Growth arrest special 5 (GAS5) is a common long non-coding RNA (lncRNA).Recent studies find that lncRNA GAS5 is down-regulated in various tumors,such as breast cancer,cervical cancer,lung cancer,gastrointestinal cancers and urologic cancers.GAS5 is closely related to the development,metastasis and prognosis of cancers,which becomes a new hotspot in cancer research.
2.Determination of tanshinoneⅡA in Rujie Xiaosan tablet

OBJECTIVE To develope an HPLC assay for the determination of tanshinoneⅡA.METHOD C18ODS column was used.The mobile phase was consisted of MeOH-water(70∶30).The detection wavelength was at 269 nm.RESULT The linear regression equation was Y=9725X-2584,r=0.9997.The average recovery was 98.29% with RSD=1.86%.(n=5).CONCLUSION The method may be used for quality control.
3.Effects of kangfengshi granules on expressions of osteoprotegerin, RANKL and M-CSF in bone tissues of rats with collagen-induced arthritis

OBJECTIVE: To observe the effects of Kangfengshi Granules (KFSG) on expressions of the mRNAs of osteoprotegerin (OPG), receptor activator of nuclear factor-kappaB ligand (RANKL) and macrophage colony stimulating factor (M-CSF) in bone tissues of rats with collagen-induced arthritis. METHODS: Forty SD rats were randomly divided into four groups: normal control group, untreated group, cyclosporine A (CsA)-treated group and KFSG-treated group. Except the rats in the normal control group, all the other rats received subcutaneous injection of collagen II to establish collagen-induced arthritis (CIA) models. Then the rats in each group were fed normal saline or corresponding drugs for four weeks. Total RNA was extracted from carpal and digital bones. The expressions of OPG, RANKL and M-CSF mRNAs were examined by real-time PCR. RESULTS: The total incidence of arthritis induced by collagen II in the rats was approximately 90%. The expression levels of RANKL and M-CSF mRNAs and the RANKL mRNA/OPG mRNA ratio in the untreated group, KFSG-treated group and CsA-treated group were all significantly higher than those in the normal control group, while the expression levels of OPG mRNA in those three groups were significantly lower than that in the normal control group. The expression level of OPG mRNA in the KFSG-treated group was obviously higher while the expression level of M-CSF mRNA and the RANKL mRNA/OPG mRNA ratio in the same group were both lower as compared with those in the untreated group. CONCLUSION: The molecular mechanism of effects of KFSG on bone erosion and destruction induced by rheumatoid arthritis is closely correlated with up-regulating the expression of OPG mRNA, down-regulating the expression of M-CSF mRNA and RANKL mRNA/OPG mRNA ratio.

Objective To constructan eukaryotic expression recombinant plasmid named pEGFP-N2-PRNP .Methods Total RNA was extracted from alzheimer (AD) disease peripheral blood ,and the PRNP gene was amplified by reverse transcription-poly-merase chain reaction(RT-PCR) .By using gene recombination technique ,human PRNP cDNA was inserted into retroviral vector pEGFP-N2 .The recombinant plasmid was identified by a pair of specified primers containing the restriction sites of Xho Ⅰ and EcoRⅠ .Results The PRNP gene could be obtained by RT-PCR ,the recombinant plasmid was identified by restriction endonucle-ase analysis ,PCR and sequence analysis ,and the expression vector pEGFP-N2-PRNP ,which could be stably expressed in SH-SY5Y cells .Conclusion The recombinant plasmid pEGFP-N2-PRNP is constructed successfully ,Which offers a basic for the further re-search on PRNP biological fuction .
5.Study of changes of free carnitine in premature newborns with hyaline membrane disease

Objective To study the serum free carnitine level and change trend in premature infantwith hyaline membrane disease (HMD) within postnatal 7 d .MethodTotally 63 preterm newbornwith gestational age of 28-32 weekin the NICU of thihospital were selected ,among them 32 caseof HMD were taken athe observation group and othe31 preterm newbornwithouHMD athe control group .The free carnitine level wameasured within 6 h aftebirth ,on 3 ,7 d by using the tandem masspectrum method .ResultThe free carnitine level within 6 h aftebirth had no statistical difference between the observation group and the control group[(23 .57 ± 7 .45)μmol/L v.(24 .34 ± 5 .73)μmol/L ,t=0 .48 ,P=0 .630] ,the free carnitine level on 3 d in the observation group wasignificantly lowethan thain the control group [(19 .21 ± 6 .83)μmol/L v.(23 .74 ± 7 .13)μmol/L ,t=2 .57 ,P=0 .010];the free carnitine level on 7 d in the observation group waalso significantly lowethan thain the control group [(16 .62 ± 7 .95)μmol/L v.(22 .83 ± 6 .56)μmol/L ,t=3 .39 ,P=0 .001] .The free carnitine level aftebirth in the control group remained stable ,which a3 time pointof 6h ,3 ,7 d had no statistical difference(F=0 .42 ,P=0 .660);whereathe free carnitine level wagradually decreased aftebirth ,which a3 time pointof 6 h ,3 ,7 had statistical difference(F=7 .13 ,P=0 .001) .Conclu-sion The free carnitine level aftebirth in preterm newbornwith HMD igradually decreased ,which needmore carnitine fopromoting the generation of pulmonary surfactan.
6.The biomechanical analysis of craniovertebral junction finite element model in atlas assimilation.

OBJECTIVETo study the biomechanical change of the craniovertebral junction in conditions of atlas assimilation.
METHODSMimics software was used to process CT data of the craniovertebral junction in a health adult to obtain the three-dimensional reconstruction and the cloudy points of C1, C2 and part of the occipital bone. Then the cloudy points were imported into the Abaqus 6. 8 software to establish the occipito-atlantoaxial finite element model in normal structure. According to the established model in normal structure, the model in conditions of atlas assimilation was set by changing the model parameters. Both models of normal structure and atlas assimilation were loaded with 1. 5 N . m static moment to simulate four motions of flexion, extension, lateral bending and axial rotation respectively. The movement characteristics,joint stress force and ligament deformation was analyzed.
RESULTSUnder 1. 5 N . m moment, in model of atlas assimilation the C1-C2 range of movement decreased from 13. 55° to 11.88° in flexion,increased from 13. 22° to 15. 24° in extension and from 4. 05° to 4. 23° in lateral bending and remained unchanged in axial rotation when compared with the normal model. In flexion movement, the contact force of the atlanto-dental joint increased from 1. 59 MPa to 3. 28 MPa and the deflection of apical ligament, tectorial membrane and alar ligament increased 129. 1%, 157. 6% and 75. 1% respectively when compared with the normal model.
CONCLUSIONSThe normal C1-C2 motion mode is destructed in conditions of atlas assimilation, leading to the changes of the range of movement,joint stress force and the ligament deformation at C1 C2 junction. The atlantoaxial instability will likely occur in flexion motion.

7.3D dynamic tongue visualization based on DirectX

Objective Tongue shape and motion is one of the important references for tongue diagnosis.However, current tongue image objectification of tongue diagnosis cannot express the characteristics of tongue.Methods In order to increase shape and dynamic information about the tongue, the methods of key frame selection and linear interpolation were used to tongue 3D dynamic visualization reconstruction model on the basis of 3D static visual model.For the convenient use of the tongue 3D dynamic visualization model, this paper put forward a model of 3D dynamic visualization interaction system which was realized based on DirectX.Results The 3D dynamic visualization model not only had a static visual model of texture and depth information, but also could intuitively describe the tongue motion.Conclusions It provides a method for the auxiliary tongue diagnosis of traditional Chinese medicine (TCM).
8.Combined screening report of hearing screening and deafness susceptibility genes screening for newborns

Objective To investigate the clinic signification of newborn hearing screening combined with deafness susceptibility genes screening. Methods 1 440 newborns(3 ~ 5 days after birth) were screened for 8 hot spot hearing loss associated mutations from GJB2, mt12S rRNA and SLC26A4. At the same time, all infants received hearing screening. Those who failed to pass two-step test were referred to further audiological assessment. Results The carrier rate of commonmutations was 1.46% for GJB2 c.235delC, 0.35% for GJB2 c.299-300delAT, 0.42% for mt12S rRNA c.1555A > G, 0.42% for SLC26A4 c.IVS7-2A > G and 0.14% for SLC26A4 c.2168A > G. The total carrier rate was 2.78%. 10 infants were diagnosed as hearing loss in the hearing screening and follow-up audiology assessment (6.94‰) and 5 were diagnosed as severe hearing loss (3.47‰). 32 hearing loss associated mutation carriers passed the hearing screening. Conclusions Genetic screening of newborn hearing screening can be helpful to find out neonates with late-onset and progressive hearing impairment, which were significant for early intervention, regular follow-up and reduction of deafness.
9.Meta Analysis of ADAM33 T1,S2 Polymorphism and the Susceptibility of Bronchial Asthma in China

Objective To investigate the correlation between ADAM33 T1, S2 gene polymorphism and Bronchial asthma risk in china. Methods We retrived the relevant published studies about ADAM33 T1, S2 gene polymorphism and bronchial asthma risk. Then we divided the population into Chinese and other Asian population. Odds ratio (OR) of Case group and control group was selected as the effect index. Stata 11.0 software was used to calculate heterogeneity test, ORs and 95%CI of two areas, and gave the forest plot and funnel plot of meta results. Results A total of 27 studies were included in this analysis,18 studies in ADAM33 T1 site were 3881 cases in case group, and 3780 cases in control group;and 14 studies in ADAM33 S2 site were 3222 cases in case group, and 3513 cases in control group. Additive model, dominant model, recessive model of ADAM33 T1 in Chinese had association with the susceptibility of bronchial asthma. The results were OR=1.488, 95% CI:1.002-2.167 in Additive model, OR=1.619, 95%CI:1.059-2.475 in dominant model;OR=2.523, 95%CI:1.910-3.333 in recessive model. Three models of ADAM33 T1 in other Asian country had no association with the susceptibility of Bronchial Asthma. Three gene model of ADAM33 S2 in Asian had no association with bronchial asthma susceptibility. Except ADAM33 T1 polymorphism in recessive model, other mode of T1, S2 had no publication bias in Chinese population. Conclusion There are association between ADAM33 T1 gene polymorphism and bronchial asthma, but ADAM33 S2 gene polymorphism and bronchial asthma have no association in Chinese population.
10.Effect of the public hospital trusteeship based on the employee satisfaction

Objective: To investigate the hospital employee satisfaction and trustworthiness in the background of the trusteeship mode and to find out the problems after trusteeship.Methods: Minnesota satisfaction questionnaire (MSQ) and the self-made questionnaire were used to conduct the survey.Results: The overall employee satisfaction (3.80±0.86) and trustworthiness (3.95±0.77)were higher.The highest level of trustworthiness concerned the cultural connotation (84.8%) followed by the management concept (82.8%) for overall satisfaction.The lowest level of employee satisfaction concerned income and workload (53.7%), followed by the working conditions and environment (55.3%).The administrative staff satisfaction was higher compared to that of medical staff (p=0.001), which showed significant statistical differences.Conclusions: The hospital has made some achievements after the trusteeship system reform, however, it still needs improvement and further strengthening in many aspects.The hospital must always keep abreast of the demands of workforce and improve the staff satisfaction so as to promote its continuous and sustainable development.
