Chinese Pharmacological Bulletin 1987;0(01):-
Aim To study the regulating effect of Tongxinluo on HIF-1?,VEGF that Generated by the Human brain microvascular endothelial cells induced by ?-amyloid 1-42. Methods Human brain microvascular endothelial cells were pre-incubated for 4h by Tongxinluo, then injured by ?-amyloid 1-42. VEGF protein and HIF-1? were detected by Western blot. Result In the A? treated Human brain microvascular endothelial cells, viable cells decreased. The expression of VEGF was Reduced, and HIF-1? elevated. After the cells were treated with Tongxinluo, all the above indexes were improved. Conclusion Tongxinluo can enhance the VEGF protein expressions by the HIF-1? and protect the Human brain microvascular endothelial cells.

In order to improve the technique for histochemical demonstration of the different lymphocyte patterns by alpha-naphthyl acetate esterase (ANAE) activity from a single microscopic slide, 10 kinds of comparative experiments were performed. The results showed as follows. The peripheral blood mononuclear cells first fixed in 0.4% glutaldehyde and postfixed in 15% formol calcium gave the best effect on preserving the enzyme activity and on connterstaning of the leucocyte nuclei. During formol calcium fixation, the erythrocytes were dissolved from the diluted blood film with PBS more easily than from the blood smear without any dilution. The lymphocytes were distributed more uniformly and densely in the former specimen than in the latter. When incubated with 0.5 mg alpha-naphthyl acetate/ml medium in hexoazotized pararosaniline for 1(1/2)~2 hrs at 37℃, the distinct ANAE patterns of circulating lymphocytes were more readily identified. In 28 healthy adults the differential counts of the lymphocyte ANAE patterns from the diluted blood specimen was similar to those found from the specimen of isolated lymphocytes. The focal pattern was about 56%, the diffuse pattern about 25% and the negative pattern about 19%; while the isolated lymphocyte specimen was compared with the diluted blood film from the same individuals, the focal pattern was decreased insignificantly, the diffuse pattern increased and the negative pattern decreased significantly.

Objective To explore the effects of different swimming training intensities on the expression of glial fibrillary acidic protein (GFAP) and basic fibroblast growth factor (bFGF) around a cerebral infarct. Methods The intraluminal thread method was applied to establish left middle cerebral artery occlusion (MCAO) for 2 h in 180 rats. They were then reperfused for 3,7 and 14 days. The rats were divided into four groups ( Ⅰ -Ⅳ) according to the intensity of the swimming training they were required to do,plus a control group and a sham operation group. The rats in training group Ⅰ swam 5 min, once daily; group Ⅱ trained for 5 min twice daily; group Ⅲ swam for 10 min once daily and group Ⅳ trained for 10 min twice daily.Neurological function was evaluated using Bederson's test, the cerebral infarction volume was calculated by tripeny tetrazolium chloride (TTC) staining. The expression of GFAP and bFGF around the infract were detected using immunohistochemistry.ResultsThe average Bederson scores of the exercise training groups on the 7th and 14th days were significantly lower than that of the control group. The infarction volumes of each exercise training group were significantly lower than that of the control group, and cells positive for GFAP and bFGF in all training groups were significantly more numerous on the 3th,7th and 14th day after MCAO,especially in training group Ⅳ.Conclusion Intensive exercise can adjust the expression of GFAP and bFGF and promote the repair of brain damage after cerebral ischemia and reperfusion.

OBJECTIVE: To establish a RP-HPLC method for the simultaneous determination of Protocatechuic acid and Protocatechuic aldehyde in Fuxuekang granules. METHODS: The chromatography was carried on Achirom Bond-1 C18 column(250 mm?4.6 mm,5 ?m). The mobile phase was composed of methanol - water (20∶80,pH=2.80 adjusted by glacial acetic acid) at a flow rate of 1.0 mL?min-1. The detection wavelengths were programmed at 256 nm for Protocatechuic acid and at 280 nm for Protocatechuic aldehyde. RESULTS: The linear ranges for Protocatechuic acid and Protocatechuic aldehyde were 5.49~73.22 ?g?mL-1(r=0.999 9) and 4.81~64.13 ?g?mL-1(r=0.999 9), respectively, and their average recoveries were 96.86%(RSD=2.52%,n=9) and 97.55%(RSD=3.82%,n=9) respectively. CONCLUSION: The method is simple, sensitive, and reproducible, and it is applicable for the quality control of Fuxuekang granules.

In the present study the effects of electroacupuncture (EA) on substance P (SP), somatostatin (SOM) and serotonin (5-HT) immunoreactivity (IR) of APUD cells and nerve fibers in the pylorus and in the skin of "Zusanli" acupo- int of rats were studied using immunohistochemical technique. Forty rats were divided into 20 pairs, one of each pair for EA (bilateral "Zusanli" acupuncture for 20 min), the other one for control. After EA the pain threshold was mark- edly increased(P

The pathogenesy of diabetic peripheral neuropathy(DPN) is approached according to collaterals diseases theory in this study, indicated that defi ciency of both vital energy and yin is the chief pathologic foundation and obstruction of collaterals by blood stasis and phlegm is the critical element in DPN.Furthermore, highlight of differentiation of symptoms and signs, therapeutic principle and diagnosis and treatment based on differentiation are illuminated. This study has supplied a new idea for precaution and treatment of DPN.

Objective To explore the structure of ribosomal protein S7 ( RPS7) gene of giant panda ( Ailuropoda melanoleuca) and investigate its homologies with other already reported sequences, including Homo sapiens,Mus musculus,Rattus norvegicus and Bos taurus. Methods The cDNA of RPS7 was cloned from the giant panda by RT-PCR. The sequence data were analyzed by GenScan software. Blast 2. 1 was used to study the homology of the obtained RPS7 sequence with the gene sequences of other species; Open reading frame ( ORF) of the DNA sequence was searched using ORF finder software; Protein structure of the RPS7 sequence cloned was deduced using Predict Protein software. Results The full length of the sequence fragment was 589 bp containing an ORF of 585 bp. The deduced protein sequence showed that the protein was composed of 194 ami- no acids and its estimated molecular weight was 22. 126 85 ?103 with a pI of 10. 09. There were 7 different pat- terns of functional sites: one N-glycosylation site; two cAMP and cGMP-dependent kinase phosphorylation sites; four casein kinase C phosphorylation sites; one casein kinase Ⅱ phosphorylation site; two N-myristoylation sites; two amidation sites and one ribosomal protein S7e signature site in the RPS7 protein. Further analysis indicated that the sequence of RPS7 and the protein encoded were highly homologous to some mammals reported. Conclusion The complete coding sequence of RPS7 gene has been cloned through RT-PCR successfully, which is the first report on the RPS7 gene from the giant panda.
