1.Separation and identification of chemical constituents of Mongolian medi-cine compound Shudage-4

Objective To study the chemical constituents of Mongolian medicine compound Shudage-4, so as to find out material bases of its pharmacodynamic effects.Methods The chemical constitutes of Shudage-4 were extracted using ethanol extraction method,and were isolated and purified using technolo-gies of silica gel,Sephadex LH-20 and reversed phase silica gel.Then the structures of the isolated com-pounds were identified by using different spectroscopic and chromatographic techniques including ultra vi-olet spectroscopy(UV),infrared spectroscopy(IR),mass spectrometry(MS)and nuclear magnetic reso-nance(NMR).Results There were 10 compounds isolated and identified from Mongolian medicine Shudage-4—1-phenyl-7-(3-methoxy-4-hydroxy) phenyl-5-ol-3-heptanone-one (1 ),stigmasterol (2 ), kaempferol(3),daucosterol(4),p-Anisic acid(5),ursolic acid(6),oleanic acid (7),dehydrocostus lactone(8),costunolide(9)and dehydrodiisoeugenol (10).Conclusion The ten compounds,except stigmasterol,were first isolated from Shudage-4.This paper provided important scientific data for the ma-terial bases of Mongolian medicine Shudage-4.
2.Chemical compositions of Mongolian medicine compound formula-Shudage-4 in rat serum

Objective To establish in vivo and in vitro the characteristic chromatograms of Mongolian medicine compound formula-Shudage-4,and affiliate and identify in vitro chemical compositions of Shudage-4 and its compositions in vivo absorbed into blood.Methods Wistar rats were intragastrically given Shudage-4 and its different single medicinal,and then serum samples were prepared and determined by using high performance liquid chromatography (HPLC).The HPLC characteristic chromatograms of in vitro samples of Shudage-4 and its different single medicinal and Shudage-4 medicated serum were established and analyzed.The chemical compositions in vitro of Shudage-4 and its compositions in vivo absorbed into blood after taken by rats were compared,affiliated and identified.Results There were totally 64 peaks found after analyzing in vitro HPLC characteristic chromatograms of Shudage-4.There were 23 chemical compositions absorbed into blood,and 17 in vitro ones absorbed directly into blood and 6 speculated as new metabolites.These 23 chemical compositions were all derived from different were derived from Gaoliangjiang (Rhizoma Alpiniae Officinarum,Galangal Rhizome),peak 14,from Muxiang (Radix Aucklandiae,Common Aucklandia Root) and peak 13,from Shichangpu (Rhizoma Acori Tatarinowii,Grassleaf Sweetflag Rhizome).The peaks 15,20,21 and 22 were derived from Gaoliangjiang,Muxiang and Shichangpu,peaks 6,10,17,18 and 19,from Gaoliangjiang and Shichangpu,peaks 7 and 16,from Gaoliangjiang and Muxiang,peak 11,from Muxiang and Shichangpu,and peaks 4,5,8 and 12,from the whole formula of Shudage-4,from which 7 chemical compositions were identified.Conclusion The chemical compositions and metabolites in blood may be the active principles of Shudage-4 with direct effects in vitro,which are derived respectively from different single medicinal in Shudage-4.
3.Effect and mechanism of effective components of Mongolian medicine compound Shudage-4 against experimental stomach ulcer

Objective To explore the protective effect and mechanism of Mongolian medicine compound Shudage-4 on gastric ulcer induced by glacial acetic acid.Methods Healthy SD rats were randomly divided into Shudage-4 high,middle and low dose (high-,mid-,and low-dose)group,positive drug groups(Anweiyang group and cimetidine group),model group and normal group.Except for the normal group,the rats in the other six groups were modeled by glacial acetic acid.After the model was estab-lished successfully,rats of each group were admistered i.g corresponding medicine for consecutive 14 days.Then the gastric tissue was resected 24 h after the last administration.Ulcer index and ulcer inhabitation rate were measured,the content of NO and MDA in gastric mucosa were detected using ELIAS kits,and pathological changes of gastric tissue was stained by HE were observed under light microscope.Results Compared with the model group,the content of NO in gastric mucosa increased significantly and MDA decreased significantly in all three Shudage-4 groups (P <0.01).The inflamma-tion decreased in all three Shudage-4 groups,in which mid-group and high-group showed more obvious effect with much less the number of inflammatory cells and improvement of the pathological condition. Conclusion Mongolian medicine compound Shudage-4 has the therapeutic effect on gastric mucosal in-jury induced by glacial acetic acid in rats.The mechanism may be related to its effect of antioxidation and vasodilation.
