1.Expression and position fixing of bFGF in retina of diabetic rabbits

Objective To investigate the expression period of bFGF and the correlation between the M?ller cells in retina and the increasing of bFGF expression in diabetic rabbits.Methods Sixteen rabbits were divided randomly into normal control and diabetic groups.After the models of diabetic rabbits were set up,the rabbits were breeded for 7 weeks.The immunofluorescence double labeling method and laser confocal microscopy were used to observe the ratina of the diabetic rabbits and the position fixing of bFGF was studied.Results In normal control group,the structure of each layer of retina was clear,the cell lined up tightly and regularly with normal shape,the location with red fluorescence was M?ller cells signaled with GFAP,labeled with bFGF,only on capillary basement membrane the red fluorescence could be found.In diabetic group,the structure of each layer of retina was clear,the cells of INL and GCL lined up loosely,the intercellular space increased,there was no significant difference compared with normal group on cell shape.Labeled with bFGF,the different intensity red fluorescence could be found in each layer cells,double labeled with GFAP and bFGF,the yellow fluorescence could be found.The retinopathy of diabetic rabbit was in BDR stage.The expression of bFGF increased in this stage.The bFGF and GFAP coexpressed in M?ller cells.Conclusion The bFGF begins to express in the M?ller cells from the BDR stage.
2.Relationship between normal weight obesity and cardiovascular risk factors

Objective To determine the relationship between normal weight obesity (NOW) and cardiovascular risk factors.MethodsA total of 940 adults who received a health examination in out hospital were recruited in a cross-sectional study,and 407 with a body mass index (BMI) of18.5 to 25 kg/m2were enrolled for further analysis.Body fat percentage ( BF% ) was measured by bioelectrical impedance analysis (BIA),and the subjects were assigned to the NOW group ( BF% ≥25% for male or BF% ≥35%for female) or the control group ( BF% < 25% for male or BF% < 35% for female).Cardiovascular risk factors and their detection rates were compared between the two groups by using independent sample t test and x2 test.The correlationbetweenNOW and cardiovascular risk factors was assessedbylogistic regression.Results The prevalence of NOW in men and women were13.1% and14.9%,respectively.The prevalence of NOW was increased with age ( x2 =6.90,P <0.05 ).Systolic blood pressure (SBP),diastolic blood pressure ( DBP ),total cholesterol ( TC ),triglycerides ( TG ),low-density lipoprotein cholesterol ( LDL-C) and serum uric acid (SUA) were significantly increased in the NOW group (t values were 2.97,2.44,2.54,5.09,2.71and 3.91,respectively; all P < 0.05 ) ; whereas high-density lipoprotein cholesterol ( HDL-C) was significantly decreased in the NOW group (t =-3.90,P < 0.05 ).The prevalence of hypertension,hyperglycemia,high triglyceride,low HDL-C,dyslipidmia and hyperuricemia was increased in the NOW group in comparison with the control group ( x2 values were 6.76,5.58,14.50,11.97,10.97 and 8.76,respectively;allP< 0.05 ).LogisticregressionshowedNOWincreasedtheriskof hypertension,hyperglycemia,dyslipidmia or hyperuricemia by 2.186,2.120,2.088 or 4.175 times.After adjustment for age and gender,the risk for hyperuricemia was decreased to 3.491,but remained statistically significant higher.Conclusions NOW may be correlated with cardiovascular risk factors,and those with NOW could be at higher risk for cardiovascular diseases.
3.Metabolic syndrome, metabolic syndrome components and thyroid nodule

Objective To evaluate the relationship of metabolic syndrome (MS) and MS components with thyroid nodule. Methods A total of 10 357 subjects ( age > 18 years old) who received physical checkup at the Second Affiliated Hospital of Dalian Medical University between June 2009 and June 2010 were enrolled in this retrospective study. Anthropometric parameter, fasting plasma glucose (FPG),serum lipid profile, blood uric acid, alanine aminotransferase and thyroid ultrasonography were measured. Results The prevalence of thyroid nodule,MS,and thyroid nodule + MS was 46. 96% ,23. 6%,and 11.6%, respectively. The prevalence of thyroid nodule was significantly higher in MS patients than in non MS patients ( 75.9% vs 38. 0%, P < 0. 05 ). Multifactor logistic analysis showed that MS, body mass index (BMI) and FBG (β vales were 0. 78,1.22,and 0. 62,respectively; odds ratios were 4. 167,3. 876,and 2. 359, respectively; all P < 0. 05 ) were independently correlated with the development of thyroid nodule. Conclusions Significantly increased prevalence of thyroid nodule could be found in MS patients. BMI and FBG may be independent risk factors of thyroid nodule.
4.Three-dimensional simulation of the structures of human N-acetyltransferase 2 protein molecule

Objective:To simulate human N-acetyltransferase 2 protein 3D model by computer technology. Methods: 3D model of human N-acetyltransferase 2 protein was simulated with protein list and computer technology,the physical,chemical,biological characteristics and the functions were analyzed.Results: The possible structure of NAT2 was simulated using ANTHEPORT5.0,a software for determining the sequence of protein,forecasting the pHi and molecular weight of NAT2 were 5.495 and(33 544) respectively.The titration curve and physics-chemistry characteristic curve of NAT2 were also obtained.A Helical Wheel chart was plotted,aiming at the fragment of NAT2 around amino acid 282 nd.Conclusion:The possible structure of NAT2 was simulated using computer,analysis and forecast was also carried out,the method was feasible to genetic analysis of NAT2.
5.Establishment of liver fibrosis in rabbit model by injecting carbon tetrachloride intraperitoneally and drinking ethanol water

Objective To explore an ideal approach to establish rabbit model of hepatic fibrosis suitable for radiological and serological research.Methods Totally 40 rabbit models of hepatic fibrosis was established by intraperitoneally injecting 5% carbon tetrachloride in oil solution or 100% carbon tetrachloride(both groups,0.1 ml/kg,once a week),and drinking 5% ethanol water.Another 8 rabbits who drank normal water and received a peritoneal injection at 0.1 ml/kg served as control.The rabbits(5 or 2 per time for model group and control) were killed at 6,8,10 and 12 weeks respectively after first injection.Their livers were resected for gross and morphological observation with HE and Masson staining.Results Death of the models usually happened within 4 weeks after first injection and became stable after 6 weeks.Mortality of 5% carbon tetrachloride group was 60%,but 25% in 100% carbon tetrachloride group.Macroscopy and microscopy indicated that liver fibrosis was observed in rat models.Conclusion Long-term intraperitoneal injection of carbon tetrachloride results in rabbit hepatic fibrosis.Injection of 100% carbon tetrachloride intraperitoneal,at 0.1 ml/kg,once a week,combined with and 5% ethanol as drinking water is a suitable approach to establish rabbit model of hepatic fibrosis with low mortality and high success rate.
6.Diagnosis and surgical treatment of functional insulinomas: report of 132 cases

Objective To investigate the diagnosis and surgical treatment of functional insulinomas.Methods One hundred and thirty two patients from January 2002 to May 2013 were retrospectively reviewed who was confirmed as functional insulinomas by pathology.Results There were 43 males and 89 females.56 cases were located in the head of pancreas,76 body and tail of pancreas.69 patients underwent open enucleation,48 patients had Laparoscopic surgery,5 patients underwent distal pancreatectomy,9 patients had Laparoscopic distal pancreatectomy,1 patients had pancreaticoduodenectomy.The difference of variant imaging examination in Checking insulinomas was statistical significant(P <0.01).Laparoscopic treatment of insulinomas was at least as safe as open treatment.Conclusions Imaging examination could improve the accuracy of preoperative diagnosis of insulinomas.Surgery was the most effective methods to cure insulinomas and Laparoscopic surgery was becoming increasingly popular in treating insulinomas.
7.Effect of advanced glycosylation end products on bFGF expression of cultured rabbit Müller cells

Objective:To observe the effects of different doses of advanced glycosylation end products ( AGEs ) on bFGF expression of cultured rabbit M üller cells in vitro.Methods:Immunocytochemistry and transmission electron microscopy methods were used identified cultured M üller cell.Immunocytochemistry method was used to semi-quantitate bFGF expression of retinal Müller cells at 640 μl/2 000 μl AGEs conditions.We observed effects of AGEs and PKC inhibitor Calphstion C on bFGF mRNA expression .Results:640 μl/2 000 μl AGEs stimulate bFGF expression of retinal Müller cell.Calphostin C inhibits bFGF mRNA increase stimulated by AGEs,and inhibition achieves strongest at concentration 50 nmol/L.Conclusion:AGEs can stimulate bFGF expression of Müller cell to exert the role of angiogenesis .bFGF mRNA expression may be regulated by activation of PKC pathway .
8.An experimental study of bio-polymer composite film as a micro-skin autograft covering

Objective To explore the promoting effects of bio-polymer composite film as a micro-skin auto-graft covering on wound healing. Methods The full thickness skin defect models were made on both sides of 30 experimental rabbits. Then, the rabbits were randomly divided into experimental group and control group. In the for-mer group, the side was covered with chitosan/glucomannan composite membrane and in the latter, the side cov-ered with acelluar porcine skin after micro-skin autograft. We obtained wound tissues at week 1 , 2, 3, 4 and 5 af-ter operation. The conditions of wound healing were observed, the rate of wound healing was calculated, HE stain-ing was made, and PCNA and CD31 were detected by immunohistochemistry. Results (1) During 2~4 weeks af-ter operation, the rate of wound healing in the experimental group was significantly higher than that of the control wound (P<0.01). (2) The amount of neutrophil in experimental group was less than that of the control after oper-ation. (3) During 1 ~ 2 weeks after operation, the expression of PCNA in the experimental group was higher than that of the control group (P < 0.01), but lower than the control wounds during 1 ~ 2 weeks after operation (P <0.01). (4) During 1 ~ 5 weeks after operation, the expression of CD31 in the experimental group was higher than that of the control group (P < 0.01). Conclusion Chitosan/glucan-mannan composite membrane as a micro-skin autograft covering may promote wound healing.
9.Risk prediction of type 2 diabetes mellitus in a health check-up population

ObjectiveTo develop a simple scoring system to identify individuals with higher risk of type 2 diabetes mellitus ( T2DM ).Methods Major risk factors of T2DM were fixed after literature review.Logistic regression was used to select risk factors of T2DM.Target populations were randomly assigned to 2 groups,and group A was used for the development of risk factor scoring method and group B for the confirmation of this new method.Results The threshold for T2DM risk was 65.0 for men and 65.5 for women ( sensitivity 90.6% and 83.3%,respectively; specificity 89.4% and 97.7%,respectively ; positive predictive value 58.8%and 80.0%,respectively;negative predictive value 98.3%and 98.2%,respectively ; area under the curve 0.955 and 0.899,respectively).There was significant difference of T2DM detection between screening and risk scoring system ( P =0.000 ).Conclusion Our T2DM risk scoring system may provide an effective tool to identify individuals with a higher risk of developing T2DM.
10.Study of the relationship among the uric acid,lipid and insulin resistance in patients with different glucose levels

Objective To study the levels of senlm uric acid(UA)in normal glucose(NC),impaired glucose regulation(IGR)and type 2 diabetes mellitus(T2DM),and investigate its relationship with insulin resistance and dyslipidemia.Method The levels of blood glucose,lipids,fasting insulin(HNS)and serum UA were measured in patients of 45 T2DM(T2DM group),20 IGR(IGR group)and 29 NC(NC group).Status of insulin resistance and insulin secretion function was evaluated by HOMA-IR and ISI.Results The levels of triglyceride(TG)and UA in T2DM group and IGR group were significantly higher than those in NC group[(3.34±8.77),(1.85±0.67),(1.26±0.38)mmoi/L and(316.71±96.20),(403.62±76.80),(325.45±94.43)mmol/L](P<0.01).HDL-C levels in T2DM group were significantly lower than those in IGR and NC group[(1.05±0.30),(1.07±0.21),(1.12±0.20)mmol/L](P<0.01).NO significant difference of FINS levels was found in the three groups.HOMA-IR level in T2DM and IGR group was higher than that in NC group(3.84,3.77,2.34)(P<0.01).ISI in T2DM and IGR group was lower than that in NC group(-4.52±0.79,-4.44±0.19,-4.03±0.58)(P<0.01).Correlation analysis indicated that the level of UA was positively related with BMI.TG and negatively related with HDL-C.Conclusion Increased UA in IGR indicates that hyperurieacidemia developes before DM.
