1.A preliminary study on understanding the multiple physiological parameters during sleep in light of traditional Chinese medicine

Objective This study aimed to understand the continuous physiological parameters during sleep under the guidance of Chinese medical theory and clinical practice and to establish the method to diagnose the physical and mental status by analyzing nocturnal sleep data.Methods More than 2000 subjects were recruited in the nocturnal sleep examination using the micro-movement sensitive mattress sleep monitoring system.Based on the analysis of the sleep indices and nocturnal physiological parameters combined with clinical data,a hypothesis was put forward that sleep cycle could reflect the circulation status of qi and blood,as well as some preliminary exploration on revealing the status of qi,blood,yin and yang.Results Sleep structure could reflect the status of qi and blood circulation in different meridians according to the traditional Chinese time unit.Sleep structure and sleep cycle could show corresponding changes if there is morbidity in some meridian,zang-viscera and fu-viscera.Heart rate,respiration rate and other physiological parameters could reflect the alternative predominance of yin and yang.Conclusion Interpreting the physiological parameters during sleep could be a supplementary diagnosis method for Chinese medicine syndrome differentiation.
2.Suture anchors fixation in the treatment of acute distal avulsion fracture of patella

Objective To investigate the feasibility and clinical effects of suture anchors technique for acute distal avulsion fracture of patella. Methods From January 2007 to July 2009, 33 patients(18males and 15 females, ranging from 28 to 72 years, with an average of 49.3 years) were treated with the suture anchors technique. The average time from injury to surgery was 3.5 days(2 to 7 d). Injury reason: traffic accident in 12 cases, injured in daily life in 11 cases, sports injury 9 cases, no clear trauma history in one case. The Bostman evaluation system, follow-up X-ray, and knee mobility were used to analyze the clinical effects of suture anchors technique. Results All patients obtained the follow-up and the average time was 16.5 months(12 to 23 months ). One patient suffered foreign body reaction, and his wound healing was good after operation. The remaining patient wound healing was good. No complications were found in all patients,such as popliteal fossa artery, tibial nerves or peroneal nerve complication. Bostman patella fracture functional score 27.3±2.3 points at the sixth months follow up, 28 cases were rated as excellent, 5 cases were good, and the excellent and good rate was 100%. Six months later, X-ray showed bone healing, knee activity recovered well. The Range of Motion was extension for -2.5°±2.1° and flexion for 122°±5.3°. Conclusion Suture anchors fixation in treatment of acute distal avulsion fracture of patella has a good efficacy, safety and reliability advantages, and without the second operation. This technique provides an alternative in lower pole patella fracture.
3.Effects of taurine on the change of apoptosis induced by IL-1?, TNF-? and IFN-? in rat pancreatic islet cells

AIM: To investigate the changes of apoptosis in isolated pancreatic islet cells, insulin secretion, expression of Bcl-xL and Bax induced by combination of IL-1?, TNF-? and IFN-?, and effects of taurine on them. METHODS: Isolated pancreatic islet cells from Wistar rat were incubated in monolayer in vitro. NO-2/ NO-3 production, NOS activity, insulin secretion, the protein expression of Bcl-xL and Bax, percentage of islet cell apoptosis and DNA fragmentation in pancreatic islet cells incubated with combination of IL-1?, TNF-? and IFN-? were measured, and the effects of taurine on the changes of them were further investigated. 4.Study on positive rate of blocking antibody in women with recurrent spontaneous abortion administered by route and frequency of paternal lymphocyte immunotherapy

Objective To investigates factors affecting the positive rate of blocking antibody treated by paternal lymphocyte immunotherapy in patients with recurrent spontaneous abortion (RSA).Methods From January 2008 to August 2012,326 RSA cases undergoing treatment in Infertility Center of Qilu Hospital were studied retrospectively.Those patients were divided into 2 groups randomly:260 cases in intradermal injection group were administered via bilateral forearm intradermal injections for immunotherapy once 21 days,then the blocking antibody was determined after 2 (23 cases),3 (73 cases),4 (74 cases),5(90 cases) times respectively,while in subcutaneous injection group,the 66 cases were administered via subcutaneous injection once 21 days,the blocking antibody measured after 3 times; In both cases,the blocking antibody was all determined 2 weeks later.The positive rate of blocking antibodies and the rate of successful pregnancy was recorded,and then followed up after the blocking antibody turning positive.Results (1)Positive rate of blocking antibodies:the positive rate of blocking antibodies were 17% (4/23),58% (42/73),72% (53/74) and 84% (76/90) in the 2,3,4,and 5 times of intradermal injection group,respectively (P < 0.05).In subcutaneous injection group,the positive rate of blocking antibodies was 38 % (25/66),which was significantly lower than that in group intradermal injection receiving 3 times immunotherapy (P <0.05).(2) The rate of pregnancy:the 176 patients out of 200 patients were pregnant when antibody was positive after immunotherapy,with 71.6% (126/176)of patients gained successful pregnancy(the length of pregnancy more than 5 months).Conclusions The route and frequency of administration of immunotherapy could influence the positive rate of blocking antibody.The rate of successful pregnancy will be increased after blocking antibody turning positive.
5.Effects of hyperbaric oxygen on C-reactive protein and tumor necrosis factor-α in patients with moderate and severe traumatic brain injury and its therapeutic efficacy

Objective To investigate effects of hyperbaric oxygen (HBO) therapy on C-reactive protein (CRP) and tumor necrosis factor-α (TNF-α) levels in patients with moderate or severe traumatic brain injury (TBI)and to analyze its therapeutic efficacy. Methods One hundred and eight patients with moderate or severe TBI were randomly divided into a control group (54 cases) and an HBO adjunctive therapy group (HBO group, 54 cases).Both groups received essential neurosurgical treatment and conventional drug treatment, and the HBO group was given one session of HBO therapy in addition. Serum CRP and TNF-α were detected, and the scores on the Glasgow coma scale (GCS) were measured before and after treatment. CRP was detected by turbidimetric immunoassay and TNF-α using ELISA. Glasgow outcome scale (GOS) scores were evaluated in a follow-up 6 months after injury. Results Average CRP, TNF-α and GCS measurements showed no statistically significant difference between the groups before treatment. After treatment, CRP and TNF-α were significantly lower and GCS scores significantly better in both groups, but patients in the HBO group were, on average, significantly better than the controls on all three measures.Six months later, GOS evaluation gave a significantly larger number of patients with a better prognosis in the HBO group compared with the controls. Conclusion HBO therapy can significantly decrease serum CRP and TNF-α after severe TBI, thus enhancing therapeutic efficacy.
6.In vitro growth characteristics of bone marrow mesenchymal stem cells derived from patients with multiple myeloma and significance of TGF-β1 gene expression

Objective To investigate the expression and significance of transforming growth factor-β1 (TGF-β1) gene in bone marrow mesenchymal stem cell (BMMSC) derived from patients with multiple myeloma (MM).Methods BMMSC of 7 MM patients and 10 patients with iron deficiency anemia were cultured in vitro.The morphology of BMMSC was observed and the growth curve was portrayed according to the daily results of BMMSC proliferation.Total RNA was extracted from BMMSCs and transcription of TGF-β1 gene in BMMSC was measured by reverse transcription-PCR.Results The proliferative activity of BMMSC was not significantly different between the two groups,but expression of TGF-β1 gene of BMMSC was higher in MM patients (0.01241±0.00419) than the control group (0.00122±0.00030) (t =3.218,P < 0.05).Conclusion The abnormally high expression of TGF-β31 gene in BMMSCs could contribute to the pathogenesis of MM.
7.Biomechanical analysis in the early period of flexor tendon healing after decimeter wave therapy

Objective To investigate the biomechanicai characteristics of flexor profundus tendons repaired after decimeter wave therapy, and to observe the effect of decimeter wave therapy on early active mobilization. Methods A total of 56 Leghorn chickens were randomly divided into a therapy group and a control group with 28 chickens in each. The 3rd and 4th toes of their left feet were employed for the establishment of a tendon injury model. The flexor profundus tendons were cut and repaired. Gypsum support was applied and fixed with an adhesive plaster after the operation. The operated sites on toes Ⅲ and Ⅳ were exposed. The external fixation was removed 3 weeks later and the chickens were left free to move. Decimeter wave therapy ( frequency 915 MHz, power 8 Watts) was ap-plied for 10 minutes once daily on the left foot of each chicken in the therapy group from day 1 until 3 weeks after the operation. Sham decimeter wave therapy was applied to chickens in the control group. Four chickens from each group were randomly selected at the 1st, 7th, 10th, 14th, 18th, 21st and 28th days for biomechanical analysis. Biome-chanical parameters including tensile strength of rupture (Pmax), elongation ratio at rupture (δimax) and the tensile adhesion strength of the rupture zone (W0>) were observed at each time point. Results At the 7th, 10th, 14th, 18th, 21st and 28th day after the operation, the differences in Pmax, δmax and W0 between the therapy and control groups were statistically significant. The results of the therapy group were better than those of the control group. Conclusions Local decimeter wave therapy after flexor tendon repair can promote intrinsic healing and reduce ex-trinsic healing. The speed and quality of healing are improved. The elasticity and tenacity of the injured tendons are enhanced. Therefore decimeter wave therapy is helpful for early active mobilization training.
8.PROGNOSTIC SIGNIFICANCE OF N1 STAGE IN NON-SMALL CELL LUNG CANCER

Objective To investigate the prognostic significance of N1 stage in non-small cell lung cancer (NSCLC) by comparing is survival with N0 and N2 stages. Methods From Jan. 1982 to Dec. 1994, 138 NSCLC patients with surgical-pathological N1 stage had received complete mediastinal lymph node dissection. Two subgroups of N1 node were identified by using New Regional Lymph Node Classification for Lung Cancer Staging. Their prognostic significances were tested and 5-year survival rates were compared with those of surgical-pathological N0 (307 cases) and N2 (176 cases) patients received radical operation during the same period. Results The overall 5-year suvival rate of N1 patients was 30.4 %. five-year survival was significantly better when N1 involvement was intralobar as compared with extralobar involvement, 50.3 % versus 26.5 % (P=0.001). Intralobar N1 5-year survival was similar to that of N0 (51.4 % vs 50.3 %), and extralobar N1 5-year survival was similar to that of N2 with singl focus (26.5 % vs 23.5 %). Conclusion N1 stage in NSCLC is a compound with two subgroups; the prognostic significance of intralobar subgroup is related to N0 stage and extralobar subgroup is related to that of single focus N2.
9.Optimization on Extraction Process of Brassicae Radix Cream by Uniform Design

This study was aimed to find out the optimum parameters to decoction extraction process of Brassicae Radix cream . With methods of integrated score and uniform design , contents of the total extract and the total polysaccharide were set as the target to investigate the optimization process of extraction. The experiment showed that the optimum extraction condition was as follows: ten folds of water, four times decoction at boiling tempera-ture , one hour for each time . It was concluded that the optimum parameters to decoction extraction of Brassicae Radix were stable and feasible, which can be used for the preparation and reference for Brassicae Radix cream in Tibetan clinics .
10.THE EFFECT OF SOYBEAN ISOFLAVONE ON LIPID PEROXIDATION AND LIVER ULTRASTRUCTURE IN OVARIECTOMIZED RATS

Objective To study the effects of soybean isoflavone(SI) on lipid peroxidation and liver ultrastructure in ovariectomized rats. Methods Seventy female Sprague-Dawley(SD) rats were randomly divided into 7 groups according to the levels of total cholesterol(TC) in serum: hyper-lipoid group,estrogen group,low-dose SI group,middle-dose SI group,high-dose SI group,sham group and normal control group.After bilateral ovaries were extirpated except sham and normal control groups for a week,the estrogen,different doses of SI or deionized water were fed with intragastric administration for 12 weeks.The rat body weight was weighted once per week and blood samples were collected at the time of ovariectomization,the 4th and 8th weeks after administeration at the time of being killed.The serum TC,triglyceride(TG),high or low density lipoprotein-cholesterol(HDL-C,LDL-C),lipoprotein(a) and antioxygen enzyme were assayed. Results LDL-C levels in SI intervention groups were significantly lower than that in hyper-lipoid groups but higher than that in normal control group;the levels of lipoprotein(a) were changed;there was almost no effect on HDL-C in serum.SI could retain integrity and ultrastructure of hepatocyte;there was similar structure in high dose SI group and normal control group.An obvious damage was detected in hyper-lipoid group.Conclusion SI can improve lipoprotein(a) and lipid peroxidation in ovariectomized rats.A continuous intervention with high-SI can reduce LDL-C to normal lever and retain integrity and ultrastructure of hepatocyte.
