1.Culture and transplantation of autologous corneal limbal stem cells

OBJECTIVE: Autotransplantation with cultured autologous limbal stem cell is viewed as the best treatment. Therefore, it is necessary to further recognize the molecular biological character of limbal stem cell and histological anatomy and to study the progression of transplantation of autologous limbal stem cell.DATA SOURCES: The papers of Medline January 1995 to December 2002 were looked up in computer by retrieved words "limbus cornea, stem cells,autotransplant" of English version. Simultaneously, the papers of January 1995 to January 2002 of Chinese version were looked up by the same retrieved words in Chinese on http:∥www.wanfangdata.com.cn.STUDY SELECTION: It is to examine initially the papers relevant with studies on histology and physiopathology of limbal stem cells and relevant with DATA EXTRACTION: More than 120 papers of relevant literatures were collected in total and 15 papers of those met the criteria and selected for synthetic analysis.cells: Stem cells of corneal epithelium present similar characters as other committed stem cells, with very strong capacity of proliferation and cell division, in poor differentiation state, long cellular cycle and unparallel divipapillary structure of limbal basal palisades of Vogt are limbal stem cells.sion of stem cells are controlled by serum-source factors and various kinds of cellular factors, the changes in factors in microenvironment affect directly the proliferation and division of limbal stem cells and result in various limbal stem cells of the healthy eye from the patient with several ocular surface diseases cultured are grafted to amniotic membrane carrier for ocular surface autotransplantation on affected eye. It is observed after operation that corneal epithelium is regenerated in 2 to 5 days and visual sensibility is remarkably increased in l month. It is visible in 4-month observation after operation that corneal epithelium is integrative and neogenetic vessels are removed persistently.CONCLUSION: Autotransplantation of cultured limbal stem cells is still at the phase of experimental research, in which, initial success has been achieved in treatment of ocular surface diseases; but the remote therapeutic effect of such transplantation is required a further clinical observation. A further study is expected on proliferation, differentiation, physiology and biochemistry of cultured limbal epithelial stem cells, on screening of unique monoclonal antibody of stem cells, on purification and culture in vitro in stem cells, as well as on how to control the differentiation of stem cells and the influence of cellular factors on stem cells. Whether is it possible to apply allogeneic transplantation of cultured limbal stem cells for disorders of both eyes with deficiency of any source of autologous limbal stem cells? Whether is it possible to reduce incidence of rejection by the possible alternations on antigenicity of cultured limbal stem cells? There would be more prospects in the study on those topics.

6.Analyze on the infection of human papilloma virus among gynecology clinical cases

Objective To analyze the epidemic situation of HPV among gynecology clinical cases and provide scientific support for control and prevention of squamous cell carcinoma .Methods Clinical cases with symptom of increased vaginal secretions and obscure vaginal bleeding and cases diagnosed with cervical squamous cell carcinoma or vaginitis and cervicitis were collected as ob-jectives for HPV testing .Samples were collected by maternity doctors for HPV DNA testing by using the reverse hybrid membrane technology gene chip .All data were analyzed by chi-square test and trend analysis(Z) method .Results 24 .64% of the 4 347 cases were positive for 23 kinds of HPV subtypes .Of the 23 subtypes ,the leading four detection rates were 16th ,43th ,58th and 52th sub-types ,which were 8 .10% (352/4 347) ,3 .93% ,3 .52% and 3 .31% respectively ,and shown significant higher than that of the other 19 subtypes .The detection rates of 6th ,11th ,18th ,33th and 56th subtypes were between 1% to 3% ,whereas the rates of other 14 subtypes were lower than 1% .The detection rates of all the age groups were between 16 .00% ~36 .72% ,and rose with the age in-creasing(Z=3 .32 ,P<0 .01) .The detection rate was the highest in symptoms of increased vaginal secretions and obscure vaginal bleeding ,accounted for 30 .76% (334/1 086) ,secondly in those of increased vaginal secretions ,obscure vaginal bleeding and men-strual were between 20 .68% and 25 .59% .All the 80 cervical squamous cell carcinoma cases were detected HPV ,97 .5% (20/80)of them were high-risk HPV type .The single infection was the most common infection in cervical squamous cell carcinoma cases ,Ac-counted for 75 .5% (60/80)of the total ,and 25% (20/80)of them were multiple infections with 2 to subtypes ,and 70% (56/80) of them were infected with 16th subtypes ,and only2 .5% of them were low-risk type(2/80) .The detection rate of inflammatory disea-ses was 4 .75% (958/3 871) ,and the detection rates were higher in cases of cervicitis and vaginitis and cervicitis complications (be-tween 23 .03% to 28 .38% ) than that of vulvitis (16 .81% )(χ2 >3 .83/6 .63 ,P< 0 .05/0 .01) .Conclusion The detection rate of 16th subtypes HPV was the highest for gynecology clinical cases ,secondly were the 43th ,58th and 52th subtypes .The detection rate of HPV was the highest in symptoms of increased vaginal secretions ,obscure vaginal bleeding and vaginitis .The detection rate rose with the age increasing .It was concluded that elderly cases with increased vaginal secretions ,obscure vaginal bleeding and vagi-nitis were the biggest targets for control and prevention of HPV and cervical squamous cell carcinoma .
7.Association between Serum C-Reactive Protein and Benign Prostatic and Patients with Prostatic Cancer Hyperplasia

Objective To explore the association between C reactive protein (CRP)with prostatic cancer (PCa)and benign prostatic hyperplasia (BPH).Methods Retrospective analysis the 110 patients of the First Affiliated Hospital of Zhejiang University In January 2010 to August 2012 whose TPSA>4 ng/ml,postoperative pathology confirmed the diagnosis of 54 cases of prostate cancer,5 6 cases of benign prostatic hyperplasia.Detected serum CRP levels by using transmission turbidim-etry and TPSA levels by using chemiluminescence immunoassay of 54 PCa and 56 BPH patients.According to the Gleason score,PCa patients were divided into high-risk and low-risk PCa two groups,the differences among high-risk PCa,low-risk PCa and BPH groups were analyzed by nonparametric statistics analysis.Results The CRP level of high-risk PCa was 4.20~2.12 mg/L,the CRP level of low-risk PCa was 1.90~0.91 mg/L and the CRP levels in BNP patients was 1.49±0.87 mg/L,the high-risk and low-risk PCa serum CRP level obviously higher than that of patients with BPH,the differences were statistically significant (P<0.05).Conclusion Serum CRP levels of PCa patients were increased significantly,espe-cially in high-risk PCa patients.

Phosphatidylserine (PS) is a phospholipid that is abundant in eukaryotic plasma membranes,has crucial biological functions.Under cell apoptosis, cells can not generate enough ATP for energy and the concentration of cytoplasmic Ca 2 +increases, resulting in PS eversion.Apoptosis and the clearance of apoptotic cells are essential processes in animal development and homeostasis.For apoptotic cells to be cleared, they must display aneat me signal, most likely PS exposure, which prompts phagocytes to engulf the cells.PS is exposed by the action of scramblase on the cell's surface in biological processes such as apoptosis and platelet activation.Once exposed to the cell surface, PS acts as an eat me signal on dead cells, and creates a scaffold for blood-clotting factors on activated platelets.The molecular identities of the flippase and scramblase that work at plasma membranes have long eluded researchers.Indeed, their identity as well as the mechanism of the PS exposure to the cell surface has only recently been revealed.We describe how PS is exposed in activated platelets and in apoptotic cells, and discuss the clearance of apoptotic cells.

In recent years, with the rapid development of antitumor targeted drug therapy, the preparation of antitumor liposomes and its targeting research have attracted attention of scholars. Anti-tumor targeted drug treatment requires the drug to reach the tumor site has a relatively high concentration and longer retention time, the tumor cells have a strong cytotoxic activity, while normal cells no significant side effects.The drug has an in vivo distribution and specificity for the target fine action.The preparation methods of liposomes include film dispersion method,reverse evaporation method,ethanol injection method,supercritical reverse phase evaporation method and freeze-drying method. Antitumor targeting liposomes can be divided into active targeting liposomes and passive targeting liposomes. Targeted liposomes can specifically target tumor cells by recognizing specific targets in the tumor tissue,thus enriching tumor cells and killing tumor cells. In this paper,the preparation of anti-tumor liposomes and the progress of its targeting,for future study and application of liposomes provide a reference.
