Objective: To investigate the genes differently expressed in the livers of Kkay diabetic and normal mice, providing data to prevent human from diabetes and its chronic complications. Methods: cDNA microarray chips containing 8 192 cDNAs were used to investigate the gene expression pattern of the liver in Kkay mouse. Results: One hundred and fifty four genes were screened out, comprising 68 complete cDNAs and expressed sequence tags, among them 40 genes were up regulated and 114 genes were down regulated. Conclusion: The pathogenesis of type Ⅱ diabetes is complicated, including the disorder of the metabolism of carbohydrate, fat and protein, and many functional abnormalities of a number of vital proteins. Objective To investigate the feasibility of a small amount of water intake at the early stage after general anesthesia in children.Methods Five hundred and seventy children underwent operations under general anesthesia were randomly divided into 2 groups:control group ( n =288) and early postoperative drinking group ( n =282).The children received routine water deprivation after operation in control group.After recovery from anesthesia and recovery of coughing and swallowing reflexes,the children were allowed to drink a small amount of water in early postoperative drinking group.The incidences of crying,bucking,aspiration and hyoxemia were recorded before water intake and at 5 min after water intake.Results Compared with control group,the incidence of crying was significantly decreased and no significant change was found in the incidence of bucking in early postoperative drinking group.No patients exhibited aspiration and hyoxemia in the two groups.Conclusion It is feasible that children drink a small amount of water at the early stage after general anesthesia.

ObjectiveTo explore the effect of bone marrow mesenchymal stem cells transplantation on the deep partial thickness derma burn injury in guinea pigs.MethodsSuspension cells of MSCs were cultured in vitro and transplanted into wounds of the female guinea pig of the deep partial thickness derma burn injuring model with the concentration of 2×106cell/ml(group A),2×107cell/ ml(group B). The rate of wound healing was observed, factor Ⅷ was detected using immunohistochemistry. Y chromosome on the wound in guinea pig was detected using PCR 15,30,50 days after MSCs had been transplanted. ResultsThe rate of wound healing of groups A and B was better than that in the control group, but there was not significantly different between groups A and B. Microvessel density of wound in groups A and B was better than those in the control group on the 15th and 30th days. There was not significantly different between the group A and B. PCR results showed that Y chromosome gene was expressed on some recoveried wound of female guinea pig. ConclusionBone marrow mesenchymal stem cells transplantation is effective in deep partial thickness derma burn injury in guinea pigs.

Objective To investigate fracture resistance and fracture patterns of maxillary anterior endodontically treated teeth restored with two different shapes of glass fiber post systems.Methods Twenty-four human sound maxillary anterior teeth of similar size were collected,and randomly divided into 3 groups,8 teeth each.After endodontical treatment,they were given the following treatments:Group A:parallel glass fiber posts (coltene parapost fiber lux) and composite core;Group B:taper glass fiber posts (coltene parapost taper lux) and composite core; Group C (control group):intact endodontically treated teeth.Teeth in Groups A and B reserved 2 mm dentin ferrule.Then the teeth were prepared and restored with IPS e.max Press all ceramic crowns.All the teeth were embedded in acrylic resin 2 mm below the cementoenamel junction (CEJ),with the silicon impression material stimulating periodontal membrane.All the specimen were loaded under a universal testing machine,at the palatal junction of incisor third and middle third,with an angle of 135 degree to the longitudinal axis of the tooth until fracture occurred,at a cross-head speed of 1.0 mm/min.Fracture loads and patterns were recorded.Statistical analysis was performed using SPSS software.Results The mean fracture loads in the three groups were as follows:(468.8±142.5) N,(440.2±202.7) N,and (459.0±147.6) N,respectively.There was no significant difference in fracture loads among groups (P＞0.05).The incidence of unfavorable fracture in Group A was higher than Groups B and C.Conclusions There is no significant difference in fracture strength between parallel and taper fiber post groups.However,the group with parallel fiber posts demonstrates a higher risk of unfavorable fracture than the group with taper fiber posts,which indicates that taper fiber posts are more favorable to preserve the remaining tooth structure.

Objective To study the relationship between homocysteine (Hcy) and hypersensitive c-reactive protein (hs-CRP) level and mental behaviors of Alzheimer's disease (AD).Methods During the period from 2014 January to 2016 January,20 subjects were selected from inpatients of the third hospital of yunnan porvince,before and after treatment,the plasma levels of Hcy and hs-CRP in patients with AD were tested by enzymatic methods.The pathological behavior rating scale (concerns-AD) was used to estimate the treatment effect.Results There were significant differences in the mental behavior score and Hcy and CRP levels between the before and after the treatment in AD patients (P＜0.01) Conclusion Plasma levels of Hcy and CRP have correlation with mental behaviors of patients with AD.

The amplified VP2 gene (PPV strain SC-1) and PCV20RF2 gene were inserted into the eukaryotic expression vector pPI-2.EGFP.Then the recombinant plasmid named pPI-2.EGFP.VP2.ORF2 was obtained.Mediated by liposome,the recombinant plasmid was transfected into Veto cells and expressed.Using immunofluorescence assay,the fluorescence of expression products were first detected at 20 h after transfection and peaked 36 h.Under electronmicroscope,virus-like particles (VLPs) can be observed in the transfected cells.To confirm the obtained VLPs to be recombinant particles,piglets were immunized using purified VLPs.The dynamic variation of blood T lymphocytes and serum antibody level of PPV and PCV2 were measured.The results showed that the ratio of CD3+,CD4+ T lymphocyte in peripheral blood lymphocyte of immunized piglets raised in a certain degree,the number of CD8+ T lymphocytes fell at 7-14 d after immunization,and then raised.Relatively high level of PP-V,PCV2 specific antibody could be detected.This indicated that the expression of recombinant plasmid pPI-2.EGFP.VP2.ORF2 was successful,the virus-like particles were formed and showed favourable immunogenicity.

Objective To observe the expression of bcl-2 gene in cell apoptosis of neonatal rats followed by hypoxic-ischemic brain damage(HIBD) and investigate the mechanism of neuronal apoptosis after HIBD.Methods Fifty-four neonatal SD rats were used in 1 sham-operated group and 8 trial groups. The models of HIBD were established in neonatal rats by inhaling the mixed gases of 92 % N 2 and 8 % O 2, the animals were sacrificed by dislocation their heads at different time points(0.5,1,3,6,12,24,48,72 h), the hippocampus were dissected for morphological analysis. The neuronal apoptosis and the expression of bcl-2 gene in hippocampus were detected by the methods of immunohistochemistry. Results The apoptotic cells appeared at the time point of 3 h, and reached the peak at 48 h, then decreased. The positive cell of bcl-2 protein increased from the time point of 30 min and reached the peak at 6 h and then decreased gradually following HIBD. Conclusion The expression of bcl-2 gene plays a role in the process of neuronal apoptosis following HIBD.
