6.In silico data mining of the human programmed cell death 5 (PDCD5) sequences
Journal of Peking University(Health Sciences) 2003;0(04):-
Objective:To lay foundation for the functional studies of programmed cell death 5 ( PDCD5 ) and develop new technical pathway for bioinformatics analysis of human functional genes. Methods:Using PDCD5 as the target molecule, intensive bioinformatics analysis of the nucleic acid and protein sequences were conducted. Data mining and comprehensive analysis by sequence against database similarity searching, ortholog structure comparison, expression profile analysis and gene “neighbor” listing were performed. Results: Two human putative pseudogenes on chromosomes 12 and 5, and one mouse putative pseudogene on chromosome 1 were identified. The methanobacterium thermoautotrophicum ortholog was classified as the same fold as ubiquitin and ribosomal protein S13. The C. elegans ortholog, ubiquitin and IAP (inhibitor of apoptosis proteins) belonged to the same expression profile cluster. This cluster was related to biosynthesis and protein synthesis. PDCD5 orthologs in various genomes were adjacent to various ribosomal proteins on the chromosome. Conclusion: The human genome contains at least two processed pseudogenes of PDCD5 . Besides the relationship with cell apoptosis, PDCD5 is predicted to have functional relationship with ubiquitin and participate in the translation regulation.
7.Epidemiologic characteristics of natural foci of plague in Qinghai province from 1999 to 2009
Chinese Journal of Endemiology 2010;29(6):663-665
Objective To analyze the epidemic characteristics of natural foci of plague in Qinghai province in the past twenty years and to provide references for prevention and control of the disease. Methods Excel software was used to analyze the database of human plague cases and the surveillance and survey data of natural foci of plague in Qinghai province from 1999 to 2009. Results Animal plague occurred in Qaidam Basin, Qinghai Plateau and central and southern Plateau year after year, which became key areas of animal plague in Qinghai province. There were many human plague cases every year except 1999, 2000, 2002, 2007, 2008 from 1990 to 2009. Cumulative incidence of human plague was 27 cases, with incidence of 73 cases, 29 cases of death, and death rate 39.73%. Epidemic areas mainly distributed in the 4 states of Yushu, Hainan, Hercynian, Haibei.Prevalent season was from May to October. The most prevalent was pneumonic type among clinical types [60.27%(44/73)]. Conclusions Animal plague has occurred actively in key areas of Qinghai province in recent years, and an outbreak of human plague in some areas is found. Monitoring and early warning should be strengthened in the key areas, and comprehensive prevention and control measures should be implemented to reduce the incidence and prevalence of the plague.
8.An experimental research on the analgesic effect and underlying mechanism of BTX-A for rat knee osteoarthritis
Chinese Journal of Rehabilitation Medicine 2017;32(6):649-653
Objective:To evaluate the analgesic effect of botulinum toxin type A (BTX-A) in osteoarthritis model and the changes of Nav1.8 protein expression in spinal ganglia changes.Method:Animal model of knee osteoarthritis (KOA) was established by intra-articular injection of 4% papain solution 0.3ml into SD rat right knee. After the formation of arthritis, they were randomly divided into two groups at the 2nd day:BTX-A group (n=10):intra-articular injection of 5μl BTX-A 0.1IU;WFI group (n=10):intra-articular injection of 5μl water. No papain or BXT-A was given to the sham group (n=10).At the 1st,3rd,5th day after injection,we tested the pain behavior, thermal pain threshold, and sodium channel 1.8 (Nav1.8) protein expression in spinal ganglion by using immunohistochemistry.Result:Analysis of spontaneous pain behavior showed abnormal gait caused by rat osteoarthritis.Comparing with the WFI group, abnormal gait caused by osteoarthritis improved significantly in BTX-A group. At day 5 it improved more significantly than day 1 and day 3.Thermal pain threshold of BTX-A group increased more than that of WFI group (P<0.05) at any time point.Abnormal high Nav1.8 protein in model rats decreased in spinal ganglia for BTX-A group. Conclusion:The intra-articular injection of BTX-A may play the analgesic effect in the model of KOA by down-regulating of the expression of Nav1.8 protein in spinal ganglia and reduction in the central sensitization to pain stimulation.
9.The correlation between vaginal intraepithelial neoplasia and cervical intraepithelial neoplasia, and their risk factors
Journal of Chinese Physician 2015;17(1):41-44
Objective To investigate the correlation and risk factors between vaginal intraepithelial neoplasia (VAIN) and cervical intraepithelial neoplasia (CIN).Methods A total of 121 cases of VAIN patients was retrospectively analyzed with examination results such as clinical manifestations, liquid based cytology (TCT),human papilloma virus infection, virus infection type, and clinical data of treatment and follow up.Results VAIN Ⅰ,VAIN],and VAIN Ⅲ] were accounted for 33.9% (41/121),38.0% (46/121),and 24.4% (34/121) ; for 40 or less years old patients,VAIN Ⅲ was accounted for 10.7% (3/28) ; for more than 40 years old patients,VAIN Ⅲ was accounted for 33.3% (31 /93).Among 121 VAIN patients,77.7% (94/121) patients had CIN history,15.7% of which (19/121) had a history of cervical cancer; 27.3% (33/121) with hysterectomy history,of which 75.8% (25/33) was due to cervical intraepithelial neoplasia and cervical cancer resection of the uterus.Papilloma viral load in patients with VAIN Ⅰ was detected with a (HPV-HC2) positive rate of 87.8% (36/41),while in VAIN Ⅱ and VAIN [patients, the positive rate was 100%,the infection of high risk type of HPV ratio was 95.9% (116/ 121).of which the mixed infection was accounted for 56.9% (66/116).There was 94.6% in oatients with VAIN (114/121),and liquid-based cytology test results suggest abnormality; 88.4% (107/121) in patients with VAIN lesions occurred in upper 1/3 of vaginal. After diagnosed cervical disease, the onset time of VAIN was (3.45 ± 2.02) years,non-total-hysterectomy was (3.24 ± 1.97) years, after hysterectomy to pathological diagnosis time for VAIN was (5.01 ± 3.95) years:for the diagnosis of VAIN disease after resection of cervical original time was (3.96 ± 2.99) years, and rather than non cervical disease resection was an average of (9.94 ±5.37) years. In the course of treatment of the 12l cases of CIN patients,6 cases of patients were progressed to invasive carcinoma. Conclusions The clinical manifestation of VAIN is similar to CIN,the principle of diagnosis and treatment of CIN can also apply to VAIN,high risk HPV infection virus, cervical intraepithelial neoplasia are the risk factors for VAIN.
