1.Experimental mouse model of Chlamydia pneumoniae strain pneumonitis

Objectives: To evaluate mice as experimental animal for Chlamydia pneumoniae, a common cause of acute respiratory infections in human. Methods: Intranasal inoculation of Icr mice with C. Pneumoniae induced a prolonged course of lung infection, as demonstrated by persistence of lung pathology(60 days). Results: Icr mice were susceptible to C. pneumoniae. Lung pathology was characterized by patchy interstitial pneumonitis with predominately neutrophil leukocyte infiltration in the early(7 days) and lymphocytes infiltration in the later stages(14 days later) of infection. Conclusions:Icr mice were susceptible to C. pneumoniae and the mouse model is useful for the investigation of the pathogenesis of C. pneumoniae infection.
2.Chemiluminescent Southern blot System for Detection of HBV Replication in vitro and Its Application for Analysis the Drugs Inhibition to HBV

Objective:To stablish of chemiluminescent Southern blot detection system for examining HBV DNA replication intermediates in HepG2.2.15,and analyse the inhibition of HBV replication with three kind of drugs with different targets.Methods:The HBV DNA replication intermediates were extracted and analyzed by Southern blot with HBV probe,which(pTHBV1047) was labelling with digoxigenin.The results of the hybridization were detected by chemiluminescent,and the condition of hybridization was optimized.After treated with lamivudine,Bay41-4109,?-Galcer in different concentration,the HBV DNA from the HepG2215 cells were detected with the system.Results:the sensitivity of the system was 1pg of pTHBV1047,and HBV specific positive signals was detected with the DNA from HepG2.2.15.The three kinds of drugs can inhibit the HBV replication obviously with chemiluminescent Southern blot detection system,the IC50 were 1.53?mol/L,0.41?mol/L,0.01?mol/L.Conclusion:The HBV replication intermediates from the cell of HepG2.2.15 can reflect the antiviral effect accurately with different targets drugs and this mothod would be used in the study of Chinese-midicine.
3.Lymphoma mimicking Beh?et's disease:case report and literature review

Objective To study the clinical char acteristics and outcome of two lymphoma patients mimicking Beh?et's disease. Methods Lymphoma was diagnosed in two patients mimicking Beh?et's disease referred to our Department in 2015. A search on published similar cases in Chinese database and the Pubmed was also performed and then analyzed. Results Eight patients were indentified in this pooled analysis, six of which were non-Hodgkin lymphoma (NHL). All of the eight cases presented with cutaneous lesion, seven cases with fever, seven cases with oral ulceration and six cases with orogenital ulceration, respectively. Ocular involvement was present in four of the eight cases, two were with a positive pathergy test. Among feverish patients, six were moderate or high fever, four were high fever, one was low-grade fever. Neutropenia was found in four patients, and lymphocytoponia in four of five patients with detailed data. All patients fulfilled the 2014 International Criteria for Beh?et's Disease (ICBD) with an average score of (5.8 ±1.5), ranging from 4 to 8. Survival period ranged from one month to 36 months, with an average of 8 months. Conclusion For patients diagnosed as Beh?et's disease are finally diagnosed as lymphoma. For patients with Beh?et's disease present-ation but also present with mediate to high fever, atypical deepseated ulcer, neutropenia or lymphocytoponia, malignancy especially lymphoma should be investigated.

5.A survey of knowledge on common cold and its treatment situation among physicians from various levels of hospitals in mainland China

Objective To investigate the cognition of the common cold and current situation of the treatment among physicians from various levels of hospitals in Chinese mainland,so as to provide evidence for future continuing medical education and rational medication.Methods A questionnaire designed for this survey was used to learn about the general information,cognitive degree of the common cold and prescription habits of physician who prescribed for cold within last three months,from various levels hospitals.Results A total of 1001 physicians were interviewed.Among them,749 physicians chose right options that the cold was the common cold and the influenza with 79.84% in resident physicians and 56.76% in chief physicians.A total of 745 physicians chose options that the course of common cold will be lasting 4 to 7 days; 895 physicians chose options that old people are the most susceptible for complication; 669 physicians thought the common cold was the most common infection in winter; 841 physicians used clinical methods to diagnose the common cold; 736 physicians thought although the cold was a kind of self-limited disease and symptomatic treatment could alleviate symptoms and improve life quality,patients should see doctor in time if it turns to severer; and 745 physicians held the opinion that treatment of the common cold should focus on relieving symptoms first. In addition, 61.60% physicians had made prescription based on clinical symptoms; 505(54.24% ) of them thought compound drugs were priority in treating the common cold.However,there were still 43 physicians prescribed antibiotics for common cold.Conclusions There is misunderstanding and discrepancy in cognition towards common cold and treatment among physicians from various levels of hospticals in mainland China.Physicians should standardize diagnosis and treatment for the common cold according to the domestic and foreign guidelines.
6.Hydrogen photoproduction from acetate by Rhodopseudomonas palustris.

Based on the characteristics of metabolism of photosynthetic bacteria and the major kinds of organic compounds produced in wastewater degradation, eleven kinds of organic compounds were chosen for hydrogen photoproduction using Rhodopseudomonas palustris Z strain. The maximal volumetric H2 productivity was obtained using acetate as the sole carbon source and electron donor. The kinetics of cell growth and H2 liberation, and the influences of several major limiting factors on photoevolution of H2 were examined using acetate as carbon source. It was shown that hydrogen production was partially correlated with cell growth. The medium composition of the preculture, the preculture time, and inoculation volume were confirmed to have big effects on hydrogen photoevolution. The time delay of H2 production was evidently shortened using the inoculum of late exponential growth phase or stationary phase using ammonium sulfate as nitrogen source or with the inoculum of middle exponential growth phase using glutamate as the nitrogen source. The identity of temperature and light intensity for H2 evolution and cell growth has significant potential application in the technology of splitting organic acid into H2 by photosynthetic bacteria. The concentrations of acetate and glutamate in the medium affected hydrogen photoevolution and cell growth significantly. The productivity of H2 increased with substrate concentrations when substrate concentrations of sodium acetate and sodium glutamate were lower than 70 mmol/L and 15 mmol/L, respectively. Hydrogen production was inhibited but the cell growth was faster when the concentration of sodium glutamate over 15 mmol/L due to forming free NH4+. The highest rate of hydrogen production was 19.4 mL.L-1.h-1 using 30 mmol/L of sodium acetate as hydrogen donor under the standard conditions, respectively. The optimal conditions for hydrogen production were 35-37 degrees C, 6000-8000 lx and pH 7.3-8.3. The effects of oxygen and inoculation volume on photoproduction of hydrogen were also discussed.

7.Study on molecular identification and pre-harvest sprouting characteristic of the transgenic anti-trxs-gene wheat line 00T89.

Pre-harvest sprouting of wheat is caused by a series of enzymes, in which alpha-amylase plays a key role. Thioredoxin (trx) is an important protein capable of reducing S-S to -SH. The authors transferred the anti-trxs gene into wheat ( Triticum aestivum L.) cultivar Wanmai 48 in 2000 in order to obtain a transgenic line with low activities of trx and alpha-amylase in wheat seeds to enhance their resistance to pre-harvest sprouting. In this work, a pair of specific anti-trxs primers were used in PCR test to validate the transgenic plants in T4 lines. A pair of primers of a wheat actin gene cDNA and a pair of trxs gene primers were used in the reverse transcription PCR test to measure the relative content of trx mRNA in the transgenic positive lines. The experiments of pre-harvest sprouting were carried out to test the resistance to pre-harvest sprouting. The results showed that there were 13 positive lines with the anti-trxs gene among 18 transgenic lines. In the maturity stage, the 8 relative contents of the trx mRNA in the 13 positive lines were reduced evidently (P < 0.01). The trx mRNA was correlated with the sprouting parameters significantly (r = 0.7181) . 6 lines with low trx mRNA showed stronger resistance to pre-harvest sprouting during the period from 30 days after anthesis to 10 days after maturity. Compared with non-transgenic plant (Wanmai 48, CK), the average sprouting time of these lines was postponed by 2.7 days (P < 0.01), the sprouting ratio of seeds on spikes and the spike sprouting degree were reduced by 35.5% (P < 0.01) and 47.5% (P < 0.01), respectively. However, 25 days after maturity, the sprouting capacity of the positive lines recovered gradually, and no significant differences were found between them (P > 0.05) . Thus, it can be concluded that the wheat pre-sprouting was controlled effectively in some 00T89 transgenic lines as a result of the expression of anti-trxs gene.

8.Effect of gastric mucosa cell turnover on the adaptive cytoprotection in chronic alcohol drinking rat.

AIMTo investigate the correlation between the gastric adaptive cytoprotection and the low concentration alcohol intake in a chronic drinking rat model and the effect of chronic ethanol exposures on the cell turnover of the gastric mucosa and its possible role in adaptive cytoprotection.
METHODSSprague-Dawley rats received the drinking water containing 6% (v/v) ethanol as their only water intake for 28 days. In the different stages of the 28 days (1st, 3rd, 7th, 14th and 28th days), the stomachs of the rats were cannulated and perfused with pure ethanol, and the severity of mucosal lesions was measured in 2 hours at the end of perfusion respectively. The cell proliferation and apoptosis in gastric mucosa of rats in different groups were analyzed by flow cytometer, immunohistochemistry and computer image analysis.
RESULTSPure ethanol caused ulcer and haemorrhagic damage in the corpus and antral mucosa of the control rats. These lesions were prevented by pretreatment of the animals with ethanol exposure in the 3 rd to 14 th days. The damage index was decreased by 80%, as compared with those in control rats. There was no significant difference in the rats exposed to the ethanol in the 1st and 28th days. Compared with control, the cell apoptosis in gastric mucosa of the rats was enhanced during they exposure to the ethanol in the 3rd to 28th days. Otherwise the cell proliferation was increased in the 3rd to 28th days, and decreased in the 28th days, respectively.
CONCLUSIONChronic adequate alcohol intake may enhance the cell turnover of gastric mucosa and lead to an adaptive cytoprotection. Long-term stimulus with the low concentration ethanol may cause the atrophy of gastric mucosa and reduce the gastric mucosal cytoprotective effect.

9.Effect of jinfu kang to experimental precancerous colon lesions and urinary metabolites in rat.

OBJECTIVE: To profile urinary metabolite variations from 1, 2-dimethylhydrazine (DMH)-induced precancerous colon rats, Jinfu Kang treated rats and healthy controls.
METHODWe used ethyl chloroformate derivatization and gas chromatography-mass spectrometry (GC-MS) based metabonomic method to analyze rat urines.
RESULTThe time-dependent variations of metabolite profile showed a progressive deviation of the metabolism in the model group from the initial pattern over time and a systemic recovery of the metabolism in the treatment group, which is consistent with the histological results. The in-depth analysis indicated that the disorder of tricarboxylic acid cycle (TCA), tryptophan metabolism, polyamine metabolism and gut flora structure were associated with DMH intervention.
CONCLUSIONMetabolic study revealed that Jinfu Kang can effectively reverse metabolic departures in DMH-induced precancerous colon rat, which is consistent with pathological results.

10.Detection of chromosomal aberration in sporadic colorectal cancer with comparative genomic hybridization.

OBJECTIVETo investigate the chromosomal aberration in sporadic colorectal carcinoma and its association with clinicopathological features.
METHODSComparative genomic hybridization(CGH) was used to screen the changes in the number of DNA sequence copies in 40 sporadic colorectal cancer patients in order to identify regions that contain genes important for the development and progression of colorectal cancer.
RESULTSIn 40 sporadic colorectal cancer, frequent gain at 20 q, 12 q, 13 q, 7 p, 7 q and 16 q were found, while loss was also found at 18 q, 5 q, 4 q, 8 pand 17 p. The number of chromosomal aberration was closely associated with tumor stage(P<0.05). No significant association was found between the number of chromosomal aberration and tumor site, histopathologic type and histologic grade.
CONCLUSIONSChromosomal aberration exists generally in sporadic colorectal carcinoma. The number of chromosomal aberration and gain of 20q are closely associated with tumor stage.
