OBJECTIVE To explore the effect of chloride channels on the neuronal injury following cerebral ischemia. METHODS Tweleve day in vitro (12dIV) neurons in rats were randomly divided into normal control, 3-morpholinosydnonimine (SIN-1, 1.0 mmol·L~(-1) for 18 h) group, SIN-1+4-acetamido-4'-isothiocyanatostilbene-2,2'-disulphonic acid(SITS, 0.5 mmol·L~(-1)) group and SIN-1+4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS, 0.1 mmol·L~(-1)) group. Drugs were added with SIN-1 simultaneously and coincubated for 18 h. The neuronal apoptosis and morphological changes were detected with Hoechst 33258. Chloride channels(ClC)-2/ClC-3 were analyzed with immunofluorescence, the chloride channel currents were recorded with whole cell patch-clamp technique. RESULTS Hoechst 33258 staining showed that the apoptotic percentages were (18.61±0.59) %, (50.43±0.56)%, (23.37±0.52)% and (23.37±0.84)% in normal control group, SIN-1 group, SIN-1+SITS or SIN-1+DIDS groups, respectively. ClC-2/ClC-3 were positively expressed in normal neurons. The currents in neurons exposed to SIN-1 were increased about 55%-56%, SITS and DIDS, two kinds of chloride channel blockers could inhibited the currents about 50%-60% and 30%-40%, respectively. CONCLUSION Voltage-dependent chloride channel maybe participate in the neuronal apoptosis induced by NO, and the activities of chloride channels are perhaps involved in the cerebral ischemic injury.

The pathogenic effects of crops from Keshan disease area were studied by rat-feeding. As double controls, another two groups were fed with crops from non-endemic area and synthetic diet. Those from disease area caused necrosis of heart muscle vascular changes of myocardial mitochondria and fatty degeneration of liver. Compared with the controls, the free radical and lipid peroxide content in heart and liver were significantly higher, while the ATP and AN contents in these organs were apparently lower. The multielement contents and its constituent patterns in the feeds, whole blood, heart, liver, kidney and brown adipose tissue of rats in experimetal group were apparently different from those in the control groups. The statistic analysis of multiple factors showed that eight elements Se, Mo, Cu, Mn, Cr, Fe, Zn and Pr might be the perilous factors, that is, under the condition of low selenium, the constituent pattern of selenium and the eight elements might be the complicated factors of endemic crops.

Aim To observe the protective effects of SITS and DIDS,two kinds of chloride channel blockers,on hippocampal neuronal damage induced by NO in culture.Methods The cultures were divided into three groups:control group,NO treatment group,NO treatment plus chloride channel blocker group. The cultures were detected with the methods of morphological stain (Hoechst 33258),and the apoptotic neurons and neuronal viabilities were observed through MTT quantitative analysis. The activated caspase-3 was analyzed with western blot.Results There were significant protective effects of SITS and DIDS on neuronal damage with dose-dependence.Conclusions Chloride channel blockers have some protective effects against neuronal injury induced by NO.

Objective To investigate the perihematomal changes of hyperacute parenchymal hematomas and the clinical value by MRI. Methods Multi-sequence MRI was performed on 4 hematomas in vitro and on 15 pigs with lobar intracerebral hemorrhage (ICH) for about 30～60 min and 3 h respectively. The integrity of blood-brain barrier (BBB) in ICH pigs was assessed by electron microscopy and Evan's blue dye technique. MR scanning was performed on 2 ICH patients proved by CT for 4 and 9 h after onsets. Results FLAIR and T 2-weighted images showed hyperintensity signal around the hematomas in vitro and in pigs with ICH within 1 h, and more obviously at 3 h. When the gelose cavity was cut, plasma was seen around the clot. The perihematomal ADC values of the pigs increased both within 1 and at 3 h after ICH. However, the BBB was intact at 3 h, which was proved by electron microscopy and Evan's blue dye technique. Water-like intensity signal was observed around the hematomas in two patients with acute ICH. Conclusion The perihematomal changes of hyperacute ICH observed on MRI are resulted from the blood clot contraction and the serum formation and extravasation, but not real cerebral edema.

Objective To observe the inhibiting activities of fibronectin (FN) and citrullinated fibronectin (cFN) on disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4),and to explore the extracellular regulative mechanisms of ADAMTS-4.Methods FN was incubated with peptidylarginine deaminase type 4 (PADI4).Western blotting analysis was used to verify the citrullination of FN.The binding activity of FN and cFN to ADAMTS-4 were investigated by enzyme-linked immunosorbent assay (ELISA).The proteolytic ability of ADAMTS-4 after binding to FN and cFN were measured with the aggrecanase activity assay kit.One-way ANOVA,LSD-t test and t-test were used for statistical analysis.Results The immunosignal of citrulline was detected in FN after incubated with PADI4,but not in the absence of PADI4.A higher absorbance at 405 nm was detected when the full-length ADAMTS-4 protein was incubated with FN (2.182±0.042) than cFN (0.624±0.033; t=50.522,P＜0.01).Additionally,the recombinant ADAMTS-4 protein with a truncation at the C-terminus displayed low absorbance at 405 nm when the enzyme was incubated with both FN(0.971±0.024) and cFN(0.934±0.012; t=2.388,P＞0.05).Large amounts of ARGxx peptide were detected with full-length ADAMTS-4 in aggrecanase activity assay [(0.908±0.088) nmol/L],but significantly less when in the presence of FN and ADAMTS-4 [(0.573±0.000) nmol/L,P＜0.05].The production of this peptide was more when full-length ADAMTS-4 was incubated with cFN [(0.830±0.020) nmol/L,P＜0.05] than with FN.The reaction containing the truncated ADAMTS-4 without FN or cFN yielded the highest concentration of ARGxx peptide [(36.420±3.673) nmol/L],peptide production was not significantly altered when FN [(41.099±0.101) nmol/L] or eFN [(41.064±0.083) nmol/L] were added to the reaction.Conclusion FN could bind to ADAMTS-4 and inhibit its proteolytic activity.After citrullinated by PADI4,the binding activity of cFN is weakened and less inhibition to ADAMTS-4.

The damage degree of neurons in perilesion at different time points was observed in order to explore the optimal operation occasion. Piglet lobar hematomas were produced by pressure-controlled infusions of 2.5 mL autonomous blood into the right frontal hemispheric white matter over 15 min, and the metabolic changes were ambulatorily detected with MRS at 3rd, 12th, 24th and 48th h after hematoma induction. Brain tissues of perihematoma were also obtained at different time points. The transcription level of Bax gene was detected by in situ hybridization and apoptosis by TUNEL technique, and the pathologic change of neurons was observed under an electron microscope. The results showed that the number of Bax positive cells reached the peak at 24 h (79.00 +/- 4.243/5 fields). There was no significant difference in A values between 3 h and 6 h, 12 h (P > 0.05), but there significant difference between 24 h and 3 h, 6 h, 12 h (P < 0.05). The number of apoptotic cells reached the peak at 24 h (P < 0.001), and there was no significant difference between 3 h and 6 h (P = 0.999). The area of the apoptotic cells showed no significant difference between 3 h and 6 h or among 3 h, 6 h and 6 h (P > 0.05). Lac peak mainly occurred at 24 h and 48 h, while on the healthy side, no Lac peak was detectable. The ratio of NAA/Cr presented a descent tendency, but there was no significant difference among the groups before 12 h (P > 0.05), there was very significant difference between 3, 6 and 24, 48 h (P < 0.01). Under electronic microscopy, the neuronal damage surrounding hematoma in 3 to 6 h was milder than in 24 h to 48 h. It was concluded that the secondary apoptosis, damage and metabolic disturbance of the neurons surrounding hematoma was milder in 3-6 h in acute intracerebral hemorrhage, while obviously aggravated in 24-48 h. An effective intervention is needed to reduce secondary damage as soon as possible.

Objective To investigate the clinical effect of homeopathic reduction with minimally invasive adjustable plate in treatment of sacral fractures.Methods A retrospective case-control study was conducted to assess the data of 89 patients with sacral fractures treated from January 2013 to January 2015.There were 49 males and 40 females,with a mean age of 37.8 years (range,18-70 years).Denis classification was type Ⅰ in 24 patients,type Ⅱ in 48,and type Ⅲ in 17.Patients were divided into three groups according to fixation methods:homeopathic reduction with minimally invasive adjustable plate group (Group A,n =30),sacroiliac screw group (Group B,n =31) and iliolumbar rod group (Group C,n =28).Operation time,blood loss,intraoperative radiographic time,and complications were recorded.Reduction quality was assessed using the Matta criteria.Bone healing was evaluated based on X-ray appearance.Functional outcome was evaluated using the Majeed score at last tollow-up.Results Operation time was significantly lower in Group A [(109.3 ± 14.4) min] and Group B [(114.2 ± 17.7) min] than Group C [(126.8 ± 15.7)min] (P ＜ 0.05),but there was no significant difference between Groups A and B (P ＞ O.05).Blood loss was significantly lower in Group A [(433.3 ± 121.3)ml] and Group B [(461.3 ± 130.8)ml] than Group C [(785.7 ±205.0)ml] (P ＜0.05),but there was no significant difference between Groups A and B (P ＞ 0.05).Radiographic time was (5.6 ± 1.9) s in Group A,(13.4 ± 3.1)s in Group B,and (8.4 ± 2.5)s in Group C,showing significant difference among the three groups (P ＜ 0.05).Excellence rate of Matta score in Group A [70％ (21/30)] and Group C [86％ (24/28)] was higher than that in Group B [32％ (10/31)],but the there was no significant difference between Group A and C (P ＞ 0.05).Fracture healing was found in all patients and no nonunion was observed.Excellent rate of Majeed score in Group A [80％ (24/30)] and Group C [82％ (23/28)] was higher than that in Group B [54％ (17/31)],but the there was no significant difference between Groups A and C (P ＞0.05).Complication rate in Group B [29％ (9/31)] and GroupC [29％ (8/28)] were higher than that in Group C [3％ (1/30)],but the there was no significant difference between Groups B and C (P ＞ 0.05).Conclusion For sacral fractures,homeopathic reduction with minimally invasive adjustable plate can reduce operation time and intraoperative radiographic time,improve reduction rate and lower incidence of complications.

Objective To investigate the clinical efficacy of trimebutine combined with Xiangsha Pingwei in the treatment of functional dyspepsia and its effect on gastrointestinal hormone.Methods One hundred and twenty cases of functional dyspepsia were randomly divided into the control group and observation group,60 cases in each group.The control group was treated with trimebutine 0.2 g,3 times/d and the observation group was treated with trimebutine 0.2 g,3 times/d and Xiangsha Pingwei 10 g,2 times/d.Both groups were treated for four weeks,a course of treatment.Clinical efficacy and the influence on gastrointestinal hormone levels were compared between the two groups.Results The symptoms scores in both groups all decreased significantly after treatment.While the postprandial fullness scores,early satiety scores and epigastric pain scores in the observation group were significantly lower than those in the control group (0.62±0.12 vs.1.85±0.25,0.56±0.14 vs.1.46±0.21,0.48±0.15 vs.1.32±0.12,t=34.36,27.62,33.87,P<0.05).The total effective rate in the observation group was significantly higher than that in the control group (90.00% vs.73.33%,χ2=5.57,P<0.05).MOT and GAS levels in both groups increased significantly.While MOT and GAS levels in the observation group were significantly higher than those in the control group (MOT: (355.2±78.1) ng/L vs.(288.7±82.4) ng/L;GAS:(96.5±17.6) ng/L vs.(88.4±16.0) ng/L,t=4.54,2.64,P<0.05).The somatostatin (SS) level in both groups decreased significantly.While the SS level in the observation group was significantly lower than that in the control group ((41.1±11.1) ng/L vs.(48.9±12.9) ng/L,t=3.55,P<0.05).Conclusion Trimebutine combined with Xiangsha Pingwei can significantly improve the clinical symptoms and gastrointestinal hormone in patients with functional dyspepsia.
