OBJECTIVE: To establish a HPLC method for the determination of the content of Demethylbellidifolin in different parts of Swertia davidi Franch. METHODS: The analysis was carried out on Hypersil C18 column (150mm?4.6mm,5 ?m) at room temperature with mobile phase consisted of CH3OH-0.5%H3PO4(56∶44) at a flow-rate of 1.0mL?min-1.The detection wavelength was set at 254 nm. RESULTS: The linear range of Demethylbellidifolin was 0.52~2.60?g (r=0.999 4) and the average recovery was 99.77%(RSD=0.95%).CONCLUSION: The method is simple, rapid, reproducible, and suitable for the determination of the content of Demethylbellidifolin in Swertia davidi Franch..
2.RP-HPLC Analysis and Pharmacokinetics Study of Serum Carthamin in Mice

OBJECTIVE:To establish an RP-HPLC analysis of Carthamin in mice and to study its pharmacokinetics.METHODS:The serum concentration of Carthamin was determined by RP-HPLC.The blood concentration-time curve was established and the main pharmacokinetic parameters were computed.RESULTS:The linear range of Carthamin was 0.558~55.8 ?g?L-1(r=0.999 2),with the lowest limit of detection at 0.005 ?g?L-1Carthamin in vivo assumed two-compartment model and rapid absorption.CONCLUSION:The proposed method is simple,sensitive and reproducible,and it met the standard for pharmacokinetic study.
3.Establishment of family health assessment index system in Shanghai urban communities

Objective To establish a standardized and feasible family health assessment index system based on the family practice system in Shanghai urban communities.Methods On the basis of literature review and special topic discussion, a preliminary index system framework was developed and an expert consultation questionnaire was compiled.Twenty one experts of general practice, medical specialties, health administration and public health management were invited and two rounds of improved Delphi expert consultation were conducted.The results of statistical analysis were summarized and the evaluation index system was established.Results The evaluation index system included 5 first-level indicators, 17 second-level indicators and 63 third-level indicators.The weight coefficients of first-level indicators in basic information, family structure, family function, family resources and health status of the family members were 0.199 6, 0.197 5, 0.201 7, 0.185 0, 0.216 2, respectively.In the two rounds of consultation, the positive coefficient of experts was both 100%, the degree of expert authority was 0.867 and 0.880, and the coordination coefficient was 0.294 and 0.415, respectively (P<0.05).The operational coordination coefficients were 0.320 and 0.402 (P<0.05), respectively.Conclusion The family health assessment index system constructed by the Delphi expert consultation method can provide reference for family doctors to carry out family health assessment in urban communities.
4.Determination of Quercitrin in Saxifrage by HPLC

OBJECTIVE:To establish a HPLC method for the determination of quercitrin in Saxifrage.METHODS:The analysis was performed on C18 column(150mm?4.6mm,5?m),the mobile phase was MeOH-0.2%H3PO4 (45∶55)with a flow rate at 1.0ml/min and wavelength at 350nm under room temperature.RESULTS:There was a good linear relationship when the sample size of quercitrin was at a range of 0.40?g~2.00?g(r=0.9 996),the recovery was 95.33%with RSD at 2.80%.CONCLUSION:This method was simple,stable,fast,and reproducible and without the interference of impurity,which can be used for the content determination of quercitrin in Saxifrage.

Objective To observe effects of Genistein and 5fluorouracil(5-FU) on human colon carcinoma cell line Colo320.Methods The MTT assay and median-effect principle were used.Results The two drugs were antagonistic at higher concentrations and synergistic at lower concentrations,The sequence and time of drug administration can influence the effects of the two drugs on Colo320.Conclusion The two drugs were cooperated at lower concentrations and antagonistic at higher concentrations.The sequence and time of drug administration were also important for their effects on the cells.
7.Simultaneous determination of three homoisoflavonoids in Ophiopogon japonicus by HPLC.

Three homoisoflavonoids, 6-aldehydo-3-ophiopogonanone A, methyl ophiopogonanone A and ophiopogonanone A from Ophiopogon japonicus were analyzed simultaneously by HPLC with acetonitrile-water containing 0.5% H3 PO4 (58:42) as the mobile phase, and the detection wavelength was set at 296 nm (0-14 min) and 275 nm (14-22 min). The mean recoveries of three homoisoflavonoids were 99.41%-99.86% (RSD 0.82%-1.05%). The linear response ranges of. 6-aldehydo-3-ophiopogonanone A, methyl ophiopogonanone A and ophiopogonanone A were 0.165-0.990 microg (r = 0.999 9), 0.153-0.918 microg (r = 0.999 9), and 0.270-1.620 microg (r = 0.999 9), respectively. This method was certified to be accurate and reliable and can be used for quality control of O. japonicus.

8.Determination of twelve ginsenosides in Panax ginseng by HPLC.

OBJECTIVETo determine the contents of twelve ginsenosides in the root of Panax ginseng by HPLC.
METHODThe analysis is carried out at room temperature on a Luna NH2 column (4.6 mm x 150 mm, 5 microm) eluted with acetonitrile and water as the mobile phases in a gradient elution. The flow-rate was 1.0 mL x min(-1), the detection wavelength was 203 nm.
RESULTTwelve ginsenosides (Rh2, Rh1, Rg2, Rg3, Rg1, Rf, Re, Rd, Re, Rb2, Rb3, Rb1) were separated at baseline within 60 min with good linearity (r > or = 0.999 5). The recovery rates were 98.1%, 95.3%, 96.1%, 95.6%, 97.3%, 98.6%, 98.0%, 96.4%, 96.1%, 97.6%, 96.8%, 96.9% (RSD < or = 3.0%).
CONCLUSIONThe method was simple,fast and could control the quality of P. ginseng effectively.

9.Effects of different concentrations of desflurane on electrophysiological stability of hearts in female patients

Objective To evaluate the effects of different concentrations of desflurane on the electrophysiological stability of hearts in female patients.Methods Forty female patients,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,aged 20-50 yr,weighing 45-77 kg,scheduled for elective surgery with general anesthesia,were divided into 2 groups (n =20 each) using a random number table method:0.6 MAC group (group D1) and 1.3 MAC group (group D2).The intravenous access was opened after admission to the operating room,midazolam 0.1 mg/kg,vecuronium 0.1 mg/kg,fentanyl 3 μg/kg and etomidate 0.3 mg/kg were injected intravenously,and mechanical ventilation was performed after tracheal intubation.The concentrations of desflurane reached 0.6 and 1.3 MAC in D1 and D2 groups,respectively.Twelve-lead electrocardiograms were collected before anesthesia induction (T1),at 5 min after intubation (T2) and at 20 min after reaching the set concentration of desflurane (T3).The QT interval and Tpe interval were measured,the QTc interval,Tp-e/QT ratio and index of cardiac electrophysiological balance were calculated,and the development of arrhythmia was also observed.Results The QT interval and QTc interval were significantly longer,and the Tp-e/QT ratio was lower at T3 than at T1,2 in two groups (P< 0.05).There was no significant difference in QT interval,Tp-e/QT ratio or index of cardiac electrophysiological balance at each time point between two groups (P>0.05).No arrhythmia was found in neither group.Conclusion The clinical concentration of desflurane affects the electrophysiological stability of hearts to some extent in female patients without causing arrhythmia.
10.Potential clinical application of Kisspeptin in reproductive endocrinology.

Gonadotropin-releasing hormone (GnRH) plays an important role in the process of reproduction. Studies have shown that a family of peptides Kisspeptin can act on GnRH-related neurons, stimulating the secretion of GnRH, and activating the hypothalamic-pituitary-gonadal axis. Both animal experiments and clinical studies have shown that exogenous administration of Kisspeptin is able to induce physiological GnRH release in healthy individuals and those with endocrine-disorders, which brings great hope for treatment of reproductive endocrine diseases. The effect of Kisspeptin is similar to the physiological process in induction of ovulation and ovum maturation, leading to high security and efficiency for women receivingfertilization. Kisspeptin is involved in trophoblast invasion, so it may be useful for predicting pregnancy outcomes. In addition, Kisspeptin is the key hormone in the onset of puberty acting as a signal transducer in metabolism and reproduction, so it provides some directions for studies of polycystic ovary syndrome, hyperprolactinemia, and other metabolic-related reproductive endocrine diseases. This article reviews the character of Kisspeptin and the prospect of its application in treatment of reproductive endocrine diseases.
