1.A study of cases with blurred vision or cecitas after cardiac or cerebral angiography

Objective To study the cause of blurred vision or cecitas after cardiac or cerebral angiography. Methods Six patients including 4 performed with cerebral angiography and 2 with cardiac angiography were analyzed. Results In those 6 patients, blurred vision happened in 4 cases, and cecitas appeared in 2 cases. Ophthalmologic examination revealed bilateral isocoria, thinning ophthalmic arteries and normal light reflex. Color Doppler flow imaging showed clearly the central retinal arteries. No cerebral infarction and brain hemorrhage were detected under CT. Conclusions Blurred vision and cecitas are the rare complications occurred with cardiac or cerebral angiography, probably with direct relationship to the concentration or dosage of the contrast media used, and the primary diseases of the patients.
2.Analysis and countermeasures of prevention and control of multidrug resistance bacteria infections

Objective To analyze the distribution of multidrug resistance bacteria infections in clinical departments and put for‐ward the corresponding countermeasures of prevention and control ,for providing basis for clinical diagnosis and treatment .Methods By carrying out the monitor of the drug resistance of bacteria ,the infection site and drug resistance strain and pathogenic bacteria distribution ,with 156 cases of multiple drug‐resistant bacteria infection ,were analyzed .Results The multidrug resistance bacteria of our hospital was distributed in the ICU unit ,accounted for 34% .Nosocomial infection of multidrug resistance bacteria was mainly the gram‐negative bacillus ,followed by the gram‐positive cocci ,and the majority of multidrug‐resistant bacteria were Escherichia co‐li(46 .2% ) ,Acinetobacter Baumannii(19 .9% ) ,Pseudomonas Aeruginosa(17 .9% ) ,Klebsiella Pneumoniae(17 .9% ) ,Staphylococcus Aureus(4 .5% ) .The priority was given to respiratory system infection ,accounted for 48 .1% ,followed by urinary system infection , accounted for 39 .0% .Conclusion The increase of multidrug resistance bacteria was very important problem that the hospital faced .It can effectively prevent and control multiple drug‐resistant bacteria infection by strengthening the monitoring of multi‐re‐sistant bacteria ,strictly executing disinfection and isolation measures ,the implementation of hand hygiene and reasonable application of antibiotics .
3.Inhibitory effect and its molecular mechanisms of MicroRNA-34a on human na-sopharyngeal carcinoma CNE-2 cell line subcutaneous xenograft tumor in nude mice

Objective:To investigate the inhibitory effect and its possible molecular mechanisms of MicroRNA-34a(miR-34a) on the human nasopharyngeal carcinoma CNE-2 cell line subcutaneous xenograft tumor in nude mice.Methods: The human nasopharyngeal carcinoma CNE-2 cell line was cultured in vitro.miR-34a and Scrambled miRNA recombinant plasmids were successfully established and stably transfected into CNE-2 cells.Fifteen six-week-old male nude mice were divided randomly into three groups:miR-34a group(5 mice) ,Scrambled miRNA group(5 mice) ,Blank control group(5 mice).Different CNE-2 cells were subcuta-neously injected on the back near right lower limb.Tumor volumes were examined every 7 days.Mice were executed on the 35 days,and the eventual average tumor volumes and weights were examined.Total RNA and protein were isolated from tumors,and the expression of miR-34a,CDK6,and Bcl-2 mRNA and protein were determined by qRT-PCR and western blot,respectively.Results: The relative expressions of miR-34a was significantly up-regulated in miR-34a transfected group compared to Scrambled miRNA transfected group (P<0.05).Eight subcutaneous xenograft tumor models were successfully established.The nude mice of miR-34a transfected group appeared a smaller tumor volume compared to the other two groups at the beginning of 21th day( P<0.05).The eventual average tumor volumes in miR-34a group,Scrambled miRNA group and blank control group were(351.37±98.19)mm3,(798.75±91.04)mm3 and
(849.62±101.32) mm3 ,respectively,and the eventual average tumor weights in miR-34a group,Scrambled miRNA group and blank control group were(0.81±0.13)g,(1.47±0.21)g and(1.58±0.37)g,respectively.Both the eventual average tumor volumes and weights in miR-34a group were lower compared to the other two groups(P<0.05).qRT-PCR results revealed that the expression of miR-34a in miR-34a transfected group was significantly higher than in the other two groups,while the mRNA and protein expression of CDK6 and Bcl-2 were lower than the other two groups ( P<0.05 ) .Conclusion: miR-34a may inhibit the growth of human nasopharyngeal carcinoma CNE-2 cell line subcutaneous xenograft tumor in nude mice by down-regulating CDK6 and Bcl-2.
4.Research and practice of management of preoperative examination based on quality control circle

Objective To investigate the effect of implementation of QCC (quality control circle) in management of preoperative examination.Methods Through analysis of the common factors of patients' preoperative examination,the QCC was used to manage every parts of preoperative examination,and the nursing process of preoperative examination for resident patients was formulated.The effect of nursing management of preoperative examination before and after the implementation of QCC was investigated and compared.Results The result showed that the degree of satisfaction was significantly higher than before,and the risk incident was significantly lower than before.Conclusions To carry out QCC could improve the efficiency of preoperative examination and the patients' safety,as well as enhance the awareness of the nursing management.

The caudal-type homeobox gene is a member of homeobox gene—Para-HOX family,including CDX1,CDX2 and CDX4.As a transcriptional factor,CDX plays an important role in embryo development,hematopoietic system formation,intestinal epithelium tissue development and so forth.The abnormal expression of CDX is usually closely related with tumorigenesis.Researching the relationship between CDX and tumors will contribute to tumor diagnosis and treatment.

7.Optimization of the Extraction Technology of Bushen Huoxue Capsules by Central Composite Design-re-sponse Surface Method

OBJECTIVE:To optimize the extraction technology of Bushen huoxue capsules. METHODS:Central composite de-sign-response surface method was used to optimize the extraction technology with the amount of water and boiling time as main fac-tors using normalized value of the contents of icariin and salvianolic acid B,the yield of dry extract as index. Validation test was al-so conducted. RESULTS:The optimal extraction technology was as follows as 15-fold water,reflux extracting for 75 min,extract-ing for 2 times. The deviation of measured value and predicted value was 0.000 3% in validation test. CONCLUSIONS:The cen-tral composite design-response surface method can be used to optimize the extraction technology of Bushen huoxue capsules with good prediction,and the technology is simple and stable.

Objective To detect the mutation of A379,we develop a TaqMan fluorogenic probe based amplification refractory mutation system (TaqMan-ARMS) and investigate whether the A379V variant and activity of Lp-PLA2 are the risk factors for CAD.Methods According to the amplification refractory mutation system(ARMS-PCR)combined with a TaqMan fluorogenic probe,we established and evaluated the assay teehnique of TaqMan-ARMS for measuring the genotype of variant.At the same time,we tested the aetivity of Lp-PLA2 in 395 patients with coronary heartdisease and 396 controls,whose clinical information [ages,CHO,GLU,TG,HDL,LDL,Hs-CRP,Lp (a)] were collected.Date was analyzed by using Independent-samples t test,Chi-square test,One-Way ANOVA,Binary Logistic Regression.Results CAD group had significantly higher Lp-PLA2 activity than the controls(31.51 nmol · ml-1 · min-1 >21.31 nmol ·ml-1 · min-1,F =16.40,P < 0.001).Comparing the highest quartile of Lp-PLA2 activitv to the bottom quartile,OR was 7.50 (95% CI:2.34-24.05) after adjustment for various traditional cardiovascular risk factors,including ages,sex,CHO,TG,Hs-CRP,Lp (a) and GLU; the genotype VV of A379V was associated with higher risk of CAD (OR =2.95; 95% CI:1.22-7.15,P < 0.05).Conclusions The TaqMan-ARMS real time PCR technique is established to analyze A379V genotype.Lp-PLA2 activity is significantly higher in CAD group and is a risk factor for CAD; the genotype VV of A379V is also a risk factor for CAD.
10.Cloning and expression of immunotoxin DT389- hbFGF

AIM: To express the DT389-hbFGF (389 amino acid residues of the N-terminus of diphtheria toxin (human basic fibroblast growth factor) fusion protein for potential targeting therapy towards posterior capsule opacification (PCO) after cataract surgery.METHODS: The DNA of inactivated diphtheria bacillus and RNA of 12-week fetal brain cortex were extracted, respectively. The fragments of truncated diphtheria toxin (containing 389 amino acids of N-terminus, DT389) )and full-length human basic fibroblast growth factor(hbFGF) sequence (encoding 18kDa protein) were amplified by PCR. The two fragments were inserted into pGEX-4T-1 prokaryotic expression vector to obtain pGEX-DT389-hbFGF prokaryotic expression plasmid. After sequence analysis, the expressing plasmid was transformed into Escherichia Coli BL21 strain and expression was induced under IPTG. The expressed fusion protein was purified and identified.RESULTS: The gene fragments encoding DT389 and hbFGF were amplified and their gene sequences were confirmed. Hybrid gene expression plasmid pGEX-DT389 (hbFGF was constructed. The fusion protein DT389-hbFGF was expressed and purified.CONCLUSIONS: The successful cloning and expression of DT389-hbFGF immunotoxin provides a foundation for targeting therapy towards posterior capsule opacification.
