1.Application of cross-linked telopeptide of type Ⅰ collagen, alkaline phosphatase in bone tumor

Objective To determine the serum levels of cross-linked telopoptide of type Ⅰ collagen (ICTP),alkaline phosphatase (ALP) in patients with primary malignant bone tumor,primary benign bone tumor and malignant tumor metastasized to the bone,and to explore the clinical value of ICTP and ALP in identification and diagnosis of bone tumor.Methods Sixteen primary malignant bone tumor patients,16 primary benign bone tumor patients and 18 malignant tumor metastasized to the bone patients in 2012 were studied.Serum ALP was assayed by SFBC rate method and ICTP by enzyme linked immunosorbent assay (EIA).Results The serum levels of ICTP was not significantly different between primary benign bone tumors and normal control group (P > 0.05),but the other between-groups had statistically significant difference (P < 0.05).The serum levels of ALP in malignant tumor metastasized to the bone was significantly higher than the rest of the group (P <0.01),but the difference between the remaining groups was not statistically significant (P > 0.05).The area under roc curve (AUC) of ICTP for diagnosis of primary benign bone tumors,malignant tumor metastasized to the bone,primary malignant bone tumor (0.923,0.926,0.874) was higher than the ALP (0.354,0.702,0.865).Conclusions Serum ICTP and ALP were sensitive and convenient biochemical indices which reflected metabolism of patients with bone tumor.Serum ICTP was more specific and sensitive than ALP and they have clinical importance for differential diagnosis as an index of bone tumor.
2.Topological characteristics of postoperative survival rate-related molecules in hepatocellular carcinoma-associated complex networks

Objective To explore the network structure of genes and microRNA(miRNA) at the system level and to study topological characteristics of survival rate related molecules by means of survival analysis.Methods Regulatory networks were established with genes of hepatocellular carcinoma( HCC) and with the expression profiling data of miRNA. Survival knowledge in nodes was studied.Results All the nodes in gene co-expression networks conformed to power-law distribution.In a network, high-degree nodes were called Hub, which enriched more survival related genes than lower degree ones.Conclusion Hub nodes in gene regulatory networks promise to be potential features for molecular subtyping.
3.Curcumin inhibits LPS-induced EMT through down-regulating NF-κB-Snail signaling in breast cancer cells

Objective To investigate whether epithelial-mesenchymal transition (EMT)is involved in the anti-invasion and anti-metastasis effects of curcumin on MDA-MB-2 3 1 breast cancer cell line and further analyze the underlying mechanisms.Methods MTT method was used to detect the anti-proliferative effect of curcumin on MDA-MB-2 3 1 cell line induced by lipopolysaccharide (LPS).The morphological changes were determined by optical and transmission electron microscopy,respectively.The expressions of Vimentin,E-cadherin,NF-κB and Snail were analyzed using RT-PCR and Western blotting.Results Curcumin could inhibit the occurrence of LPS-induced EMT of MDA-MB-2 3 1 breast cancer cell line, decrease the expression of LPS-induced EMT marker Vimentin and increase the expression of E-cadherin, resulting in the inhibition of in vitro cell motility and invasiveness.These actions were mediated by inactivating NF-κB-Snail signaling pathways.Conclusion Our data provide a new perspective of the anti-invasion mechanism of curcumin,indicating that the effect is in part due to its ability to inhibit the EMT process.
4.The safety and efficacy of radiofrequency ablation for treating large hepatic hemangiomas

Objective To assess the safety and efficacy of radiofrequency ablation (RFA) for the treatment of large (≥5 cm in diameter) hepatic hemangiomas.Methods Clinical data of 50 patients with large hepatic hemangiomas (≥5 cm in diameter) treated with RFA between October 2007 and December 2012 were analyzed.Patients were divided into two groups (5-10 cm and ≥ 10 cm) according to tumor size.Results Thirty-two patients had 36 hemangiomas of 5-10 cm in diameter and 18 patients had 19 hemangiomas of ≥ 10 cm in diameter.Technical success,complications related to RFA,completed ablation,symptom relief,change in size of ablation zone and recurrence of the residual tumor were analyzed.The average diameters of the two groups were 7.1 ± 1.2 cm and 13.2-± 2.4 cm separately (t =-12.57,P < 0.01) ; the technical achievement ratios of the two groups were both 100% ; Seven of 32 patients with hemangiomas 5-10 cm and all the 18 patients with hemangiomas ≥ 10 cm had 13 and 61 complications related to RFA,the incidence of complications were 21.88% and 100% respectively (x2 =28.13,P < 0.01); 94.55% hemangiomas (52/55) acquired complete ablation,the complete ablation rates of 5-10 cm hemangiomas and ≥10 cm hemangiomas were 100% (36/36) and 84.21% (16/19) respectively (P =0.014).The mean diameters of ablation zone were respectively decreased to 5.3 ± 1.0 cm and 10.62±1.8 cm (t =-14.30,P <0.01).Conclusions RFA for hepatic hemangiomas 5-10 cm in diameter is safe and effective; while its complication for ablation of hemangiomas ≥ 10 cm is high.
5.The etiology, molecular epidemiology and clinical characteristics of Clostridium difficile infections in Xiangya Hospital

Objective A preliminary study on the etiology , the gene typing , the PCR-ribotyping and the clinical features of Clostridium difficile from clinical isolates at Xiangya Hospital could improve the isolation rate and provide the basis for effectively prevention of C.difficile.Methods A prospective observational study was performed.A total of 452 stool samples were collected during June to December 2012 at Xiangya Hospital.All stools were anaerobic cultured by selective medium and identified by API 20A for C.difficile.The positive isolates were detected the toxin genes ( tcdA, tcdB, cdtA, cdtB ) and ribotyping (16S-23S internal spacer region ) by PCR.The clinical data of all patients were collected and analyzed through Logistic regression to discover the risk factors for the development of C.difficile infection ( CDI ) . Results The rate of CDI occurrence was 13.94%(63/452), among them, 42.86%(36/63) were A-B+strains and only 14.29%(9/63) were obtained from community acquired-CDI.No binary toxin was detected in any of the isolates.Eleven different PCR ribotypes were identified , the dominant ribotype CD017 accounted for 22.22%(14/63).Logistic regression analysis showed that the risk factors for CDI included age>55(P=0.016;OR=4.45;95%CI:1.33-14.91), diarrhea frequency(P=0.007, OR=0.03;95%CI:0.002 -0.38 ) and the duration of diarrhea ( P =0.015; OR =7.86; 95%CI: 1.50 -41.16 ) . Conclusions C.difficile is the main pathogens of diarrhea patients and is mainly from hospital infections with higher detection rate of A -B+ in Xiangya Hospital.Ribotyping exist comparative advantages type CD017.No evidence suggests outbreak of C.difficile infection.
6.In vitro analysis of the antibacterial activity of Micafungin against Pseudomonas aeruginosa

Objectives Selecting and constructing the biofilm -model of Pseudomonas aeruginosa in vitro.Observing the antibacterial activity of using Micafungin alone , or combined with Meropenem against Pseudomonas aeruginosa ( plankton-grown and biofilm-grown ) . Methods Ten clinical isolates of Pseudomonas aeruginosa were collected in July 2012, constructing the biofilm-model by microwell plate from Xiangya Hospital, Central South University.The ability of biofilm-formation of these strains was estimated by crystal violet colorimetric method, and optical microscope was used to observe the shape of the biofilm .MICs of Micafungin and Meropenem against plankton -grown and biofilm-grown Pseudomonas aeruginosa were tested by broth microdilution method, and the changes of MICs were compared.Using broth microdilution method, and connecting with the crystal violet colorimetric method , to observe the antibacterial effect of using Micafungin alone, or combined with antibiotics in the inhibition of the biofilm formation and destruction of mature biofilm of Pseudomonas aeruginosa.SPSS18.0 and t-test were used in comparing the differences between both treatment group and control group.P <0.05 showed the difference was statistically significant . Results Ten strains of Pseudomonas aeruginosa were successful in forming biofilms.Comparing with their planktonic counterparts, biofilms became more resistant to Meropenem , with the MIC raised 4-128 times. However, MIC of Micafungin could not be measured.Micafungin can inhibit the formation of biofilm in 9 experimental strains (PA1-PA9), where the minimum effective concentration of Micafungin were 156.25, 625, 10 000, 2 500, 1 250, 2 500, 1 250, 625 and 10 000 mg/L respectively.The absorbance values of the minimum effective concentration group and its positive growth control group were 0.342 ±0.020 vs 0.491 ±0.027, 0.512 ±0.018 vs 0.627 ±0.043, 0.862 ±0.021 vs 1.155 ±0.027, 0.731 ±0.028 vs 0.863 ± 0.017, 0.311 ±0.003 vs 0.447 ±0.021, 0.435 ±0.021 vs 0.597 ±0.011, 0.520 ±0.012 vs 0.605 ± 0.027, 0.611 ±0.059 vs 0.734 ±0.017, 0.223 ±0.011 vs 0.343 ±0.037 respectively, where the P values were 0.02, 0.03, 0.00, 0.01, 0.01, 0.00, 0.03, 0.01 and 0.03 respectively.The differences are statistically significant.Micafungin can damage the mature biofilm of 7 strains (PA1, PA2, PA4 -PA8), where the minimum effective concentration of Micafungin were 2 500, 2 500, 5 000, 2 500, 5 000, 2 500, 5 000 mg/L respectively.The absorbance values of the minimum effective concentration group and its positive growth control group were 1.459 ±0.014 vs 1.534 ±0.020, 1.279 ±0.020 vs 1.431 ±0.007, 1.365 ±0.024 vs 1.467 ±0.065, 1.322 ±0.028 vs 1.530 ±0.090, 0.920 ±0.004 vs 1.047 ±0.013, 1.860 ±0.005 vs 1.953 ±0.055, 1.407 ±0.005 vs 1.553 ±0.045 respectively, where the P values were 0.01, 0.01, 0.02, 0.01, 0.00, 0.03, 0.02.The difference is statistically significant.Micafungin combined with Meropenem applied in multiple drug resistant strains , which can inhibit the formation of biofilm better.Conclusions Micafungin can inhibit the formation Pseudomonas aeruginosa biofilm and damage the mature biofilms.Micafungin combined with Meropenem can act on multiple drug resistant strain , which may get a higher inhibition rate of the biofilm.
7.Effects of rolling manipulation with different durations on erector spinae muscle tonus and low back pain in patients with lumbar muscle strain

Objective:To observe the effects of rolling manipulation with different durations on erector spinae muscle tonus and low back pain in patients with lumbar muscle strain and to explore the best duration for rolling manipulation in treating lumbar muscle strain. Methods:A total of 75 patients who met the inclusion criteria were randomized into a 5-minute rolling manipulation group,a 10-minute rolling manipulation group,and a 15-minute rolling manipulation group using the random number table method,with 25 cases in each group.All three groups were treated with the same rolling manipulation,while the treatment time was 5 min,10 min,and 15 min,respectively.The treatment was performed 3 times a week for 2 consecutive weeks.The muscle tonus of bilateral erector spinae and visual analog scale(VAS)score for low back pain were compared among the three groups before and after treatment. Results:A total of 63 patients completed the study.Before treatment and after the last treatment,there were no statistical differences in the muscle tonus of the left erector spinae and right erector spinae among the three groups(P>0.05).After the last treatment,bilateral erector spinae muscle tonus dropped compared with the baseline in all three groups(P<0.01).Before treatment and after the last treatment,there were no significant differences in comparing the low back pain VAS score among the three groups(P>0.05).After the last treatment,the VAS score for low back pain in all three groups decreased(P<0.01). Conclusion:Under the condition that the forward swing force is 50-70 N,the backward swing force is 20-40 N,and the frequency is 138 times/min,the 5-minute rolling manipulation can significantly reduce the muscle tonus of erector spinae and relieve low back pain.

Objective:To observe the effect of Yi Jin Jing(Sinew-transforming Qigong Exercises)intervention on anxiety in asymptomatic patients with Corona Virus Disease 2019(COVID-19)infection during quarantine.Methods:A total of 160 asymptomatic patients with COVID-19 infection were stratified by gender and divided into an observation group and a control group by the stratified randomization method,with 80 cases in each group.The control group was given basic nursing in the cabin,and the observation group was given additional Yi Jin Jing exercises once a day,20 min each time,and trained continuously until 1 d before leaving the cabin.The Hamilton anxiety scale(HAMA)score was compared between the two groups.Results:A total of 154 cases were included for data analysis in this study,including 74 cases in the observation group and 80 cases in the control group.After intervention,the HAMA scores in both groups increased(P<0.05),while the score in the observation group was lower than that in the control group(P<0.05).In the control group,the HAMA score of females after intervention was higher than that of males.The scores of mental anxiety and somatic anxiety in both groups were higher than those before intervention,while the score of mental anxiety in the observation group was significantly lower than that in the control(P<0.05).However,there was no statistical difference in the somatic score between the two groups(P>0.05).Conclusion:Patients with COVID-19 infection will get anxious during quarantine.Yi Jin Jing exercise can effectively reduce the aggravation of anxiety in asymptomatic patients with COVID-19 infection during concentrated quarantine.

10.Risk factors for coronary microvascular disease in elderly female patients

Objective To investigate the risk factors of coronary microvascular disease(CMVD)in elderly female patients.Methods Clinical data of 109 consecutive female elderly patients who re-ceived coronary angiography and coronary flow reserve(CFR)in our hospital from September 2020 to June 2022 were retrospectively collected in this study.According to CFR value,they were divided into CMVD group(CFR<2.0,n=55)and non-CMVD group(CFR ≥2.0,n=54).The basic clinical data were compared between the two groups,and influencing factors associated with CMVD were analyzed.Results Significantly higher BMI and elevated UA,hs-CRP,TG,LDL and Lp-PLA2 levels,while lower HDL level was observed in the CMVD group than the non-CMVD group(P<0.05,P<0.01).Lp-PLA2 and hs-CRP were negatively correlated with CFR(r=-0.816;r=-0.399,P<0.01).Spearman correlation analysis indicated that CRV value was nega-tively correlated with Lp-PLA2 and hs-CRP levels(r=-0.816,-0.399,P<0.01).Logistic re-gression analysis showed that hs-CRP and Lp-PLA2 were independent risk factors for CMVD in elderly female patients(P<0.05).ROC curve analysis revealed that the AUC value of Lp-PLA2 for diagnosing CMVD in elderly female patients was 0.745(95%CI:0.655-0.836),with a sensi-tivity of 69.1%and a specificity of 67.0%.And,the AUC value of hs-CRP was 0.752(95%CI:0.661-0.844).Conclusion Lp-PLA2 and hs-CRP levels are quite higher in elderly female CMVD patients,and can predict the occurrence of CMVD in elderly female patients.
