1.Study of relationship between scattered or single lesion on diffusion-weighted imaging and arterial stenosis in acute cerebral infarction

Objective To investigate the relationship between scattered or single lesion of acute cerebral infarction in middle cerebral artery territory on diffusion-weighted imaging (DWI) and stenosis of middle cerebral artery (MCA) or internal carotid artery (ICA). Methods With exclusion of cardioembolism, 73 consecutive patients with acute cerebral infarction of the unilateral MCA territory on DWI were analyzed. All patients got magnetic resonance imaging (MRI) and angiography (MRA) within 24 hours after onset, and 7 patients also had digital subtraction angiography (DSA). The patients were classified into single lesion group or scattered lesions group according to the DWI findings. The incidence of stenosis or occlusion of ipsolateral MCA, intracranial and extracranial ICA were compared between the two groups. Results 42 patients had scattered lesions and 31 patients had single lesion. The scattered-lesions group had a high incidence of ipsilateral extracranial ICA or MCA occlusion or severe stenosis ( 25.6%versus 0, x2 = 10.6, P = 0.001 ) and a high incidence of ipsilateral intracranial ICA or MCA moderate or mild stenosis (31.0% versus 9.7% ,x2 =4.717, P =0.03 ). A positive correlation was found between the scattered lesions and severe or multifocal stenosis of ipsilateral ICA and MCA ( OR: 13.7, 95% CI: 3.6 to 52.5). There was a low incidence of absence of extra- and intracranial stenosis on MRA or DSA in the scattered-lesions group ( 11.9% versus 32.3%, x2= 4.526, P = 0.033 ). A negative correlation was found between the scattered lesions and absence of large-artery stenosis ( OR: 0.284, 95% CI: 0.09 to 0.94).Conclusions ( 1 ) Patients with acute cerebral infarction and scattered lesions on DWI were more likely to suffer from stenosis or occlusion of ICA or MCA, especially over the extracranial ICA. (2) Patients with single lesion were less likely to have severe or multiple stenosis of MCA and ICA, indicating the relevance of small-vessel pathogenesis.

Objective To investigate the protective mechanism of trace element-selenium(Se) on the myocardium by against free radical superoxidation during myocardial ischemia/reperfusion(I/R) period in patients undergoing pulmonary bypass.Methods 46 patients with ASD or VSD were divided into control(23 patients) and Se administration(23 patients) groups.Before heart surgery,patients in the Se group orally received 400?g Se supplementation per day,for 7 days.The blood and myocardium samples were collected before clamping and after release respectively during pulmonary bypass.Biochemistry,electron spin resonance spectrometer(ESR),atomic absorption,RT-PCR and cDNA sequencing,were used to measure Se concentrations,MDA level,glutathione peroxidase(GPX) activity,GPX mRNA expression and GPX cDNA sequence in blood and myocardium.Results In Se group,before clamping,Se contents in blood cells and plasma, were not changed,but at reperfusion 30 minute,myocardial Se concentrations was evidently increased by about 1.1-fold,contrary to reduction 41% in blood cells.In Se administration group,at myocardial reperfusion 10 minute,free radical levels of the coronary sinus blood were significantly decreased 28%.No significantly changes in myocardial GPX activity or MDA level were found till reperfusion 30 minute.However,Se group had a higher myocardial GPX mRNA expression than that in controls in both before ischemia and after reperfusion.Conclusions Se supplementation can enhance Se content,expression of GPX-mRNA in myocardial tissue,and protect myocardium from radical lipooxidation damage.
3.Total flavonoids from arachniodes exilispromotes osteogenic differentiation of hUCMSCs

Aim To study the role of the total flavonoids from arachniodes exilis(TFAE) in osteogenic differentiation of human umbilical cord mesenchymal stem cells(hUCMSCs).Methods hUCMSCs were isolated and cultured by tissue explants adherent method, hUCMSCs at passage 3 were used to do the experiment, hUCMSCs were treated by different concentrations of TFAE, and cellular viability was measured by CCK 8 assay;alkaline phosphatase(ALP) activity was tested by AMP method;calcium nodule formation was detected by alizarin red staining;expression level of osteogenesis-related gene type I collagen enzyme a1(collagen type Ia1, Col1a1), osteopontin(OPN), Runx2, Osterix(Osx) mRNA was measured by RT-PCR;expression level of Col1a1,OPN protein was measured by Western blot.Results Certain concentration of TFAE(1 mg·L-1, 5 mg·L-1) promoted cellular proliferation, calcium nodules were more, ALP activity was enhanced;expression level of osteogenesis-related gene Col1a1, OPN, Runx2, OsxmRNA and Col1a1, OPN protein was up-regulated.Conculsion A certain concentration of TFAE promotes proliferation and osteogenic differentiation of hUCMSCs.
4.Inhibitory effect of silymarin on human osteosarcoma Saos-2 cells and its mechanism

Aim To study the effect of silymarin on proliferation and apoptosis of human osteosarcoma saos-2 cells and to explore its possible mechanisms .Meth-ods Control group and different concentration groups of silymarin were set , and Saos-2 cells were treated with silymarin .Celluar morphologic changes were ob-served by inverted phase contrast microscope .Prolifer-ation of cells was tested CCK-8 assay .Apoptosis rate of cells was analyzed by flow cytometry . ERK1/2、p-ERK1/2 and cleaved caspase-3 protein expression were measured by Western blot .Results Silymarin inhibi-ted the proliferation of Saos-2 cells in a time-dependent and concentration-dependent manner . Silymarin in-duced the apoptosis of Saos-2 cells in a concentration-dependent manner . Silymarin decreased p-ERK1/2 protein expression and increased cleaved caspase-3 protein expression in a concentration-dependent man-ner, while ERK1/2 protein expression had no obvious change.Conculsions Silymarin can inhibit the prolif-eration of human osteosarcoma saos-2 cells and induce apoptosis of them , and the mechanism may be related to inhibition of ERK signaling pathway and upregulated caspase-3 protein expression .
5.The Quantitative Analysis of Trace Germanium in 18 Kinds of Chinese Medicinal Herbs

By digestion with concentration sulfate acid and fuming nitri c acid under refluxing coprecipitation with Fe(OH)3, colorimetry with phenylfluorone-cetyl trimethyl ammonium bromide (CTMAB), germanium (Ge) contents of 18 kinds of Chinese medicinal herbs were determined. The Ge contents in Ganoderma lucidum and oldenladia diffusa were higher than those of others (2.46 and 0.35 ?10-8, respectively).In this method, the recoveries of Ge added to various herbs were in the range of 91%~93%. The detection limit and coefficient variation were 198 ng/g and 0.89%, respectively.
6.Experimental Study on the Role of CGRP in the Pathogenesis of Neonatal Necrotizing Enterocolitis

Objective To investigate the pathogenesis of neonatal necrotizing enterocolitis (NEC), to explore the laboratory basis for endogenous protective substance. Method Experimental models of both ischemia/reperfusion (I/R) group and ischemic preconditioning (IPC) group were set up using healthy male Wistar neonatal rats. Serum CGRP concentration, activity of LDH, the contents of MDA and the ratio of wet weight/dry weight (WW/DW) were measured respectively. Pathological sections of small intestines were prepared and observed under microscope. 7.Effects of oral administration of ?-blockers on the level of serum CK-MB in patients with coronary heart disease after intervention treamtent

Objective To study the protective function of ?-blockers to injury myocardial cell after PCI.Methods 60 cases were divided into two groups randomly,group A(30 cases) was control group,group B(30 cases) took ?-blockers orally before PCI.Serum levels of CK-MB were measured in 60 cases undergoigg coronary interventions in preprocedure,1st hour and 6th hour after operation,we judged the effect of ?-blockers.Results No difference was found in proprecedural level of CK-MB of two groups.There was significant difference between group A and group B 1h and 6h after operation(P=0.012 and 0.002).In the control group,we saw that the levels of postoperative CK-MB were higher than preprocedure (P=0.033 and 0.022).In hospital MACE did not occur in both groups.Conclution ?-blockers decrease the level of CK-MB in 30 cases undergoing coronary interventions.Thus it can decrease the mortality.
8.Relationship between Leukoaraiosis and Blood Lipid Levels

Objective To investigate the relationship between blood lipids levels and leukoaraiosis (LA). Methods 322 cases were designed into non-LA group (n=141) and LA group (n=181) according to MRI. LA group was further designed into mild (Fazekas Ⅰ, n=57),moderate (Fazekas Ⅱ, n=52), and severe (Fazekas Ⅲ, n=72) groups according to Fazekas classification. Their blood lipids levels were compared.Results The level of total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) increased (P<0.05) with the level of LA,while the level of triglycerides (TG) increased in all the LA groups (P<0.05), but no significant difference among them (P>0.05). There was no significant difference among all the groups in high-density lipoprotein cholesterol (HDL-C) (P>0.05). Conclusion TC, TG, and LDL-Cincrease in those with LA, and the TC and LDL-C increase with the severity of LA. The HDL-C do not relate with LA.
9.Rapid Identification of Sanjiu Weitai Granule and Quantitative Analysis of Moisture by Near-infrared Spec-troscopy

OBJECTIVE:To establish a method for the rapid identification of Sanjiu weitai granule and quantitative analysis of moisture. METHODS:Near-infrared (NIR) spectroscopy was adopted. 38 batches of Sanjiu weitai granule were collected,NIR spectrum was determined by integrating sphere diffuse-reflectance. Conformity test model was performed by comparing consistency index(CI)value and CI limit,and quantitative model of moisture was established using partial least squares(PLS)algorithm. RE-SULTS:When CI value was less than 6,the established model can accurately distinguish Sanjiu weitai granule,the model was proven feasible;the correlation coefficient (r2) of quantitative model was 97.44,the root mean square error of cross validation (RMSECV) was 0.453,the root mean square error of prediction (RMSEP) was 0.748. CONCLUSIONS:The method is simple and rapid without destroying,which can be applied to fast screening of Sanjiu weitai granule in site and fast determination of mois-ture content.
10.Curative Efficacy of Metformin Combined with Jinlida Granules in Treatment of Gestational Diabetes Mellitus and Its Effects on Serum VEGF, APN and Hcy Levels

Objective:To study the curative efficacy of metformin combined with Jinlida granules in the treatment of gestational diabetes mellitus and its effects on the serum vascular endothelial growth factor (VEGF),adiponectin (APN) and homocysteine(Hcy) levels.Methods:94 patients of gestational diabetes mellitus who were treated from July 2014 to July 2016 in our hospital were selected.According to random number table,those patients were divided into the observation group (n=47) and the control group (n=47).On the basis of routine treatment,such as control diet,reasonable exercise and healthy diet,etc,the control group was treated with metformin,while the observation group was combined with Jinlida granules on the basis of the control group.The changes of blood glucose,blood lipid and serum VEGF,APN and Hcy before and after treatment were compared between the two groups,the incidence of maternal complications and neonatal adverse outcomes were compared.Results:Compared with before treatment,the blood glucose,blood lipid of both groups after treatment were significantly improved (P <0.05),the fasting plasma glucose (FBG),postprandial 2h blood glucose (2hPG),glycosylated hemoglobin (HbAlc),total cholesterol (TC),triacylglycerol (TG),low density lipoprotein cholesterol(LDL-C) of observation group were significantly lower than those of the control group,the serum high density lipoprotein cholesterol (HDL-C) level was significantly higher than that of the control group(P<0.05);after treatment,the serum VEGF,APN and Hcy levels were significantly improved than those before treatment in both groups (P<0.05),and the serum VEGF,and Hcy levels of observation group were lower than those of the control group,the serum APN level was higher than that of the control group (P < 0.05);the incidence of gestational hypertension,hydramnios,cesarean section and premature delivery of observation group was significantly lower than that of the control group (P <0.05);the incidence of giant child,neonatal Jaundice and neonatal respiratory distress in the observation group was significantly lower than that of the control group (P<0.05).Conclusion:Metformin combined with Jinlida granules was effective for the gestational diabetes mellitus,which could effectively control the blood glucose,blood lipid levels and might be related to the regulation of serum VEGF,APN and Hey levels.
