1.Negative acute phase proteins in patients with chronic urticaria

Objective To investigate the relationship between negative acute phase proteins and chronic urticaria (CU).Methods Fifty patients with CU were enrolled into this study,and divided into three grades,i.e.,mild (grade 1),moderate (grade 2) and severe (grade 3) according to symptoms.Twenty-eight health checkup examinees served as the control group.Immunoturbidimetry was performed to determine serum levels of prealbumin (PA) and transferrin (TRF).Enzyme-linked immunosorbent assay was conducted to measure serum levels of insulinlike growth factor-1 (IGF-1) and tumor necrosis factor-α (TNF-α).Statistical analysis was carried out to assess differences in these indices between these two groups,the relationship among these indices and between these indices and disease severity.Results Compared with the control group,the patients with CU showed reduced serum levels of PA ((229.99 ± 54.16) vs.(272.06 ± 36.42) mg/L,t =3.667,P < 0.05) and IGF-1 ((177.23 ± 46.48) vs.(239.88 ± 45.16) μg/L,t =5.748,P< 0.05),but higher serum levels of TNF-α ((25.39 ± 11.01) vs.(14.13 ± 6.12) ng/L,t =4.989,P< 0.05),and similar serum levels of TRF ((2.48 ± 0.49) vs.(2.48 ± 0.25) g/L,P> 0.05).The serum level of PA showed a significant negative correlation with that of TNF-α (r =-0.312,P < 0.05),as well as with disease severity (r =-0.635,P < 0.01),whereas the serum level of TNF-α showed a significant positive correlation with disease severity (r =0.409,P < 0.01),and no statistical correlation was found between the remaining indices (all P > 0.05) in the patients with CU.Conclusions Serum levels of some negative acute phase proteins decrease and negatively correlate with disease severity in patients with CU.Acute phase response may be involved in the occurrence of CU.
2.Biomarkers for acute-phase response and coagulation/fibrinolysis in patients with chronic urticaria: a correlation study

Objective To evaluate the relationship of acute-phase response and coagulation/fibrinolysis with chronic urticaria (CU).Methods Fifty-three patients with CU and twenty-five healthy human controls were enrolled in this study.Venous blood samples were collected from all of these subjects.Then,enzyme-linked immunosorbent assay was carried out to measure the serum levels of interleukin-6 (IL-6) and amyloid protein A (AA),and immunoturbidimetry to determine the levels of serum high sensitive C-reactive protein (Hs-CRP) and plasma D-dimer.The patients with CU were classified into three groups according to the severity of clinical symptoms.The levels of serum IL-6 and plasma D-dimer were given in mean ± standard deviation,and those of serum AA and Hs-CRP in mean (25th percentile,75th percentile).Rank sum test and t test were performed to compare these parameters between the patients and controls,and Spearman rank correlation analysis was conducted to study the relationship among these parameters as well as between these parameters and symptom severity in these patients.Results The levels of serum IL-6,serum AA and plasma D-dimer were (10.70 ± 4.94) ng/L,4.92 (8.22,12.51) μg/L,and (222.32 ± 163.69) μg/L respectively in the patients with CU,significantly higher than those in the healthy controls ((7.49 ± 3.41) ng/L,2.11 (1.21,2.83) μg/L,(104.72 ± 43.12) μg/L,respectively,all P< 0.05),while no significant differences were observed between the patients and controls in the level of serum HsCRP (0.30 (0.10,1.40) mg/L vs.0.30 (0.10,0.55) mg/L,P > 0.05).In patients with CU,the levels of serum IL-6 were unrelated to those of serum Hs-CRP,serum AA,or plasma D-dimer (all P > 0.05),whereas a positive correlation was observed between the levels of serum Hs-CRP and AA (r =0.67,P < 0.01),serum Hs-CRP and plasma D-dimer (r =0.49,P < 0.01),serum AA and plasma D-dimer (r =0.38,P < 0.01).Further more,the levels of serum Hs-CRP,AA and plasma D-dimer were significantly correlated with symptom severity in patients with CU (r =0.63,0.62,0.47,respectively,all P < 0.01).Conclusions Acute-phase response coexists with the activation of coagulation system in patients with CU,suggesting a potential association between acute-phase response and coagulation system activation.
3.Balloon vertebroplasty reinforced by injectable calcium sulfate cement versus posterolateral fusion for treatment of thoracolumbar burst fractures

Objective To compare the clinical curative effects of balloon vertebroplasty reinforced by injectable calcium sulfate cement(CSC) and posterolateral fusion after short-segment pedicle screw fixation through the fractured vertebra in treatment of thoracolumbar bumt fractures.Methods From February 2012 to May 2015,68 cases patients with single thoracolumbar burst fracture were admitted in Orthopaedics and Traumatology Department of Traditional Chinese Medicine Hospital of Gaozhou,and were randomly divided into two observation group and control group,each group with 34 cases.Observation group were treated by short-segment pedicle screw fixation through the fractured vertebra followed by transpedicular balloon reduction of the endplate and CSC injection,the control group were underwent posterolateral fusion in addition to short-segment pedicle screw fixation through the fractured vertebra.The 2 groups were compared at preoperation,and the last follow-up in terms of visual analog pain score(VAS),Oswestry disability index(ODI),anterior height ratio of the injured vertebra,middle height ratio of the injured vertebra,the vertebral wedge angle and the sagittal Cobb angle.Results The patients were followed up from 12 to 54 months.All the measure indexes at the last follow-up were significantly improved than those at preoperation in all the patients(P<0.05),there was no significant between two groups difference in the VAS score or ODI at last follow-up(P>0.05).At the last follow-up,the observation group showed a significantly less loss than the control group regarding the corrections of anterior height ratio of the injured vertebra((93.6±3.4) vs.(81.8±4.8)),middle height ratio of the injured vertebra((90.7±3.8) vs.(78.9±5.1)),the veaebral wedge angle((5.7±1.9) ° vs.(8.8±3.0) °) and the sagittal Cobb angle((5.2±2.1) ° vs.(7.9±2.4) °)(t=4.143,5.786,4.819,4.465,P<0.05).Conclusion In treatment of thoracolumbar burst fractures,compared with posterolateral fusion,balloon vertebroplasty reinforced by injectable CSC can better restore the shape of injured vertebral body and stability of the anterior and middle columns in the early period,can also reduce losses of corrections,and keep the function of the spinal motion segment.
5.Recent curative effect evaluation of double Endobutton technique and clavicular hook plate in patients with Tossy grade Ⅲ acromioclavicular separation

Objective To compare recent curative effect between double Endobutton technique and clavicular hook plate (CHP) in patients with Tossy grade Ⅲ acromioclavicular separation.Methods One hundred and forty-nine patients with Tossy grade Ⅲ acromioclavicular dislocation were divided into control group (78 cases,treated with CHP) and observation group (71 cases,treated with double Endobutton technique) according to treatment method.The postoperative injured shoulder visual analogue score (VAS),abduction-rise activity and flexion-rise activity,Constant-Murley score and Karlsson grade were compared between 2 groups.Results All the patients successfully completed the operation,and the postoperative follow-up time was 9-40 (15.8 ± 5.2) months.The injured shoulder abduction-rise activity and flexion-rise activity postoperative 9 months in observation group were (100.9 ± 13.3)° and (131.6 ± 13.4)°,in control group were (81.4 ± 8.4)° and (96.7 ± 10.9)°; the injured shoulder VAS in observation group and control group were (2.2-± 0.8) and (3.0 ± 0.9) scores,and there were statistical differences between 2 groups (P < 0.01).The Constant-Murley score in observation group and control group were (85.4 ± 6.4) and (82.5 ± 6.2) scores,the excellent and good rate of Karlsson grade in observation group and control group were 84.5%(60/71),91.0% (71/78),and there were no statistical differences (P > 0.05).Conclusions The recent curative effect of double Endobutton technique and CHP in patients with Tossy grade Ⅲ acromioclavicular separation is similar.
6.Comparison of Treatment Outcomes of Ticagrelor and Clopidogrel among Patients Undergoing Percutaneous Coronary Intervention: A Meta-analysis

We performed a meta-analysis of randomized controlled trials (RCTs) to investigate the efficacy and safety of ticagrelor (TIC) vs.clopidogrel (CLO) in patients undergoing percutaneous coronary intervention (PCI).In Jun 2016,a literature search was started and all the studies were conducted from 2010 to 2015.We systematically searched the literature through the MEDLINE database,Cochrane library,and EMBASE database.Quality assessments were evaluated with Jadad quality scale.Data were extracted considering the characteristics of efficacy and safety designs.Six RCTs enrolling 26 244 participants and satisfying the inclusion criteria were finally analyzed.There was a significant decrease of all-cause mortality (MD=0.83,95%CI=0.74-0.93,P=0.001) and myocardial infarction (MI) (MD=0.78,95%CI=0.70~.88,P=0.000).There were no significant differences in stroke (MD=1.34,95%CI=0.99-1.79,P=0.06),total bleeding (MD=0.97,95%CI=0.84-1.12,P=0.66),minor or major bleeding (MD=l.06,95%CI=0.94-1.19,P=0.35) in patients undergoing PCI after treatment with TIC vs.CLO.TIC could be more significant in decreasing all-cause mortality and MI than CLO,but there were no significant differences between TIC and CLO in inhibiting stroke,major bleeding,major or minor bleeding in patients undergoing PCI.
7.Rapid production of tissue-engineered bone based on two kinds of adult stem cells in bone fusion

BACKGROUND:Despite tissue repair with a kind of stem cels has achieved great outcomes, but the combination of two or more kinds of stem cels to construct tissue-engineered bone is stil rare.
OBJECTIVE:To evaluate the efficacy of tissue-engineered bone constructed based on combining adipose-derived stem cels with bone marrow mesenchymal stem cels and alogenic bone in rabbit posterior intertransverse lumbar interbody fusion models.
METHODS:Seventy-five New Zealand rabbits were randomly divided into five groups. L5,6 interbody fusion models were made in each group. Then, these five groups were respectively treated as folows (1) group A: bone marrow mesenchymal stem cels/alograft iliac bone with adipose-derived stem cels; (2) group B: bone marrow mesenchymal stem cels/alograft iliac bone with normal saline; (3) group C: alograft iliac bone with adipose-derived stem cels; (4) group D: alograft iliac bone with normal saline; group E: autologous iliac bone alone. Each group was detected by PET/CT to contrast the standard uptake value at 1, 3, 5 weeks postoperatively.
RESULTS AND CONCLUSION: PET/CT images showed varying degrees of bone fusion and bone metabolism enhancement in the five groups. With time, the standard uptake value was increased in each group, and except the group D, the standard uptake value was higher in al the groups at 3, 5 weeks than at 1 week (P < 0.05). There was no difference in the standard uptake value of al the five groups at weeks 3 and 5 after operation (P > 0.05). The standard uptake value of the group A was better than that in the group B and group E (P < 0.05). Taken together, the combination of adipose-derived stem cels and bone marrow mesenchymal stem cels for rapid production of tissue-engineered bone has good osteogenesis and vascularization.
8.Five cases report of Ebola virus disease

Objective To analyze the clinical characteristics ,treatment and outcome of Ebola virus disease so as to provide early clinical recognition and treatment for this disease .Methods The clinical manifestations and treatment of 5 cases of Ebola virus disease in Ebola Holding Center of Sierra‐Leone China Friendship Hospital from 15 March 2015 to 15 May 2015 were retrospectively analyzed .And the clinical characteristics and possible effective treatment were discussed combined with related literature . Results Five patients were diagnosed with Ebola virus disease by polymerase chain reaction and 4 cases of 5 patients had confirmed contact history ,while 1 case had no clear contact history .All the 5 cases presented with low fever ,headache and joint pain .Three cases then progressed into severe gastrointestinal symptoms such as nausea ,vomiting ,diarrhea and hypovolemic shock .The patients presented with fast heart rate and shortness of breath and other inflammatory response syndrome in acute phase .One patient rapidly progressed to liver pain ,jaundice and anuria ,then died .Three severe cases recovered after treated with fluid resuscitation ,circulation maintenance and electrolyte balance in acute phase .Conclusions The early symptoms of Ebola virus disease are low fever ,joint pain and nausea .Frequent vomiting ,diarrhea , low blood pressure and electrolyte disorder indicate severe conditions .Shock and electrolyte disorder are deadly complications .Early recognition ,diagnosis and treatment are the key to improve the prognosis .
9.Determination of equilibrium solubility and n-octanol/water partition coefficient of pulchinenosiden D by HPLC.

OBJECTIVETo determine the equilibrium solubility of pulchinenosiden D in different solvents and its n-octanol/water partition coefficients.
METHODCombining shaking flask method and high performance liquid chromatography (HPLC) to detect the n-octanol/water partition coefficients of pulchinenosiden D, the equilibrium solubility of pulchinenosiden D in six organic solvents and different pH buffer solution were determined by HPLC analysis.
RESULTn-Octanol/water partition coefficients of pulchinenosiden D in different pH were greater than zero, the equilibrium solubility of pulchinenosiden D was increased with increase the pH of the buffer solution. The maximum equilibrium solubility of pulchinenosiden D was 255.89 g x L(-1) in methanol, and minimum equilibrium solubility of pulchinenosiden D was 0.20 g x L(-1) in acetonitrile.
CONCLUSIONUnder gastrointestinal physiological conditions, pulchinenosiden D exists in molecular state and it has good absorption but poor water-solubility, so increasing the dissolution rate of pulchinenosiden D may enhance its bioavailability.

10.Effects of Jisuikang on Nogo-NgR gene expression in spinal cord rats with injury.

OBJECTIVETo study the effects of Jisuikang (Chinese characters) on Nogo-NgR gene expression, and to explore the protective effects and mechanism of Jisuikang (Chinese characters) on spinal cord injury in rats.
METHODSOne hundred eighty female rats were randomly assigned to 6 groups(30 rats per group). Sham group: T10 lamina was resected only and spinal cord was untreated. Model group: spine cord injury (SCI) was created with a modified impinger of Allen's by impacting on the T10 spinal cord. Prednisolone group: Prednisolone (0.06 g/kg) was given by intragastric administration at a time interval of 24 hours after operation. The Jisuikang (Chinese characters) high, moderate and low dose groups: Jisuikang (Chinese characters) was supplied with different dose (50 g/kg, 25 g/kg, 12.5 g/kg) by intragastric administration in rats after operation,for the first time at 30 min after surgery. Animals were killed 3, 7, 14 days after surgery. The expression levels of Nogo-A and NgR were observed by Western Blot and Real-time PCR.
RESULTSThe expression of Nogo-A and NgR was at the basic level at all time points in sham group. Compared with model group, the protein expression levels of Nogo-A and NgR in sham, prednisolone, Jisuikang (Chinese characters) moderate dose groups were statistically significant at all time points (P < 0.05). No difference was found in Jisuikang (Chinese characters) high and low dose groups (P > 0.05). Three days after surgery, the mRNA levels of Nogo-A and NgR in treatment group were significantly lower than that in model group (P < 0.01); 7 days after surgery,Nogo-A and NgR mRNA expression were dramatically upregulated and peaked; 14 days after operation, the expression was decreased, but still significantly higher than that in other treatment groups (P < 0.01). Prednisolone and Jisuikang (Chinese characters) moderate dose groups showed the most significant effects among all groups,but there was no statistically significant difference between two groups (P > 0.05).
CONCLUSIONThe decoction Jisuikang (Chinese characters) can promote the nerve cell regeneration by regulating Nogo-A and NgR gene expression, activating Nogo- NgR signaling pathways after acute spinal cord injury.
