2.Protective effects of exogenous leptin on liver injury induced by severe acute pancreatitis

Objective To investigate the effect and mechanism of exogenous leptin on liver injury in severe acute pancreatitis rat .Methods Thirty male SD rats were randomly divided into the sham operation group ,SAP group and leptin intervention group . The SAP rat models was established by retrograde injection of 3 .5% sodium taurocholate into the biliopancreatic duct .The leptin intervention group was intraperitoneally injected with leptin 20 μg/kg .The rats in each group were sacrificed at 12 h after model‐ing .The pancreas and liver tissues were taken for HE staining and detecting the nuclear factor kappa B (NF‐κB) .The cell apoptosis in situ labeling method was adopted for detecting the liver tissue cell apoptosis index .ALT ,AST and AMY were detected . Results Compared with the sham operation group ,the liver tissue pathology score in SAP group and leptin intervention group were significantly increased(P<0 .05) .The liver tissue pathology scores in the leptin intervention group were lower than those in the SAP group(P<0 .05) .The NF‐κB expression of liver tissue in the SAP group and leptin intervention group was obviously increased compared with the sham operation group ,the expression in the leptin intervention group was decreased compared with the SAP group (P<0 .05) .The liver cell apoptosis index in the leptin intervention group and SAP group was significantly higher than that in the sham operation group (P<0 .05) ,and which leptin intervention group was decreased compared with the SAP group (P<0 .05) . The results of ALT ,AST and AMY in the SAP group and leptin intervention group were increased significantly compared with the sham operation group(P<0 .05) ,while which in the leptin intervention group was decreased compared with the SAP group (P<0 .05) .Conclusion The exogenous leptin may play the protective effect on SAP complicating liver damage by lowering the liver tis‐sue NF‐κB expression and reducing the liver cell apoptosis index .
3.Optimizing Alcohol-extraction Process for Fushuning Granules by Orthogonal Design

Objective To optimize the method and condition s for the extraction process of Fushuning Granules.Methods Different factors and levels were in vestigated by orthogonal design wit h yields of oleanolic acid and tanshi noneⅡ A as the markers.The yields of effective com ponents by percolation and reflux we re compared.Results The extraction ratio by re-flux was higher than that by percolation.The primary factors influencin g extraction ratio by reflux were the pulverization degree of medicinal materials,concentration of alcohol,refluxing times and refluxing perio d.Conclusion The opti-mum extraction process was:refluxing and extracting the pulverized med icinal materials for 3times(1hours each time )with 6-fold 90%alcohol added in the f irst time and then 5-fold in the secon d and the third time.
4.Impact of HMME-PDT on Pg of the surface of implants and cementum tablets

Objective To evaluate the bactericidal effect of diode laser on Porphyromonas gingivalis (Pg),and to explore an optimized protocol for a safe dose of photodynamic therapy (PDT) to eliminate periodontal pathogens as well as the impact on the implant surfaces,so as to provide theoretical and experimental basis for PDT in periimplantitis therapy.Methods Artificial in vitro models were formatted by culturing Pg standard strain and ITI (International Team for Implantology) implants together in CDC broth.Then artificial in vitro models were treated by different doses of hematoporphyrin monomethyl ether (HMME) and different energy density of laser (EDL) for 60 s.The cultures were counted by colony form unit (CFU),and SPSS 17.0 statistical software was used for data statistical analysis to select the best EDL and HMME dose.Finally,ITI implants were observed by scanning electron microscope (SEM) to evaluate the impact of HMME-PDT on Pg of implant surfaces.Results When EDL was 12 J/cm2 and mass concentration of HMME was 25 μg/ml,SEM observations showed that PDT could effectively kill Pg ((13.00±5.00) CFU)without damaging the implant surfaces.Conclusions PDT therapy combining 630 nm diode laser with photosensitizer HMME have good bactericidal effect on Pg,and the EDL and HMME dose is as small as the clinical applicable safe dose.
5.ICP-MS method for the determination of uranium concentration and 235U/238U ratio in urine and its uncertainty evaluation

Objective To establish a method of analyzing total uranium and 235U/238U ratio in urine using inductively coupled plasma mass spectrometry (ICP-MS) and uncertainty assessment.Methods Urine sample was digested with HNO3 and H2O2,and total uranium was determined using ICP-MS method directly.The digested urine sample was separated to concentrate uranium with tributyl phosphate (TBP) column,and 235U/238U ratio was analyzed using ICP-MS.The uncertainty was evaluated through sample pre-treatment,measurement and standard curve calculation.Results The recovery of total uranium in urine was 98.4%-102.4%,detection limit was 0.002 μg/L.The relative expanded uncertainty of total uranium concentration in urine was 0.26 (k =2).235 U/238 U ratio was 0.001 1 (k =2).Conclusions This study offers a low detection limit,good recovery and precision method for rapid determination of total uranium and 235U/238 U ratio in urine samples.It is potential used for both in occupational exposure assessment and nuclear emergency situation.The uncertainty evaluation of total uranium and 235U/238U ratio in urine are reliable.
7.Length changes of knee lateral collateral ligaments following anterior cruciate ligament injury

BACKGROUND: Lateral collateral ligaments play an important role in maintaining knee stability.Motion reduction of knee joint can be realized and the changes laws of medial and lateral collateral ligaments' length after anterior cruciate ligament(ACL)injury during weight-bearing flexion can be obtained via 2D/3D image registration technique.OBJECTIVE: To study in vivo stability of length changes of the medial and lateral collateral ligaments of ACL injury knee during weight-bearing flexion.METHODS: Eight volunteers with unilateral ACL rupture and contralateral normal knees,was captured CT images and 2orthogonal images of the knee at 0,15°,30°,60°,and 90° of weight-bearing flexion.These orthogonal images were used to recreate the in vivo knee positions at each of the targeted flexion angles by the method of 2D/3D image registration.Through the bone insertion of medial and lateral collateral ligaments,the elongation changes of medial and lateral collateral ligaments were obtained.RESULTS AND CONCLUSION: At 0°,15° and 30°,the length of medial collateral ligament of ACL injury knees was longer than normal knees,but the lateral collateral ligaments length of ACL injury knee was shorter than that of normal knees.All the differences have statistical significances(P < 0.05).The findings demonstrated that,at 0°,15° and 30°,the medial collateral ligament length of ACL injury knees was longer than normal knees,but lateral collateral ligaments length of ACL injury knees was shorter than normal knees.
8.Application of computer-aided technique in pelvic precise model preparation and individualized prosthesis implantation

BACKGROUND:Bone tumors around the Ⅱ section of pelvis are difficult to treat due to complicated anatomic structures.Using computer-aided technique,the excision range and prosthesis preparation can be individualized,which may obtain notable therapeutic efficacy in treating pelvic fractures in the clinic.OBJECTIVE:To discuss the application and the clinic effect of computer-aided technique in bone tumors therapy around the Ⅱsection of pelvis.METHODS:The pelvis model was generated with its CT data by rapid prototyping.Simulated bone resection and reconstruction were performed on the models.Then we designed surgical extension and made hemi-pelvic.Eight cases received resection of pelvic tumor and reconstruction based on computer-aided technique.RESULTS AND CONCLUSION:The resection of tumor and implantation of prosthesis were easily accessible.Two cases relapsed and 1 case loosened at 2 years after operation.According to Harris scoring criteria after total hip replacement,the scores of cases were well.The simulated resection and reconstruction of bone tumors around the Ⅱ section of pelvis based on computer-aided technique makes the operation easy and reconstruction precise,which produces good clinic results and offers a good promise for the application.
9.Effect of high-flux hemodialysis on immune state in maintenance hemodialysis patients

ObjectiveTo study the changes of serum immune state in maintenance hemodialysis (MHD) patients with high-flux hemodialysis. MethodsSixty MHD patients were divided into high-flux hemodialysis group (group T, 30 cases, receiving high-flux hemodialysis, 3 times per week and routine drug therapy) and conventional dialysis group (group C, 30 cases, taking conventional dialysis, 3 times per week and routine drug therapy) by random number table. They were treated for 12 weeks. Serum IgG,IgA, IgM,C3, C4, T lymphocyte subsets and albumin, prealbumin, hemoglobin were detected at the experiment onset and 12 weeks later, and compared with those of 20 normal medical examination adults (group N ), meanwhile the ratio of infection was calculated. ResultsThe levels of IgG,IgA,IgM,C3,C4,CD3+,CD4+,CD4+/CD/8+ in group C had no significant changes before and after treatment (P > 0.05 ), but there were significant changes in group T[(12.20 ± 3.98) g/L vs.(6.18± 1.45) g/L, (1.89 ±0.58) g/L vs. (0.63 ±0.15) g/L, (1.29 ± 0.47)g/L vs. (0.51 ± 0.13) g/L, (0.94 ± 0.36) g/L vs.(0.58 ± 0.20) g/L, (0.28 ± 0.06) g/L vs. (0.11 ± 0.04) g/L,(63.11 ± 9.43 )% vs. (53.26 ± 9.08 )%, ( 38.21 ± 6.15 )% vs. ( 31.56± 6.02 )%, 1.48 ± 0.37 vs. 1.25 ± 0.43](P< 0.05), and there were significant differences between group T and group C after treatment (P< 0.05).The ratio of infection in group T was decreased obviously [46.7% (14/30) vs. 133.3% (40/30), P < 0.05], but there was no significant difference in group C [126.7%(38/30) vs. 136.7%(41/30),P> 0.05]. Conclusion High-flux hemodialysis can improve the immune state and the nuuitional state in MHD patients, and it could decrease the infection rate.
10.Determination of uranium in drinking water in the vicinity of nuclear power plants by ICP-MS

Objective To ascertain the concentrations of uranium in drinking water around nuclear power plants.Methods A total of 106 water samples were collected from June 2009 to March 2010 in Jiangsu,Zhejiang,Liaoning and Shandong provinces.Inductively coupled plasma-msgs spectrometry(ICPMS)was applied to determine uranium content in local water source and drinking water.The detection limit of U was 0.8 ng/L.The recovery was 100.9%.Results The uranium concentrations in all samples were less than 15μg/L which was the limit given by World Health Organization(WHO).Conclusions The concentration of uranium in water sources was as follows:Liaoning>Shandong>Jiangsu>Zhejiang.The concentration of uranium in drinking water W88 maximal in Shandong Province and minimal in Zhejiang Province.
