To improve the inulinase application in biotechnology, the characteristic of inulinase from Kluyveromyces marxianus YX01 was investigated. The inu gene of K. marxianus YX01 was transformed into Pichiapastoris GS115 host cells with molecular biology techniques. Then we achieved the heterologous expression of exo-inulinase whose molecular mass was about 86.0 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Furthermore, six His-tag was added to the inulinase and a two-step method was applied in the purification of inulinase, including concentration via dialysis by polyethylene glycol 20 000 and metal Ni-NTA Agarose affinity adsorption. The purification factor of purified protein was 3.6 and the recovery rate of enzyme activity was 33.1%. We characterized the purified inulinase. The optimum temperature was 60 degrees C and pH was 4.62. When inulin and sucrose were used as substrates, the K(m) and V(max) values were 80.53 g/L vs 4.49 g/(L x min) and 183.10 g/L vs 20.20 g/(L x min), respectively. In addition, metal ions including Mn2+, Ca2+, Cu2+, Zn2+ and Fe2+ exhibited different degrees of inhibition on the enzyme activity, and Cu2+, Zn2+ and Fe2+ exhibited the most significant inhibition. Our findings might lay a good foundation for industrial application of inulinase.

Objectives To assess the relationship between amniotic fluid erythropoietin(EPO)and neona-tal adverse outcome in fetal growth restriction(FGR)pregnancy labored during 28-36 gestational weeks. To explore the clinical application in timing of delivery. Methods The retrospective research had recruited 87 patients with single pregnancy complicated FGR,of which the gestational weeks range from 28 weeks to 36 weeks. All subjects were collected from amniotic fluid at cesarean section or within a week of cesarean section. Amniotic fluid EPO were detected according to the classical definition. We categorized EPO < 27 IU/L as an normal state,whereasE-PO≥27 IU/L as an abnormal state. The relationship between amniotic fluid EPO with biophysical profile, the flow velocity waveform/blood gas parameters of the umbilical artery, and the neonatal adverse outcome were observed. Results For FGR pregnant women who chose 28-36 weeks for delivery, the incidence of neonatal adverse out-comes was significantly higher in the amniotic fluid EPO increased group than that in normal concentration group (χ2= 9.49,P = 0.002). Pearson analysis showed that amniotic fluid EPO concentration was negatively correlated with umbilical artery pH(P<0.001,r=-0.908)and base excess(P<0.001,r=-0.624).However,it was pos-itively correlated with PCO2(P<0.001,r=0.631),whereas there was no significant correlation between amniotic fluid EPO concentration and PO2(P=0.068,r=-0.197).In addition, neither biophysical profile nor flow velocity waveform has difference in amniotic fluid EPO concentration. Conclusions The abnormal increased amniotic fluid EPO in FGR pregnant women who delivered before 36 gestational weeks were closely related to the adverse out-come of the newborn. The amniotic fluid EPO is expected to be an additional indicator of fetal hypoxia, which can help determine the time of birth.

OBJECTIVE: To explore the combined effect of noise and other occupational hazards on hearing impairment and electrocardiograph(ECG) of workers in automobile manufacturing enterprises. METHODS: A judgment sampling method was used to select 3 434 workers as study subjects from an automobile manufacture enterprise. According to the exposure to different types of occupational hazardous factors, they were divided into noise series group, noise dust group, noise welding dust group, noise benzene series group, noise nitrogen oxide group and control group, with 716, 693, 1 540, 45, 195 and 245 cases in each group, respectively. The subjects were examined with pure-tone hearing test and ECG. RESULTS: The total rate of high frequency hearing loss was 11.2%(385/3 434), and high frequency hearing loss was the main type of hearing loss among workers in each group. The simple high-frequency hearing loss rates of noise group, noise dust group, noise welding dust group, noise benzene series group, noise nitrogen oxide group and the control group were 10.3%, 11.5%, 12.3%, 26.7%, 10.3% and 4.1% respectively, and the rate of total hearing loss were 13.7%, 14.6%, 14.6%, 31.1%, 16.9% and 6.5% respectively. Compared with the control group, the rate of simple high-frequency hearing loss and total hearing loss were higher in the other 5 groups(P<0.005). The rate of high-frequency hearing loss in noise group was lower than that in noise benzene series group(P<0.005). The rate of arrhythmia was 20.9%(717/3 434). Arrhythmia was the main type of ECG abnormality in all groups. The rates of arrhythmia in noise group, noise dust group, noise welding dust group, noise benzene series group, noise nitrogen oxide group and control group were 16.9%, 16.6%, 27.9%, 17.8%, 13.8% and 6.9%, respectively. The rate of arrhythmia in noise welding dust group was higher than that in noise group(P<0.005). CONCLUSION: The combined effect of noise and benzene series increases the risk of high-frequency hearing loss in workers. The combined effect of noise and welding dust increases the risk of arrhythmia in workers.
