1.Role of compound traditional Chinese medicine Yiqiling in regulation of immune-related genes expression in SHR

Objective To investigate the role of compound traditional Chinese medicine Yiqilin in the regulation of immune-related genes expression in spontaneously hypertensive rats(SHR).Methods Ten SHR were randomly divided into test group(fed with Yiqilin and ordinary food at the ratio of 1:9 for 6 months,n=5)and control group(fed with ordinary food for 6 months,n=5).Total RNA was extracted from myocardial tissues of SHR after 6 months,and gene chip hybridization technique was employed to analyse the expression of immune-regulated genes of myocardial tissues in two groups.The genes with differentiation expression were screened,and were verified by Real-time PCR.Results The expression of 32 immune-related genes were up-regulated in the test group,including interleukin(IL)6 receptor,IL-6 signal transduction factor,chemotatic factor,antigen presenting molecule,antibody receptor,heat shock protein(Hsp), etc.No immune-related gene was down-regulated in test group.The results of 2 immune-related genes(Hsp 105 and Hsp 90)detected by Real-time PCR were consistent with those by gene chip hybridization.Conclusion Part of the immune-related genes in myocardial tissues in SHR may be up-regulated by Yiqiling,which indicates that Yiqiling may play a role in the regulation of expression of immune-related genes in SHR.
3.Currents status and strategies for the teaching of selective course of neuroanatomy for medical graduates

Selective courses are different from compulsory courses in teaching objective,students' study demand,teaching content.Different teaching designs should be used according to different teaching objectives in selective courses for graduates.Targeted scientific thoughts and methodological training should be designed with respect to the fact that students will begin their experimental studies after the selective lessons.We analyzed the current status of selective course of neuroanatomy for graduates and proposed some solutions.
4.Clinical study of Ludloff osteotomy and artificial metatarsophalangeal arthroplasty in the treatment for severe hallux valgus

Objective To analyze the clinical outcomes of Ludloff osteotomy and artificial metatarsophalangeal arthroplasty in the patients with severe hallux valgus and metatarsophalangeal joint osteoarthritis.Methods From Apr.2013 to Aug.2015,120 feet with severe hallux valgus were treated by operation in Beijing Friendship Hospital,Capital Medical University.Before operation,measured some angles,such as hallux valgus angle,intermetatarsal angle,proximal articular surficial intrinsic angle and so on in the two groups.And evaluated the patients condition by Maryland scale at the same time.Through the date,we make sure that there was no significant difference between the two groups before operation.According to the order of hospitalization,all patients were randomly divided into experimental group (n =60) and control group (n =60).The cases of experimental group were treated by Ludloff osteotomy and artificial joint replacement.The cases of control group were treated by Ludloff osteotomy and Akin osteotomy.We used the visual analogue scale,the core quality of life scale-36 and Maryland scale to record the date,such as pain,the range of motion and appearance of the metatarsophalangeal joint and the ability of daily life and sports before and after operation.Compared above categorical variables using paired T-test between before and after operation and using T-test between the two groups.Compared the rate of operative complications by Fisher's exact test between the two groups.Results Through the Maryland scale,the total excellent rate of the experimental group (91.7%) was higher than the total excellent rate of the control group (75.0%),with statistically significant (P < 0.05).The operative complication rate of experimental group (1.7%) was lower than the control group (8.3%),but without statistically significant (P >0.05).The scores of quality of life of the experimental group after treatment were significantly higher than those of the control group,with statistically significant (P < 0.05).The visual analogue scale pain score (1.61 ± 0.12) of the experimental group was significantly lower than the control group (3.68 ± 0.58),with statistically significant (P < 0.05).Conclusion The clinical outcomes of Ludloff osteotomy and artificial metatarsophalangeal arthroplasty in patients with severe hallux valgus and metatarsophalangeal joint osteoarthritis is better,high safety,less trauma and faster recovery.It is worthy of promotion and use.
5.Efficacy of esomeprazole and flupentixol and melitracen in treatment of non-erosive gastroesophageal reflux disease with depression and anxiety

Objective To investigate the efficacy of esomeprazole (Esomeprazole) and flupentixol and melitracen (Deanxit) in treatment of non-erosive gastroesophageal reflux disease (NERD) with depression and anxiety.Methods The diagnosis of NERD was based on the results of esorneprazole scale (reflux diagnostic ques- tionnaires,RDQ) and endoscopy,the degree and frequency of symptoms were graded and scored.Hamilton de- pression scale was used to evaluate depression and anxiety status.Sixty-three patients were randomly divided into group A (esomeprazole 20 mg qd),B (Deanxit 1 tab qd),C (esomeprazole 20 mg qd and Deanxit 1 tab qd) with each group 21 patients,and treated for four weeks.The efficacy was evaluated by the symptom scores.Results There were some effects of esomeprazole or Deanxit alone on the improvement of symptoms of NERD.Although the effect of Deanxit was not as good as esomeprazole,there was not difference for relieving heartburn and sub- sternal pain between two groups in time point of each week after treatment (P>0.05).esomeprazole was better for relieving acid regurgitation than Deanxit (P<0.01).Combined treatment of esomeprazole with Deanxit was better and faster for relieving the symptoms of NERD than esomeprazole or Deanxit alone (relieving rate 81% in group C,23.8% in group A and 14.3% in group B,P<0.01).The total efficacy rate of group C (100%) was higher than that of group A (80.9%) or group B (61.9%,P<0.01),but there were not difference of relieving rate and total efficacy rate between group A and B (P>0.05).Conclusions There was better efficacy of com- bined treatment of esomeprazole and low-dose Deanxit in treating NERD with depression and anxiety.These re- sults suggest that depression and anxiety have some effects on the pathogenesis and progress of NERD.
6.Preliminary of research of effect of artesunate on invasion of human colon cancer Lovo cells

Aim To investigate the effect of artesunate on the invasion of human colon cancer Lovo cells and the possible mechanisms. Methods After Lovo cells were treated with different doses of artesunate(20,80, 160 μmol·L - 1 ), the soft agar colony formation test was adopted to observe the anchorage-independent pro-liferation of Lovo cells. Transwell assay was used to determine the effect of artesunate on the invasion abili-ty of Lovo cells. And the protein expressions of HMGB1 and MMP-2 were investigated by western blot. Results Artesunate could significantly inhibit both proliferation and invasion ability of Lovo cells in a dose-dependent manner(P < 0. 01). The experimental group treated with artesunate significantly down-regula-ted the protein expressions of HMGB1 and MMP-2 compared with control group(P < 0. 05). Conclusion Artesunate could inhibit the invasion of human colon cancer Lovo cells by down-regulating HMGB1 and MMP-2 expressions.
7.Effect of whole-body hypothermia on cardiac hemodynamics neonatal piglet after hypoxia-ischemia

Objective To determine the effect of mild hypothermia on neonatal piglet cardiac hemodynamic function after hypoxia-ischemia (HI).Method Twenty five 7-day-old piglets were used for hypoxic ischemic brain damage (HIBD) model by the method of temporary occlusion of the bilateral carotid arteries and followed by mechanical ventilation with low concentration of oxygen (FiO_2=6%) for 30 minutes.The piglets were randomly divided into three groups:group A (normothermia with body temperature to 39℃,n=9),group B (body temperature to 36℃for 72 hours,n=8),and group C (body temperature to 34℃for 72 hours,n=8).Mild hypothermia was initiated at 4 hours after HI,the systolic and diastole function were evaluated by Doppler echocardiography at pre-HI,post-Hi 4 hours and post-HI 72 hours.Results There were no significant differences in left ventrieular ejection time/left ventrieular ejection time (LPEP/LVEF),right ventricular ejection acceleration time/right ventricular ejection time (RACT/RVET) and CO at post-HI with hypothermia 72 hours in three groups,but the heart rate decreased in B and group C group.Compared with nonnothermia,mild hypothermia treatment showed no significant differences in MAP,LPEP/LVET,RACT/RVET,CO,SV at post-HI with hypothermia 72 hours.Conclusions Body temperature decreased by 3~5℃for 72 hours will not aggravate hemodynamic abnormity.
8.Regulating effect of moxibustion pretreatment on Th1/Th2 imbalance of athletes during course of heavy load training.

OBJECTIVETo explore the effect of moxibustion pretreatment on the immune function of athletes during the courses of heavy load training.
METHODSTwelve male middle-distance race athletes were randomly divided into an observation group (n = 6) and a control group (n= 6). In observation group, mild moxibustion was applied at Guanyuan (CV 4), Zusanli (ST 36) or Guanyuan (CV 4), Sanyinjiao (SP 6) alternatively once daily, for 4 weeks. In control group, there was no any intervention. The expressions of gamma-interferon (IFN-gamma) and interleukin-4 (IL-4) were detected by realtime fluorescent quantitative PCR method before heavy load training, after 3 weeks of treatment, after 1 week adjusting training.
RESULTS(1) The ratios of Testosterone/Cortisol (T/C) of (0.017 +/- 0.003) in the control group and of (0.021 +/- 0. 007) in the observation group after heavy load training for three weeks were significant lower than 0.036 +/- 0.009 and 0.037 +/- 0.013 before heavy load training in the same group, respectively (both P < 0.01) (2) IFN-gamma mRNA of (2.76 +/- 1.28) and ratio of IFN-gamma mRNA/IL-4 mRNA of (3.28 +/- 1.76) in the control group after heavy load training for three weeks were significant higher than 1.75 +/- 0.44 and 1.52 +/- 0.76 before heavy load training, respectively (both P < 0.05), and ratio of IFN-gamma mRNA/IL-4 mRNA of (0.86 +/- 0.66) after adjusting training for 1 week reduced significantly as compared with that after heavy load training for three weeks (P < 0.05). (3) In the observation group, IFN-gamma mRNA, IL-4 mRNA and ratio of IFN-gamma mRNA/IL-4 mRNA increased firstly and then restore with the time, but these changes had no significant differences (all P > 0.05).
CONCLUSIONMoxibustion pretreatment can prevent Th1/Th2 imbalance of male middle-distance race athlete during the course of heavy load training.

9.Study on the relationship between promoting apoptosis effect of artesunate and Wnt/β-catenin pathway in colon cancer cells

Aim To investigate the promoting apoptosis effect of artesunate( ART) on human colon cancer Lovo cells and its mechanisms. Methods MTT assay was performed to determine the anti-proliferative effect of artesunate. Flow cytometry assay and electron micros-copy( EM) were used to evaluate the apoptotic effect of artesunate. Luciferase reporter assay was introduced to measure the activation of Wnt/β-catenin pathway. Western blot was used to detect the pathway-related protein levels of β-catenin, GSK-3β,c-Myc and apop-tosis-related protein level of casepase-3 . Results Compared with the control group, the inhibitory rate of cell proliferation at 72 h and 320 μmol·L-1 ART was (78. 99 ± 1. 95 )% ( F =898. 301, P =0. 000 ); the cell apoptotic rate at 24 h and 160 μmol · L-1 ART was(19. 00 ± 0. 05)% and morphological signs of cell apoptosis were found by EM;the transcriptional activi-ty of TCF4/LEF at 24 h and 160 μmol·L-1 ART was (0. 36 ± 0. 30)%(F =470. 954,P <0. 01); the ex-pressions of caspase-3 and GSK-3β were significantly increased, whileβ-catenin and c-Myc were significant-ly decreased when treated with different concentrations of ART for 48 h ( P <0. 01 ) . Conclusion ART may significantly inhibit proliferation and promote apoptosis of Lovo cells probably by inactivating Wnt/β-catenin pathway.
10.Three-dimensional tissue engineering scaffolds with electrospinning technique:application and prospects

BACKGROUND:The electrospinning technique has been used to prepare biological scaffolds to simulate nano-fiber structure of extracelular matrix; therefore, widespread attention has been paid to the electrospinning technique in the field of regenerative medicine and tissue engineering. OBJECTIVE: To review the articles about increasing electrospun nanofiber scaffold porosity, enlarging pore diameter, promoting cel infiltration with related technologies, in order to discover the most practical and economical technology. METHODS:The first author retrieved CNKI database, Wanfang database and PubMed with the keywords of “cel infiltration, 3D scaffold, electrospinning” in Chinese and English, respectively. Literature retrieval period was from January 2004 to October 2014. RESULTS AND CONCLUSION:Electrospinning technology is the most effective method for preparation of nanofiber scaffolds. Electrospinning scaffolds as tissue engineering scaffolds have become an issue of concern in the basic research year by year. However, the internal nano-scale pore of nanofiber scaffolds limits the cels to grow on the surface, so recent research has been focused on highly porous three-dimensional structure which can promote the permeable growth of cels instead of two-dimensional scaffolds. Several techniques have been used, which go from the adjustment of materials and speed of electrospinning to the applications of various kinds of complicated machines. However, the existing researches are stil not mature and stable, the majority of which are applied onlyin vitro as cel implantation or subcutaneous implantation in smal animals. The above-mentioned methods stil need long-term comparative studies to confirm the feasibility in the tissue-engineered repair of organs.
