Objective To explore the effects of rosiglitazone on the expression of TNF-? in visceral adipose tissue of insulin-resistant rats induced by high-glucose diet. Methods Sprague-Dawley (SD) rats were fed with high-glucose diet for 6 months to reproduce the non-obesity insulin-resistant rat model (abbreviated as IR rats). The model rats were then randomized into insulin resistan (IR) group and rosiglitazone (ROS) group (10 each). Ten rats fed with normal diet were enrolled as controls (CN group). Rats in ROS group were administered with 10?mol/(kg?d) of rosiglitazone for 6 weeks from the 7th week of experiment, and those in CN and IR groups were administered with same amount of distilled water. At the 6th and 12th week, the systolic pressure, body weight, and biochemical parameters (fasting and 2-hour post-glucose load plasma glucose, insulin, free fatty acid, triglyceride, high density lipoprotein cholesterol) were measured, and insulin resistance index (HOMA-IR) was calculated. At the end of the 12th week, the visceral adipose tissue was collected to calculate the ratio of visceral fat to body weight (VF/BW),and to determine the protein and mRNA expressions of tumor necrosis factor-? (TNF-?) with inmmunohistochemical assay and RT-PCR. Results As compared with CN group, the systolic pressure,HOMA-IR,plasma insulin, triglyceride and free fatty acid increased, whereas high density lipoprotein cholesterol decreased (P0.05). Inmmunohistochemical examination showed positive expression of TNF-? in intercellular substance, but not in visceral adipocytes. The expressions of TNF-? protein and mRNA in visceral adipose tissue were dramatically higher in IR group than in CN and ROS groups (P0.05). Conclusion Rosiglitazone may significantly abate insulin resistance, reduce the abnormality of plasma lipid and blood pressure, and inhibit the over expression of TNF-? in visceral adipose tissue in the non-obesity insulin-resistant rats induced by high-glucose diet.

Background:Blood oxygenation level-dependent magnetic resonance imaging (BOLD-MRI) is a functional MRI technique which involves using the paramagnetic properties of deoxyhemoglobin to image the local tissue oxygen concentration. The purpose of this study was to investigate whether BOLD-MRI could evaluate hypoxia and angiogenesis of breast invasive ductal carcinoma (IDC).Methods:Ninety-eight female patients with IDC were retrospectively included in this research. All patients underwent breast BOLD-MRI at 3.0 T before surgery.R2* values of BOLD-MRI were measured. The expression of carbonic anhydrase IX (CA IX) and vascular endothelial growth factor (VEGF) was analyzed by immunohistochemistry.Spearman's correlation analysis was used to correlate R2* value with CA IX and VEGF levels. Results:Heterogeneous intensity on BOLD-MRI images was the main finding of IDCs.The mean R2* value was 52.8 ± 18.6 Hz.The R2* values in patients with axillary lymph node metastasis were significantly higher than the R2* values in patients without axillary lymph node metastasis (t =2.882,P =0.005).R2* values increased with CA IX level and positively correlated with the level of CA IX (r =0.616,P < 0.001);however,R2* value had no significantly correlation with the level of VEGF (r =0.110,P =0.281).Conclusion:BOLD-MRI could noninvasively evaluate chronic hypoxia of IDC,but not angiogenesis.

OBJECTIVETo investigate the effect of four traditional Chinese medicines distributed along lung meridian, namely Ephedrae Hebra, Zingiberis Rhizoma, Scutellariae Radix and Mori Cortex, on TLR2 and NF-κB expressions in mice with lung heat syndrome, in order to study the intervention effect of the four traditional Chinese medicines (TCMs) on the lung heat syndrome.
METHODOne hundred KM mice were randomly divided into the normal control group, the model group, the Ephedrae Hebra group, the Zingiberis Rhizoma group, the Scutellariae Radix group and the Mori Cortex group (20, 10 g x kg(-1)), nasally dripped with streptococcus pneumoniae to establish the mouse lung heat syndrome model, and then administered with different TCMs. The expressions of TLR2, NF-κB p65 proteins in lung tissues were analyzed by the immunohistochemical method. The expressions of TLR2, NF-κB p65 mRNA were measured by real time PCR.
RESULTCompared with the normal control group, the expressions of TLR2 and NF-κB p65 proteins in lung tissues in the model group were higher (P < 0.01), and the expressions of TLR2 and NF-κB p65 mRNA in lung tissues were up-regulated (P < 0.01 or P < 0.05). Compared with the model group, Ephedrae Hebra high and low dose groups, the Zingiberis Rhizoma low dose group and the Scutellariae Radix high dose group showed decreased expression of TLR2 protein (P < 0.05 or P < 0.01); Ephedrae Hebra high and low dose groups, the Zingiberis Rhizoma low dose group, Scutellariae Radix high and low dose groups and Mori Cortex high and low dose groups showed reduced expression of NF-κB p65 protein (P < 0.05 or P < 0.01). Ephedrae Hebra high and low dose groups, Zingiberis Rhizoma high and low dose groups, Scutellariae Radix high dose group and Mori Cortex high dose group showed down-regulated expression of TLR2 mRNA (P < 0.01 or P < 0.05).
CONCLUSIONEphedrae Hebra, Zingiberis Rhizoma, Scutellariae Radix and Mori Cortex can induce the TLR2/NF-κB inflammatory signal pathways by down-regulating the expressions of TLR2 and NF-κB p65 in protein and mRNA, so as to alleviate the lung tissue injury in mice with lung heat syndrome.
