A tumor suppressor gene at 10q23.3, designated PTEN, encoding a dual specificity phosphatase with lipid and protein phosphatase activity, has been shown to play an important role in the pathogenesis of a variety of human cancers and sporadic tumor. In this paper, we reviewed the construction and function of PTEN, and the mechanism of activation in cancer especially in thyroid carcinoma.
2.The influence of Jiuganning decoction on the content of CYP2E1 and cytokine IL-6,TNF in rats models of alcoholic hepatic injury

Objective To explore the influences of Jinganning decoction on the content of CYP2E1 and cytokine IL-6,TNF in rats models of alcohdic hepatic injury.Methos 72 male Wistar clean level rats were randomly divided into 6 groups:blank control group,Jiuganning drug toxicity test group,alcoholic hepatic disease model group,Yiganling treatment group,Jiuganning low dose group,and Jiuganning high dose group.Blank control group was treated with physiological saline;Jiuganning drug toxicity test group was treated with Jiuganning decoction to observe toxicity and side effects.Jiuganning low and high dose treatment group were treated with 17.5 g/kg and 23.5 g/kg crude drug.Yiganning treatment group were treated with 0.206 g/kg crude drug.After 4 weeks'treatment,the content of TNF,IL-6,MDA as well as the activity of CYP2E1 and SOD were measured.Results Compared with the alcoholic hepatic disease model group,the content of IL-6(F=46.281,9.101),TNF and the activity (F=62.537)of CYP2E1 of all treatment group were significantly improved(P<0.01).The content of SOD(F=19.210)in the rats serum of all treatment group was significantly increased while the MDA(F=34.952)was decreased.All the indexes in the treatment group had significant differences with the alcoholic hepatic disease and model group(P<0.01).Conclusions The mechanism of Jiuganning decoction can effectively reduce the content of CYP2E1 and IL-6,TNF-a,with the changes of cell physiopathology and the influences of relevant enzymes,reveals the functional routines of the decoction treating hepatic injury:anti-lipid peroxidative,adjusting cytokine's activity,protecting hepatocyte,palliating the hepatocyte inflammation,and easing fibration.
3.The ankle-brachial index of type 2 diabetes

Objective To study the ankle-brachial index of the diabetic gangrene. Methods ABI of the sixty cases with type 2 diabetes were measured. The results were divided into four groups: normal, gently abnormal moderate abnormal severely abnormal group. Results 30 cases were normal, occupying 50% (P>0.05), gently and moderate abnormal attained 5 cases (8.33%) and 4 cases (6.67%) respectively, and the rests 21cases were severely abnormal, occupying 35% (P>0.05).Conclusion The normal ABI was not always indicating type 2 diabetes without the gangrene or tendency.
4.The nutrition and health status of children should be improved by increasing milk and its product consumption in their diets.

5.Effect of a disintegrin and metalloprotease 17 deletion on production of reactive oxygen species and mitochondrial function in nasopharyngeal carcinoma cells

Objective To investigate the effects of a disintegrin and metalloproteinase 17(ADAM17) deletion on the production of reactive oxygen species(ROS) and mitochondrial function in nasopharyngeal carcinoma(NPC) cells.Methods Three groups of ADAM1 7 interfering plasmid ADAM17 shRNA and empty plasmid ADAM17-shRNA-NC were transfected into NPC cell line(CNE1) and detected for the interference efficiency by RT-PCR and Western blot to select shRNA with the best interference effect for the follow-up experiments.The cell proliferation was detected by CCK-8 assay,while the cell growth by clone formation test,the apoptosis and changes in mitochondrial membrane potential(MMP) by flow cytometry,the level of mitochondrial oxidative damage product ROS by fluorescence microscope,the contents of oxidative stress markers MDA and SOD by malondialdehyde(MDA) kit and superoxide dismutase(SOD) kit and the expression of mitochondrial damage markers Bax/Bcl-2,cleaved-caspase 9/caspase 9,cleaved-caspase 3/caspase 3 and c-Myc by Western blot.Results ADAM17-shRNA2 group showed the best interference effect.Compared with shRNA-NC group,the proliferation rate of cell in ADAM17-shRNA 2 group decreased significantly(t=8.964,P=0.036);the number of colonies were significantly reduced(t=10.351,P=0.014);the number of apoptosis increased significantly(t=11.25,P=0.008);the fluorescence intensity representing ROS level in cells increased obviously;the mitochondrial membrane potential decreased significantly(t=9.233,P=0.013);the SOD content decreased(t=7.233,P=0.034) and MDA content increased(t=7.415,P=0.038) significantly;the levels of Bax/Bcl-2,cleaved-caspase 9/caspase 9 and cleaved-caspase 3/caspase 3 significantly increased(t=8.985,9.021 and 7.789,P=0.023,0.011 and 0.031,respectively),while the expression of c-Myc proteins significantly decreased(t=10.352,P=0.004).Conclusion Interfering with ADAM1 7 induced SOD decrease and MDA increase by promoting oxidation,thereby alleviating oxidative damage of cell membrane,which also promoted the expression level of ROS in mitochondrion,reduced MMP,inhibited cell proliferation in vitro,and promoted apoptosis.
6.Probe and Practice of Fitting the Education of Handicapped Children to Community-based Rehabilitation

The education and development of the handicapped children has been regarded as part of the community-based rehabilitation now. Our purpose is to give those handicapped children opportunities to be educated, take part in social life, and come back to the society. The author summarized and analyzed some successful methods about community-based rehabilitation in western poverty region of China by international project, and suggest that great efforts has to be taken to improve the inclusive education of handicapped children by community-based rehabilitation.
7.Prognosis and effect of rectovaginal fistula operation on patients' life quality

ObjectiveTo investigate prognosis and effect of rectovaginal fistula operation on patients' life quality.MethodsMedical data of 18 cases with rectovaginal fistula were analyzed retrospectively,and a follow-up was also made.ResultsAmong 18 cases,types were anal canal (1 case),low rectum (5 cases) and high rectum (12 cases). Three different kinds of surgical repair procedures were performed on these patients respectively,with 13 cases obtaining the first surgical repair successfully and 2 cases having the second surgical repair because of the fistula relapse. After operation,the general life quality and sexual life of patients improved.ConclusionThe rectovaginal fistula operation can deliver a good therapeutic effect,and evidently enhanced the life quality of patients.
8.Discussion on clinical significance of the coagulation function detection of non small cell lung cancer patients

Objective To observe the changes and clinical significance of the coagulation function detection of non small cell lung cancer patients .Methods Selected 148 cases of non-small cell lung cancer admit of our hospital from 2011 January to 2013 April as the study group ,and 120 healthy persons as control group ,two groups were detected plasma prothrombin time (PT ) ,activated par-tial thromboplastin time(APTT) ,fibrinogen(Fib) ,platelet count(PLT) and D-dimer(D-D) .Results In the study group:PT exten-sion 8 cases(5 .4% ) ,APTT extension 27 cases(18 .2% ) ,Fib increased 80 cases(54 .1% ) ,PLT increased 25 cases(16 .9% ) ,PLT reduced 4 cases(2 .7% ) ,D-D increased 88 cases(59 .5% ) .APTT ,Fib ,PLT and D-D level in the study group was significantly high-er than the control group(P<0 .05) .No significant differences between the two groups of PT (P>0 .05) .Fib and D-D levels in stage Ⅳ NSCLC were significantly higher than patients with stage Ⅰ ~ Ⅲ lung cancer tumor metastasis ,Fib and D-D levels of pa-tients were significantly higher than those without metastasis patients (P<0 .05);for tumor size ,pathological type ,tumor differenti-ation degree unrelated to APTT ,Fib ,PLT ,D-D levels ,there were no difference statistical significance(P>0 .05) .Conclusion Pa-tients with lung cancer APTT ,Fib ,PLT ,D-D level increased significantly ,there is a close relationship between the prognosis of pa-tients w ith Fib and D-D levels and lung cancer .
9.Construction,expression,purification and quality identification of kanamycin resistant strain of recombinant human interleukin-1 receptor antagonist

Objective To construct a recombinant human interleukin-1 receptor antagonist(rhIL-1Ra)strain of kanamycin resistance,express,purify and identify rhIL-1Ra protein in order to reduce the risks of β-lactam antibiotics.Methods The ampicillin-resistant rhIL-1Ra(A-rhIL-1Ra)plasmid and PET-28a plasmid were used as templates to prepare the linearized vector and kanamycin gene fragment by PCR.The kanamycin resistant rhIL-1Ra plasmid(K-rhIL-1Ra)was constructed by homologous recombination,which was transformed into E.coli BL21(DE3)to construct the recombinant engineered bacteria after correct sequencing.The engineered bacteria were induced by IPTG and then purified by CM Bestarose Fast Flow and DEAE Bestarose Fast Flow column chromatography.The purified products were collected,and then detected for the purity by 15% SDS-PAGE,size-exclusion high-performance liquid chromatography(SEC-HPLC),and reversed-phase high-performance liquid chromatography(RP-HPLC),determined for the relative molecular mass by tandem mass spectrometry,identified for the specificity by Western blot,measured for the biological activity by reporter gene assay,and compared for the related impurities by liquid chromatography-mass spectrometry(LC-MS)analysis.Results Colony PCR and sequencing results showed that K-rhIL-1Ra plasmid was constructed correctly.The expressed K-rhIL-1Ra protein had a relative molecular mass of about 17 000,mainly existing in soluble form,and the expression amount accounted for more than 30% of the total bacterial proteins.The purity of K-rhIL-1Ra protein purified by two steps was 97% and 99%,the monomer content was 99.33%and 100%,and the chromatographic purity was 91.86% and 96.96%,respectively.The mass spectral molecular mass was consistent with that of the standard(A-rhIL-1Ra protein),and the protein reacted specifically relative with mouse antihuman IL-1Ra monoclonal antibody.The biological activity of the purified K-rhIL-1Ra protein was 1.29 × 105U/mg,and the chemical modification types of related proteins were the same as those of the standard(A-rhIL-1Ra protein).Conclusion The K-rhIL-1Ra strain was successfully constructed,and the expressed and purified protein was in line with the characteristics and quality standards of rhIL-1Ra,which lays a foundation for the study of rhIL-1Ra strain change and comparability
10.Full-scale setback and thinking in R&D of Alzheimer's disease drug.

During the past 30 years, Alzheimer's disease (AD) drug R&D aiming at a variety of potential targets has undergone tremendous setback although a large amount of resources have been invested by pharmaceutical companies, governments, academic institutions and venture capitals globally. There is no doubt that uneasy early diagnosis, unsatisfying therapeutic strategies and genetic polymorphism of AD patients bringing variety of responses to the drugs contribute to the failure of AD drug therapy, but the most important reason might be that people have insufficient understanding of the very complex diseases including AD. Continuing to make efforts in the previous way to find AD drugs might not be a good way. Development of systems biomedicine opens an avenue to understand the disease networks and pursue multi-target drugs R&D based on systems pharmacology theory; neural stem cell transplantation, and neurogenesis affected by small molecules might bring new hope for AD therapy; discovery of new biomarkers of AD will help the study of the pathogenesis and diagnosis of the disease and finally the discovery of new types of AD drugs.
