2.Effect of beclin1 on vincristine-induced dopaminergic neurons injury in zebrafish.

To investigate vincristine-induced dopaminergic neurons toxicity and mechanism, and explore the molecular target to reduce the toxicity, zebrafish was chosen as a model animal, based on RT-PCR, Western blotting, whole mount in situ immunofluorescence and other technical means. The results showed that the transcription levels of tyrosine hydroxylase gene and dopamine transporter protein gene were inhibited. Furthermore, the number of dopaminergic neurons was decreased by vincristine. Autophagy was suppressed and beclin1 gene expression was inhibited in a dose-dependent manner by vincristine in larval zebrafish. Up-regulated beclin1 partly reduced vincristine-induced neurotoxicity, and down-regulated beclin1 increased toxicity. Beclin1 plays an important role in vincristine-induced dopaminergic neurons toxicity.

3.Effects of intensive insulin therapy on non-specific inflammatory factors in patients of critical illness complicated with hyperglycemia

Objective To observe the effects of intensive insulin therapy on C-reactive protein (CRP) ,interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α ) in the patients of critical illness complicated with hyperglycemia and its incidence of side effect. Methods Two hundred and nine patients of critical illness complicated with hyperglycemia were randomly divided into intensive insulin therapy group (106 patients,blood glucose maintained at a level of 4.4-6.1 mmol/L) and conventional insulin therapy group ( 103 patients, blood glucose maintained at a level of 9.0-11.1 mmol/L). Serum levels of CRP,TNF- α and IL-6 were determined on 0,24,48,72 h respectively after ICU admission. Results The levels of blood glucose of both groups reached the target level. The incidence rates of the hypoglycemia had no significant difference between two groups [6.60 % (7/106) vs. 4.76% ( 3/63 ),P > 0.05]. After 72 h treatment, serum level of CRP in intensive insulin therapy group was significantly lower than that in conventional insulin therapy group (P < 0.05 ). After 24,48 and 72 h treatment, serum level of IL-6 in intensive insulin therapy group was significantly lower than that in conventional insulin therapy group (P < 0.05 ). After 48 and 72 h treatment, serum level of TNF-αin intensive insulin therapy group was significantly lower than that in conventional insulin therapy group (P < 0.05). Conclusion Intensive insulin therapy can significantly decrease the levels of non-specific inflammatory factors in patients of critical illness complicated with hyperglycemia, which brings beneficial effect to the patients.
4.Single nucleotide polymorphisms in the open reading frame 26 (ORF26) gene of human herpesvirus 8 (HHV-8) in Kaposi's sarcoma

ObjectiveTo study the single nucleotide polymorphisms(SNPs) in the ORF26 gene of HHV-8 in Kaposi's sarcoma(KS),and to assess their correlations with the clinical phenotype and mucosal invasion of KS.MethodsHHV-8 DNA was extracted with phenol-chloroform-isoamyl alcohol from paraffin-embedded tissue specimens obtained from 32 cases of KS(including 26 classic and 6 AIDS-related KS).The ORF26 gene of HHV-8 was amplified by nested-PCR followed by bidirectional sequencing.The software DNAStar and program Clustal W were used to assess the SNPs in the ORF26 gene.Statistical analysis was carried out by using the Fisher's exact probability test.ResultsHHV-8 DNA was detected in 30 of the 32 tissue specimens,and in all of the 6 AIDS-related specimens.The predominant SNPs were 981 T/C(n =12),1086 C/T(n =12) and 1139 A/C(n =12) in the ORF26 gene of the 30 strains of HHV-8.No significant difference was observed in the distribution of SNPs in ORF26 between different phenotypes of KS or between KS with and without mucosal invasion.ConclusionThe ORF26 SNPs of HHV-8 seem unrelated to the clinical phenotypes or mucosal invasion of KS.
5.CT features of lung alveolar echinococcosis

Objective To analyze the CT features of lung alveolar echinococcosis and assess the value of CT scanning for the diagnosis of this disease. Methods The CT features of lung alveolar echinococcosis in 26 patients diagnosed pathologically or clinically were reviewed retrospectively and correlated with histopathology.Results Lung alveolar echinococcosis appeared as multiple lesions bilaterally in 18 patients, as multiple lesions unilaterally in 6 patients, and as single lesion bilaterally in 2 patients. Seventeen cases presented as masses and nodules,8 patients presented as nodules,1 patient presented as a mass. Lobulation and spiculation were showed in most lesions, irregular cavity or bubble-like opacity in 13 patients,calcification in 18 patients,a patchy area or fibrotic cord around lesion in 14 patients. Pleural thickening adjacent to the lesion in 20 patients. Conclusion Lung alveolar echinococcosis has characteristic features on CT, and CT can provide important information for the diagnosis of lung alveolar echinococcosis.
6.Impact of mild hypothermia on changes of somatosensory evoked potential and synaptophysin mRNA level following traumatic brain injury

Objective To investigate effect of mild hypothermia on changes of somatosensory evoked potential (SEP) and synaptophysin mRNA level after traumatic brain injury (TBI) and determine hypothermia-induced neuroprotection.Methods Forty-five SD rats were allocated into mild hypothermia group,TBI group and sham operation group with 15 rats per group according to the random number table.Left-side fluid percussion impact was performed to induce models of TBI.Rats were exposed to hypothermia environment (32-35℃) for 6 hours in mild hypothermia group after TBI.Rats in sham operation group were treated by only drilling on left side of the head,rather than hitting.To evaluate function outcome,modified neurological severity score (mNSS),SEP and synaptophysin mRNA level were measured at 6 hours,24 hours and 7 days postinjury.Results The mNSS in mild hypothermia group lowered compared with TBI group,especially at 24 hours and 7 days (P < 0.05).SEP in mild hypothermia group was significantly shortened at 6 and 24 hours compared with TBI group (P < 0.05),but SEP revealed no significant difference among the 3 groups at 7 days (P > 0.05).Level of synaptophysin mRNA in mild hypothermia group increased at 6 hours postinjury compared with TBI group [(0.08 ± 0.02) vs (0.12 ±0.04)],with further increase at 7 days postinjury[(0.06 ± 0.01) vs (0.33 ± 0.10)] (P <0.05).Conclusion The shortage of nerve conduction time of the injured side and promotion of nerve regeneration suggest the neuroprotective role of mild hypothermia following TBI.
7. Structural characterization of Guiqi polysaccharides AAP-2A

Objective: To isolate the Guiqi (Angelicae Sinensis Radix and Astragali Radix) polysaccharides (AAP) and study the structural characterization of AAP-2A. Methods: Hot water extracting and ethanol precipitating method was employed to isolate the AAP. A new fraction (AAP-2A) of AAP was obtained after AAP was purified by deproteination, gel chromatography on DEAE-52 cellulose, gel chromatography on Sephadex G-100, and other means. Sugar content of AAP-2A was determined using phenol sulfuric acid method. Chromatography techniques were used to determine the ultraviolet properties, infrared properties, relative molecular weight, absolute molecular weight, molecular conformation and molecular weight distribution, and monosaccharide composition with the molar ratio of AAP-2A. The connection, main chain and branched-chain structures, and the condition of branching point were studied by methylation and NMR methods. Results: The total sugar content of AAP-2A, which was absent of proteins and nucleic acids, was 98.98%. Infrared spectroscopy showed that AAP-2A had the typical signals of polysaccharides. The relative and absolute molecular weights of AAP-2A were above 6.68 × 105 and 2.252 × 106; AAP-2A was a neutral sugar and composed of rhamnose (Rha), galactose (Gal), arabinose (Ara), and glucose (Glc) with a molar ratio of 1 : 2.13 : 3.22 : 6.18. AAP-2A proved to be a heteropolysaccharide, with 1, 3-α-L-Rha, 1,3-β-D-Gal, 1,3-, 1,5-, and 1,3,5-α-L-Ara, 1,4- and 1,4,6-α-D-Glc residues in backbone and 1-α-D-Glcp residues in branches. Conclusion: AAP-2A is a new neutral heteropolysaccharide from Guiqi polysaccharides with a highly branched conformation.
8.Iodine nutrition survey of children aged 8 to 10 in coastal and non-coastal areas of Rizhao city Shandong province

Objective To investigate the iodine nutritional status of children aged 8 to 10 in both costal and non-costal areas of Rizhao city Shandong province,and provide a basis for scientific supplementation of iodine.Methods In 2009 and 2010,townships of Lanshan and Donggang were chosen as survey points which situated close to the coast,the counties of Wulian and Juxian that were more than 50 kilometers away from the coast were selected as controls.The iodine concentration of drinking water in every village and water supply point was determined.At the county level,5 to 9 towns were selected according to their sub-area positions of east,west,south,north and center,4 villages were selected in each chosen township,8 - 15 households were selected in each chosen village,edible salt from the households was collected; 5 primary schools were selected in each chosen township,60 students aged 8 - 10 were selected to take thyroid examination in each chosen school.Twenty copies of urine samples were collected from the 60 students to detect the iodine concentration.Iodine in drinking water was tested by cerous sulfate catalytic spectrophotometry,in iodized salt by direct titration,in urine by arsenic-cerium contact method,and thyroids were examined by palpation.Results Totally 3483 copies of drinking water samples,1164 copies of edible salt samples,and 476 copies of urine samples were tested,and a total of 1200 children aged 8 to 10 were investigated; and the ratio of water iodine frequency distribution ≤ 10 μg/L (in costal and non-costal area) accounted for 90.03% ( 1011/1123 ) and 91.10% (2150/2360),respectively,the medians of water iodine were 5.6 and 4.2 μg/L,respectively; the iodine medians of edible salt were 29.03 and 29.99 mg/kg,respectively; the consumption rates of qualified iodized salt were 96.77% (569/588) and 97.05% (559/576),respectively; total goiter rates were 1.17%(7/600) and 1.33%(8/600),respectively; the medians of urinary iodine were 144.05 and 159.15 μg/L,respectively; the percentages of urinary iodine that bellow 100 μg/L were 26.58% (63/237) and 22.59% (54/239),respectively; between 100 - 300 μg/L were 66.75% (158/237) and 64.02% (153/239),respectively;above 300μg/L were 6.75% (16/237)and13.39% (32/239),respectively.ConclusionsThe costal of Rizhao is an iodine deficient area.The current popularity of household's consumption of qualified iodized salt is suitable.The levels of iodine nutrition of children aged 8 to 10 is appropriate,which met the national standard of elimination of iodine deficiency disorders (IDD).There is no iodine excess.There are no significant differences in levels of iodine nutrition between costal and non-costal areas on the premise that the same iodized salt is supplied in both the areas,and there is no need to make differentiated supply of iodized salt.
9.Application of case - based learning teaching mode in ophthalmology clinical teaching

AIM: To evaluate the case-based learning ( CBL ) in ophthalmology teaching practice.
METHODS: A total of 88 students were taught by the combination of lecture-based teaching with CBL form of sensory organs in Jinzhou Medical Teaching Classes. After class, the questionnaire was filled by each student to evaluate the CBL teaching mode.
RESULTS:The description of the data was analyzed as a percentage of each option. All the students were taught by CBL at the end of the class, most of the students could accept the teaching mode, it was helpful for students to understand the principle of clinical treatment, increased the interest in the process of teaching. And it was conducive to review the knowledge and exercise clinical case analysis ability. And it could recognize the ophthalmic diseases and special auxiliary examination. CBL teaching mode could stimulate students' interest in learning ophthalmology, it was beneficial to the cultivation of theory combined with practice and clinical thinking.
CONCLUSION: CBL teaching mode can be used as an auxiliary teaching lecture-based learning teaching.
10.Transport efficiency and safety evaluation by process management during the handover of patients with tracheal intubation post-operation

Objective To evaluate transport efficiency and safety by process management during the handover of patients with tracheal intubation post-operation. Methods Prospective studies were performed between patients with or without process management during the handover. The time of handover were recorded and compared. The difference between systolic and diastolic blood pressure, heart rate, and arterial oxygen saturation were also recorded and compared. The adverse events during the handover were also investigated. Results It costed less time in the handover of patients under process management with significant difference[(4.75±0.54) min vs. (7.05±0.88) min, t=-17.21, P<0.01]. The incidence rate of harmful cases in the handover of patients under process management was significantly declined than that without process management[ 1.67%(1/60) vs. 13.33%(8/60),Χ2=4.324 3,P<0.05 ]. Conclusions Process management may facilitate the handover of patients with tracheal intubation post-operation and improve its safety.
