1.Ozone injection combined with radiofrequency ablation for the treatment of far lateral lumbar disc herniation:observation of the curative effect

Objective To investigate the clinical efficacy of ozone injection combined with radiofrequency ablation (RFA) for the treatment of far lateral lumbar disc herniation (FLLDH). Methods A total of 60 patients with FLLDH, who were admitted to authors’ hospital during the period from March 2013 to March 2014, were randomly and equally divided into the study group (n=30) and the control group (n=30). Simple ozone injection treatment was employed for the patients of the control group, while ozone injection combined with radiofrequency ablation was adopted for the patients of the study group. The same other adjunctive medication was used in both groups. All the patients were followed up at one week as well as at one, 3 and 6 months after the treatment. The clinical effect and the complications of both g roups were recorded. Results Successful operation was obtained in all 60 patients, no perioperative complications occurred, and the technical success rate was 100%. The excellent clinical results evaluated at one week, and one, 3 and 6 months after the treatment in the control group and in the study group were 73.3%, 76.7%, 70%, 60% and 70%, 76.7%, 83.3%, 90% respectively. The 3-month and 6-month clinical results of the study group were significantly better than those of the control group. Conclusion For the treatment of FLLDH, ozone injection combined with radiofrequency ablation is safe and effective. This technique should be recommended in clinical practice.

3.Changes in circulating microparticles in mice with ventilator-induced lung injury

Objective To investigate the changes in serum level of microparticles (EMPs) in mice with ventilator-induced lung injury (VILI), and explore its significance in VILI. Methods Forty-eight grade SPF male C57BL/6J mice were randomly divided into two groups, with 24 mice in each group: the mice in mechanical ventilation (MV) group were given high tidal volume (VT 30 mL/kg) MV for 4 hours after tracheal intubation, and those in spontaneous breathing group were spontaneously breathed for 4 hours. The apical blood of 12 mice in each group were collected, and serum levels of interleukins (IL-1β, IL-6) and tumor necrosis factor-α (TNF-α) were determined by enzyme linked immunosorbent assay (ELISA), and serum EMPs levels were determined by flow cytometer. The correlations between EMPs and IL-1β, IL-6, and TNF-α were analyzed by linear regression analysis. The lung tissues of other 12 mice in each group were harvested, and wet/dry weight (W/D) ratio was assessed. After hematoxylin-eosin (HE) staining, the morphological changes in lung tissue were observed under light microscope. After double staining of uranium acetate and lead citrate, the ultrastructural changes in lung tissue were observed with electron microscope. Results Compared with spontaneous breathing group, the levels of lung W/D ratio in MV group was significantly increased (5.47±0.14 vs. 4.34±0.11), the levels of IL-1β, IL-6, TNF-α and EMPs were also significantly increased [IL-1β (ng/L): 42.4±4.4 vs. 7.7±3.6, IL-6 (ng/L): 1 239.5±66.3 vs. 21.7±4.6, TNF-α (ng/L):237.6±25.8 vs. 37.1±19.1, EMPs (cells/μL): 28.6±1.8 vs. 5.9±1.8, all P < 0.01]. It was shown by correlation analysis that EMPs were positively related with IL-1β, IL-6, and TNF-α (r value was 0.968, 0.932, 0.945, respectively, all P = 0.000). It was shown by fitting linear regression analysis that when EMPs increased by 1 cell/μL, IL-1β increased by 2.4 ng/L [95% confidence interval (95%CI) = 1.9-2.8, P < 0.001], IL-6 increased by 34.5 ng/L (95%CI = 25.1-44.0, P < 0.001), and TNF-α increased by 13.6 ng/L (95%CI = 10.3-16.9,P < 0.001). It was shown by light microscope that the structure of lung tissue and alveolar of mice in spontaneous breathing group appeared normal, while the shrinks of alveolar and disappearance of alveolar architecture were found in MV group. It was shown by electron microscopy that alveolar wall edema and thickening and broken alveolar septa were found in MV group, by contrast, the structure of alveolar was normal in spontaneous breathing group. Conclusion 30 mL/kg VT ventilation for 4 hours could induce VILI with increase in EMPs, suggesting EMPs closely related to VILI, and EMPs level may be putative biomarker of VILI.
4.Adventitious root induction and in vitro culture of Panax notoginseng.

OBJECTIVETo investigate the induction and culture of adventitious root of Panax notoginseng.
METHODThree ways, induction from the explants of three-year-old P. notoginseng. The explants of regenerated shoots and calluses, were used to induce adventitious roots. The effects of 2, 4-dichlorophenoxyacetic acid, indole-3-butyric acid and naphthylacetic acid on adventitious root induction were investigated respectively. The effects of four modes of separating adventitious roots from the parent tissues on culture in vitro were compared.
RESULTAdventitious roots were successfully induced by three methods, of which the young flower bud callus was the best material for the induction of adventitious root. Indole-3-butyric acid possessed the strongest potency for induction. The liquid culture system was established by continuous culture of adventitious roots together with their parent tissues before separated.
CONCLUSIONThe acquisition and culture in vitro in liquid culture system of adventitious roots of P. notoginseng lay a foundation for the next investigation.

6.Predictive value of serum uric acid on type 2 diabetes mellitus

Objective To evaluate the predictive value of uric acid (UA) on type 2 diabetes mellitus (DM).Methods 711 subjects aged 45-60 years old with normal fasting blood glucose (FBG) were studied in 1992.The subjects were divided into 2 groups according to their 1992 UA data in the normal UA group and in the hyperuricemia (HUA) group.We analyzed the prevalence of DM-2 in 2007 according to data on UA in 1992.Relative risk (RR) of the cohort was calculated under x2 test and logistic regression analysis.Results FBG and the prevalence rate of DM in 2007 in the HUA group were statistically higher than those in the normal group.The relative risk (RR) of HUA to DM was 3.749 (P=0.000),with 95% Confidence interval (CI) as 2.387-5.890.Data from the logistic regression analysis,after adjusting for other risk factors,the RR of UA to DM was 1.426 (P=0.003),with 95% CI as 1.173-1.705.Conclusion The abnormal UA was closely related to glucose metabolism disorder while hyperuricemia appeared to be associated with increased risk of type 2 DM.
7.5-HT2C receptor subtype modulate production of secretory beta-amyloid protein precursor in incubated rat hippocampal slices.

AIMTo determine whether serotonin, a major neurotransmitter in brain, can modulate the production of secretory beta-amyloid protein precursor (sAPP) by activation of serotonin 5-HT2C receptor.
METHODSThe hippocampal slices of rats were incubated with various concentrations of serotonin, M-110, or L-107. sAPP released into the incubation medium were assayed by Western blot analysis assay with monoclonal antibody 22C11 for 2 h.
RESULTSVarious concentrations of serotonin (1.0 x 10(-2) - 1.0 x 10(3) micromol x L(-1)), M-110, a serotonin 5-HT2C agonist (1.5 x 10(-6) - 1.5 x 10(3) micromol x L(-1)), showed positive effect on the production of sAPP while L-107, a serotonin 5-HT2C antagonist (1.0 x 10(-9) - 1.0 x 10(3) micromol x L(-1)), showed negative effect on the production of sAPP over controls.
CONCLUSIONSerotonin modulates production of secretory amyloid beta-protein precursor through serotonin 5-HT2C receptor in incubated rat hippocampal slices.

8.Research progress in medicinal plant cell suspension culture.

China consumes and exports traditional Chinese medicinal resources the most in the world. However, we cannot anchor our hope on field production of traditional Chinese medicinal materials and their active ingredients, due to limited land resources. Therefore, the development of biotechnology is of great importance for China to solve the problem of traditional Chinese medicinal resources. Plant cell culture is an important approach for the sustainable development of precious medicinal resources. This essary summarizes the optimization of conditions for medicinal plant cell culture, the regulation of secondary metabolic pathways and cell bioreactor culture, and realizes that the authentic commercial production of more medicinal plants requires efforts from all aspects.

9.The influence of Astragalus Membranaceus on Calcium and Phosphorus Metabolism and FGF2 3-Klotho Axis in Rats with Chronic Kidney Disease

OBJECTIVE To observe the expression change of FGF23-Klotho axis on calcium and phosphorus metabolism in rats with chronic kidney disease,and the influence of Astragalus membranaceus on them.METHODS 30 SD rats were ran-domly divided into three groups(control group,model group and AM group),after adaptive feeding for 1 week,250 mg/kg ade-nine suspension by gastrogavage and 1.2% phosphorus content by dietary were executed to molding for 4 weeks.Then,ade-nine gavage was given once in 2d in the following 2 weeks;at the same time,the rats in AM group were given AM decoction 1 2.5 g/(kg·d)gavage and control group & model group were given normal saline equivalent.Before dosing,selected 3 rats randomly in each group to detect the level of blood urea nitrogen(BUN),creatinine(Scr),calcium(Ca),phosphorus(P)via heart puncture blood.Meanwhile,the parathyroid hormone(PTH),FGF23,Klotho and 25(OH)D3were detected by ELISA.After 6 weeks,the histological changes in renal were observed by HE staining,the level of BUN,Scr,Ca,P in serum were detected via abdominal aortic blood.Meanwhile,PTH,FGF23,Klotho and 25(OH)D3were detected by ELISA.RESULTS Compared with the control group,the qualities of kidney and theBUN、Scr、Ca、P levels significantly increased in model group(P<0.01,P<0. 05),while the PTH level not markedly elevated.Compared with the model group,the qualities of kidney and theBUN、Scr、Ca、P levels were decreased in AM group,while the PTH level not markedly elevated,and the BUN、Ca、P levels were statistical difference(P<0.01,P<0.05).HE staining showed that the number of normal glomerular improved,compared with the model group,the expansion degree of renal tubular decreased,the number of inflammatory cells decreased,and the crystallization of adenine decreased in the BHD group.Compared with the control group,the FGF23 level increased in model group,while the Klotho and 25(OH)D3decreased.Compared with the model group,the FGF23 levelnot markedly elevated(P<0.05),and the Klotho not markedly declined(P<0.05).CONCLUSION Astragalus membranaceus could alleviate the disorder of calcium and phosphorus metabolism in CKD rats,inhibit the high expression of FGF23,and up-regulate the expression of Klotho,thus improving histology of kidney,kidney function,and the balance metabolism of Ca and P in CKD rats.
10.Effect of ablation of complex fractionated atrial electrogram on vagal modulation in dogs.

BACKGROUNDClinical observations have shown that the complex fractionated atrial electrogram (CFAE) associates with ganglionated plexus activity in the cardiac autonomic nervous system. This study aimed to investigate the impact of CFAE ablation on vagal modulation to atria and vulnerability to develop atrial fibrillation (AF).
METHODSTen adult mongrel dogs were involved. Cervical sympathovagal trunks were decentralized and sympathetic effects were blocked. CFAE was color tagged on the atrial 3-dimensional image and ablated during AF induced by S1S2 programmed stimulation plus sympathovagal trunk stimulation. Atrial effective refractory period (ERP) and vulnerability window (VW) of AF were measured on baseline and at vagal stimulation at 4 atrium sites. Serial tissue sections from ablative and control specimens received hematoxylin and eosin staining for microscopic examination.
RESULTSMost CFAE areas were localized at the right superior pulmonary quadrant, distal coronary sinus (CS(d)) quadrant, and proximal coronary sinus (CS(p)) quadrant (21.74%, separately). Sinus rhythm cycle length (SCL) shortening did not decrease significantly after ablation at the sites, including right atrial appendage, left atrial appendage, CS(d), and CS(p) (P > 0.05). ERP shortening during vagal stimulation significantly decreased after ablation (P < 0.01); the VW to vagal stimulation significantly decreased after ablation (P < 0.05). The architecture of individual ganglia altered after ablation.
CONCLUSIONSCFAE has an autonomic basis in dogs. The decreased SCL and ERP shortening to vagal stimulation after CFAE ablation demonstrate that CFAE ablation attenuates vagal modulation to the atria, thereby suppressing AF mediated by enhanced vagal activity. CFAE ablation could suppress AF mediated by enhanced vagal activity.
