1.Effect of Cryptoporus polysaccharide on eotaxin mRNA and TNF-? mRNA expression in lung tissue of sensitized mice

Objective To study the time-course relationship of eotaxin mRNA and TNF-? mRNA expression in the lung tissue of sensitized mice after antigen challenge and the effects of Cryptoporus polysaccharide (CP) on these expression. Methods Eotaxin mRNA and TNF-? mRNA expression was determined by semi-quantitative RT-PCR. The function implications of eotaxin mRNA and TNF-? mRNA expression were examed by detecting the number of total leucocyte and eosinophils in bronchoalveolar lavage fluid (BALF). Results Peak level of TNF-? mRNA expression appeared at 8 h and eotaxin mRNA expression at 24 h in the lung tissue of sensitized mice after antigen challenge. Compared with the control group, total numbers of leukocyte and eosinophil in BALF increased in sensitized mice. CP 3, 10, and 30 mg/kg inhibited eotaxin mRNA and TNF-? mRNA expression, and inflammatory cells accumulation in airway of the sensitized mice in a dose-dependent manner. Conclusion The mechanism of inhibitory effects of CP on eotaxin mRNA and TNF-? mRNA expression may be related to the anti-allergic inflammation.
2.Comparison of cognitive function between patients with hemodialysis and peritoneal dialysis

Objective:To evaluate whether dialysis modality will affect cognitive function in dialysis population.Methods:This was a cross-sectional study. Chronic dialysis patients in our center was screened from July 2013 to July 2014. All of the subjects received brain magnetic resonance imaging (MRI) examination and comprehensive cognitive function evaluation.Results:A total of 189 chronic dialysis patients were enrolled in this study, 122 cases on hemodialysis (HD) and 67 cases on peritoneal dialysis (PD). There was no significant difference in age between HD and PD groups [(56.4±13.2) years vs (56.4±16.1) years, t=0.004, P=0.997]. The dialysis vintage and serum albumin of HD patients was higher than those of PD patients[58.0(16.8, 107.5) months vs 31.0(7.0, 67.0) months, Z=-3.490, P<0.001; (39.6±3.9) g/L vs (35.3±3.8) g/L, t=7.328, P<0.001, respectively]. The prevalence of cerebral small vessel diseases (CSVDs) was comparable between HD and PD groups (all P>0.05). Compared with HD patients, PD patients presented a 11.90-fold risk of immediate memory impairment (95% CI 1.40-101.08, P=0.023) and a 6.18-fold risk of long-delayed memory impairment (95% CI 2.12-18.05, P=0.001). After adjusting for age, educational lever, dialysis vintage, serum creatinine, and CSVDs, the influence of dialysis modality on memory still worked. PD patients presented a 43% risk of executive function impairment of HD patients ( OR=0.43, 95% CI 0.17-1.04, P=0.061). Conclusions:HD patients manifested better memory than PD patients, while PD probably performed better in executive function than HD patients. There was no significant difference in language function between the two groups. The difference in cognitive function may not be related to CSVDs.
3.Comparative Study on the Protective Effects of Different Effective Components of Astragali Radix against Ionizing Radiation-induced BMSCs DNA Damage

OBJECTIVE:To compare the protective effects of different effective components of Astragali radix against DNA damage of human bone marrow mesenchymal stem cells (BMSCs)induced by ionizing radiation. METHODS :2 Gy X-rays were used to directly irradiate BMSCs to establish a radiation model. CCK- 8 method was used to detect the effects of different mass concentrations(25,50,75,100 μg/mL)of astragalus polysaccharide ,astragalus saponin and astragalus flavonoids for 1 day before radiation + 1 to 5 days after radiation on the proliferation of BMSCs. The dose concentration and the duration of intervention after radiation were selected. The irradiated BMSCs were divided into radiation group ,astragalus polysaccharide group ,astragalus saponin group and astragalus flavonoids group. The last three groups were treated with appropriate dosage of corresponding drugs before and 2 days after radiation ,and a blank groupwas set for comparison. Cytoplasmic division arrest qq.com micronucleus method was used to detect micronucleus cell rate and cell micronucleus rate after appropriate time of was used to detect th e number of 53BP1 foci in cells after appropriare time of intervention following radiation ;the number of 53BP1 foci were compared among different time points (0.5,2,12,24 h). RESULTS :Compared with blank group ,OD values of BMSCs were decreased significantly in radiation group (P<0.05 or P<0.01). Compared with radiation group ,the OD values of BMSCs were significantly increased when 50 μ g/mL astragalus polysaccharide,astragalus saponin and astragalus flavonoids continuously intervened radiation for 2-3 days,there was significant difference in other groups at some time point (P<0.05 or P< 0.01). After consideration ,drug concentration was determined to be 50 μg/mL,and the continuous intervention time was 2 days after radiation. Compared with blank group ,the micronucleus cell rate and cell micronucleus rate of radiation group ,astragalus polysaccharide group ,astragalus saponin group and astragalus flavonoids group increased significantly ,and the number of 53BP1 focus cluster in radiation group and astragalus polysaccharide group increased significantly (P<0.01). Compared with radiation group and astragalus flavonoids group ,the micronucleus cell rate ,cell micronucleus rate and the number of 53BP1 focus cluster (continued intervention for 0.5,2,12 h)in the astragalus polysaccharide group and astragalus saponin group were significantly reduced,and the micronucleus cell rate and cell micronucleus rate in the astragalus polysaccharide group were significantly lower than astragalus saponin group (P<0.05). 53BP1 focus cluster could not be detected 24 h later (P<0.05). CONCLUSIONS : Astragalus polysaccharide and astragalus saponin both have protective effects on BMSCs DNA damage induced by radiation ,and the protective effect of astragalus polysaccharide is better than that of astragalus saponin ;astragalus flavonoids has no protective effect on radiation-induced DNA damage.

The Ehlers-Danlos syndrome(EDS)is a rare inherent connective tissue disorder. The prevalence of EDS in the population is estimated at one out of ten thousand to one out of a hundred thousand. The vascular EDS(vEDS) are rare among the subtypes but are the worst in prognosis. The article reports a case of vEDS admitted to the hospital. The patient was a young man complaining of a sudden onset of aphasia in right hemiparalysis and severe left abdominal pain for unknown reasons. The diagnosis was made after the genetic testing. The patient suffered from vEDS. Then, the multi-disciplinary team(MDT)made a treatment plan tailored to this young patient. The complexity in classification and delusive presentations of the EDS make the correct diagnosis very challenging. This article hopes to report this case and to share the experiences to the better understanding of this disease.
