1.Determination of Total Polyphenols in Guhong Injection

Objective To establish determination method for total polyphenols in Guhong Injection, and provide reference for internal quality control of Guhong Injection.[Methods]Using the Folin-Ciocalteu colorimetric method, according to the consistent degree of the maximum absorption wavelengths of hydroxysafflor yel ow A and kaempferol relative to Guhong Injection, we selected the appropriate reference and color reaction condition to content determination.[Rsults]With hydroxysafflor yel ow A as the standard, the content of total polyphenols in 10 batches of samples were 9.94, 9.55, 9.75, 9.67, 9.84, 10.03, 9.81, 9.52, 9.88, 10.09mg·mL-1. The average recovery of total polyphenols was 98.11% and RSD was 1.68%(n=6). [Conclusion]The established Folin-Ciocalteu method is simple, accurate, sensitive, and is reliable for detecting total polyphenols in Guhong Injection.
2.Identification of Host Factors Interacting with the Movement Protein of Apple Chlorotic Leaf Spot Virus by Yeast Two-Hybrid System.

In order to identify host factors which interact with the movement protein (MP) of Apple chlorotic leaf spot virus (ACLSV), ACLSV MP was cloned into the bait vector pGBKT7 and used to screen a cDNA library of Malus sylvestris cv. R12740-7A, which had previously been constructed by yeast two-hybrid sequencing transformation. The protein functions of the identified host factors were determined according to their gene annotations in GenBank. The result showed that the bait plasmid pGBKT7-MP showed no virulence or self-activating effect on yeast strain Y2H Gold. Sixty-nine interactor proteins were identified, which were divided into the following 10 classes according to their described functions: hydrolases; pathogenesis-related proteins; DNA binding proteins; phosphatases; ligases; proteins with catalytic activity; phenylalanine ammonialyases; peroxidases; NAD binding proteins; and proteins of unknown function. Bioinformatic analysis of gene homology suggested that phosphatases, pathogenesis-related proteins and glyceraldehyde-3-phosphate dehydrogenase A may play an important role in the interaction between virus and host. This study may provide a theoretical basis for the further study of viral pathogenesis and virus-host interaction mechanisms.

3.A Meta-analysis of randomized controlled trials on Xipayi Mouth Rinse for the treatment of halitosis

Objective:To evaluate the effectiveness of Xipayi Mouth Rinse in the treatment of halitosis.Methods:PubMed/MEDLINE,China National Knowledge Infrastructure (CNKI) (1979-2015) and Weipu Database for Chinese Technical Periodicals (1989-2013) were systemically searched.The methodological quality of the included trials was assessed based on the Jadad scale and the available data were analyzed with RevMan software (version 5.3).Results:8 studies involving 608 participants satisfied the minimum criteria for Meta-analysis.The evidence showed that the Organoleptic Score (OS) of Xipayi Mouth Rinse compared with placebo for treatment of halitosis was [WMD =-0.43,95% CI (-0.57,-0.29)],plaque index (PLI) [WMD =-1.59,95% CI (-1.67,-1.51)];the OS of Xipayi Mouth Rinse compared with western medicine [WMD =0.16,95% CI (0.05,0.28)],PLI [WMD =0.10,95% CI(0.03,0.18)];the total efficiency of Xipayi Mouth Rinse compared with normal saline was[RR:1.64,95% CI(1.11,2.42)].Conclusion:The current clinical evidence showed that the effectiveness of Xipayi Mouth Rinse in the treatment of halitosis is higher than the Placebo,but lower than western medicine.
4.Study on Sedative and Hypnotic Effects of Different Eluents of Shuangxiatang

This article was aimed to study the sedative and hypnotic effects of different eluents of Shuangxiatang (SXT). The effects of SXT water decoction, water eluent, 20%, 70% and 95% alcohol eluent on spontaneous ac-tivity and the sleeping induced by subthreshold dose of pentobarbital sodium were measured. The results showed that the SXT decoction, 20% and 95% alcohol eluent can significantly decrease the number of rearing in mice with the percentage of 78.5%, 78.3% and 62.5%, respectively. SXT water eluent and 70% alcohol eluent can significantly decrease the spontaneous activity of mice (P < 0.01), the number of rearing (P < 0.01) and grooming time (P < 0.05). SXT water decoction can significantly shorten sleep latency (P < 0.05), prolong sleep time (P <0.05), and increase rates of sleeping in mice. SXT water eluent can significantly shorten sleep latency in mice (P< 0.05), increase rates of sleeping in mice. SXT water decoction and water eluent have the sedative and hypnotic effects. And the effects are more than alcohol eluents.
5.Isolation and identification of a Pp-LIM only A gene expressed during cold acclimation in Physcomitrella patens.

Cold acclimation can improve freezing tolerance. Here cDNA amplified fragment length polymorphism (cDNA-AFLP) was used to isolate differentially expressed cDNAs and a Pp-LIM only A cDNA was isolated and identified in the cold acclimation of Physcomitrella patens. Real-time RT-PCR indicated it is obviously up-regulated at 6 h, 12 h, 24 h, 48 h and 72 h after cold acclimation. After comparing the cDNA with the gene sequence, seven introns and eight exons were identified in the cDNA. The cDNA putatively encodes a protein of 345 amino acid residues and only contains one LIM domain which has highly similarity with the PDZ/LIM domains of the protein family in animals. We proposed that Pp-LIM only A be a new gene coding for the LIM-domain containing protein and enhance stability of cell membrane via their effects on cytoskeleton during cold acclimation in P. patens.

6.Targeted deletion of mouse Rad1 leads to deficient cellular DNA damage responses.

The Rad1 gene is evolutionarily conserved from yeast to human. The fission yeast Schizosaccharomyces pombe Rad1 ortholog promotes cell survival against DNA damage and is required for G(2)/M checkpoint activation. In this study, mouse embryonic stem (ES) cells with a targeted deletion of Mrad1, the mouse ortholog of this gene, were created to evaluate its function in mammalian cells. Mrad1 (-/-) ES cells were highly sensitive to ultraviolet-light (UV light), hydroxyurea (HU) and gamma rays, and were defective in G(2)/M as well as S/M checkpoints. These data indicate that Mrad1 is required for repairing DNA lesions induced by UV-light, HU and gamma rays, and for mediating G(2)/M and S/M checkpoint controls. We further demonstrated that Mrad1 plays an important role in homologous recombination repair (HRR) in ES cells, but a minor HRR role in differentiated mouse cells.

7.A randomized double-blinded placebo-controlled clinical trial of minodronate tablet in postmenopausal Chinese women with osteoporosis

Objective:To verify the efficacy and safety of daily oral minodronate in postmenopausal women with established osteoporosis.Methods:In this randomized, double-blinded, placebo-controlled trial, 262 postmenopausal women were enrolled. Patients were randomized to receive daily oral minodronate 1 mg with supplements of 500 mg calcium and 200 U vitamin D 3 ( n=130) or placebo ( n=132) with daily supplements of 500 mg calcium and 200 U vitamin D 3, for 48 weeks. The primary endpoint was the average bone mineral density (BMD) change in the lumbar vertebrae 48 weeks post-treatment. Secondary outcome measures was the incidence of vertebral fractures. Safety assessments included the rate of adverse events. Results:At the end of 48 weeks treatment, the average BMD change rate from baseline were: full analysis set results: (3.52±4.82)% in the minodronate group and (2.00±5.74)% in the placebo group; per-protocol set results: (3.99±5.05)% in the minodronate group and (2.07±6.20)% in the placebo group; the differences were all significant (all P<0.05). Vertebral fracture occured in 3 patients (2.3%, 3/132) in the placebo group, and 1 case (0.8%, 1/130) in the minodronate group ( P>0.05). The incidence of adverse events was 71.5% (93/130) in the minodronate group and 78.0% (103/132) in the placebo group ( P>0.05). Conclusion:Minodronate is effective and safe in the treatment of postmenopausal osteoporosis without severe side effects.
8.A benzenesulfonic acid-modified organic polymer monolithic column with reversed-phase/hydrophilic bifunctional selectivity for capillary electrochromatography

Here,a styrene-based polymer monolithic column poly(VBS-co-TAT-co-AHM)with reversed-phase/hydrophilic interaction liquid chromatography(RPLC/HILIC)bifunctional separation mode was success-fully prepared for capillary electrochromatography by the in situ polymerization of sodium p-styrene sulfonate(VBS)with cross-linkers 3-(acryloyloxy)-2-hydroxypropyl methacrylate(AHM)and 1,3,5-triacryloylhexahydro-1,3,5-triazine(TAT).The preparation conditions of the monolith were optimized.The morphology and formation of the poly(VBS-co-TAT-co-AHM)monolith were confirmed by scanning electron microscopy(SEM)and Fourier transform infrared spectroscopy(FT-IR).The separation perfor-mances of the monolith were evaluated systematically.It should be noted that the incorporation of VBS functional monomer can provide π-π interactions,hydrophilic interactions,and ion-exchange in-teractions.Hence,the prepared poly(VBS-co-TAT-co-AHM)monolith can achieve efficient separation of thiourea compounds,benzene series,phenol compounds,aniline compounds and sulfonamides in RPLC or HILIC separation mode.The largest theoretical plate number for N,N'-dimethylthiourea reached 1.7×105 plates/m.In addition,the poly(VBS-co-TAT-co-AHM)monolithic column showed excellent reproducibility and stability.This novel monolithic column has great application value and potential in capillary electrochromatography(CEC).
9.One Case of Hand-foot Syndrome Recurrence Induced by Sequential Paclitaxel Liposome for Injection with Doxorubicin Hydrochloride Liposome Injection and Literature Review

OBJECTIVE:To investigate the role of clinical pharmacists in the diagnosis and treatment of liposome-induced hand-food syndrome (HFS),and to provide reference for rational use of liposome preparation in clinic. METHODS :One case of elderly female patient with breast cancer ,admitted to our hospital suffered from HFS (grade 2)after treated with Doxorubicin hydrochloride liposome ;after successful therapy ,the patient had skin symptoms (grade 3)again due to Paclitaxel liposome ,and clinical pharmacist judged the recurrence of HFS. For symptomatic treatment ,stopping the treatment and external use of hormone was suggested ,and whole-process pharmaceutical care was provided. The pathogenesis ,differential diagnosis ,risk factors and therapeutic drugs of HFS were summarized based on literature review and 2 case reports in the database. RESULTS :The physicians adopted the suggestion of clinical pharmacists ;the patient ’s symptoms improved significantly on the third day and disappeared after 1 week. Combined with literature analysis and 2 case reports ,doxorubicin liposome metabolized more slowly than non liposomes in palms and soles of feet ,resulting in accumulation of doxorubicin in sweat duct and stratum corneum ,aggravating skin damage and leading to HFS. Sequential paclitaxel in liposome form may also lead to the accumulation in eccrine duct ,further caused skin damage and induced HFS. CONCLUSIONS :Clinical pharmacists actively participate in the diagnosis and treatment of ADR , which is conducive to the rehabilitation of patients. At same time ,combination or sequential of Paclitaxel liposome with PLD should be avoided ,as it can lead to ADR as HFS.
