intervention were compared, and the occurrence of adverse reactions between the two groups during intervention was statistically analyzed.Results:After the intervention, the dimension scores of psychological resilience scale and the dimension scores of quality of life scale in the study group were higher than those in the control group, and the differences were statistically significant ( P<0.05). The incidence of gastrointestinal reactions in the study group was lower than that in the control group, and the difference was statistically significant ( P<0.05) . Conclusions:Time-sensitive incentive nursing can improve the psychological resilience of MM patients, reduce the occurrence of gastrointestinal reactions caused by chemotherapy and improve the quality of life.
10.Effect of composite salvia injection on platelet parameters in children with anaphylactoid purpura.

OBJECTIVETo explore the effect of composite salvia injection (CSI) on platelet parameters in children with anaphylactoid purpura (AP) and its clinical significance.
METHODSOne hundred and fifty children with AP were assigned to two groups, 80 in Group A and 70 in Group B. They were treated, respectively, with conventional therapy only or conventional therapy combined with CSI. Their platelet parameters, including blood platelet count (BPC), mean platelet volume (MPV), platelet distribution width (PDW) and plateletcrit (PCT) were determined at the acute stage and convalescent stage, respectively.
RESULTSAt the acute stage, the BPC in AP children of both groups was in the normal range, but significant abnormality was shown in the levels of MPV, PDW and PCT. As for comparisons of these parameters at the convalescent stage, significant difference between the two groups was also shown in terms of MPV, PDW and PCT (P<0.05 or P<0.01).
CONCLUSIONAlthough platelet shows no quantitative change in the pathogenic process of AP, important functional changes are surely existent. CSI could promote the normalization of platelet function.
