1.Gelatin-chitosan crosslinked film in prevention of adhesion after craniotomy

Objective:To study the hemostatic and adhesion-prevention properties of gelatin-chitosan crosslinked film after craniotomy. Methods: Gelatin-chitosan crosslinked film was prepared by casting the mixed solution of dialyzed chitosan and gelatin after crosslinking with glutaraldehyde in freeze dryer. Bleeding time was recorded for bilateral back incisions (5 incisions on each side) in 4 New Zealand white rabbits. Incision on one side was treated with gelatin-chitosan crosslinked film, and on the other was treated with gauze. Hemoglobin amount in films or gauzes was measured to determine the hemostatic effect after bleeding. Bilateral craniotomies were performed at calvaria in 16 adult rabbits. A piece of gelatin-chitosan crosslinked film was put in one randomized side and the other side served as a control. The scar formation was evaluated grossly and histologically on week 2,4,8,12. 2.Association between Interleukin-8 and Apolipoprotein E Gene Polymorphism and Late-onset Alzheimer's Disease

[Objective] To investigate the relation among the polymorphism of interleukin-8 (IL-8) gene A-251T and apolipoprotein E (ApoE) gene with late-onset Alzheimer's disease (LOAD). [Methods] Polymerase chain reaction and restrietive fragment length polymorphism (PCR-RFLP) were used to detect the polymorphism distribution of IL-8 gene A-251T and ApoE gene of 185 people in Han ethnicity of Guangdong province (study group: 88 LOAD patients, eontrolled group: 97 people). The correlation of these genes was analyzed. [Results] (1) The differences of genotypes and alleles of IL-8 gene A-251T between study group and controlled group had no statistical significance. (2) The frequency of ApoE 64 allele in study group was significantly higher than that in controlled group, as risk factor of LOAD (OR=2.272, P=0.003). (3) Study group and controlled group were both divided into two groups by carrying ApoE ε4 allele or not. The differenees of IL-8 genotypes were no significant whether in carrying ApoE ε4 allele group or not (carrying ε4 group: P=0.965; no carrying ε4 group: P=0.523). [Conclusion] It is suggested that lL-8 gene A-251T polymorphism perhaps has no relation with LOAD and has no interaction with ApoE ε4 allele.
3.Single-leg Static Postural Balance in Individuals with or Without Unilateral Functional Ankle Instability

Objective To explore the static postural balance in individuals with or without functional ankle instability (FAI)by identifying and quantifying the spatial characteristics of center of pressure (COP),as well as rambling and trembling trajectory properties.Methods Twelve FAI patients was selected into the FAI group,while another 14 healthy students were chosen into the control group.The single-leg standing test with and without vision was conducted on the unstable side of the FAI group and the non-dominant side of the control group 3 trials with each trial lasting 30 s.The displacement of COP and the time and frequency domain of rambling and trembling were analyzed and compared between the two groups.Results There was no significant difference in the displacement of COP with opened eyes(P>0.05).The area and the anterior-posterior length of COP of the FAI group were larger than those of the control group with eyes closed (P<0.05).For the rambling and trembling trajectory,no significant differences were found in all the time domains between the two groups with eyes open or closed(P>0.05),except the mean sway amplitude(MSA)(P<0.05).Moreover,the mean square root and standard deviation of MSA of the FAI group were significantly bigger than the control group(P< 0.05).Significant differences were found in the MSA frequency between open and closed eyes(P>0.05),but not between the FAI and control groups(P<0.05).Conclusions The FAI patients are weaker than healthy people not only in the peripheral control,but also in the central control of the static postural balance.Therefore,injuries in the central neural system must also be considered when analyzing the reason for FAI.The imbalance of the FAI patients can be avoided by opening eyes.
4.A study on quantification assessment of segmental volume after thigh liposuction with three-dimensional scanning technology

Objective:To investigate the quantification assessment of segmental volume after thigh liposuction utilizing three-dimensional(3D) scanning technology.Methods:This retrospective study was performed with the analysis of 3D scanning images of patients who had undergone bilateral thigh liposuction in Body Contouring & Fat Grafting Center, Plastic Surgery Hospital, Chinese Academy of Medical Sciences from January 2018 to September 2022. Preoperative and postoperative 3D scanning were performed to build visual 3D models of bilateral thighs. From top to the bottom, bilateral thighs were segmented into seven sections with a space of 5 cm in acquired 3D model. Certain measurements and calculation of preoperative and postoperative volume and volume change rate of the overall thigh and each segment were conducted, to validate the efficiency of liposuction (EOL). Additionally, EOL of each thigh segment was computed and the symmetry of bilateral thighs was analyzed before and after surgery. The volume differences were computed using the paired Wilcoxon rank-sum test, and intraclass correlation coefficient (ICC) was exerted to assess the symmetry of bilateral thighs before and after surgery.Results:A total of 36 female patients were included in the study, with an age range of 18 to 49 years and a mean age of (28.6±7.6) years. Follow-up duration ranged from 23 to 1 133 days postoperatively, with a mean follow-up period of 274.7 days. The results of 3D scanning measurements indicated significant changes ( P<0.01) in both the overall volume of the thigh and the volumes of each segment before and after surgery. The highest volume change rate and EOL were observed in the uppermost segment, and the volumetric change rate and EOL exhibited a descending trend across the segments of the thigh from the uppermost to the lowermost segments. The ICC of the volume of each segment consistently surpassed 0.950 whether preoperatively or postoperatively, indicating a high level of symmetry between the bilateral thighs, and the ICC of overall volume showed a notable increase from 0.992 preoperatively to 0.997 postoperatively. Conclusion:3D scanning technology can be exerted to quantify the volume changes before and after thigh liposuction. This study provided quantitative and objective evidence to confirm the efficacy of thigh liposuction procedure, elucidating that the most significant liposuction effects observed in the uppermost segment of the thigh. Moreover, postoperative assessments reveal a further enhancement in bilateral thigh symmetry.

6.Construction and Application of National Rare Diseases Registry System of China

Rare diseases have been a major challenge for clinical medicine and public health challenge in China. One of the effective measures is to conduct proactive research on rare diseases to deal with the disease burden of the diseases. However, low prevalence, disperse distribution of patients, lack of knowledge about the disease course, and phenotype heterogeneity hamper the development of research for rare diseases. Recently, it has been found that patients registry is effective in understanding the course of the disease and accu- mulating the cases and data of clinical research or clinical trial design. At present, most of developed countries or regions in the world have promoted clinical research and clinical trials of new medications on rare diseases by using the registration of rare disease. In 2016, Peking Union Medical College Hospital established China's first registry system at the national level-National Rare Disease Registry System of China(NRDRS). NRDRS has accumulated 68 137 cases data registered by the researchers from China's 101 collaborating hospitals in 29 provinces/municipalities/autonomous regions, covering 171 different, and forming 188 cohorts. To date, NRDRS complete the initial stage of resources buildup.Nex stage will be focused on clinical research and clinical trials related to rare diseases based on NRDRS. This article is on the process of building NRDRS, the potential support for conducting clinical research and clinical trials related to rare diseases, and the challenges will be faced.
