1.Schistosomiasis control effect of agricultural integrated development in Tie-ban Marshland of Yangtze River

Objective To evaluate the schistosomiasis control effect of the comprehensive measures that focused on beach?land smoothing and tillage cultivation in Tieban Marshland,Wuhan Section of the Yangtze River. Methods From 2011 to 2015, before and after the comprehensive control,the Oncomelania hupensis snail situations in Tieban Marshland were investigated in spring annually,and the residents aged 6-65 years in Huayuan Community which was next to the marshland were examined to un?derstand their schistosome infection status. In late July,2014,the sentinel mouse surveillance was carried out. Moreover,the eco?nomic effectiveness of comprehensive development was evaluated. Results The schistosome infection rates of residents in 2011 (before the comprehensive control),2012(after the comprehensive control),and 2013 were 0.72%(3/414),0.37%(2/536)and 0.31%(1/326),respectively,and no schistosomiasis patients were found in 2014 and 2015. The snail area,the occurrence rate of frames with snails,and the average density of living snails in Tieban Marshland in 2005 decreased by 22.18%,97.83% and 98.25%,respectively compared to those in 2011,and no schistosome infected snails were found. No positive mice were found in the sentinel mouse surveillance. The annual net income of Tieban Marshland increased by 233.33%compared to that before the comprehensive development. Conclusion The endemic situation of schistosomiasis and snail situation declined significantly af?ter the comprehensive development in Tieban Marshland,and the economic and social benefits are significant.
2.Content of L-carnitine and its acylates in plasma of patients with diabetes and diabetes complications

Aim To investigate the changes of content of L-carnitine (LC ), acetyl-L-carnitine (ALC ) and propionyl-L-carnitine (PLC )in patients with diabetes and its complications.Methods By replicating meth-od of pre-column derivatization HPLC,the contents of plasma LC,ALC,PLC were detercted in normal sub-jects and in patients of diabetes mellitus,diabetic reti-nopathy, diabetic nephropathy, diabetic peripheral neuropathy,diabetic hypertension,diabetic coronary heart disease.Results The concentration of LC in normal subjects and in patients of diabetes mellitus, diabetic retinopathy, diabetic nephropathy, diabetic peripheral neuropathy,diabetic hypertension,diabetic coronary heart disease were (41.01 ±8.05 )μmol · L-1 ,(36.72 ±8.23 )μmol · L-1 ,(33.3 1 ±6.26 )μmol·L-1 ,(33.81 ±5.61 )μmol·L-1 ,(33.57 ± 6.67 )μmol · L-1 , (33.67 ±5.36 )μmol · L-1 , (32.87 ±6.05 )μmol · L-1 respectively.The plasma LC concentration in diabetic group was significantly lower than that of normal control group (P<0.05 ). Moreover,the LC concentration in other groups of dia-betic complications was lower than that in diabetic group, and LC concentration showed no significant difference in various groups of diabetic complications. There was no significant difference in the plasma con-centrations of ALC and PLC of diabetes group and the diabetic complication groups too.Conclusion In Chi-nese normal subjects,patients with diabetes and dia-betic complications,the plasma concentrations of LC in diabetic patients are lower than normal,and plasma concentration of LC in diabetic complications are lower than that in patients with diabetes mellitus.
3.Effects of microRNA-294 on inflammatory factor of sepsis by targeting triggering receptor expressed on myeloid cells-1

Objective To investigate the effects of microRNA-294 (miR-294) on tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6) and high mobility group box 1 (HMGB 1) secretion in sepsis by targeting triggering receptor expressed on myeloid cell-1 (TREM-1).Methods miRNA-294 was predicted to regulate TREM-1 specially through bioinformatics analysis.Mice macrophage cell lines RAW264.7 were cultured in vitro,the cells were divided into non-inflammatory stage and inflammatory stage,and the cells in the two stages were subdivided into five groups as follows:normal control (NC),NC mimic transfection (NCm),NC inhibitor transfection (NCi),miR-294 mimic transfection (miR-294m) and miR-294 inhibitor transfection (miR-294i) groups.The ability of miR-294 was confirmed with dual-luciferase activity assay.At non-inflammatory stage,the cells were transfected with mimic or inhibitor of miR-294 or NC using TurboFectTM siRNA Transfection Reagent for 48 hours,mRNA expression of TREM-1 was detected by real-time reverse transcription-polymerase chain reaction (RT-PCR).At inflammatory stage,6 hours after stimulation by lipopolysaccharide (LPS,1 mg/L),the concentrations of TNF-α,IL-6 and HMGB1 were determined by enzyme linked immunosorbent assay (ELISA),the protein expression of TREM-1 was determined by Western Blot.Results ① Dual-luciferase activity assay demonstrated that TREM-1 was the target of miR-294.② In non-inflammatory stage,the expression of TREM-1 mRNA (2-ΔΔ~) in miR-294m group was significantly lower than that of the NC and NCm groups (0.673 ± 0.049 vs.1.000 ± 0.003,0.915 ± 0.039,t1=2.184,t2=5.421,both P<0.001),the expression of TREM-1 protein (gray scale) was (50.00 ± 1.19)% of NCm group (t=41.586,P<0.001).③ In inflammatory stage,the concentrations of TNF-α (ng/L) in miR-294m group was significantly lower than that of the NC group (1 547.18 ±47.18 vs.2 702.11 ± 327.20,t=4.212,P=0.010),the concentrations of IL-6 (ng/L) was significantly lower than that of the NC and NCm groups (505.28 ± 33.33 vs.837.66 ± 69.43,918.72 ± 119.39,t1 =4.382,P1=0.015; t2=5.451,P2=0.021),the level of TREM-1 protein (gray scale) was (51.33 ±0.88)% of NCm group (t=63.368,P<0.001).Conclusion miR-294 reduce TNF-α and IL-6 secretion in LPS-induced RAW264.7 through inhibiting the expression of TREM-1 specifically.
4.Expression of Yes-associated protein in colorectal carcinoma and its effect on tumor cell proliferation

Objective To evaluate the expression of Yes-associated protein (YAP) in colorectal carcinoma and analyze its influence on tumor cell proliferation.Methods The expressions of YAP in 94paired colorectal carcinomas and pericancerous normal tissues were detected by using immunohistochemistry method.The expressions of YAP in colorectal carcinoma cell line HCT116 were inhibited with a YAP-spe-cific siRNA.Cell proliferation was then determined by methyl thiazolyl diphenyl-tetrazolium bromide (MTT) assay.Results The positive rate of YAP in colorectal carcinomas was significantly higher than that in pericancerous normal tissues [69.1% (65/94) vs 22.3 % (21/94),P < 0.001].The expression of YAP was associated with tumor Node Metastasis(TNM) stage and lymph node metastasis(P <0.05),but not associated with gender,age,tumor location and histological grade(P >0.05).After YAP-specific siR-NA was transfected into HCT116 using lipofectamine,the expression of YAP mRNA and protein in the experimental group were reduced by (78.2 ±2.1)% and (81.7 ± 1.5)%,respectively,with a statistically significant difference (t =67.55,91.601,P <0.01).The growth of HCT116 was significantly inhibited and the reduced rate of cell proliferation was (28.1 ± 1.6) %,(34.7 ± 2.4) % and (24.7 ± 1.2) % at the time point of 48 h,72 h and 96 h,respectively.Conclusions Expression of YAP was upregulated in colorectal carcinomas and downregulation of YAP expression could inhibit growth of colorectal carcinoma cells.YAP can be used as a new candidate target for diagnosis and treatment of a colorectal carcinoma.
5.Study of absence of theta oscillation of working memory dysfunction in temporal lobe epilepsy

Objective Temporal lobe epilepsy (TLE),frequently accompanied by working memory dysfunction,is an important form of adult epilepsy syndrome.Study on neural mechanisms of TLE with working memory dysfunction has important scientific value and clinical significance.Theta oscillations are synchronous activity of the brain in the 4-8 Hz frequency range.It is well recognized that there is a close relationship between the theta oscillations and working memory behavior.TLE patients with working memory dysfunction were taken as the research subjects,and the theta oscillations absence in TLE with working memory behavioral disorder was carried out,which provide the reference for further research into the neural mechanisms of TLE with working memory dysfunction.Methods The 34-channel electroencephalographs (EEGs) were recorded from TLE group (18 TLE patients) and control groups (18 healthy subjects) while subjects were performing visual working memory (delayed matching-tosample) tasks.The EEGs during the 3 s delay period was analyzed as experimental data.Fourier transform was used to assess the EEGs spectrum.The channel with the strongest spectrum was selected as the feature channel.Short-time Fourier transform algosiths was employed to calculate the time-frequency representation of the feature channel for the TLE and control groups.Frequency band with the strongest spectrum was selected for control group as the feature frequency band in working memory.Then,the topographical maps of the feature frequency band spectrum were calculated for the TLE and control groups.Results Compared with the control group,the working memnory behavioral performance of the TLE group was lower:accuracy of the TLE group was significantly lower than that of the control group (P<0.05),and reaction time of the TLE group was significantly longer than that of the control group (P<0.001).The feature channel in working memnory was frontal midline (Fz) and the feature frequency band was theta band.The Fz spectrum of the feature frequency for TLE was lower than that for control (P<0.05).The frontal spectrum (seven channels) of the feature frequency for TLE was lower than that of control (P<0.01).Conclusions Theta oscillations for TLE with working memory behavioral disorder is absent,which maybe one of the possible neural mechanism of TLE with working memory dysfunction.
6.Survey on cognitive attitude and participation willingness of public hospitals social evaluation based on stakeholders theory

Objective To survey the cognitive attitude and participation intention for stakeholders of public hospitals,in order to provide basis for public hospitals social evaluation.Methods 621 stakeholders of public hospitals were subject to the questionnaire survey.Results 76.7%(475/619) stakeholders held it as necessary to independently launch the public hospitals social evaluation,and held that the evaluation subjects selection should be professional(83.8%),independent(67.7%),authoritative (55.6%),and proactive(46.1%).The survey also recommended that social evaluation of public hospitals should have such indexes as government investment (80.2%),social supervision (76.0%),and information disclosure system(7 1.1%).The overall willingness rate of stakeholders to participate in the public hospital social evaluation was 7 1.6%.Conclusions Development of the social evaluation index system of public hospitals and diversified evaluation subj ects are key to developing the theory and practice system of public hospitals social evaluation.
7.Influence of exercise rehabilitation combined diet guidance on patients after heart valve replacement

Objective:To observe influence of exercise rehabilitation combined diet guidance on cardiac function and quality of life (QOL) in patients after heart valve replacement .Methods:A total of 120 patients after heart valve replacement were selected and randomly divided into control group (n=60 ,received routine health education ) and experiment group (n=60 ,received diet guidance and exercise rehabilitation based on routine health education ) .After discharge ,patients re‐ceived outpatient follow‐up for six months ,body weight ,cardiac function and QOL on six months after surgery were fol‐lowed up and observed in two groups ,and the results were statistically compared .Results:Compared with control group six months after discharge ,there were significant rise in left ventricular ejection fraction [ (51.75 ± 9.30)% vs .(57.73 ± 6.13)% ] andoxygenmetabolicequivalent [(7.05±1.85)METsvs.(11.34±2.18)METs],andeachitemscoreofQOL and total score of QOL [ (4.98 ± 0.40) scores vs .(6.49 ± 0.53) scores] ,P<0.05 or <0.01;and significant reduction in body weight [(67.75 ± 10.67) kg vs .(61.25 ± 10.34) kg] in experiment group ,P<0.01. Conclusion:Exercise rehabili‐tation combined diet guidance can avoid rapid increase in body weight after surgery ,promote recovery of cardiac function and improve quality of life after surgery in patients with heart valvular diseases .
8.Establishment of a porcine model of penetrating-suture pancreaticojejunostomy

Objective To establish a porcine model of penetrating-suture pancreaticojejunostomy.Methods Ten domestic pigs were selected,and after general anesthesia they underwent laparotomy and the pancreas was visualized.Then the pancreas was dissected at the level of superior mesenteric vascular,and the proximal pancreatic stump was sewed up.The anastomosis between the distal pancreatic stump and the intestinal wall adopted penetrating-suture pancreaticojejunostomy; the digestive tract was reconstructed by Roux-en-Y.Results The anastomosis with penetrating-suture pancreaticojejunostomy was successful in ten domestic pigs.The mean pancreatic stump diameter was 2.5 cm,and the mean pancreatic duct diameter was 1.5 mm,the mean time for operation was 1.0 ~ 2.5 h,and the average time of pancreaticojejunostomy was 8 minutes.The mean blood loss was 25 ml.After operation,diarrhea occurred in 2 pigs and wound infection occurred in 1 pig,and all were cured with appropriate management.No pig died intra-operatively,and no pancreatic fistula or death occurred after operation.Conclusions A porcine model of penetrating-suture pancreaticojejunostomy is successfully established.
9.Proteomic identification of malignant transformation-related proteins in esophageal squamous cell carcinoma

Objective:To identify differentially expressed proteins related with malignant transformation of esophageal squamous cell carcinoma (ESCC) using proteomic analysis. Methods:Two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization timE-of flight mass spectrometry (MALDI-TOF-MS) in combination with protein database searching were used to determine and identify differentially expressed proteins in esophageal cancer cell lines (EC1, EC18, and EC109) and immortal cell line (NECA-E6E7-hTERT). Western blotting and immunocytochemistry were used to verify the differential expression of annexin 2 in esophageal cancer cell lines and immortal cell line (NECA-E6E7-hTERT). Real-time fluorogentic quantitative PCR(RFQ-PCR) was performed to analyze the expression level of annexin A2 mRNA.Results: A total of 15 differentially expressed proteins were identified with more than 5 folds difference. Among them three proteins were down-regulated and 12 proteins were up-regulated. Western blotting and immunocytochemical analysis verified the down-regulation of annexin A2 protein in ESCC cell lines. However, differential expression pattern of annexin A2 mRNA was not consistant with its protein expression in ESCC cell lines and immortal cell line (NECA-E6E7-hTERT). Conclusion:The findings provide important clues for identifying the candidate biomarkers for high-risk population screening and early diagnosis of ESCC. Post-translative regulation/modification contributes to the down-regulation of annexin A2 protein.
10.Twenty-six clinical case analysis of defect repairing after the resection of basal cell carcinoma on external nose.

OBJECTIVE:
To study a surgical and repairing method for defects after the resection of basal cell carcinoma of external nose.
METHOD:
There are 26 cases of basal cell carcinoma that tumors have been resected completely after operation. For defect repairing in those 26 cases, 2 cases adopt direct suture method; 2 cases use skin graft repairing methods; 18 cases employ naselabial skin flap repairing method; 4 cases choose forehead pedicle skin flap repairing methods.
RESULT:
the wound of all the 26 cases was primary healed. Additionally, skin flap and skin graft were all survived. Follow-up studied in patients 1 to 3 years after the surgery showed that the local scar was not obvious and no tumor recurred or transferred.
CONCLUSION
Different surgical and repairing methods are performed to obtain a satisfactory results based on the area of defect and its location in nose. Naselabial skin flap is especially an ideal method to repair defects.
