Objective To explore the effect on Harris hip score of osteoprotegerin for bone mineral density in patients with hormone os-teonecrosis. Methods Sixty patietns with glucocorticoid-induced avascular necrosis admitted in our hospital were randomly divided into two groups,30 cases in the control group received routine calcitonin treatment for a period of 4 weeks treatment,30 cases in the experimental group treated by femoral head subperiosteal injection for bone protection (0. 5 mg/kg) based on conventional treatment for a period of 4 weeks. Then the bone mineral density,pain index and Harris hip score of 2 groups before and after treatment were compared. Results The bone mineral density of 2 groups has increased after treatment,compared with control group[(0.84 ±0.22) g/cm2,(0.81 ±0.23)g/cm2], thebonemineraldensityofexperimentalgroupincreasedobviously[(0.99±0.20) g/cm2,0.98±0.25)g/cm2].Thepainindexscoreof2 groups has decreased,the pain index score of experimental group (0. 76 ± 0. 62) point was lower than that of control group (1. 56 ± 1. 02) point. The pain index level of experiment group was more excellent than that of control group. The Harris score of experimental group (92. 10 ± 12. 23) point was obviously higher than that of control group(83. 15 ± 14. 63) point. The level of Harris score in experimental group was more excellent than that in control group. The difference was statistically significant (P<0. 05). Conclusion Osteoprotegerin can obviously improve the patient' s clinical indicators,which has guiding significance for hormone osteonecrosis in clinic.

Objective To investigate the therapeutic efficacy of Aegisy retrievable filter in inferior vena cava interventional treatment for acute deep venous thrombosis of lower extremity. Methods Acute or sub acute unilateral lower extremity deep vein thrombosis ( deep vein thrombosis, DVT) patients in our hospital were selected, and Aegisy filter was put into the inferior vena cava through the femoral, and thrombolytic therapy was carried out in the next day. Limb conditions was observation after 10 to 15 days, confirmed by vascular ultrasound and angiography of lower limb vein after no fresh or free thrombus. The filter was removed with protector, anticoagulant therapy after review the venography of lower limb postoperative. Results All Aegisy filter were put into patients, and there were no obvious forward jumping and drawing back occurred during the release of the filters, and all Aegisy filter had a better position. Placement of a filter everage time was (16. 8 ± 1. 3) days, 89 cases were successfully recovery filter. The effct of Aegisy filter on the patients with lower extremity deep venous thrombosis after treatment is obvious effect, no pain, and no symptom of pulmonary embolism. Conclusion Aegisy retrievable inferior vena cava filter has many advantages in clinical use. The filter can be placed to the scheduled position accurately,and it is very powerful in captu-ring the thrombus,it can be hold in the inferior vona cava for long time before it is planed to be retrieved,and its retrieval success is very high. Moreover,with high technieal successful rate the implantation of Aegisy retrievable inferior vena cava filter is very safe and effective for the interventional treatment of acute deep venous thrombosis of lower extremity.

AIM:To investigate the histone modification changes of topoisomerase Ⅱα( TOPOⅡα) promoter regulatory factor Sp1 in the patients with chronic benzene poisoning .METHODS:The bone marrow samples were collect-ed from 25 chronic benzene poisoning cases and 25 controls.The chromatin immunoprecipitation assay was carried out to study the possible mechanism of TOPOⅡαpromoter regulatory factor Sp 1 expression changes .The mRNA expression of Sp1 was detected by RT-PCR.RESULTS:Compared with the controls , the histone H4 acetylation and histone H3 acetyla-tion of Sp1 in the chronic benzene poisoning patients significantly decreased (P<0.01), and histone H3K9 methylation level of Sp1 increased (P<0.01), but the histone H3K4 methylation level of SP1 was not obviously changed (P>0.05). The mRNA expression of Sp1 in the chronic benzene poisoning patients was significantly lower than that in the controls (P<0.05).CONCLUSION:In chronic benzene poisoning patients , the histone acetylation and methylation modification changes of TOPOⅡαpromoter regulatory factor Sp 1 accompanied with the changes of mRNA level are observed .Histone H4 and H3 acetylation and H3K9 methylation modification of Sp1 may play an important role in the benzene ’s hematopoiet-ic toxicities.

Objective:To explore the risk factors influencing the prognosis for patients with hepatocellular carcinoma (HCC) recurrence after liver transplantation and summarize the relevant diagnostic and therapeutic experiences.Methods:The clinicopathological features with diagnosis and treatment plan of 102 recurrent HCC patients fulfilling the Fudan Criteria were compared for survival rate (univariate analysis) and independent prognostic indicators were obtained by Cox multivariate analysis.Results:The 1/3/5-year overall survival rates were 92.2%, 48.6% and 34.6% and the 1/3/5-year survival rates with tumor were 63.2%, 31.0% and 16.7% respectively. Cox regression analysis indicated that patient age, whether tumor can be surgically resected or not and personalized diagnostic & therapeutic plan based upon targeted therapy were independent prognostic factors affecting the overall survival rates and survival rates with tumor.Conclusions:Although HCC recurrence and metastasis after liver transplantation seriously influence patient prognosis, satisfactory outcomes may be obtained for some patients through active, effective and precise managements.

Cytokine-activated T cells (CATs) can be easily expanded and are widely applied to cancer immunotherapy. However, the good efficacy of CATs is rarely reported in clinical applications because CATs have no or very low antigen specificity. The low-efficacy problem can be resolved using T cell antigen receptor-engineered CAT (TCR-CAT). Herein, we demonstrate that NY-ESO-1 HLA-A*02:01-specific high-affinity TCR (HAT)-transduced CATs can specifically kill cancer cells with good efficacy. With low micromolar range dissociation equilibrium constants, HAT-transduced CATs showed good specificity with no off-target killing. Furthermore, the high-affinity TCR-CATs delivered significantly better activation and cytotoxicity than the equivalent TCR-engineered T cells (TCR-Ts) in terms of interferon-γ and granzyme B production and in vitro cancer cell killing ability. TCR-CAT may be a very good alternative to the expensive TCR-T, which is considered an effective personalized cyto-immunotherapy.

ObjectiveTo observe the effect of ginsenoside Rg₁ （G-Rg₁） on the biological activity of cryopreserved Schwann cells （SCs） of the rat sciatic nerve and explore the feasibility of G-Rg₁ in reducing the cryopreservation-induced injury in SCs. MethodBilateral sciatic nerves of SD rats were randomly divided into a fresh group， a blank group， and five G-Rg₁ groups of different doses （1×10^-7， 1×10^-6， 1×10^-5， 1×10^-4， and 1×10^-3 mol·L^-1）. The nerves in the blank group and the G-Rg₁ groups were preserved in liquid nitrogen solutions containing 0， 1×10^-7， 1×10^-6， 1×10^-5， 1×10^-4， and 1×10^-3 mol·L^-1 G-Rg₁ for four weeks. The apoptosis of SCs was detected by TdT-mediated dUTP-biotin nick end labeling （TUNEL）/S100 immunofluorescence staining. The expression of cysteinyl aspartate-specific protease （Caspase）-9， Caspase-3， major histocompatibility complex （MHC）-Ⅰ， and MHC-Ⅱ was detected by Western blot. Subsequently， all nerves were cultured in the incubator at 37 ℃ with 5% CO₂ for 7 days. The expression of glial cell line-derived neurotrophic factor （GDNF） and nerve growth factor （NGF） was detected by Western blot. In addition， the above cryopreserved nerves in the blank group and the 1×10^-6， 1×10^-5， and 1×10^-4 mol·L^-1 G-Rg₁ groups were transplanted to the Wistar rats by allografting （blank transplantation group and the 1×10^-6， 1×10^-5， and 1×10^-4 mol·L^-1 G-Rg₁ transplantation groups）， and fresh sciatic nerve allograft and isograft control group were set up. Sixteen weeks after transplantation， compound muscle action potential （CMAP） and nerve conduction velocity （NCV） were measured by electrophysiology. Nerve filament （NF）200 immunofluorescence staining， transmission electron microscopy， and toluidine blue staining were used to analyze the histology of the regenerated nerves. ResultCompared with the fresh group， the blank group and the G-Rg₁ groups showed increased expression of Caspase-9， Caspase-3， and the apoptosis of SCs （P<0.05，P<0.01） and decreased expression of GDNF， NGF， MHC-Ⅰ， and MHC-Ⅱ （P<0.01）. Compared with the results in the blank group， the expression of Caspase-9 and Caspase-3 decreased in the 1×10^-7， 1×10^-6， 1×10^-5，1×10^-4 mol·L^-1 G-Rg₁ groups （P<0.01）， and the apoptosis of SCs was reduced in the 1×10^-7-1×10^-4 mol·L^-1 G-Rg₁ groups（P<0.05，P<0.01） and increased in the 1×10^-3 mol·L^-1 group （P<0.05）， while the expression of GDNF and NGF increased in the 1×10^-7， 1×10^-6， 1×10^-5，1×10^-4 mol·L^-1 G-Rg₁ groups and decreased in the 1×10^-3 mol·L^-1 group （P<0.05）. There was no statistical significance in the expression of MHC-Ⅰ and MHC-Ⅱ between the blank group and the G-Rg₁ groups. Compared with the 1×10^-7 mol·L^-1 and 1×10^-3 mol·L^-1 G-Rg₁ groups， the 1×10^-6 1×10^-5， 1×10^-4 mol·L^-1 G-Rg₁ groups showed decreased expression of Caspase-3 and the apoptosis of SCs （P<0.05，P<0.01） and increased expression of GDNF and NGF （P<0.05，P<0.01）. There was no statistical significance in MHC-Ⅰ and MHC-Ⅱ expression among G-Rg₁ groups. Sixteen weeks after transplantation， compared with the isograft group， the blank transplantation group and the G-Rg₁ transplantation groups showed decreased CMAP， NCV， myelin sheath thickness， and number of myelinated nerve fibers （P<0.01）， and the 1×10^-6 and 1×10^-4 mol·L^-1 G-Rg₁ transplantation groups showed decreased NF200 （P<0.01）. Compared with the allograft group， the blank transplantation group and the G-Rg₁ transplantation groups showed increased CMAP， NCV， NF200， myelin sheath thickness， and number of myelinated nerve fibers （P<0.05，P<0.01）. Compared with the blank transplantation group， the G-Rg₁ transplantation groups showed increased CMAP， NCV， NF200， myelin sheath thickness， and number of myelinated nerve fibers （P<0.05，P<0.01）. Among all groups of G-Rg₁ transplantation， each index of the 1×10^-5 mol·L^-1 G-Rg₁ transplantation group was superior to that of the 1×10^-4 and 1×10^-6 mol·L^-1 G-Rg₁ transplantation group （P<0.05）. ConclusionG-Rg₁ at a certain centration can maintain the biological activity of cryopreserved SCs of rat sciatic nerve， alleviate the cryopreservation-induced injury of rat sciatic nerve， and promote nerve regeneration after allograft.
