AIM: To construct retroviral expression vector pLXmIL-12SN containing mouse IL-12 (mIL-12) gene and detect the gene expression in B16F10 cells. METHODS: Vector pGCp35-IRES-p40SN was digested to obtain p35-IRES-p40 (mIL-12 gene fragment) and retroviral vector pLXSN was digested by restriction enzyme, and then they were linked by ligase. The recombinant vector that mIL-12 gene fragment had correctly been inserted into pLXSN was identified by sequencing. The recombination vector with mIL-12 gene was transfected into B16F10 cell and was detected by RT-PCR. RESULTS: Expression vector pLXmIL-12SN was successfully constructed. mIL-12 gene was confirmed to express in B16F10 cells at the level of mRNA. CONCLUSION: Acquired vector pLXmIL-12SN and confirmed mIL-12 gene expression lays a foundation for mIL-12 gene therapy to eye diseases.

Methods Seventy-two patients were randomly divided into a treatment group (37 patients) or a control group (35 patients) by using a random number table. Patients in the treatment group and in the control group were treated with combination of cervical manipulation or cervical manipulation only, respectively. At the end of treatment and 3 months, treatment effects were assessed by Neck Disability Index (NDI) and Visual Analog Scale (VAS). Results NDI scores in both of the treatment group and the control group after treatment were significantly lower han those before treatment (t=4.471 in the treatment group, t=3.599 in the control group, all P<0.05), and NDI score in the treatment group were significantly lower than that in the control group after treatment (t=-2.085,P<0.05). Follow-up at 3 months, NDI score in the treatment group was significantly lower than that in the control group (t=-4.549, P<0.05). Proportions of improved patients, as assessed by NDI items, including pain intensity, personal care, lifting, reading, headaches, concentration, work, sleeping, driving and recreation in the treatment group were significantly higher than those in the control groups (Z=-3.521,-3.331,-3.003,-3.001,-3.265,-3.198,-3.016,-3.000,-2.102,-2.971, respectively, all P<0.05). VAS scores in both of the treatment group and the control group after treatment were significantly lower than those before treatment (t=7.986 in the treatment group, t=6.507 in the control group, all P<0.05), and VAS score in the treatment group were significantly lower than that in the control group after treatment (t=-2.426,P<0.05). Follow-up at 3 months, the VAS score in the treatment group was significantly lower than that in the control group (t=-7.065, P<0.05). Proportions of improved patients in the treatment group was significantly higher than that in the control group (91.89% vs 74.29%; χ2=4.014, P=0.045). Conclusion Combination of cervical manipulation with cervical conditioning exercise may be superior to cervical manipulation only for the treatment of cervical spondylosis of neck type or nerve root type.

Objective:To investigate the role of heme oxygenase-1(HO-1) in molecular mechanism of experimental allergic encephalomyelitis (EAE). Methods: Seventy-eight healthy female Wistar rats were randomly divided into 3 groups(n=26): rats in control group received no treatment; rats in EAE group were induced with complete Freund's adjuvant and Guinea pig spinal cord homogenate(CFA-GPSCH); and rats in pyrrolidine-dithiocharbamate (PDTC) group received PDTC (100 mg/kg), a specific inhibitor of NF-kB, 1 h before and after(once a day for 7 d) CFA-GPSCH treatment. HO-1 mRNA expression were analyzed with reverse transcription polymerase chain reaction(RT-PCR) on 1 d,7 d,14 d,and 21 d after EAE induction, respectively. The relationship between HO-1 and symptoms of EAE was also investigated. Results: The expression of HO-1 mRNA was very low in the brains of the control group (0.27?0.05), whereas enhanced gradually with onset and development of EAE in EAE group, peaked on d 7 (1.11?0.12), kept at a high level till d 14(1.06?0.18) and decreased on d 21 (0.37?0.1). HO-1 mRNA expression change was in parallel with severities of EAE. In PDTC group, the EAE symptoms were mitigated markedly and the expression of HO-1 mRNA reduced noticeably compared with that of EAE group. Conclusion: Brain HO-1 mRNA expression may play an important role in the pathogenesis of EAE,and application of some inhibitors of NF-kB may be one of the potential therapies for prevention and treatment of EAE.

To obtain non-pathogenic rabies virus glycoprotein(RV-G),we expressed RV-G in Saccaromyces Cerevisiae(S.cerevisiae).In our study,tat-G fusion gene was cloned into the expression vector pYes2.0,which allows expression of a foreign gene in the yeast cells under the control of GAl1 promoter.Transforma-tion was performed by using lithium-treated yeast cells and several Ura+-tranformants were isolated.Ac-cording to the relative mobility in SDS-PAGE,we know probably two forms(designated as yGI and yGⅡ) of RV-G analogues produced in S.cerevisiae, their molecular weights were estimated as 66 kD and 56 kD,respectively. On the other hand, there was a specific band about 56 kD shown in western blot result.Com-bining precursors’ achievements,we will draw a conclusion that trans-membrane domain(TD) and cyto-plasmic domain have a negative regulation on RV-G antigen immunogenicity in S.cerevisiae.

Journal of Clinical Hepatology 2019;35(6):1374-1376
Primary liver cancer is a common malignant tumor with complex pathogeneses and has the features of insidious onset, high degree of malignancy, and poor prognosis. The change in glycolytic pathway is one of the most important differences between tumor cells and normal cells, and tumor cells prefer to generate energy from glycolysis. Aerobic glycolysis is often associated with the progression and poor prognosis of primary liver cancer. Long non-coding RNAs (lncRNAs) can influence the glycolysis pathway in many tumors by regulating glucose uptake and the expression and activation of glycolytic enzymes and thus play an important role in the development and progression of primary liver cancer, which suggests that lncRNAs can be used as a therapeutic target for liver cancer. This article summarizes the influence of lncRNAs on primary liver cancer and glucose metabolism and related mechanisms, so as to find potential and effective targeted therapies for primary liver cancer.
