Background The combination procedure of the removal of silicon oil and cataract extraction+intraocular lens (IOL) implantation is the main method of treating cataract in silicone oil tamponade eye.However,the conventional measuring method of the IOL power is limiting in eyes with silicone oil tamponade.The IOL-Master system is confirmed to be an ideal non-contact optical measuring instrument in vivo.Objective The aim of this study is to measure the preoperative IOL power,evaluate the accuracy of biometry with the IOL-Master system and compare the relationship between different factors and refractive error.Methods IOL power was measured in 29 silicone oil tamponade eyes of 29 patients prior to the silicone oil removal combining IOL implantation with the IOL-Master system according to different factors such as placement time,axis oculi,complications and so on.The reasons of visual acuity recovery and measurement error were discussed.Results Postoperative visual acuity was improved in all of the patients.The average predictive refractive error was 0.329±0.846 (-1.5--2D).Classified factors for the cause of disease for axis oculi included rhegmatogenous retinal detachment (RRD)(t=0.478,P=0.637),macular hole(t=0.135,P=0.895),myopia(t=0.435,P=0.667)and oil-placed time (good for less than 1 year).These factors showed an indirect relationship with errors.Conclusion Silicone oil removal combined with IOL implantation can improve visual acuity.The IOL-Master biometry is accurate,safe and convenient in measuring the IOL power in silicon oil temponade eye,but future research should be performed to decrease the measuring error.

Objective:The pathogenesis of endometriosis(EMT)was explored by detecting the relationship of progesterone receptors(PR)and apoptosis-associated factors with EMT.Methods:Immunohistochemical imaging analysis was used to quantify the expression of PR,Bcl-2 and Bax in ectopic and eutopic endometrium in patients with EMT.Results:Expressions of PR and Bcl-2 in eutopic endometrium are higher than those in ectopic endometrium(P<0.05).Expression of Bax in ectopic endometrium is higher than that in eutopic endometrium(P<0.05).There is no statistical difference of PR,Bcl-2 and Bax expression between proliferative and secretary endometrium.Conclusions:Either in proliferative or secretary phase,expressions of PR in eutopic endometrium are higher than those in ectopic endometrium,thus eutopic endometrium is more liable to proliferate in low level continuously,migrate and implant.Expressions of Bcl-2 and Bax in eutopic and ectopic endometrium have no relationship with endometrium cycling.The apoptosis of ectopic endometrium is weakened and its proliferative power is continuously enhanced.

Objective To investigate the application of high frequency ultrasound in knee joint lesions of elderly patients with rheumatoid arthritis.Methods Fifty patients with rheumatoid arthritis were collected in Yangpu Hospital Affiliated to Tongji University from March 2015 to December 2016 as test group.Fifty healthy elderly people without knee joint lesion were randomly selected as control group.High frequency ultrasound was used to examine the patients in the test group and the Results of this examination were used to compare with those of the control group and observe four indexes: patellofemoral effusion,synovial thickness,cartilage thickness and blood flow in the lesion area.Results There was only a small amount of synovial fluid in the articular cavity in the control group,the thickness was (2.11± 0.94) mm,while the thickness of the test group was (10.33±2.74) mm,the difference was statistically significant (t=17.03,P<0.05);in the control group,there was no synovial hyperplasia,the thickness was (1.11±0.24) mm,while the thickness of the test group was (5.48±2.74) mm which was significantly higher than that of the control group (t=13.18,P<0.05);in the control group,the cartilage tissue was striated without echo band,the thickness was (1.76±0.29) mm,while the cartilage tissue of the experimental group had lesion,the thickness was (1.12±0.12) mm lower than that of the control group and the difference was statistically significant (t=6.44,P<0.05);in the control group,there was no blood flow signal in the joint cavity,while low-speed blood flow signal could be seen in the joint cavity of the test group,the difference was statistically significant (0 vs.96%,χ2=6.44,P<0.05).Conclusion High frequency ultrasonography is effective in the treatment of senile rheumatoid arthritis with high specificity,high sensitivity,low cost,high efficiency and simple operation and it is not subject to external conditions.Therefore,it is suitable for early examination of senile rheumatoid arthritis and clinical promotion.

Thermal Shift Assays was applied for high-throughput screening optimized buffer system used in the gel filtration chromatography for purification of recombinant TBEV serine protease. The optimization included kinds of salt and pH. It was optimized new buffer remarkably improved the quality of the purified TBEV serine protease by increasing its monomer from 75% to 99%. This result may improve future crystal studies of this protein.

Objective To study the effects of TGFβ-activated kinase-1 (TAK1)mediated cell autophagy after global cerebral ischemia-reperfusion (IR ) in rats.Methods Seventy-two male Kunming rats were randomly divided into six groups:control group (group C),sham operation group (group S),ischemia-reperfusion group (group IR),TAK1 shRNA lentivirus group (group T),nega-tive lentivirus group (group Y)and normal saline group (group NS)(n = 12 each).The rats in groups T,Y and NS received cerebral ventricles injection of TAK1 shRNA lentivirus,negative lenti-virus and normal saline 10 μl two weeks before preparing animal model.Using thread embolism of the right middle cerebral artery occlusion (MCAO)to cause focal ischemia for 2 h and released for 24 h for reperfusion in groups IR,T,Y and NS.The common carotid arteries were separated but not liga-ted in group S,the rest of the procedure as the same as group IR.The rats of each group were evalua-ted by neurological severity scores (NSS)24 h after reperfusion,the cerebral infarct volume was measured with the method of TTC and the expression of TAK1,LC3Ⅱ/LC3Ⅰ,Beclin1 and p62 pro-tein in rat hippocampus were determined by using Western blot.Results The infarct volume and NSS in groups IR,T,Y and NS were significantly higher than those in group C (P <0.05).The infarct volume and NSS in group T were significantly lower than those in group IR (P <0.05).TAK1 pro-tein of hippocampus in groups IR,Y and NS was significantly higher than that in group C (P <0.05).TAK1 protein of hippocampus in group T were significantly lower than that in group IR (P <0.05).LC3Ⅱ/LC3 Ⅰand Beclin1 protein of hippocampus in groups IR,T,Y and NS were signifi-cantly higher than those in group C,and the p62 protein of hippocampus in groups IR,T,Y and NS was significantly lower than that in group C (P <0.05).The LC3 Ⅱ/LC3 Ⅰand Beclin1 in group T were significantly lower than those in group IR,and the p62 protein of hippocampus in group T was significantly higher than that in group C (P < 0.05 ).Conclusion TAK1 mediated cell autophagy takes part in the mechanism of brain ischemia-reperfusion injury in rats.

Abstract: Objective To explore the relationship between peripheral blood and pleural effusion tuberculosis (TB) infection effector T cells, and to further evaluate the value of combined pleural effusion adenosine deaminase (ADA) for rapid diagnosis of tuberculous pleurisy. Methods The test data of 80 cases of tuberculous pleurisy and 70 cases of nontuberculous pleurisy treated in the Sixth People's Hospital of Nantong City from January 2017 to December 2020 were analyzed. The TBinfected effector T cells were also detected simultaneously in the peripheral blood and the pleural effusion by the T-SPOT technique, and the pleural effusion ADA was detected by the rate method. The subject operating characteristic curve (ROC) was applied to take the optimal pleural effusion ADA threshold to compare the sensitivity and specificity of different critical values. Person phase analysis was applied to analyze the correlation between peripheral blood and pleural effusion T-SPOT.TB. Data of peripheral blood, pleural effusion T-SPOT.TB and ADA were integrated. Results When pleural effusion ADA>45 U/L, the sensitivity and specificity for the diagnosis of tuberculous pleurisy were 50.0% and 94.3%, respectively; when ADA > 25.15 U/ L, the sensitivity and specificity were 80.0% and 72.9%. When ADA > 45 U / L, pleural/ blood T-SPOT.TB spot ratio (spot forming cells, SFCs) > 2 times, the specificity for the diagnosis of tuberculous pleurisy was 100% (highest); when 25.15 U/L< pleural effusion ADA ≤ 45 U/L, pleural/blood T-SPOT.TB spot ratio > 2 times, the specificity for the diagnosis of tuberculous pleurisy was 92.3% (second). When pleural effusion ADA ≤ 25.15 U/L, and the pleural effusion/blood T-SPOT.TB spot number ratio > 2 times, with 83.3% specificity (the lowest of the three groups). Conclusions The level of pleural effusion ADA is one of the most used methods for diagnosing tuberculous pleurisy. Further combination of pleural effusion and blood T-SPOT.TB, if the ratio of pleural effusion / blood T-SPOT. TB spots is greater than 2 times, it can further improve the diagnosis rate of tuberculous pleurisy.

Objective To investigate the expression and signification of B-cell lymphoma/leukemia-2(bcl-2),bax and ER proteins in epithelial cells in different depth of adenomyosis.Methods Expression and correlation of bcl-2,bax and ER proteins were detected by immunohistochemistry staining in 36 adenomyosis cases with superficial (＜ 1/2 depth of penetration) and deep (≥ 1/2 depth of penetration)ectopic endometrium and eutopic endometrial tissues matched with 30 cases with benign ovarian tumor,uterine septum,pelvic floor dysfunction without adenomyosis as controls.Results (1) The expression of ER protein in superficial and deep ectopic tissues (5.04 ±0.24,4.91 ±0.16) were found significantly lower than 6.06 ± 0.36 in eutopic tissues (P ＜ 0.01) and higher than 3.70 ± 0.58 in control group (P ＜ 0.05).There was no statistical difference of ER expression in superficial and deep ectopic endometrium (P ＞ 0.05).(2) The level of bcl-2 protein of 5.6 ± 0.4 in superficial and 6.0 ± 0.3 in deep myometrium of ectopic tissues were significantly higher than 3.6 ± 0.4 in eutopic tissues and 1.9 ± 0.4 in control group (P ＜ 0.01).(3) The level of bax protein of 3.50 ± 0.28 in superficial and 4.80 ± 0.29 in deep myometrial ectopic and 4.43 ± 0.37 in eutopic tissues were significantly lower than 6.18 ± 0.65 in control groups (P ＜0.05).The expression of bax in superficial myometrium is significantly lower than deep myometrium in ectopic tissues (P ＜0.01).(4) The positive correlation between the expression of ER and bcl-2 protein at superficial myometrium of ectopic tissues (r =0.720,P ＜ 0.01).And there was no significant correlation with the expression of ER and bax protein at superficial myometrium (r =0.008,P ＞ 0.05).As well as,there was not significant correlation with the expression of bcl-2,bax and ER protein at deep myometrium (r =0.089,r =-0.023,P ＞ 0.05).The expression of bax protein in ectopic endometrium was positive correlation with the depth of adenomyosis penetration (r =0.736,P ＜ 0.01).There was positive correlation between the expression of ER and bcl-2 protein at normal endometrium (r =0.453,P ＜ 0.05).And there was negative correlation between the expression of ER and bax protein at control group (r =-0.514,P =0.05).Conclusion The bax protein expression of ectopic endometrium in deep adenomyosis was higher than superficial adenomyosis.
