1.Therapeutic potential of recombinant adenovirus expressing p53 on hepatocellular carcinoma cells possessing different p53 functional status

Objective To determine the effect of exogenous wild-type p53(WT-p53) on the growth of hepatocellular carcinoma (HCC) cell lines possessing different p53 functional status.Methods With recombinant adenoviral vector expressing WT-p53(Ad-p53),p53 gene was transfected into HCC cell lines:Bel-7402,QGY-7701, and PLC/PRF/5.The cytotoxicities of Ad-p53 to these cell lines were measured by MTT assay,cell growth properties and cell cycle pattern were assessed by flow cytometry.Results The OD of PLC/PRF/5, Bel-7402,and QGY-7701 cells transfected with Ad-p53 of 40,200,and 800 MOI were 0.02?0.00 vs 0.98?0.16(P0.05),compared with control groups respectively. Analysis of flow cytometry showed higher G2/M? lower S and apoptosis ratio in PLC/PRF/5 cells,while higher G0/G1 ratio and lower S ratio in Bel-7402 cells. ConclusionReplication-deficient adenoviral vector expressing WT-p53 may be useful for gene therapy in HCC regardless the status of endogenous p53.
3.Analysis of risk factors for local tumor progression after radiofrequency ablation of hepatocellular carcinoma

ObjectiveTo evaluate prognostic factors affecting local tumor progression after radiofrequency ablation (RFA) of hepatocellular carcinoma (HCC).MethodsA total of 246 HCC patients (343 lesions) underwent RFA treatment in our department and were enrolled into this study.The average tumor size was 3.7 cm ( range 0.9 ~ 3.7 cm).Regular follow-up with enhanced CT was performed to evalutate the treatment results.Kaplan-Meier model and log-rank test were used in univariate analysis and COX regression model was used in multivariate analysis to identify risk factors for local tumor progression.ResultsThe local tumor progression rate was 11.4% (39/343 lesions),and the average time from initial RFA to local tumor progression was 12.0 months.Univariate analysis indicated tumor size ( P <0.001 ),close to intrahepatic vessels ( P <0.001),tumor boundary ( P =0.020),pathological grade( P =0.010) and CEUS before RFA ( P =0.001) as risk factors for local progression.The following factors were identified as independent prognostic factors for local tumor progression by multivariate model:tumor size (P < 0.001),isolated or close to intrahepatic vessels( P <0.001) and CEUS before RFA(P =0.018).ConclusionsTumor size,CEUS before RFA and close to intrahepatic vessels are the most important factors for local progression after RFA.Being awaring of possible risk factors for local tumor progression may increase the treatment efficacy and help to promote the use of RFA technique.
4.Immunoregulatory Effects of Polysaccharide of Mudan Cortex on Immunosupressive Mice

Objective To study immunoregulatory activity of polysaccharide of mudan cortex (PSM) on immunodepressive mice. Method The immunodepressive mice model was established with cyclophosphamide treated and tumor-bearing. PSM was orally administered to mice for 10 d. MTT method was used to observe the prolifernation of lymphocytes. The function of macrophage in abdominal activity was measured. Sectrophotometric determination was adopted to evaluate the ability of plaque forming cell. Result PSM could enhance the function of macrophage, increase the content of plaque forming cell, and promote the prelifernation of lymphocytes. Conclusion PSM could enhance immunofunction of immunodepressive mice.
5.Effect of concentrations of sucrose on Streptococcus mutans gbpC expression

Objective To assess the influence of concentrations of sucrose on Streptococcus tautens (S. mutans) gbpC expression. Methods Total RNA was extracted from S. mutans after cultured in 0.5%, 1.0%, 5.0% sucrose and then reversely transcripted to cDNA. By TaqMan RT-RCR technique, we checked gbpC expression. Results Compared with 0.5% sucrose, 1.0% sucrose significantly upregulated gbpC expression(P<0.05); gbpC expression of more-adherent S. mutans strains was higher than that of less-adherent strains, Conclusion The upregulation of gbpC expression by sucrose may be one of the mechanisms underlying the increased adherence; There might be some correlation between gbpC expression andS. mutans adherence.
6.Clinical analysis of 40 elderly patients with malignant tumor undergoing venous thromboembolism

Objective To study the influencing factors,treatment and prevention of venous thromboembolic (VTE) in elderly patients with malignant tumor.Methods The clinical findings in forty elderly patients with malignant tumor combined with VTE from our hospital within 5 years were analyzed retrospectively.The risk,site,time,prevention and treatment of VTE in these tumor patients were analyzed and summarized.Results After active treatment,the positive efficacy response was obtained in 26 cases (65.0%),no-response in 14 cases (35.0%).Specifically,14 cases died,7 cases cured,and 19 cases were improved.Besides cancer and aging,the elderly tumor patients were considered as the high-risk group because of the risk factors including hypertension,diabetes,hyperlipidemia,pulmonary infection,surgery,radiation,chemotherapy and so on.D-dimer level was significantly increased in elderly malignant tumor patients with VTE.Venous thrombosis could occur at any stage.Venous thrombosis was found before tumor in 1 case.The most common site of DVT was the lower limbs,especially the left lower extremities (14 cases,35%).Most of internal carotid vein and upper limb vein thrombosis were associated with peripherally inserted central catheter (PICC) Conclusions The elderly patients with malignant tumor is the extremely high risk groups for venous thrombosis,and their quality of life and survival will be seriously impacted.Anticoagulant therapy through the whole process of treatment in patients with VTE is beneficial.The prevention and treatment of VTE play an important role in the tumor patients with risk factors such as chronic diseases,high D dimer level,radiation and chemotherapy,which should attract the attention of clinicians.

Human proximal tubule epithelial cell line,HK-2 cells,were cultured with various concentrations of insulin for 48 h.Human urate transporter (hUAT) mRNA was detected by realtime quantitative PCR.hUAT mRNA levels were down-regulated by insulin (5,25,125,500 μIU/ml)in a dose-dependent manner (relative expression median were 0.95,0.40,0.24,and 0.23).In vitro,the expression of hUAT mRNA in HK-2 cells is associated with the concentration of insulin.

Objective To establish a recombinant eukaryotic expression vector containing human interferon-?(IFN-?) cDNA,and investigate the expression of IFN-? gene in transfected hepatocellular carcinoma (HCC) cell lines. Methods pcDNA3-IFN-?,established by subcloning IFN-? cDNA into a mammalian expression vector pcDNA3,was mediated into HCC cell lines SMMC-7721 and QGY-7701 by lipofectamine. The transfected HCC cells were selected in RPMI1640 containing G418(400~700?g).RT-PCR analysis and ELISA assay were used for mRNA transcription and protein expression of IFN-? gene. Results IFN-? mRNA was detected only in pcDNA3-IFN-? transfected cell lines, and high levels of IFN-? protein were detected only in the supernatants of IFN-? gene-modified cell lines. 9.Analysis of correlation factors between peripapillary duodenal diverticulum and choledochectasia by CT scan

Objective:To analyze the correlation factors between peripapillary duodenal diverticulum (PDD) and choledochectasia by CT scan.Methods:The clinical data of 220 patients with duodenal diverticulum detected by multi-slice spiral CT scan and confirmed by gastrointestinal angiography or endoscopic retrograde cholangiopancreatography (ERCP) in Dahua Hospital, Xuhui District of Shanghai City were retrospectively analyzed. The correlation of the PDD, the contact of common bile duct (CBD), length of contact and exudation with choledochectasia in patients with PDD were analyzed.Results:A total of 236 duodenal diverticulum were found in 220 patients. Among them, there were 152 PDD, 41 diverticulum located superior to the duodenal papilla, 28 diverticulum located inferior to the duodenal papilla, 3 diverticulum located lateral to the duodenal papilla, and 12 diverticulumlocated in the horizontal portion. The incidence of choledochectasia in patients with PDD contacted with CBD was significantly higher than that in patients with PDD not contacted with CBD: 59.35% (73/123) vs. 37.93% (11/29), and there was statistical difference ( P<0.05); the incidence of choledochectasia in patients with contact length of PDD and CBD ≥1.5 cm was significantly higher than that in patients without contact of PDD and CBD and patients with contact length of PDD and CBD <1.5 cm: 82.43% (61/74) vs. 24.49% (12/49) and 37.93% (11/29), and there was statistical difference ( P<0.05); the incidence of choledochectasia in PDD patients with exudation was significantly higher than that in PDD patients without exudation: 10/11 vs. 52.48% (74/141), and there was statistical difference ( P<0.05). Conclusions:The patients with contact length of PDD and CBD ≥1.5 cm and patients with PDD combined with exudation could be prone to choledochectasia.
