4.The application of optogenetics in the treatment of retinal degeneration disease

Optogenetics is a novel technique which combines optics with genetics.Using genetic means,a selected opsin protein is ectopically expressed in target neurons,which are then stimulated by light to moderate the neuronal circuit,as a consequence to regulate the animal's behaviors.Retinal degeneration like retinitis pigmentosa and aged macular degeneration causes visual impairment and eventual blindness.Optogenetics techniques have opened the door to creating artificial photoreceptors in the remaining retinal circuits of retinal degeneration retinas via gene therapy.However,there are still limitations in optogenetics technique,for example,potential risk in virus infection,the choice of target cells and the low visual resolution of the experiment animal.It has been reported that vision was successfully restored to a certain extent in animal model using optogenetics technique.With higher photosensitivity of opsin protein,longer activation kinetics and higher transfection efficiency of virus vector,optogenetics techniques' application in ophthalmology will be improved.
6.The Application of Low Dose MSCT in Diagnosing the Pharyngeal Disease

Objective To study the best parameters of low dose MSCT in examining the pharyngeal disease.Methods 20 volunteers were scanned by MSCT using 10 mm collimation,10 mm interval,120 kV,200 mA,100 mA,50 mA,25 mA parameters,respectively.All of the data was reformed(collimation 2.5 mm,interval 1.3 mm) on ADW 4.0 workstation.The image quality in different dose were scored respectively by 4 experienced radiolagists. Results The best appropriate parameter was 10 mm collimation,10 mm interval,120 kV,50 mA.Conclusion In examining the pharyngeal disease,the application of low dose(120 kV,50 mA) scan mode not only can assure the image quality but can decrease the exposure to patients.
7.Study of low dose and dynamic multi-slice CT about obstructive sleep apnea syndrome in sleeping

Objective To perform Low dose dynamic MSCT( multi-slice CT) in sleeping obstructive sleep apnea syndrome ( OSAS) patients correcting the imprecise measure values in waking state, and to exactly analyse the location and extension of the dynamic changes about the condition. Methods Sixteen OSAS patients were scanned both in waking and naturally sleeping period ( end phase of inspiration and expiration). Measured at the narrowest part of the retropalatal ( RP) and retroglossal ( RG) and 5 mm under the tip of epiglottis at the epiglottal ( EPG) at the end period of inspiration in sleeping, respectively, and compared the accurate pos(?)on of the narrowest or occlusive level in 3 phases. All patients were also scanned using cine mode at the narrowest level at the end period of inspiration in sleeping to show the pharyngeal cavity changes during sleep. 8.Abnormal expression of costimulatory molecules CD_(28)and CD_(137)on PBMC from patients with chronic nephritis

Objectives To investigate the expression features and roles of the costimulatory molecules and T lymphocyte subsets from patients with chronic nephritis.Methods The expression of the costimulatory molecules CD 28 and CD 137 on PBMC (peripheral blood mononuclear cells) and T lymphocyte subsets from 52 patients with chronic nephritis were studied by immunophenotyping and flow cytometry analysis.Results The T lymphocyte subsets from patients with chronic nephritis showed an obvious imbalance with reversing CD 4/CD 8 ratio,decreasing CD + 4T cells and increasing CD + 8T cells.The expression of the costimulatory molecule CD 28 on T cells was significantly lower compared with normal controls (P
9.Clinicopathological analysis of malignant triton tumor of the posterior mediastinum

Objective To investigate the pathologic appearances,immunohistochemical features,and genetic changes of malignant triton tumor(MTT). Methods One case of MTT was studied pathologically and immunohistochemically,and the related literatures were reviewed. Results A huge mass,demonstrated in the thorax by X ray and CT scan was seen in the posterior mediastinum in the surgery.Histologically,the tumor was composed of spindle cells with significant atypia.Some of the tumor cells had dense eosinophilic cytoplasm.Immunohistochemical staining revealed positive for myoglobin,desmin and S-100 in most of the tumor cells.The pathological diagnosis was MTT of the posterior mediastinum. Conclusion Cases of MTT in the mediastinum are very rare,with less specific clinical and imaging manifestations.The diagnosis is mainly made on the basis of pathological examination and immunohistochemical staining.
10.Clinical study of the insulin resistance and pancreaticβ-cell function in patients with gestational diabetes

Objective To investigate the insulin resistance and the changes of pancreaticβ-cell function in order to improve the clinical diagnosis and therapy .Methods 99 cases of pregnant women in the late ministers were randomly selected for the study and divided into three groups(normal glucose tolerance group ,impaired glucose tolerance group ,and gestational diabetes group) .The in-sulin resistance and isletβ-cell function of the three groups were evaluated by HOMA model and its related indicators .Results The blood pressure and blood lipid level was not significant different among the three groups(P>0 .05) .Pairwise comparisons of glu-cose infusion rate ,the first phase insulin secretion ,the second phase insulin secretion ,fasting glucose ,fasting insulin ,HOM A-IR , and HOMA-βwere made among the three groups ,and there were difference with statistical significance(P<0 .05) .Conclusion The situation of insulin resistance and the dysfunction of pancreaticβ-cell in trimester pregnant women would become bad associated with the abnormal glucose tolerance .
