Objective To evaluate the application of domestic probe fluorescence in situ hybridization (FISH) in prenatal diagnosis on uncultured amniocytes aneuploid. Methods One thousand three hundred and sixty-nine uncultured amniocytes (16-24 gestational weeks) from 37 hospitals in China were selected for prenatal diagnosis. 5 chromosomes (21, 13, 18, X and Y) were detected with muhicolor FISH. In the mean time, cytogenetic karyotype analysis was performed as control. Results Of all the samples, 1361 samples were successfully tested by FISH, the rate of successful detection was 99.42% (1361/1369). Thirty-five samples were shown with abnormal karyotypes by domestic FISH probe, the abnormal rate is 2. 57% (35/1361 ), including trisomy 21 (22 samples), trisomy 13 (4 samples), trisomy 18(6 samples), X0 (1 sample) and XXY (2 samples). Results of both FISH and cytogenetic karyotype analysis exhibited extreme concordance. Conclusion Domestic FISH probe used in prenatal diagnosis on uncultured aminiocytes showed the following advantages, such as highly efficient, low cost, small amounts of samples needed and reliable results.

Objective To investigate the global and regional dyssynchrony of left ventricle in patients with coronary artery disease before and after revascularization by tissue synchronization imaging(TSI).Methods Subjects with the left anterior descending coronary artery(LAD)stenosis>75% who underwent revascularization(n=26)and normal controls(n=30)were studied with TSI.Echocardiography was performed one day before revascularization, then repeated one month and three month after the operation, respectively. The sample volumes were located at the mid-myocardium of base and middle segments in apical four-,two- and three-chamber view of left ventricle. The time-to-peak velocity(Tp)of all myocardial segments were examined to access the regional dyssynchrony, and the standard deviations of Tp of 12 segments(TSD)were calculated to evaluate the global dyssynchrony. Results Before revascularization,Tp of segments in the anterior wall and the interventricutar septum in patients were more delayed than those of control group(P<0.01),and the color coding of ischemic segments were red and yellow. After operation,Tp of delayed segments were significant improved, and the color coding turn to yellow or even green, but there was still significant difference of Tp between disease group and control group(P<0.01 or P<0.05).TSD decreased gradually on the preoperatative, early postoperative, and later postoperative echocardiogram, and it on each time point of the disease group were all longer than that of control group. An improvement of TSD was observed after revascularization, especially for the three month examination(P<0.01),but the difference between early and later postoperation was not significant(P>0.05).Conclusions The asynchronous motion of LV is very obvious in CAD patients. After revascularization, both global and regional dyssynchrony were improved gradually. Moreover,TSI is a convenient and non-invasive way to quantitate left ventricular asynchrony with the parameters.

Objective:The significant role of long non-coding RNAs (lncRNAs) in early diagnosis and predicting prognosis has been recognized in various cancers recently. However, the prognostic value of HOXA transcript at the distal tip (HOTTIP),a vital lncRNA in tumorigenesis, remains unclear. In this study,we evaluated its prognostic value by analyzing the correlation of HOTTIP expression with overall survival (OS),lymph node metastasis (LNM) and distant metastasis (DM) in different cancer types by meta-analysis.Methods:We performed a systematic search in PUBMED,MEDLINE,Web of Science and Cochrane Library update to November of 2016.A total of 604 patients from 6 studies were included in final analysis and went through a quantitative meta-analysis by Review manager 5.3.Results:We demonstrated that high expression of HOTTIP had a significant correlation with poor OS (hazard ratio [HR] =2.37,95% confidence interval [CI] =1.81-3.10,p<0.001),high LNM rate (odds ratio [OR]=2.29,95%CI=1.54-3.40,p<0.001) as well as more DM occurrence (OR=3.30,95%CI=1.78-6.12,p<0.001).Conclusion:Our results indicated that long non-coding RNA HOTTIP may serve as a potential prognostic biomarker in cancer progression.

Objective To explore the clinical value of wave intensity(WI) curve of the carotid artery in patients with thyroid diseases and its value in evaluating cardiovascular function. Methods All 85 patients with thyroid diseases, including 45 hyperthyroidism and 40 hypothyroidism, were enrolled as case groups, while 270 healthy volunteers were considered as control. All the case groups and control were taken WI test under quiescent condition, and six continuous curves of the carotid artery diameter changes were recorded. Then the parameters of accelerating wave intensity (W1), decelerating wave intensity ( W2), negative area (NA) ,corrected R-W1 and W1-W2,elasticity modulus(Eρ) ,stiffness index(β) ,arterial compliance(AC) and pluse wave velocity(PWV) were calculated. Relationship between those indices and FT3 ,FT4 and TSH were analyzed by Pearson correlative analysis. Results The WI curve appeared differently among different groups, but the vessel elasticity indices did not have statistical significance. Correlation only existed between FT3 and part of the WI parameters. FT3 positively correlated with W1 ( r = 0. 951, P <0.01 ), NA( r =0.813, P <0.01) and W1-W2' ( r = 0. 887, P <0.01 ). And R-W1 ' negatively correlated with FT3( r =-0.878, P <0.01 ). Conclusions The WI curve could represent different function status of thyroid, and could be considered as a noninvasive examination for further clinical research, which may help explain the corresponding cardiovascular changes.

Objective To investigate the effect of meridian-unblocking and mind-regulating acupuncture pretreatment on neurobehavioral functions and cerebral water content and explore its mechanism for modulating miRNA664 and MMP9 expressions in cerebral ischemia/reperfusion rats. Methods One hundred and thirty Wistar rats were randomized into group A (electroacupuncture pretreatment) of 30 rats, group B (moxibustion pretreatment) of 30 rats, group C (aspirin pretreatment) of 30 rats, group D (model) of 30 rats and group E (blank control) of 10 rats. Group A received electroacupuncture; group B, suspended moxibustion with moxa sticks; group C, an oral gavage of aspirin 10 mg/kg. A model of cerebral ischemia/reperfusion was made in every group after seven days. Rat neurobehavioral functions were observed and cerebral water content and relative miRNA664 and MMP9 expressions in the cortical region were determined at 24 hours after reperfusion. Results There was a statistically significant difference in the neurobehavioral score between group A, B, C or D and group E (P<0.01), between group A or B and group C (P<0.01) and between groups A and B (P<0.01). There were statistically significant differences in cerebral water content and relative miRNA664 and MMP9 expressions between group A, B, C or D and group E (P<0.01,P<0.05), between group A, B or C and group D (P<0.01), between group A or B and group C (P<0.01,P<0.05) and between groups A and B (P<0.01,P<0.05).Conclusions Meridian-unblocking and mind-regulating acupuncture pretreatment can effectively decrease the neurobehavioral score and cerebral water content and reduce relative MMP9 expression through modulating miRNA664 expression in cerebral ischemia/reperfusion rats. One of the mechanisms of Du meridian-unblocking and mind-regulating acupuncture pretreatment for protecting the brain may be reducing relative MMP9 expression, inducing tolerance to cerebral ischemia and relieving cerebral edema by the modulation of miRNA664 expression.
