1.Apoptin induces apoptosis in tumor cells
Journal of International Oncology 2008;35(5):330-333
Apoptin,a chicken anemia virus-derived protein.selectively induces apoptosis in trans-formed or tumor cells but not in normal cells. The tumor specificity of apoptin is thought to be related to its cel-lular localization. The apoptin-induced apoptosis is p53-independent and can not be blocked by overexpressionof Bcl-2.But activation of Caspase-3 is essential for apoptin-induced apoptosis. These features make apoptin a potential candidate as a novel therapeutic and diagnostic tool in cancer treatment.
2.Reconstruction of combined anterior cruciate ligament and posterior cruciate ligament injuries by using LARS artificial ligament under arthroscope
Chinese Journal of Trauma 2008;24(4):263-265
Objective To explore the treatment method and clinical effect of LARS artificial ligament in treatment of combined anterior cruciate ligament(ACL) and posterior crueiate ligament (PCL) injuries of the knee under arthroscope. Methods Thirteen cases of ACL and PCL injuries were reconstructed with LARS ligament. The patients were followed up for 12-36 months. The International Knee Documentation Committee (IKDC) and Lysholm knee score scale were used for functional evaluation. In the meantime, KT-1000 was employed to check anterior and posterior laxation of the knee. Results No knee infection or limitation of knee extension occurred. Knee flexion was 105° -125° (average 117°). At termination of follow up, 10 cases (77%) was graded as A and 3 (23%) as B according to IKDC. KT-1000 examination at 25° flexion showed that the difference of anterior laxation was less than 2 mm in 12 cases and 3-5mm in 1. KT-1000 examination at 70°flexion showed that the difference of anterior laxation was less than 2mm in 12 cases and 2-4 mm in 1. The Lysholm score was (63.8±2.9) points (49459 points) before operation and (91.1 ±2.7) points (88-95 points) at follow up, with statistical difference (P <0.01). Conclusion Reconstruction of ACL and PCL by using LARS ligament under arthroscope is a kind of minimally invasive, safe and effective method to treat ACL and PCL injuries of the knee, with good short term outcome.
3.Establishment of osteosarcoma cell models:effects of NS-398 on invasive ability of MG-63 cells under hypoxic condition
Chinese Journal of Tissue Engineering Research 2015;(40):6408-6412
BACKGROUND:In anoxic environment, invasive ability of tumor cels significantly increased. The increase in above invasive ability is possibly associated with the changes in urokinase or matrix metaloproteinases. Studies on hypoxia and tumor therapy especialy the effect on chemotherapy have made great progress, and found that hypoxia-inducible factor 1 is a key nuclear factor mediating celular hypoxia response, and also an important factor for regulating cel apoptosis during hypoxia. OBJECTIVE:To observe the effects of cyclooxygenase-2 inhibitor NS-398 on invasive ability of MG-63 cels under hypoxic condition in cel models of hypoxia osteosarcoma. METHODS: Chemical hypoxia inducer cobalt chloride was used to establish chemical hypoxia environment of concentration gradient of 0, 100, 200, 400 μmol/L. Under 200 μmol/L cobalt chloride, concentration gradient of 30, 60, 90 μmol/L of NS-398 were set. Osteosarcoma cels were determined after 48 hours of culture. Quantitative PCR was used to detect the expression of matrix metaloproteinases 9 mRNA and hypoxia-inducible factor 1α mRNA. Western blot assay was applied to measure hypoxia-inducible factor 1α expression. Flow cytometry was utilized to examine cel apoptosis in each group. RESULTS AND CONCLUSION:PCR and western blot assay results demonstrated that with increased concentration of cobalt chloride, matrix metaloproteinases 9 mRNA and hypoxia-inducible factor 1α mRNA and protein expression increased (P < 0.05), showing a concentration-dependent manner. Under cobalt chloride concentration of 200 μmol/L and hypoxia, with increased concentration of NS-398, matrix metaloproteinases 9 mRNA and hypoxia-inducible factor 1α mRNA and protein expression gradualy reduced, showing a concentration-dependent manner (P < 0.05). Flow cytometry results showed that under 400 μmol/L cobalt chloride concentration, cel apoptosis noticeably reduced. Under cobalt chloride concentration of 200 μmol/L and hypoxia, with increased concentration of NS-398, cel apoptosis increased (P < 0.05). Results confirm that anaerobic environment is an important factor for increased invasive ability of osteosarcoma. Under the anaerobic environment, NS-398 could effectively suppress the invasive ability of osteosarcoma and promote cel apoptosis.
4.Molecular mechanism of long non-coding RNAs in tumor drug resistance
Journal of International Oncology 2015;(6):439-441
Long non-coding RNAs (lncRNAs)are closely related to the tumorigenesis, proliferation, metastasis and clinical outcome, whihc play important roles in the process of drug resistance of many tumor cells.Recently, research indicates that many lncRNAs regulate drug resistance of tumor cells through a variety of signaling pathways both in vitro and in vivo.Furthermore,it is also found that after blocking the relevant tar-gets can reverse this chemoresistance.

Journal of Kunming Medical University 2013;(10):93-95
Objctive To evaluate therapeutic effect of periocline ointment as an auxiliary treatment of adult serious chronic periodontitis. Methods 56 patients were divided into the test group and the control group in register sequence. Following initial therapy, patients in the test group were treated with PERIOCLINE into the periodontal pocket once a week for 4 weeks, while patients in the control group were treated with iodine glycerine. At the baseline level, the indexes of PLI,SBI,PD,AL were recorded and compared between the two groups on the 1st month, 3rd month and 6th month. Results All the indices of both groups were improved, compared with the baseline ( <0.05) . The results of the test group were better than those of the control group, and the differences were statistically significant (<0.05) . Conclusion Periocline ointment as an auxiliary treatment of adult serious chronic periodontitis is superior to compounded iodine glycerine in the amelioration of all the indices.
6.Meta-analysis of Chinese Medicine Treatment in Endometriosis
Chinese Journal of Information on Traditional Chinese Medicine 2006;0(04):-
Objective To assess the effectiveness of Chinese medicine treatment in endometriosis(EMT) by evidence-based medicine methods. Methods Retrieve articles mainly from Wanfang data,VIP information,CNKI and China TCM Database.Bring the articles report into research on the basis of strict quality evaluation.Consolidate cure rate and effective rate for meta-analysis.Results Based on 8 articles report, cure rate of Chinese medicine treatment is superior to western medicine treatment, and effective rate is equivalent to western medicine treatment. There is no side effect and other harmful consequence in evidence. Conclusions Chinese medicine treatment is superior to western medicine treatment in treating EMT.However, existence of some data methodological problems maybe directly affect the authenticity of the findings. So,more rigorous, randomized controlled trials should be designed in order to draw more reliable conclusions.

To study the effects of growth hormone on function of T lymphocytes. Methods: Tne expression plasmid PcDNA-GHcDNA was constructed, then the plasmids were transfected into Jurkat cells,on the other hand,Jurkat cells were treated with different doses ofrhGH, to investigate the effect of overexpression GH and rhGH on function of T lymphocytes. Results: Overexpression GH could increase the ex-pression of IL-2R(140%) and secretion of IL-2(160%) and IFN-γ( 198% ) in Jurkat cells after PHA activation, but these data were broughtdown by the treatment of GH antibody. rhGH could also enhancer the function of T lymphocytes after PHA activation. Conclusion: These resultssupport the idea that GH is an autocrine and paracrine factor, and can regulate the function of T lymphocytes.

Objective To explore the effect of ginseng combined with transplantation of bone marrow mesenchymal stem cells(BMSCs)on functional recovery of rats with spinal cord injury(SCI). Methods Forty-eight female Wistar rats were randomly assigned to following four groups(n=12 per group):the sham operation group was treated by opening the vertebral lamina and exposing spinal cord without SCI;the SCI model group was reproduced by using improved Allen bump method and afterwards no treatment was given;methyl prednisolone(MP)group was treated by MP pulse treatment after SCI,including intravenous injection of MP 30 mg/kg immediately after SCI and 4 hours later the same injection was repeated, and then the same intravenous injection 2 times daily,3 days in total; The ginseng+BMSCs group was treated by orally taking ginseng ultra-microgranules 300 mg/kg after SCI,twice a day for 20 days and BMSCs 5μL(concentration 1×107 cell/μL)transplantation was carried out in SCI region on the 7th day after SCI. In the above 4 groups, the ethological observation(BBB scores)was done regularly and on the 30th day after operation, silver staining was applied to investigate the changes of spinal cord, and neuro-electrophysiological tests including somatosensory evoked potential(SEP)and motor evoked potential(MEP)were performed. Results In sham operation group, after surgery the movement of both hind limbs became temporarily sluggish and on the 7th day their functions recovered to approximately normal. In SCI model group, after injury paralysis of both hind limbs occurred, while in the MP group and ginseng+BMSCs group, different degrees of functional recovery of the injured limbs developed, and the recovery in ginseng + BMSCs group was more significant. Compared with sham operation group, after surgery the BBB score was reduced markedly at various time points in SCI model group;compared to the SCI model group, the BBB scores in MP and ginseng+BMSCs groups were increased significantly, especially more remarkable in ginseng+BMSCs group(all P<0.05),and beginning from the 12th day after operation, the difference became obvious (5.23±1.22 vs. 3.61±1.03, P<0.05). Histological detection showed that in sham operation group, the structure of spinal cord was complete, neurons distributed evenly in the gray matter and a large number of silver staining positive nerve fibers paralleled to each other and arranged regularly;in SCI model group, fragmented construction was present and the defects of gray and white matters were prominent. Compared with the SCI model group, the extents of tissue necrosis in MP and ginseng + BMSCs groups were ameliorated. The neuro-electrophysiological tests demonstrated that in SCI model group, the loss of normal wave form occurred. Compared with SCI model group,in MP and ginseng+BMSCs groups, after treatment latent periods(ms)were shortened prominently in different degrees of SEP and MEP,and their peak-to-peak values(mV)were increased obviously;the improvement in potential in ginseng+BMSCs group was greater than that in MP group〔SEP:latent period(ms):3.31±0.36 vs. 4.66±0.33, peak-to-peak value(mV):0.10±0.01 vs. 0.05±0.01,MEP:latent period(ms):3.40±0.13 vs. 4.24±0.31, peak-to-peak value(mV):41.12±0.56 vs. 16.46±2.83,all P<0.05〕. Conclusion A combined treatment of ginseng and BMSCs transplantation can effectively promote the recovery of neural function for rats with SCI.
