1.The effects of FL、rIL-6、rIL-6R in vitro expansion and differentiation of human cord blood CD34~+ cells

Objective:To determine the effects of FL,rIL 6,rIL 6R in vitro expansion and differentiation of human cord blood CD34 + cells.Methods:Human cord blood CD34 + cells were enriched by magnetic cell isolation system CD34 + cells were incubated in the presence of FL,rIL 6,rIL 6R and their various companision.The total number of cells and colonies were counted at various time intervals.Results:The expanded cells by FL,rIL 6,rIL 6R showed colonies were CFU GM mainly but rIL 6+rIL 6R and FL+rIL 6+rIL 6R,showed some BFU E and CFU Mix but no CFU MK.Erythrocyte cells were conformed and total cells were increased 6.4 folds by FL+rIL 6+rIL 6R.Conclusion:Human cord blood CD34 + cells could expanded and differentiated by FL+rIL 6+rIL 6R.
2.A decrease in proliferative ability and directional differentiation ability of autologous bone marrow mesenchymal stem cells in a rat model of steroid-induced femoral head necrosis

BACKGROUND:In recent years, stem cel therapy for early osteonecrosis of the femoral head has become an alternative method, but the quality of stem cels is a key to the therapeutic outcomes. OBJECTIVE:To evaluate the proliferative ability and directional differentiation ability of autologous bone marrow mesenchymal stem cels in a rat model of steroid-induced femoral head necrosis. METHODS:Twenty Sprague-Dawley rats were randomly divided into control and observation groups with ten in each group. An animal model of steroid-induced femoral head necrosis was built in the observation group, and then bone marrow mesenchymal stem cels from rats in both two groups were isolated and cultured. Cel counting kit-8 was used to detect proliferation of passage 3 cels. Bone marrow mesenchymal stem cels at passage 3 were selected in the two groups for osteogenic and adipogenic induction. Alkaline phosphatase staining and alizarin red staining were adopted at 7 and 14 days of osteogenic induction, and oil red O staining as performed at 21 days of adipogenic induction. RESULTS AND CONCLUSION:The absorbance values of bone marrow mesenchymal stem cels were lower in the observation group than the control group at 1, 3, 5 days of culture, but there was no significant difference between two groups (P > 0.05). Until the 7th day of culture, the absorbance value and alkaline phosphatase activity in the observation group were significantly lower than that in the control group (P < 0.05). Additionaly, there were fewer calcium nodules and lipid droplets in the observation group compared with the control group. These findings suggest that the proliferative ability and directional differentiation ability of autologous bone marrow mesenchymal stem cels from a rat model of steroid-induced femoral head necrosis are both decreased.

4.Exosomal miR-21 derived from Schwann cells promoting the repair of sciatic nerve injury by targeting SPRY2

Objective:To investigate the effect and potential mechanism of exosomal miR-21 derived from Schwann cell(SC)in the repairment of sciatic nerve injury(SNI). Method:SC was infected with negative control lentivirus and lentivirus that expression of miR-21,and its exosome was collected and identified respectively.The SNI rat model was established by nerve stump anasto-mosis.After surgery,the rats were randomly divided into the model group(n=10),the SC-derived exosome group(n=10)and the SC-derived exosome group(n=10)that high expression of miR-21.The SC-derived exosomes group and SC-derived exosome group that high expression of miR-21 were treated with local injec-tion of exosome collected in vitro.After 3 weeks,the functional recovery was detected by behavioral experi-ment.The nerve regeneration was evaluated by immunofluorescence staining.RT-qPCR was used to detect the expression of miR-21 in serum exosome and miR-21 and SPRY2 in sciatic nerve.The targeting interaction be-tween miR-21 and SPRY2 was verified by dual-luciferase reporter gene experiment. Result:Compared with the model group,the functional recovery and regeneration of sciatic nerve were signifi-cantly improved in the other two groups.The expression of miR-21 was significantly increased,while the ex-pression of SPRY2 was significantly decreased.The improvement in the SC-derived exosome group with high expression of miR-21 were more significantly than those in the SC-derived exosome group. Conclusion:Exosomal miR-21 derived from SC can significantly promote the repair of sciatic nerve injury by targeting SPRY2.
5.Toddalia asiatica attnuates cerebral ischemia-reperfusion injury in rats through modulating M1/M2 polarization of microglia

AIM:Observation of neuroprotective effects of Toddalia asiatica(TA)on cerebral ischemia-reper-fusion injury(CIRI)in rats by investigating the effects and mechanisms of drugs on the polarization of microglia M1/M2 subtype and the Toll-like receptor 4(TLR4)/myeloid differentiation factor 88(MyD88)/nuclear factor-κB(NF-κB)path-way.METHODS:The modified thread occlusion method was used to establish a rat model of CIRI,and the rats were ran-domly divided into the model group,Toddalia asiatica(1.08 g/kg)group,donepezil hydrochloride(0.45 mg/kg)group,and sham group,with 16 rats in each group.Based on the assessment of neurofunctional changes in each group of rats,HE and Nissl staining were used to observe the pathological changes in brain tissue.TUNEL staining was performed to de-tect neuronal apoptosis,immunohistochemistry was used to detect the expression of M1 microglia marker ionized calcium-binding adapter molecule 1(Iba1),M2 microglia marker arginase 1(Arg1),and TLR4 localization.Western blot was used to detect the expression of microglia polarization proteins and proteins related to TLR4/MyD88/NF-κB pathway in the hippocampus region.RESULTS:Compared with sham group,the model group rats had higher neurological function scores(P<0.01),and neuronal arrangement in the hippocampus and cortex was loose and disordered,Nissl bodies de-creased,and neuronal apoptosis increased.The numbers of M1 microglia marker Iba1-,M2 microglia marker Arg1-,and TLR4-positive cells were significantly increased.In addition,the protein levels of TLR4,MyD88,p-NF-κB p65,NF-κB p65,p-NF-κB inhibitory factor(p-IκB),Iba1,interleukin-6(IL-6),and Arg1 in the hippocampus were elevated(P<0.05),while IL-4 and IL-10 expression were decreased(P<0.01).Compared with model group,the Toddalia asiatica group and Donepezil hydrochloride group showed increased protein expression of Arg1,IL-4 and IL-10(P<0.05),while the other indicators were decreased(P<0.05,P<0.01).CONCLUSION:Toddalia asiatica possesses neuroprotective effects on CIRI rats,which may be attributed to its ability to regulate M1/M2 polarization and inhibit the TLR4/MyD88/NF-κB-mediated inflammatory pathway.
6.Clinical characteristics and pregnancy outcomes of non-criteria obstetric antiphospholipid syndrome

Objective:To study clinical characteristics and pregnancy outcomes under anti-coagulation therapy of non-criteria obstetric antiphospholipid syndrome.Methods:Patients suspected of obstetric antiphospholipid syndrome(OAPS) were recruited through Chinese Rheumatism Data Center from 2015 to 2019 consecutively. Patients fulfilling 2006 Sydney revised antiphospholipid syndrome criteria were classified as OAPS. Patients fulfilling definition of non-criteria OAPS(NCOAPS) by expert consensus on diagnosis and management of obstetric antiphospholipid syndrome of China were classified as NCOAPS. Clinical characteristics and laboratory results of two groups were compared. Live birth rates and pregnancy outcomes under anti-coagulation therapy were studied.Results:A total of 88 patients were enrolled, including 56 patients (63.6%) as OAPS, 32(36.4%) as NCOAPS. Live births were only reached in 16.1% (9/56) in OAPS patients and 12.5%(4/32) in NCOAPS. Fetal losses after 10 weeks of gestation and pre-eclampsia before 34 weeks were more common in OAPS group compared to NCOAPS group [78.6%(44/56) vs. 18.8%(6/32), P<0.001; 25.0%(14/56) vs. 3.1%(1/32), P=0.020, respectively]. After enrollment, 15 pregnancies were recorded in OAPS, 10 in NCOAPS, all of whom were treated with low-dose aspirin (LDA) combined with low-molecular weight heparin (LMWH). Live birth rates saw dramatic improvements compared to baseline levels in OAPS [16.1% (9/56) vs. 11/15] along with NCOAPS [12.5% (4/32) vs. 7/10]. Conclusion:Though NCOAPS and OAPS patients differ in antiphospholipid antibody spectrum and pattern of pregnancy morbidities, both groups benefit from LDA combined with LWMH treatment, as live birth rates improve. Non-criteria OAPS patients are recommended to receive anti-coagulation therapy during pregnancy.
