2.Toxicity of a Heavy Metal Removal Engineering Strain to Mice

Objective To test the toxicity of genetically engineered strain R32 released to the environment.Methods Acute,chronic and cumulative toxicity test on mice,micronucleus assay of mice bone marrow cell and mice sperm malformation test were conducted to assess the toxicity of strain R32.Results R32 did not cause acute,chronic,cumulative and genetical toxic effect on mice before it absorbed heavy metal chromium.Although the mice bone marrow micronucleus frequency increased and some cumulative toxic effect emerged after R32 absorbed chromium,it did not cause acute and chronic toxicity effect on mice and mice sperm malformation rate did not increase neither.Furthermore,whether R32 absorbed heavy metal chromium or not,the effect of its secretion was not significant in all experiments compared with the control.Conclusion R32 can not cause toxic effect on mice before it absorbed chromium.Whether R32 absorbed chromium or not,the effect of its secretion was not significant in all toxicological experiments conducted in the present paper.Large content of R32 may cause some cumulative toxic effect in mice after it absorbed chromium.
3.Study on Extraction Technology of Sun-screening Constituents from Radix Scutellaria

Objective:To optimize the extraction technology of radix scutellariae. Methods: The extraction of radix scutellariae was scanned by ultraviolet spectrophotometry from 200 to 400nm. The content of baicalin was determined by HPLC. The ultraviolet ab-sorption, baicalin content and extraction rate were used as the indices, and the optimal extraction conditions were investigated by single factor experiments and orthogonal design tests. Results: The optimal extraction conditions were as follows: the ethanol concentration was 60%, the solid-liquid ratio was 1∶40, ultrasound extraction time and temperature was 40 min and 60℃, respectively. Conclusion:The extraction of radix scutellariae has good sunscreen with promising ultraviolet absorption in UVB. Ultrasound extraction has high ex-traction yield with short time, which can be used to extract sun-screening constituents from radix scutellariae.
4.Improvement of Quality Standard for Pingyou Granules

To establish the quality standard for Pingyou granules. Methods: Paeoniae radix, Carthamus tinctorius, Coicis semen and Nutgrass galingale rhizome were qualitatively identified by TLC. The content of peoniflorin was determined by HPLC. The separation was performed on a Lichrospher-C18 (250 mm × 4. 6 mm, 5 μm) column with the mobile phase of methanol-acetic acid (24∶76). The detection wavelength was 232nm. Results:Paeoniae radix, Carthamus tinctorius, Coicis semen and Nutgrass galingale rhizome could be identified by TLC without any interference from the negative control. The linear range of peoniflorin was 6. 560-104. 920 μg·ml-1(r=0. 999 9) with the average recovery of 98. 77%(RSD=2. 73%,n=9). Conclusion:The qualitative identifi-cation is specific and reproducible, and the quantitative method is simple, accurate and reliable, which can be used in the quality con-trol of Pingyou granules.
5.Content Determination of Paeoniflorin and Icariin inQixue Shuangbu Oral Liquid by HPLC

Objective To improve the quality standard of Qixue Shuangbu Oral Liquid;To provide guarantee for the quality of oral liquids.Methods HPLC was used to simultaneously determine the contents of paeoniflorin and icariin in the oral liquid. The determination was performed on a Agiltnt TC-C18 (2) column (250 mm × 4.6 mm, 5μm) with the mobile phase of acetonitrile and 0.1% H3PO4 gradient mode. The flow rate was 1.0 mL/min;the column temperature was set at 25℃; the detection wavelength was 232 nm.Results The linear ranges of paeoniflorin and icariin were obtained between 0.542-8.677μg (r=0.999 9) and 0.185-2.963μg (r=0.999 9), with the average recoveries (n=6) of 99.29% (RSD=1.06%) and 98.91% (RSD=1.97%), respectively.Conclusion This method is accurate and feasible, which can be used conveniently in quality control ofQixue Shuangbu Oral Liquid.
6.Study on the Quality Standard for Naoxin Luotong Pills

Objective:To establish the quality standard for Naoxin Luotong pills. Methods:Astragali radix, bombyx batryticatus and scorpio were identified under a microscope. Peach kernel, astragali radix, paeoniae radix alba, rheum, paeonia suffruticosa and salvia miltiorrhiza were identified by TLC. The content of total ferulic acid was determined by HPLC with a Sapphire C18 column (250 mm × 4. 6 mm,5 μm). The mobile phase was acetonitrile-0. 1% phosphoric acid(12 ∶88)and the detection wavelength was 316 nm. The column temperature was 35℃ and the flow rate was 1. 0 ml·min-1 . Results:The microscopic characteristics of astragali radix, bombyx batryticatus and scorpio were obvious to be observed easily. The TLC spots of peach kernel, astragali radix, paeoniae radix al-ba, rheum, paeonia suffruticosa and salvia miltiorrhiza were clear and specific. Ferulic acid had a good linear relationship within the range of 5. 1-81. 6μg·ml-1(r=0. 999 9), and the average recovery was 103. 30% with RSD of 1. 96% (n= 9). Conclusion:The standard can be used for the quality control of Naoxin Luotong pills.
7.Risk factors for delayed gastric emptying after pancreaticoduodenectomy

Objective To analyze the related risk factors for delayed gastric emptying ( DGE) after pancreaticoduodenectomy .Methods Clinical data on 308 patients who underwent pancreaticoduodenectomy at Ningbo Lihuili hospital from January 2009 to December 2014 were retrospectively analyzed , and patients were divided into DGE group and non-DGE group.Univariate analysis and multivariate logistic regression analysis were used to study the risk factors associated with DGE during perioperative period .Results DGE occurred in 55 patients (17.9%).The incidences of grade A, grade B and grade C DGE were 7.1%(22/308), 6.2%(19/308) and 4.5%(14/308), respectively.The univariate analysis showed the method of pancreatic digestive tract reconstruction ( pancreaticogastrostomy or pancreaticojejunostomy ) , postoperative pancreatic fistula, postoperative biliary fistula and postoperative intraabdominal infection were risk factors for DGE after surgery. Multivariate analysis indicated that the method of pancreatic digestive tract reconstruction (OR=1.19, P=0.046), postoperative pancreatic fistula ( OR=1.33, P=0.014), postoperative biliary fistula (OR=1.43, P=0.047) and postoperative intraabdominal infection (OR=1.51, P=0.001) were independently associated with DGE . Postoperative pancreatic fistula (OR=3.692, P=0.021) and intraabdominal infection (OR=3.725,P=0.003)were also the independent risk factors for Grade B and Grade C DGE. Conclusions DGE after pancreaticoduodenectomy was strongly related to the postoperative complications .Postoperative pancreatic fistula , biliary fistula and intraabdominal infection were associated with increased risk of DGE , while pancreaticogastrostomy reduced the incidence of DGE by decreasing the incidence of pancreatic or biliary fistula .
8.Quality Control for Tongsai Yinao Oral Liquids

Objective:To improve the quality standard of Tongsai Yinao oral liquids for the quality control. Methods: TLC was used for the qualitative identification of Rhizoma corydails, Rhizoma chuanxiong and Rhizoma acori tatarinowii. HPLC was used to sim-ultaneously determine the content of total ferulic acid in the oral liquids. The determination was performed on a Lichrospher-C18 (250 mm×4.6 mm,5 μm)column with the mobile phase of methanol-0.1% H3PO4(30∶70). The flow rate was 1.0 ml·min-1,the col-umn temperature was at 30℃, the detection wavelength was 321nm and the injection volume was 20μl. Results:The spots on the TLC plates were clear without the interference of negative control. The linear range of ferulic acid was obtained between 2. 24 and 35. 84 μg ·ml-1(r=0. 999 9), and the average recovery was 102. 54%(RSD=0. 85%, n=6). Conclusion:The improved method is accu-rate and feasible. It can be used very conveniently in the quality control.
9.Ultrasonic research on the correlation between aortomitral angle and left ventricular systolic function in the patients with ischemic cardiomyopathy

Objective To evaluate the relationship between the change of the aortomitral angle (AMA) with left ventricular systolic function in patients with ischemic cardiomyopathy (ICM) by echocardiography.Methods Thirty-one patients were enrolled in the ICM group,and 59 healthy subjects were selected as the control group.On the parasternal left ventricular long axis plane,AMA were measured at the R wave apex (R-AMA),J-point(J-AMA),ST-segment midpoint(ST-AMA),T-final wave (T-AMA)and P-final wave (P-AMA).The angle difference(⊿ θ) =AMAmax-AMAmin,the angle changing rate =⊿ θ/AMAmax.The global left ventricular longitudinal strain (GLS) and global circumferential strain (GCS) were obtained by 2D-speckle tracking echocardiography.Left ventricular ejection fraction(LVEF),left ventricular end-diastolic volume(LVEDV) and left ventricular end-systolic volume(LVESV) were measured using Simpson biplane method.Results The J-AMA was the largest in the control group,while the ST-AMA was the largest in the ICM group.The levels of LVEDV,LVESV and AMA in ICM group were significantly higher than those in control group,while LVEF,GLS,GCS,⊿ θ/AMAmax and ⊿ θ were decreased (P <0.05).In the control group,there was a correlation between T-AMA and LVEF (r =-0.349,P =0.007),and ⊿ θ was negatively correlated with GLS (r =-0.372,P =0.004).In the ICM group,⊿1 θ/AMAmax and ⊿ θ were correlated with LVEF (r =0.424,P =0.018;r =0.490,P =0.005).Conclusions AMA in ICM patients is significantly increased.The angle difference and the rate of its change are closely related to the LVEF,which is a manifestation of three-dimensional structure change of the myocardial.
10.Study on Effects of Chromium on Heavy Metal Removing Strains

Effects of Cr 6+ concentration and culture time on four heavy metal removing strains,stability of these four strains removing Cr 6+,configuration changes inside or outside their cells,effects of Cr 6+ on soluble reductive sugar inside their cells,and effect of several factors on these strains had been studied,and the Cr 6+ resistance mechanisms of these strains have been discussed elementarily. The results showed that the Lethality of these strains caused by Cr 6+ was similar with one another, namely, increasing at first, then decreasing, and finally increasing again as culture time passed. Acclimatization of Candida sp. was better than Sporobolomycetaceae sp.,and the Cr 6+ resistance of Sporobolomycetaceae sp. 7-3 was the best of the four. The research also demonstrated that the metabolic activity of these strains had been influenced by Cr 6+ in a certain extent. Scanning electron microscopy,transmission electron microscopy,and atomic force microscopy observations approved that removal of Cr 6+ by Candida sp. was depended on both surface adsorption and intracellular accumulation. Effects of Cr 6+ concentration, pH, culture time, nitrogen source, carbon source and adsorption time on these strains are not the same.
