2.Correlation of MRI and histopathology after partial resection of normal brain: an experimental study in rabbits

Objective To investigate the manifestations of MR imaging and histopathology of early postoperative normal brain, and to define the correlation between MRI and histopathology. Methods Thirty six New Zealand rabbits weighing 2.0 to 3.0 kg were divided into 10 groups according to the different postoperative days: 1 to 10 day. Six animals were in groups 3, 5, 7, 10, and 2 were in the other groups. A partial resection of the parietooccipital region was performed under the usual aseptic conditions after the animals were anesthetized intravenously with 3% pentobarbital (30 mg/kg). MR imaging procedures consisted of pre and postcontrast scanning were carried out on postoperative 1 to 10 day respectively. Brain tissue samples were obtained to be prepared for examinations immediately after MR scanning. Histopathological study was made under microscope and electron microscope. The findings of MRI were compared with pathology findings. Results (1) Surgical margin contrast enhancement on MR images could be seen 24 hours after surgery. (2) The degree of contrast enhancement increased gradually before 5 days postoperatively, and no remarkable changes were present from 5 to 10 days. (3) The disruption of blood brain barrier(BBB) is the main cause of contrast enhancement during the postoperative 3 days. After that period, mechanism responsible for contrast enhancement is the formation of neovascularity and broken BBB, an increase in the number of neovascularity play a predominant role in the contrast enhancement in normal postoperative brain tissue. Conclusion The characteristics of enhanced MRI present at the surgical margin followed a typical time course during the early postoperative period. The role of neovascularity and BBB disruption in the formation of contrast enhancement at surgical margin vary with time. Knowledge of the characteristics on postoperative MR images of the normal brain can help in differentiating benign changes from malignant gliomas residual.

AIM:To investigate the effects of Arg-Gly-Asp-Ser (RGDS) tetrapeptide on proliferation,apoptosis and caspase 3 expression in FN-stimulated HSCs in vitro. METHODS:[ 3H]-thymidine incorporation,Annexin-V/Propidium Iodide double-labeled flow cytometry(FCM),TUNEL,scanning electron microscope and transmission electron microscopy were employed to estimate the influence of RGDS on proliferation and apoptosis of HSCs. 4.Clinical Observation of TACE Combined with HIFU in the Treatment of Middle and Advanced Primary He-patocellular Carcinoma

OBJECTIVE To observe therapeutic efficacy and safety of transcatheter arterial chemoembolization (TACE) com-bined with high intensity focused ultrasound (HIFU) in the treatment of middle and advanced primary hepatocellular carcinoma (HCC). METHODS:76 patients with middle and advanced primary HCC were randomly divided into treatment group(36 cases) and control group(40 cases). Control group was given TACE alone,and treatment group was additionally given HIFU 2-3 weeks after TACE. Clinical efficacy,the content of alpha-fetoprotein(AFP)before and after operation,survival rate,survival period and ADR were compared between 2 groups. RESULTS:The efficiency rate and total effective rate of treatment group were 61.1% and 94.4%,which were significantly higher than those of control group(35.0%,77.5%),with statistical significance(P<0.05). The content of AFP in 2 groups decreased significantly after operation,with statistical significance(P<0.05);there also was statistical significance between 2 groups(P<0.05). Survival rate of treatment group was higher than that of control group 24 and 36 months after operation,with statistical significance (P<0.05). The median survival time was 11.3 months in treatment group and 9.2 month in control group,with no statistical difference(P>0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:Com-pared with TACE alone,TACE combined with HIFU in the treatment of middle and advanced primary HCC can improve long-term survival rate and the short-term efficacy,with good safety.
5.Clinical characteristics of gastroesophageal reflux disease in the aged patients

Objective To analyze clinical characteristics of gastroesophageal reflux disease(GERD) in aged patients for improvement of diagnosis and treatemcnt. Methods The reflux disease questionnaire was performed in patients diagnosed as GERD based on Montreal definition and classification as well as Rome Ⅲ criteria.All patients were divided into elderly group (≥65 years) and control group(<65 years). The incidence of hita[ hernia (HH), the frequencies of esophagitis (based on Los Angeles classification), clinical features, and quality of life were compared between two groups. Results There was no difference between two groups in male/female ratio and morbidity of HH(P>0.05). In comparison with control group, the frequency of esophagitis graded as LC or LD increased and extra-esophageal symptoms were higher in elderly group (P< 0.05), but the lower typical symptoms (heartburn and regurgitation) were seen in the elderly group(P<0.05). The scores of role physical, bodily pain and role emotional were higher in elderly group than those in control group (P<0.05). There was no significant differences between two groups in physical function, vitality,social functioning, mental health, and general health. Conclusion The elderly GERD patients often have lower score of typical reflux symptoms (heartburn and regurgitation) and high incidence of severer esophagitis, but their quality of life is not significantly influenced.
6.Expression and significance of Oct4 B1 in colorectal cancer stem cells

Objective:To investigate the expression and its possible role of Oct4B1 subtype of Embryonic stem cell transcription factor Oct4 in colorectal cancer stem cells. Methods: 3D microspheres were cultured by suspension culture to human colorectal cancer cell line SW480 cells. The 3D microspheres and SW480 cells were used as the research objects. Whether 3D microspheres were enriched cancer stem cells,we used the methods of cell differentiation experiments,soft agar cloning experiments,and the expression levels of cancer stem cells markers CD133,CD44 detected by flow cytometry. The expression levels of Oct4B1 mRNA were detected by RT-qPCR. Results:3D microspheres could differentiate into normal cancer cells. Compared with the parental SW480 cells,in vitro colony formation was significantly enhanced(P<0. 01),the percentage of positive cells of CD133 and CD44 were significantly increased ( P < 0. 01 ), the expression levels of Oct4B1 mRNA were obviously higher ( P < 0. 01 ) in 3D microspheres. Conclusion: Oct4B1 subtype of Embryonic stem cell transcription factor Oct4 in 3D microspheres enriched human colorectal cancer stem cells,which may be involved in the regulation of colorectal cancer stem cells.
7.Application of Planar Microelectrode Array Modified by Nano-structure Titanium Nitride on Dual Mode Neural Information Recording

The nano-structure TiN was modified on the laboratory self-made planar microelectrode array pMEA by magnetron sputtering method. The performance of modified pMEA was investigated. Research on neuroelectrical and neurochemical recording was studied in vitro. The impedance of the modified pMEA was decreased almost one order of magnitude, and the background noise level was reduced to ±6 μV. In the same testing environment, the signal-to-noise ratio (SNR) of modified electrodes was 1. 7 times of bare electrodes. The SNR of neuroelectrical recording on the brain slice of SD rats reached 10:1 , and the weak signal such as ±12 μV was separated easily. For neuroelectrical recordings, the detection limit of dopamine ( DA) solution reached 50 nmol/L with the 2:1 (S/N). During the concentration range of 0. 05-100 μmol/L, the linearly correlation coefficient of the DA oxidation currents was 0 . 998 . The modification of nano-structure TiN on pMEA reduced pMEA impedance and background noise level, meanwhile the SNR was increased. The weak signals of neuroelectrical and neurochemical recording were successfully recorded.
8.Proteome analysis in the process of inducing C2C12 cells differentiation towards osteoblast by BMP-2 based on iTRAQ technology

Objective To apply iTRAQ technology to observe changes in protein expression group in the process of inducing C2C12 cells differentiation towards osteoblast by BMP-2.Methods The myoblast C2C12 cells were seeded in BMP-2 induced differentiation system for differentiation induction.In the 7th day,differentiation protein was extracted and labeled with iTRAQ reagent.Then,mass spectrometric detection,data analysis of differentially expressed proteins,and analysis of biological information were carried out.Results 23 significantly differentially expressed protein spots were screened by BMP-2-induced myoblast C2C12 differentiated cell protein expression profile analysis,where the protein was labeled with iTRAQ reagent.8 protein points were up-regulated,and 15 protein points were down-regulated.Trend classification found that the above differential protein had differential expression in each period of C2C12 cell osteogenic differentiation (1-7 days).Part of up-regulated protein in the early differentiation period showed high expression level;part of up-regulated protein in the late differentiation period showed high expression level;similarly,part of down-regulated protein in the early differentiation period presented low expression level;part of down-regulated protein in the late differentiation period showed low expression level.Preliminary identification showed SERCA3,Cytochrome bS,S100A4,ATPase inhibitor and ATPIF1 presented dynamic changes,which suggests that these proteins may be related to inducing osteogenic differentiation mechanism.Conclusion The results of differential protein expression trend show the necessity of full monitoring of C2C 12 cells osteogenic differentiation and indicate that iTRAQ technology is an effective method of studying protein changes of cellular molecule.Five proteins including SERCA3,Cytochrome b5,S100A4,ATPase inhibitor and ATPIF1 can be used as candidate targets for osteogenic differentiation mechanism research.
9.The composition of vaginal microbiota in patients with cervical precancerous lesion

Objective To investigate the relationship between the composition of vaginal microbiota and the course of cervical precancerous lesion.Methods A total of 64 vaginal swabs were collected from 22 healthy women, 18 CINⅠ patients and 24 CINⅡ/Ⅲ patients who visited Obstetrics and Gynecology of the Second Xiangya Hospital of Central South University during July 2014 and July 2015.The Bacterial genomic DNA was extracted and the V3 and V4 hypervariable regions of 16S rRNA were amplified and high-throughput sequenced.The abundance and composition of vaginal microbiota were analyzed by Uparse, Mothur and LefSe statistical software.Results There was no significant difference in Alpha diversity index between CINⅡ/Ⅲ group(Chao:63±32;ACE:72±38;Simpson:0.70±0.27;Shannon:0.70±0.63) and control group ( Chao:48±24;ACE:54±25;Simpson:0.71±0.27;Shannon:0.65±0.58)(W=192,P=0.11;W=189,P=0.10;W=281,P=0.72;W=241,P=0.62).The ACE(85±37) and Chao(66±25) values of CINⅠgroup were significantly different from those of the control group (ACE:54±25;Chao:48±24)(W=99,P=0.006;W=113,P=0.02).At the phylum level, 78.69%(309 020/392 722) of the vaginal microbiota in the control group was Firmicutes, 16%(62 846/392 722) was Actinobacteria.Firmicutes was reduced to 64.86%(208 422/321 318) and Actinobacteria increased to 27.71%(89 040/321 318) in CINⅠgroup.The composition of vaginal microbiotain in CINⅡ/Ⅲ group was similar to those of control group.At the genus level, the composition of vaginal microbiota were similar between CINⅡ/Ⅲ group and control group, with Lactobacillus as predominant genus[71.81%(307 658/418 424)], Gardnerella[12.91%(55 299/428 424)], others such as Prevotella, atopobium were less.In the CINⅠ group, the abundance of Lactobacillus was decreased to 56.26%(180 787/321 318), Gardnerella was increased to 19.62%(63 057/321 318), and Listeria was increased to 7.7%(24 746/321 318).The composition of vaginal microbiota in the most samples was classified as CSTⅢ and CSTⅠ, with Lactobacillus inersand and Lactobacillus crispatus were dominant respectively.There was no significant difference in the composition of vaginal microbiota between the three groups(χ2=2.72, P=0.949).LEfSe analysis showed that the abundance of bacteria in CIN group and control group were varied.At the genus level, there were significant differences in the abundance of Geobacter, Atopobium and Ureaplasma (P<0.05, P<0.05, P<0.01, respectively).At the species level, there was significant difference in the abundance of Ureaplasma urealyticum serotype 9 (P<0.01).Conclusion The diversity and the composition of vaginal microbiota were similar between CIN patients and healthy women, but the abundances of some bacteria were varied, with Ureaplasma increased in patients with CIN.
10.Epidemiological characteristics of measles in Yichang from 2009-2021

Objective To investigate the epidemic situation of measles in Yichang from 2009 to 2021, and to provide scientific evidence for the control and elimination of measles. Methods Descriptive epidemiological methods were used to analyze the case data of measles cases reported by the notifiable infectious disease reporting system and measles surveillance system in Yichang from 2009 to 2021. Results A total of 364 cases of confirmed measles were reported in Yichang from 2009 to 2021, with an average annual incidence rate of 0.68/100 000. During these years, the incidence rates fluctuated from 0.5 to 28.87/100 000. The epidemic peak of measles was from January to July. Most cases were concentrated in five regions including Xiling, Wujiagang, Yiling, Dangyang and Yidu, accounting for 66.48% of all cases. More male cases were reported. The age of these patients ranged from < 1 month to 68 years, in which a great majority were infants (32.14%) and adults between 20 and 49 years old (48.08%). The five most common occupations of measles patients were scattered children, farmers, housekeeping or unemployed, workers and students, with the scattered children being the highest proportion accounting for 42.58%. There were 72 cases (19.78%) with the history of measles vaccine immunization, 113 cases (31.04%) without the immunization history, and 179 cases (49.18%) with unknown immunization history. Conclusion Incidence rates of measles in Yichang remains at a low level from 2009 to 2021. Young age groups and adults are the key groups for measles prevention and control. Strategies and measures such as immune strengthening for key areas and key populations will help improve the effect of measles prevention and control.
