China Medical Equipment 2016;13(6):53-55,56
Objective:To analyze the differences and common differential diagnostic value on the fourth ventricle magic resonance imaging (MRI) findings of primary tumors, diffusion-weighted imaging (DWI) and minimum apparent diffusion coefficient (ADC).Methods:A total 100 cases with the fourth ventricle of the primary tumor who were given MRI examination, were selected from March 2011 to April 2012 in our hospital, recording an average minimum ADC value of tumor parenchyma. Results: 43 cases of medulloblastoma showed cerebellar hemisphere pressed shift or tonsillar herniation and restricted diffusion on DWI tumors showed high signal tumor. The minimum ADC value was (0.43±0.15)×10-3mm2/s. 33 cases of astrocytomas showed significant mass effect and the minimum ADC value was (1.58±0.58)×10-3mm2/s. 24 cases of ependymoma MRI showed typical “melting wa×” degeneration and tumor minimum ADC value was(0.93±0.18)×10-3mm2/s. The minimum ADC value of the three different forms of tumor cell lines was calculated by variance test and twenty-two row-LSD-t test, and the results were statistically significant difference (F=21.985,P=0.001). DWI high signal is limited to an average minimum ADC value of 0.875×10-3mm2/s as a threshold value. Compared with the pathology, the specificity of the fourth ventricle medulloblastoma was 98.5%, the sensitivity was 97.3%, the negative predictive value was 98.5%, and a positive predictive value was 100%. The suppression test was carried out between all the results above and the results of histological findings and Kappa value was 0.912.Conclusion: The primary tumor of the fourth ventricle was diagnosed with the ADC, DWI and conventional MRI, which showed that the specificity, sensitivity, negative predictive value and positive predictive value were high. It had obvious diagnostic effect and great value.

Chinese Journal of Behavioral Medicine and Brain Science 2016;(2):128-133
Objective To observe the protective effect of grape seed proanthocyanidin extract ( GSPE) on learning disabilities after radiation injury brain in rats. Methods 120 male Wistar rats were divided into control group, model group, high and low dose GSPE groups. The radiation injury brain models were established using a method of linear accelerator irradiation in 22 Gy.The morphological changes of neu-rons in hippocampus were observed with HE staining; the expression of phosphorylated extracellular signal regulated kinase 1/2 ( ERK1/2) and growth associated protein-43( GAP-43) were detected by immunohisto-chemistry;the contents of malonalde hyde(MDA) and superoxide dismutase(SOD) were measured respec-tively by thiobarbituric acid( TBA) and xanthine oxidation( XTO) methods;the learning ability was assessed with shuttle box. Results Compared with the model group, the GSPE groups showed a decreased degree of nerve cell morphological injury; and increased ERK1/2 activities((13.20±1.45)/view,(27.40±2.52)/view,(19.80±1.30)/view),GAP-43 expression level((52.93±2.07)/view,(66.50±0.77)/view,(73.05± 2.40)/view) and SOD activities((79.82±5.26) U/mg,(76.20±6.86) U/mg,(80.12±5.76) U/mg)(P<0.05);and decreased the contents of MDA((71.62±1.88) μmol/g,(76.41±1.94) μmol/g,(72.32±1.98)μmol/g)(P<0.05).Shuttle box testing showed that the active avoidance reaction rate was increased(7 d:(56.23±2.56)%;14 d:(44.66±2.79)%;28 d:(50.40±2.16)%)(P<0.05)and passive avoidance latency was shorted(7 d:(34.11±1.57)s;14 d:(45.52±1.88)s;28 d:( 39.52±1.79)s)(P<0.05) in GSPE groups. The above mentioned changes were more significant in high dose of GSPE(P<0.05) .Conclusion GSPE has protective effect on learning disabilities of radiation injuries brain, which is related to enhance ERK1/2 activ-ity and the expression of GAP-43.
8.Effect of Grape Seed Proanthocyanidin on Growth Associated Protein-43 through Extracellular Signal Regulated Kinase 1/2 Pathway in Rats with Brain Radiation Injury
Chinese Journal of Rehabilitation Theory and Practice 2015;(12):1397-1401
Objective To observe the effect of grape seed proanthocyanidin extract (GSPE) on growth associated protein-43 (GAP-43) through extracellular signal regulated kinase 1/2 (ERK1/2) pathway in rats with brain radiation injury. Methods 72 3-month male Sprague-Dawley rats were divided into control group (n=18), model group (n=18), high dose GSPE group (n=18) and low dose GSPE group (n=18). The brain radiation injury models were established by linear accelerator irradiation with 22 Gy. The learning ability was assessed with shuttle box. The morphological changes of neurons in hippocampus were observed with HE staining;the expression of GAP-43 was de-tected by RT-PCR; and the expression of phosphorylated ERK1/2 was detected by Western blotting. Results Compared with the model group, the active avoidance reaction rate in the shuttle box test increased and the passive avoidance latency shortened in GSPE groups (P<0.001); the nerve cell morphological injury reduced and the expression of GAP-43 mRNA and phosphorylated ERK1/2 increased in the GSPE groups (P<0.001), especially in the high dose GSPE group (P<0.001). The GAP-43 mRNA level positively correlated with phosphory-lated ERK1/2 level in the model group (r=0.764, P<0.001), the low dose GSPE group (r=0.814, P<0.001) and the high dose GSPE group (r=0.822, P<0.001). Conclusion GSPE could promote the expression of GAP-43 through ERK1/2 pathway in rats, and prevent the brain from radiation injury.
9.Current research of retinal ischemia model in rodent
Chinese Journal of Experimental Ophthalmology 2013;(5):513-516
Retinal ischemia is a common cause of blindness and visual impairment. Some eye diseases, such as glaucoma, retinal artery occlusion and diabetic retinopathy, are associated with retinal ischemia. It is very important to build an ideal animal model which can imitate human retinal ischemia disease for investigating the pathological mechanism and treatment of retinal ischemia.Rodent is a popular choice for retinal ischemia model due to its availability and the similarity to human retinal blood supply. To provide a supportive evidence of retinal ischemia research, some literature concerning rodent models is reviewed and various methods to build the model are summarized, such as elevated intraocular pressure, optic nerve ligation, ligation of vessels,photo-chemistry,endothelin-1 injection and middle cerebral artery occlusion intraluminal suture, etc. Here, the specific protocols, the pathological changes and molecular mechanism of each model are reviewed.
