1.Effects of methylflavonolamine on arachidonic acid metabolism pathway of platelet

Effects of methylflavonolamine (MFA) on arachidonic acid(AA) metabolism pathway were studied. MFA (iv 40 mg ? kg-1) lightened the acute pulmonary thromboenblism signs in mice and reduced the mortality induced by AA. MFA(12. 5~200?mol ? L-1) in vitro dose-dependently inhibited rabbit platelet aggregation induced by AA. MFA(0.1~0. 4mmol ? L-1) in vitro dose - dependently inhibited rabbit platelet malondiadehyde(MDA) formation in-duced by AA. MFA(0. 4mmol?L-1) inhibited platelet MDA formation in rabbits induced by thrombin and AA. While propranolol inhibited MDA formation induced by thrombin but not by AA. MFA (0.4 mmol ? L-1) did not affect cAMP content in rabbit platelet. These results suggest that inhibitory effect of MFA on platelet AA metabolism may be one of the mechanism by which MFA inhibited platelet aggregation.
2.Intravitreal injection of Conbercept combined with PPV in the treatment of proliferative diabetic retinopathy

AIM:To observe the value of intravitreal injection of Conbercept in the treatment of proliferative diabetic retinopathy(PDR).
METHODS: Totally 64 patients(75 eyes)with PDR admitted to the hospital between January 2016 and October 2019 were recruited and divided into observation group(32 cases, 38 eyes)and control group(32 cases, 37 eyes)by random number table method. The observation group received vitrectomy and intravitreal injection of Conbercept, while the control group received simple vitrectomy. The best corrected visual acuity(BCVA)was detected before treatment, at 1wk, 1mo, and 3mo after operation. Changes in central macular thickness(CMT)before treatment and at 3mo after operation were determined. Aqueous humor or vitreous humor samples were collected before and after treatment to measure concentrations of vascular endothelial growth factor(VEGF), placental growth factor(PIGF)and basic fibroblast growth factor(bFGF)by enzyme-linked immunosorbent assay. The operation time, intraoperative blood loss and improvement time of symptoms(retinal edema, fundus hemorrhage, exudation)of the 2 groups were statistically analyzed. Incidences of complications in the 2 groups were recorded.
RESULTS: The observation group had better BCVA than the control group at 1wk, 1mo and 3mo after operation(P<0.05), and had smaller CMT than the control group at 3mo after operation(P<0.001). The concentrations of VEGF, PIGF and bFGF in observation group during operation were lower than those in the control group(P<0.001). The operation time, absorption time of retinal edema, fundus hemorrhage and exudation, intraoperative blood loss, and the total incidence of complications in the observation group were shorter and lower than those in the control group(P<0.05).
CONCLUSION: Intravitreal injection of Conbercept before vitrectomy can degrade new blood vessels, and shorten the operation time. It can not only help improve vision and relieve macular edema but also reduce surgical complications and promote postoperative recovery.
3.Design and validation of synchronous exposure control system of dental cone beam CT.

In the design of oral cone beam CT, cooperation between synergic control of X-ray source, real-time acquisition of flat detector and motion of mechanical structure affects the CT image quality. Based on the full analysis of the flat detector's timing signal characteristics, this research was carried out with microprocessor controller (MCU), complex programmable logic device (CPLD), and light couplings to design and realize synchronous exposure control system. To evaluate whether the design of the synchronous exposure control system in this project could reach the required imaging accuracy, we employed the projected images in the system to analyze its stability, linear consistency, signal to noise ratio and precede the FDK construction.

4.Evaluation of application effect of evidence- based nursing in reducing ventilator - associated pneumonia of patients with severe head injury

Objective To evaluate the application effect of evidence-based nursing in reducing ventilator-associated pneumonia (VAP) of patients with severe head injury. Methods 100 patients with severe head injury using mechanical ventilation were randomly divided into the control group and the observation group with 50 patients in each group.The observation group explored evidence from nursing of ventilator tube,oropharyngeal and sound sects care,airway care,balloon sleeves care to prevent biofilm on the endotracheal tube(ETT-BF) to fall off,enteral nutrition care,decubitus care,ward environment and the hand disinfection of medical staff and was given evidence- based nursing.The control group took routine care.The incidence of VAP,off-line time and mortality rate of the two groups were compared during the same time period. Results The incidence of VAP,off-line time,mortality rate showed significant difference between the two groups.The observation group showed better effect. Conclusions Implementation of evidence-based nursing can significantly reduce the off-line time of patients with severe traumatic brain injury using mechanical ventilation.It can decrease the incidence of VAP and mortality rate,improve the prognosis and reduce medical cost.
5.Study on Pathology and Neurotransmitters of Esophageal Sphincter after Radiofrequency Thermocoagulation in Dogs

Objective To investigate the effects of radiofrequency thermocoagulation (RFT) on pathological features and the expressions of choline acetyltransferase (ChAT), vasoactive intestinal peptide (VIP) and nitric oxide synthase (NOS) at lower esophageal sphincter (LES) in family dogs. Methods A total of 15 dogs were randomly divided into three groups. Sham group underwent gastroscopy and was fed for 3 months (n=5). Dogs were given RFT and were fed for 24 h after RFT (n=5, RFT+24 h group). Dogs were given RFT and were fed for 3 months after RFT (n=5, RFT+3m group). The pathological changes of LES were observed after HE staining in three groups. The expressions of ChAT, VIP and NOS were detected by immunohistochemical method in three groups. Results Results of HE staining showed nearly the same tissues in Sham group and control group. There were active inflammatory reaction and structural damage in RFT+24 h group. The chronic in-flammatory reaction and structural remodeling were found in RFT+3m group. Immunohistochemistry showed that ChAT was significantly increased in RFT+3m group compare than that of Sham group. Values of VIP and NOS were significantly de-creased in RFT+3m group compare than that of Sham group (P<0.01). Conclusion The thickness and increased pressure of LES were found after RFT,which also caused changes in neurotransmitters of local tissues in dogs.
6.Preliminary study on identification of algicidal active substance from mangrove bacteria Flavobacterium sp. and its algicidal effect on red tide algae Alexandrium tamarense

Objective To study on algicidal bioactive substances from mangrove bacteria and its algicidal effect on red tide algae Alexandriam tamarense.Methods A red algicidal bioactive substance in Guangxi mangrove bacteria was isolated and purified by Silica-gel Column Chromatography,and its structure was determined on the basis of UV,IR,1H NMR,13C NMR and HREIMS spectroscopic analyses and comparison with the literatures.Its algicidal affect was primarily studied in this paper.Results and Conelusion The results showed that the red algicidal bioactive substance was prodigiosin,which had an inhibitory effect on the cells growth of Alexandrium tamarense,and the inhibitory effect increased with the increase of prodigiosin concentration.The prodigiosin stimulated the production of MDA and reduced the content of sulfhydryl group.In addition,plasma membrane permeability of leaked cell solution was increased.These indicated the extinguishing mechanism: the cell membrane was damaged followed by the inhibition of the cell growth.

Effects of thioproline on thrombus formation and platelet function were studied. Thioprolline given in vitro ( 1 g/L ) or ex vivo ( iv 25mg /kg ) shortened length of thrombi and decresed the wet and dry weight of thrombi in rabbits or rats. Thioproline given iv vitro ( 1 g/L ) or ex vivo ( iv 50mg/kg ) inhibited platelet adhesion and aggregation induced by ADP in rabbits.
8. Influence analysis of temperature-holding nursing in the anesthesia and stress state during the recovery period of general anesthesia

Objective:
To discuss the influence of temperature-holding nursing in the anesthesia and stress state during the recovery period of general anesthesia for patients with thoracoscopic lung surgery.
Methods:
120 patients with thoracoscopic lung surgery underwent the general anesthesia from January 2017 to July 2018 in our hospital were selected and randomly assigned to two groups, 60 cases in each group. At the recovery period, the control group was treated with conventional nursing; the observation group was treated with conventional nursing and temperature-holding nursing. At each time period, the body temperature, stress response and postoperative rehabilitation conditions were probed.
Results:
At the end of the operation 30 minutes, 60 minutes and the end of the operation, the body temperature of the observation group was (36.39±0.34)°C, (36.50±0.38)°C, (36.56±0.38)°C, and the control group was (35.49±0.31)°C, (35.63±0.41) °C, (36.17±0.52)°C, the difference between the two groups was statistically significant (
9.The effect of TNF-alpha and G-CSF on the intracoronary transplantation rate of rabbit mesenchymal stem cells in vivo.

To investigate effects of tumor necrosis factor-alpha(TNF-alpha) and granulocyte-colony stimulation factor(G-CSF) on the transplantation rate of rabbit mesenchymal stem cells (MSCs) through intracoronary injection. Japanese white ears rabbits (2-3 months old) were used as myocardial injury models by intravenously injecting adriamycin, then they were divided into three groups randomly: (1) Blank group, (2) TNF-alpha group; (3) G-CSF group. There were 7 rabbits each group. Before transplantation, MSCs and rabbit models were neither intervened by cytokine in blank group; in TNF-alpha group, rabbits were injected TNF-alpha (5 microg/kg) through ear-edge vein one day before transplantation, MSCs were pretreated by TNF-alpha (10ng/ml)12 hours before transplantation. In G-CSF group, rabbits were injected G-CSF(10 microg/kg) intramuscular each day 3 days before transplantation. MSCs were pretreated by G-CSF(10 ng/ ml) 12 hours before transplantation. Separated by density gradient centrifugation and labeled by 4,6-biamidine-2-phenyliodole (DAPI) after cells expanded in vitro, MSCs were then transplanted through the root of aorta when the ascending aorta occluded above the sinus aortae. Myocardial pathological sections were made after 4 weeks and count transplanted cells under laser scanning confocal microscope and statiscally analyze. The amount of cells transplanted in rabbit myocardial were much more in TNF-alpha group G-CSF group than in blank group, there was statistically significant difference (P < 0.001). It sugested that TNF-alpha and G-CSF intervened the cultured MSCs, and animal model of myocardial injury could elevate the transplantation rate of intracoronary injection rabbit mesenchymal stem cells. The mechanism may have relation to that cytokine can enhance the attachment of MSCs to cardiac microvascular endothelium (CMVE).

10.Effects of cyclosporin A and tacrolimus on biological behaviors of lung cancer A549 cells in nude mice

Objective To explore the effects of cyclosporin A (CsA) and tacrolimus (Tac) on biological behaviors of lung cancer A549 cells in nude mice.Methods Thirty-six models of transplanted tumor in Balb/c mice were established by using lung cancer A549 cells and divided into three groups:control group,without given any immunosuppressant; CsA group,intraperitoneally given CsA; Tac group,intraperitoneally given Tac.The transplanted tumor growth curve was drawn according to the transplanted tumor volume,and the influencing ratio was calculated according to the final tumor weight.The changes in cell migration ability were observed by using Transwell system.Terminal deoxynucleotidyl transferase mediated UTP nick end labeling (TUNEL) assay was used to examine the apoptosis index of the transplanted tumor.Quantitative RT-PCR was used to detect the expression levels of Bcl-2 mRNA and Bax mRNA.Results The growth of transplanted tumor in CsA and Tac groups was faster than in control group.Final tumor volume and final tumor weight in CsA and Tac groups were greater than those in control group.The influencing ratio in CsA and Tac groups was 19% (P<0.05) and 25% (P<0.05),respectively.The migration ability was greater in CsA and Tac groups than in control group (P<0.01).The apoptosis index of the transplanted tumor in CsA and Tac groups was lower than in control group (P<0.05).The expression level of Bcl-2 mRNA was higher in CsA and Tac groups than in control group (P<0.05),and that of Bax mRNA was lower in CsA and Tac groups than in control group (P<0.05).Conclusion Both CsA and Tac can promote the growth of transplanted tumor in nuce mice bearing 549 cells and enhance the invasion forces,which is probably related with the apoptosis induction of tumor cells.
