AIMTo clone the gene of staphylococcal enterotoxin C2 and express it in the form of a soluble fusion protein in E. coli. Then the activation of SEC2 on mice lymphocyte and its lethal effects on tumor cells were studied.
METHODSStaphylococcus aureus SEC2 gene was cloned into GST gene fusion vector pGEX-4T-1. The resultant plasmid pGEX-4T-SEC2 was used to transform E. coli BL21, where the GST-SEC2 fusion protein was expressed efficiently. The rSEC2 protein was purified with Glutathione Sepharose 4B affinity column and digested with thrombin. The in vitro culture system was utilized to observe the activation of the SEC2 on mice lymphocyte and the lethal effects on tumor cells of the activated mice lymphocyte.
RESULTSThe proper gene of SEC2 was cloned and purified rSEC2 was obtained. The MTT results indicated that rSEC2 have strong ability to stimulate mice lymphocyte to proliferate with a dose-dependent manner. With the proliferation of mice splenic lymphocyte, rSEC2 has a strong lethal effect on tumor cells B16, K562 and K562-AD.
CONCLUSIONIn this study, the gene of SEC2 was cloned and the rSEC2 protein was obtained, which had strong lethal effect on tumor cells B16, K562 and K562-AD.

BACKGROUND:Significant increasing of intercel ular adhesion molecule 1 expression can promote the occurrence of inflammatory response and increase brain tissue injury.
OBJECTIVE:To investigate the effect of eye acupuncture point and non-point therapy on intercel ular adhesion molecule 1 expression in rat hippocampus of acute cerebral ischemia-reperfusion injury model.
METHODS:Forty Sprague Dawley rats were divided into normal group, sham-operation group, model group, eye acupuncture point group and eye acupuncture non-point group. Rats in the model group, eye acupuncture point group and eye acupuncture non-point group were used to establish the acute cerebral ischemia-reperfusion injury model with suture method, the suture was deep for 1.8-2.2 cm. In the eye acupuncture point group, the acupuncture points of hepatic region, upper energizer area, lower energizer area and kidney area were selected when cerebral ischemia-reperfusion occurred immediately and 30 minutes before drawing materials, then horizontal needling the acupuncture points around the orbit for 20 minutes. In the eye acupuncture non-point group, the puncture site was selected at 3 mm away from eye acupuncture non-point area, and then needled the puncture site with the same method in the eye acupuncture point group.
RESULTS AND CONCLUSION:After eye acupuncture point therapy, the neurological deficit scores were decreased, and the intercel ular adhesion molecule 1 expression in rat hippocampus was significantly decreased (P<0.01). After eye acupuncture non-point therapy, there were no significant changes in neurological deficit scores and the expression of intercel ular adhesion molecule 1 protein and mRNA in rat hippocampus. The results indicate that eye acupuncture can significantly improve the rat cerebral ischemia-reperfusion injury, and the mechanism may relate with reducing the intercel ular adhesion molecule 1 expression in rat hippocampus.

Objective To investigate and analyze the related factors influencing the quality of life in patients with epilepsy, and provide a theoretical basis for taking appropriate measures to improve the quality of life in patients with epilepsy. Methods Sixty-five patients with epilepsy visiting our hospital from July 2007 to December 2008 were chosen in our study. The table-31 for quality of life in patients with epilepsy (Chinese version) was performed on them. The age, gender, education degree, seizure types, course of disease, cognitive function, medication compliance and fear of attack were considered as the independent variable; related factors in the table-31 for quality of life were adopted as dependent variable; multiple linear regression analysis was performed on these 2 variables. Results Education degree and course of disease could affect the memory of patients with epilepsy (standardized regression coefficients were 0.380 and 0.264, respectively). Age could affect the social activities of the patients (standardized regression coefficient was -0.303). Gender could affect the attack of the patients (standardized regression coefficient was 0.332). Conclusion The influencing factors of quality of life in patients with epilepsy are age, gender, education degree, course of disease; and education degree and course of disease enjoy the greatest influence.

Objective To investigate the associations between epidermal growth factor receptor (EGFR) gene mutations and serum tumor markers in advanced lung adenocarcinomas.
Methods We investigated the association between EGFR gene mutations and clinical features, including serum tumor marker levels, in 97 advanced lung adenocarcinomas patients who did not undergo the treatment of EGFR tyrosine kinase inhibitors. EGFR gene mutation was detected by real-time PCR at exons 18, 19, 20, and 21. Serum tumor marker concentrations were analyzed by chemiluminescence assay kit at the same time.
Results EGFR gene mutations were detected in 42 (43%) advanced lung adenocarcinoma patients. Gender (P=0.003), smoking status (P=0.001), and abnormal serum status of carcinoembryonic antigen (CEA, P=0.028) were significantly associated with EGFR gene mutation incidence. Multivariate analysis showed the abnormal CEA level in serum was independently associated with the incidence of EGFR gene mutation (P=0.046) with an odds ratio of 2.613 (95%CI:1.018-6.710). However, receiver operating characteristic (ROC) curve analysis revealed CEA was not an ideal predictive marker for EGFR gene mutation status in advanced lung adenocarcinoma (the area under the ROC curve was 0.608, P=0.069).
Conclusions EGFR gene mutation status is significantly associated with serum CEA status in advanced lung adenocarcinmoas. However, serum CEA is not an ideal predictor for EGFR mutation.
