1.Expression of E-cadherin and nm23 in papillary thyroid carcinoma and its clinical significance

Purpose:To investigate the expression of E cadherin and nm23 in papillary thyroid carcinoma (PTC) and its clinical significance. Methods:The expression of E cadherin and nm23 were detected by SP immunohistochemical technique in 70 paraffin embedded tissue specimens of PTC.Results:The positive rates of E cadherin and nm23 were 44.3%(31/70) and 62.9%(44/70) respectively. There were close relations between the expression of E cadherin and nm23 and pathological grade, TNM stage and lymph node metastasis, and no difference could be found between the expression of E cadherin and nm23 and size of tumor, sex and age of patients. There was distinct correlation between the expression of E cadherin and nm23. The metastatic rate in the group, which the expression of E cadherin and nm23 were both positive, was 12%(3/25), but in the all negative group, the metastatic rate was 75%(15/20). 2.Study on effect of scutellarin in resisting liver fibrosis in rats.

Totally 80 rats were randomly divided into the control group, the model group, low, middle and high dose (25, 50, 100 mg x kg(-1)) scutellarin( SC) groups and the colchicine ( Col) group. Apart from the blank group, all of the remaining groups were intraperitoneally injected with 0.5 mL pig serum twice every week for consecutively 13 weeks and orally administered with the corresponding drugs since the 9th week. The blank group and the model group were orally given equal volume of normal saline once every for consecutively four weeks. After the experiment, efforts were made to detect the contents of alanine aminotransferase (ALT), aspertate aminotransferase (AST), albumin (ALB), total protein (TP), total bilirubin (TBIL), hyaluronic acid (HA), laminin (LN) and collagen type IV (CIV), collect liver tissues of fixed positions, observe the pathological changes through hematoxylin-eosin (HE) staining, conduct the pathological grading for liver fibrosis, determine the expressions of hepatic collagen type I and III (C I, C III) and calculate their color rendering index. Compared with the model group, low, middle and high dose (25, 50, 100 mg x kg(-1)) SC groups could decrease the contents of ALT, AST, TBIL, HA, LN, CIV, increase the contents of ALB, TP in serum and reduce the contents of C I, C III in liver tissues. In conclusion, scutellarin has a certain therapeutic effect on immune liver fibrosis in rats induced by pig serum.

5.Effects of Naoshuning on neurological outcome following traumatic brain injury in rats

ObjectiveTo observe the protective effects of Chinese medicine Naoshuning on neurological deficits following traumatic brain injury (TBI) in rats.MethodsCerebral contusion was produced by dropping weight,the beam-walking task was employed to observe neurological functional changes,water content in the brain tissue and neuropathological changes were also evaluated.ResultsWithin one week the ability to process in beam-walking task was significantly lessened,the outcome of beam-walking task of Naoshuning group was significantly better than that of the model group.Water content in the brain tissue and neuropathological changes were also alleviated in Naoshuning group compared with that in model group.ConclusionEarly alleviating brain edema,protecting brain tissue may be one of the mechanism of Chinese medicine to ameliorate the behavior deficits following TBI in rats.
6.Effect of aquaporin 4 on traumatic brain edema

ObjectiveTo investigate the dynamic change of cerebral aquaporin 4 (AQP4) expression level during traumatic brain injury (TBI) in rats and the relationship between AQP4 expression and traumatic brain injury and edema. MethodsThe experimental TBI model was established by bumpiness of free falling body according to Feeney's. The changes of brain water content (BWC) and permeability of blood-brain barrier (BBB) were measured by the methods of wet and dry weight and Evans blue fluorometry. Immunohistochemistry was used to determine the change of expression of AQP4 during different periods after TBI. Results6 h after TBI, water content of brain tissue and EB content in the contusion side significantly increased, and the protein expression of AQP4 in contusion marginal region began to increase. They reached summit 24 h after operation and did not come back normal level 5 d after operation. There was significantly positive correlation between protein expression of AQP4 and change of EB content (r=0.957486,P<0.05). ConclusionThe dynamic change of cerebral AQP4 expression level closely related to the destruction of BBB and the formation of cerebral edema during traumatic brain injury.
7.The roles of traditional Chinese medicine in gene therapy.

The field of gene therapy has been increasingly studied in the last four decades, and its clinical application has become a reality in the last 15 years. Traditional Chinese medicine (TCM), an important component of complementary and alternative medicine, has evolved over thousands of years with its own unique system of theories, diagnostics and therapies. TCM is well-known for its various roles in preventing and treating infectious and chronic diseases, and its usage in other modern clinical practice. However, whether TCM can be applied alongside gene therapy is a topic that has not been systematically examined. Here we provide an overview of TCM theories in relation to gene therapy. We believe that TCM theories are congruent with some principles of gene therapy. TCM-derived drugs may also act as gene therapy vehicles, therapeutic genes, synergistic therapeutic treatments, and as co-administrated drugs to reduce side effects. We also discuss in this review some possible approaches to combine TCM and gene therapy.

9.Comparation of dezocine and morphine for postoperative analgesia combined with Flurbiprofen after gynecological surgery

Objective To compare the efficiency and safety of dezocine and morphine combined with flurbiprofen for gynecologic postoperative analgesia.Methods Ninty patients for elective hysterectomy,ASA (American Society of Anesthesiologists) Ⅰ - Ⅱ,were randomly divided into three groups,given postoperative intravenous analgesic pump:dezocine 50 mg + flurbiprofen 100 mg( Group D) ;morphine 50 mg + flurbiprofen 100 mg ( Group M) ;dezocine 25 mg + morphine 25 mg + flurbiprofen 100 mg ( Group DM ).Anesthesia were induced by midazolam,etomidate,fentanyl and cisatracurium,maintained with propofol and remifentanil.At 15 -30 min before the end of surgery,flurbiprofen 50 mg was given,if pain was not relieved statsifically,morphine 5mg/per time was given additionally.VAS Pain relief scores,Ramsay sedation Score,usage of morphine and side effects such as nausea,vomiting and itch of skin were recorded at 4,8,24,48 h after surgery.Results At 4,8,24 and 48 h after surgery,VAS score at sedation was ( 2.27 ± 0.64 ),( 2.17 ± 0.65 ),( 1.97 ± 0.67 ),and ( 1.60 ± 0.56) in Group D,and ( 2.50 ± 0.63 ),( 2.40 ± 0.62),( 2.20 ± 0.61 ) and ( 1.87 ± 0.57 ) in Group DM at sedation,which were all significantly low than those of ( 3.10 ± 0.76),( 3.00 ± 0.74 ),( 2.80 ± 0.71 )and (2.40 ±0.72)in Group M.At 4,8,24 and 48 h after surgery,VAS score at active was (3.10 ±0.76),(2.97 ±0.76),(2.70 ±0.84) and (2.17 ±0.70)in Group D,(3.43 ±0.63),(3.30 ±0.65),(3.03 ±0.76)and (2.43 ± 0.68 )in Group DM,which were all significantly lower than those of (4.13 ± 0.94),(3.93 ±1.05),(3.60 ± 1.05 ) and ( 3.03 ± 0.96 ) in Group M ( Ps < 0.05).And the VAS scores of Group DM was significantly higher than those of Group D.Sedation score at 48 h after surgery in group D was better than that in Group M ( x2 =4.812,P < 0.05 ),The side effects at 48 h after surgery was 26.7% in Group D,46.7% in Group DM and 80% in Group M,and there significant difference among these 3 groups (P < 0.05 ).Conclusion Compared to Morphine and or Morphine combined with Dezocine,Dezocine 50 mg + flumazenil 100 mg group is better to release the postoperative pain,and withless adverse effects.
10.Application of pressure-controlled ventilation-volume guaranteed in obese patients with obstructive sleep apnea syndrome

Objective To explore the clinical effectiveness of pressure-controlled ventilation-volume guar-anteed(PCV-VG)in obese patients with obstructive sleep apnea syndrome(OSAS)during uvulopalatopharyngo-plasty. Methods 40 obese patients(BMI≥30 kg/m2)with OSAS scheduled for uvulopalatopharyngoplasty under general anesthesia were randomly divided into two groups of volume-controlled ventilation(group V,n = 20)and pressure-controlled ventilation-volume guaranteed(group P,n=20). The heart rate(HR),mean arterial pressure (MAP),arterial partial pressure of oxygen(PaO2)and arterial partial pressure of carbondioxide(PaCO2)were recorded before induction of anesthesia without oxygen inhalation(T0),30 min(T1)and 1 h(T2)after tracheal intubation,and 30 min after extubation(T3). The peak airway pressure(Ppeak),airway resistance(Raw),thoracic compliance (CL),oxygenation index (OI) and respiratory index (RI) were also calculated at T1 and T2 under observation of recovery. Results There were no obvious differences between the two groups of patients before anesthesia and after recovery. Compared with the group V ,PaCO2 ,PPEAK ,Raw at T1 ,T2 and RI at T1 ~ T3 of the group P decreased(P<0.05),while CL at T1,T2 and PaO2,OI at T1~T3 increased(P<0.05). There were no sig-nificant differences in HR ,MAP at the above time points. Conclusions Compared with volume-controlled venti-lation,PCV-VG can effectively enhance thoracic compliance,lower inspiratory pressure and airway resistance ,and decrease intrapulmonary shunt ,which is conductive to improve arterial oxygenation and gas exchange in obese patients with OSAS.
