Acta Universitatis Medicinalis Anhui 2015;(6):723-726
Objective To observe the effect of puerarin preconditioning on hippocampal CA1 region neuronal injury and further explore its mechanisms. Methods 30 male Sprague-Dawley rats were randomly assigned into sham, model, and puerarin preconditioning ischemia-reperfusion groups. The middle cerebral artery was occluded for 60 min by an intraluminal filament before reperfusion to induce transient focal cerebral ischemia reperfusion rats. Puer-arin preconditioning groups rats received puerarin(100 mg / kg,i. p)1 h before ischemia. The histological changes of hippocampal CA1 region neurons were measured by HE staining on the 5th day after reperfusion. The expressions of Caspase-3 and Caspase-9 proteins in the CA1 region were examined by immunohistochemistry staining. Results The results of HE staining showed that, compared with the model group, puerarin preconditioning significantly re-duced the number of cerebral ischemia induced injury of hippocampal CA1 region neurons(P < 0. 05). Immunohis-tochemistry data showed that the expressions of Caspase-3 and Caspase-9 in the hippocampal CA1 region of puerarin preconditioning group were significantly lower than that of model group(P < 0. 05). Conclusion The results indi-cate that puerarin preconditioning 1 h before ischemia exerts neuroprotective effects on focal cerebral ischemia-reperfusion induced neuronal injury. The neuroprotective mechanisms of puerarin may attribute to inhibiting the ex-pressions of Caspase-3 and Caspase-9 protein.

Objective To explore the role of extracellular signal-regulated kinase (ERK)in central nucleus of amygdala (CeA)in the mechanism of fentanyl-induced hyperalgesia (OIH)in rats. Methods Step 1:12 healthy male Sprague-Dawley rats, weighing 60-100 g,were randomly divided into OIH and Control group. The mechanical paw withdrawal threshold (PWT)and the thermal paw withdrawal latency (PWL)were tested at pre-and post-OIH induction. Then the level of p-ERK in the CeA was analyzed by Western blotting.Step 2:After successful induction of OIH and catheterization in CeA,another 30 SD male rats were randomly divided into 5 groups (n = 6 each):Group OIH;Group OIH+U0124;Group OIH+U0126(0.1 5 nmol);Group OIH+U0126(0.45 nmol)and Group OIH+U0126 (1.5 nmol),then 0.3 μl of DMSO,U0124 (1.5 nmol),U0126 (0.1 5 nmol,0.45 nmol,1.5 nmol)was given through the catheter separately.PWT and PWL were tested before cathe-terization,at pre-OIH induction,post-OIH induction and 0.5 h after CeA drug administration. After the last test of pain threshold, the rats were sacrificed and CeA tissues were sampled for analyzing the expression of p-ERK by western blot. Results In step 1 compared with control group,PWT and PWL of OIH group were sharply decreased post-OIH induction (P <0.05),concomitant increase of the expression of p-ERK in CeA in OIH group was also observed. In step 2,both PWT and PWL were sharply decreased post-OIH induction (P <0.05).Intra-CeA U0126 injection, but not U0124, reversed both behavioral hyperalgesia and molecular activation of ERK in CeA in a dose-dependent manner (P <0.05).Conclusion ERK plays a pivotal role in the maintenance of fentanyl-induced hy-peralgesia.Targeting inhibition of ERK activation in CeA can alleviate fentanyl-induced hyperalgesia.

As an important material in pharmaceutical and chemical industry, β-alanine was mainly produced by chemical methods. L-aspartate-α-decarboxylase could catalyze the α-decarboxylation from L-aspartate to β-alanine. Determinations for specific activities of PanDs from Escherichia coli, Corynebacterium glutamicum and Bacillus subtilis were performed in this study (0.98 U/mg, 7.52 U/mg and 8.4 U/mg respectively). The optimal temperature and pH of PanDs from C. glutamicum and B. subtilis were 65 degrees C, pH 6.5 and 60 degrees C, pH 6.5 respectively. According to our research, PanD from B. subtilis could be more appropriate for industrial application because of the higher activity and thermostability when compared to PanDs from E. coli and C. glutamicum which had been the most studied. We also analyzed and discussed the special post-translation self-cleavage phenomenon and the mechanism based inactivation.

Objective To investigate the clinical efficacy of colonoscopic metal stenting and ileus tube catheteriza-tion on left colorectal cancer with acute obstruction. Methods Clinical data of 80 patients meeting the diagnostic criteria for acute obstruction of the left half of colorectal cancer were included in this study. Patients were randomly divided into met-al stent group (n=40) and ileus tube group (n=40). The metal stent group was treated by metal stent and the ileus tube group was treated by ileus tube. Both were carried on by colonoscopy and X-line. After the relief of obstruction ( 7-10 d), patients were underwent colorectal cancer radical resection and anastomosis. The technical operations, improvement of obstruction and the efficacy of surgical treatment were compared between two groups. Results The success rate was 87.5%in metal stent group and the 97.5%in ileus tube group. There was no significant difference in the success rate between two groups (P>0.05). No complications were found in two groups. The operation time and the treatment cost were higher in metal stent group than those in ileus tube group. The relief rates of obstruction were 100%and 95%for metal stent group and ileus tube group, and there was no difference between them. Two cases were operated for emergency because of the failure of obstruc-tion relief. The relief time of obstruction and the difference between C-reactive protein values were much better in metal stent group than those of ileus tube group. After the obstruction relief, patients underwent a radical resection of the tumor and anastomosis, no anastomotic leakage was found in two groups. There were no significant differences in the operation time, in-cision infection and hospitalization time between two groups. Conclusion Anal ileus tube catheterization has a better eco-nomic value in the treatment of acute obstruction, but the efficacy of metal stent is better.

Objective To investigate the expression patterns of major bone growth factors and bone morphogenetic protein-2 (BMP2) in the distracted calluses following transfected gene during mandibular distraction osteogenesis in a rabbit model. Methods Bilateral mandibular osteotomies were performed in New-Zeland rabbits. After a latency of 3 days, the mandibles were elongated using distractors for 7 days. After the completion of distraction, the rabbits were randomly divided into 5 groups.Three animals each were sacrificed at the end of the delay phase,at 7,14,and 28 days after completion of distraction, respectively. The lengthened mandibles were harvested and processed for immunohistochemical detection of BMP2 expression, the mean optic densities and integral optical density of BMP2 positive cells were measured by computerized image analyzer. Results Elevated cellular expression of BMP2,in the distraction gap,was observed following mandibular distraction.BMP2 staining was mainly located in inflammatory cells, and the connective tissues arrounding the new bone.Their strongest expression was the 7th day, some of those growth factors expressed weakly or negatively. Conclusions Electroporation-mediated gene transfection can promote BMP2 expression effectively, which plays an important role in cell differentiation and proliferation during distraction osteo genesis. The BMP2 stimulates extracellular matrix synthesis, induces the proliferation and differentiation of fibroblasts and osteoblasts, which then promotes the new bone formation and repair.
