3.The Neuropsychological Analysis of Agraphia After Basal Ganglia Infarction

Objective:To investigate the neuropsychological characteristics of Chinese agraphia caused by basal ganglia(BG)infarction.Methods:The writing abilities of 40 patients with BG infarction were detected by Chinese agraphia battery(CAB),and all the writing scores and agraphia quotient were calculated.The head CT/MRI images in agraphia and non-agraphia groups were standardized,the infarction were revealed and the superposition of two-dimensional arrays were performed,so that the central tendency of infarction was visually displayed.Results: Among the 40 patients,21 had left BG infraction,and 17 had agraphia;19 had right BG infraction,and 4 had agraphia.The two-dimensional superimposing neuroimages showed that BG infarctions caused agraphia was mostly in the left BG,including the left putamen,the head and body of the caudate nucleus,but there were fewer infarctions in the right putamen and the body of the candate nucleus.BG infarction caused agraphia was mostly aphasic agraphia,which was characterized by the orthographic disorders,paragraphia,and grammar mistakes.Conclusions: BG infarction may result in aphasic agraphia,which suggested that BG is involed in writing and processing,and it is the subcortical center of this advanced neurofunction in writing.
4.Construction, Expression and Evaluation of the System Reporting Streptococcus pneumoniae Virulent Gene by GFP in vivo

Streptococcus pneumoniae(S.pn) is an opportunity pathogenic bacteria,environmental factors play a key role in the pathogenicity of S.pn. It is important to study virulent gene in vivo. The S.pn suicide plasmid containing gfp reporter was constructed by fusing the genes pneumolysin and gfp,in which gfp is an excellent molecule probe in vivo. The plasmid was integrated to No.22 S.pn by homologous recombination. The recombinant S.pn was gained and evaluated in aspects of fluorescence excitation, biological character and physio-activity. The results showed it is efficient and available to report the expression of virulent genes in vivo and in vitro, which will provide a new easy method for evaluating and screening the virulent genes of S.pn in vivo.
5. Mechanism and experimental verification of antioxidant action of Heyan Kuntai Capsule based on systematic pharmacology

Objective: To explore and verify the potential antioxidation mechanism of traditional Chinese medicine Heyan Kuntai Capsule (HKC). Methods: Gene targets of 21 ingredients of HKC were searched for through Traditional Chinese Medicine Systems Pharmacology (TCMSP) database and analysis platform, followed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis with Database for Annotation, Visualization and Integrated Discovery (DAVID) tools. Further, the anti-oxidative function of HKC was validated in an in vitro H2O2-induced oxidative stress model in MII oocytes. Results: A total of 151 genes were obtained as targets to 21 components of HKC, whose GO were further significantly enriched in mitochondrion, superoxide dismutase activity and energy pathways, while KEGG showed the enrichment in PI3K/AKT, mTOR and ErbB1 signaling pathways. Afterwards, it was confirmed that HKC increased ATP production in H2O2-treated MII oocytes by improving mitochondrial function and reduced the dissipation of mitochondrial membrane potential (ΔΨm) to exert the antioxidation effect. Conclusion:s HKC exerts anti-oxidative effect by improving mitochondrial function.

7.Brain microvascular basement membrane damage and the expression of uPA and uPA mRNA in brain areas after focal cerebral ischemia/reperfusion in renovascular hypertensive stroke-prone rats

AIM: To explore the structural changes of brain microvasculature and mechanism in microvascular lesion after focal cerebral ischemia with reperfusion. METHODS: Using the techniques of immunohistochemical staining, in situ hybridization,optical microscopy and transmission electron microscopy, the expression of uPA, uPA mRNA, and changes in miocrovascular structure were examined in ischemic focus and perifocal areas after focal cerebral ischemia 2 hours with various time points of reperfusion in stroke-prone renovascular hypertensive rats (RHRSP). RESULTS: The brain edema and hemorrhage were severe 12 hours to 3 days after reperfusion. Ultrastructural change showed that the damage characterizations of the basement membrane were degradation, defection, and exfoliated of basement membrane, while uPA, which attack the basememt membrane around cerebral capillaries and extra-cellular matrix, and uPA mRNA expression increased significantly in ischemic and perifocal areas 12 hour to 3 day after reperfusion. CONCLUSION: The main pathologic mechanism of brain edema and hemorrhage after cerebral ischemia with reperfusion may result from the basement membrane lesion of brain microvasculature. The increase in the expression of uPA in reperfusion area may be the main cause of the basement membrane lesion .
8.A Toxicological Study of Polyunsaturated Fatty Acids and Its Application and Research

Objective To provide the toxicity basis for safety evaluation of Polyunsaturated Fatty Acids.Methods Acute toxicity test,micronucleus test of born marrow in mice,Ames test,sperm shape abnormality and thirty-day feeding test in rats were conducted in this study.Results The oral LD50 of Polyunsaturated Fatty Acids in both mice and rats were more than 20.0g/kg.BW,so the Polyunsaturated Fatty Acids was classed as actual non-toxicity.The results of micronucleus test of born marrow in mice,Ames test,sperm shape abnormality and thirty-day feeding test in rats were negative.The thirty-day feeding test in rats demonstrated that it had no obvious toxic effects on routine blood,body weight,and biochemical index.No obvious adverse effects dose of this sample was 6.66g/kg.BW.Conclusion Polyunsaturated Fatty Acids toxicity test results showed no obvious toxicity and was safe for edibility.
9.Nasal Colonization by Opportunistic Pathogens among Health Care Workers:A Survey

OBJECTIVE To investigate the prevalence of nasal colonization among health care workers(HCWs).METHODS Nasal swabs from 93 ICU workers and 98 other clinical workers were cultured and isolated and the tests of antibiotic susceptibility were performed by using paper diffusion method.RESULTS In total,214 isolates of 8 species from 191 health care workers were recovered,of which 187 isolates were coagulase-negative Staphylococcus(CNS)(the carriage rate of 93.71%) and 8 isolates were Staphylococcus aureus(the carriage rate of 4.19%).While the total Gram-negative bacteria carriage rate was 14.14%(27 isolates).The most frequent CNS species were S.epidermidis and S.haemolyticus.The antibiotic susceptibility profiles of S.aureus and CNS differed sharply: all 8 S.aureus strains were resistant to penicillin but were fully susceptible to oxacillin,in contrast,most of CNS were resistant to both penicillin and oxacillin.The carriage rate of CNS(60.2%)and Gram-negative bacteria(26.9%)in HCWs of ICU were higher than other HCWs(P

Objective: To investigate the effects of tea polysaccharide (TP) on blood glucose and diabetic symptoms in mice. Methods: Alloxan-induced diabetic mice were divided into 4 groups, and administered orally with saline, TP at doses of 1.0, 2.0, 4.0 g/kg body weight, respectively for 4 weeks. Water intake and body weight were recorded daily and weekly respectively.Fasting blood samples were collected for measurement of glucose at the end of 2nd and 4th week.Results: Symptoms of diabetic mice fed TP were improved obviously. Their blood glucose concentration and water intake decreased significantly. On the other hand, their body weight increased obviously. There was positive correlation between blood glucose and water intake, but negative correlation between water intake and body weight. Conclusion: TP can decrease blood glucose concentration and improve diabetic symptoms in mice.
