3.POTENTIATING EFFECTS OF ROYAL JELLY ON IMMUNO LOGICAL FUNCTION IN MICE

Royal jelly (2.5g/kg/d igX8d) increased the clearance rate of iv charcoal particles in normal mice, and completely antagonized the decrease of clearance rate caused by cortisone acetate. It also potentiated the delayed type hypersensitivity ( DTH ) reaction of footpad induced by SRBC in normal mice, and entirely antagonized the inhibition of DTH reaction caused by eye lophosphamide. Royal jelly (2.5 g/kg/d ig X 7 d ) had no marked influence on hemolysin formation in normal mice and immunosuppressed mice caused by eye lophosphamide.
4.INFLUENCE OF bPTH ( 1-34 ) ON PGI_2 RELEASE FROM HEART CELLS

In order to explore the mechanism of cardiac damage by PTH, the release of PGI2 from heart cells was measured. The results showed that bPTH ( 1-34) increased the release of 6-keto-PGFla from heart cells in a dose dependent manner. Calcium inophone A23187 also increased the 6-keto-PGF1? release, while EDTA and verapamil reduced it. These suggest that the PGI2 synthesis in heart cells was affected by intra and extracellular calcium. The significance of bPTH( 1-34) induced increase of PGI2 synthesis might be associated with interference of energy metabolism, and then, cell damage.
5.The effect of ( - )Daurisoline on cytoplasmic free calcium concentration of rabbit lymphocytes

This paper reported the estimation of free calcium concentration in rabbit resting lymphocytes, and the effect of (-) Daurisoline (8701) on the cyto-plasmic free calcium concentration by using fluorescent indicator Fura -2/Am estimating technique. Results were compared with the effect of the Ca2-ionophore A23187.The results indicated that the free calcium concentration of resting lymphocytes was 109. 6nmol ? L-1, after 10-5 mol? L-1,10-4.5 mol ? L-1,and 10-4 mol ? L-1-)Daurisoline administration, the cyto-plasmic free calcium concentration increased to 178. 8%, 204. 4%and 817. 5% respectively, while same concentration A23187 only led the increase of 164. 6% , 198.4% and 327% respectively.Results suggested that ( - )Daurisoline may be a Ca2- agonist and may induce the cytotoxicity effect on lymphocytes at high concentr ation.
6.Percutaneous Endoscopic Lumber Discectomy Versus Minimally Invasive Transforaminal Lumbar Interbody Fusion for Lumbar Disc Herniation:a Prospective Randomized Controlled Study

Objective To compare clinical outcomes of percutaneous endoscopic lumber discectomy ( PELD ) versus minimally invasive transforaminal lumbar interbody fusion ( Mis-TLIF ) under the Quadrant system in the treatment of lumbar disc herniation. Methods From January 2010 to December 2013, 60 patients with lumbar disc herniation and failed to conservative treatment were enrolled in this study.According to the random number table, the patients were divided into two groups, with 30 patients in each group.There were no significant differences in age, gender, segment of disease, clinical diagnosis, and imaging results between the two groups (P >0.05).The surgery of PELD under the TESSYS endoscopic system or Mis-TLIF under the Quadrant system was performed by a same surgical team.Follow-up was conducted for 12-24 months (mean, 16.2 months).The operation time, blood loss, hospital stay, visual analogue scores (VAS) for back and leg pain, Oswestry disability index (ODI), Japanese Orthopaedic Association ( JOA ) scores, MacNab criteria, and complications were compared between the two groups. Results As compared with the Mis-TLIF group, the PELD group had significantly shorter operation time [(72.0 ±18.7) min vs. (137.0 ±48.3) min, t=-6.857, P=0.000], less intraoperative blood loss [(28.0 ±14.7) ml vs.(314.0 ±13.6) ml, t=-11.831, P=0.000], and shorter hospitalization stay [(4.0 ±1.0) d vs.(10.0 ±3.0) d, t=-9.298, P=0.000].The scores of VAS of back and leg pain at 2 weeks and 3 months postoperation in the PELD group was lower than the Mis-TLIF group (P<0.05). The scores of ODI and JOA at 3 and 12 months postoperation in the PELD group were superior to the Mis-TLIF group (P<0.05).No significant difference was recorded in the MacNab criteria at 12 months postoperation between the two groups (P>0.05).There wasno significant difference in complication rate between the two groups. Conclusions Both PELD and Mis-TLIF are safe and effective for lumbar disc herniation.PELD has smaller incision, shorter operation time, and less blood loss as compared with Mis-TLIF.
7.Hepatitis B virus (HBV) S gene-specific antisense locked nucleic acid (LNA) significantly inhibits HBV replication in vitro

Objective To investigate the inhibitory effects of hepatitis B virus (HBV) S gene-specific antisense locked nucleic acid (LNA) on HBV replication and expression in HepG_22.2.15 cells,and to screen the effective short sequence of LNA.Methods Four different lengths of short sequence of antisense locked nucleic acid which were complementary to the initiator of HBV S gene were designed,synthesized and transfected by cationic liposomes into HepG_22.2.15 cells.The HBsAg and HBV DNA of supematant was tested by enzyme linked immunoadsorbent assay(ELISA) and real-time fluorescent quantitative PCR(FQ-PCR) at 24,48 and 72 hours after treatment.LNA's cyto-toxicity on cell was evaluated by MTT method.Results Four different lengths of short sequence of LNA inhibi-ted the expression of HBsAg and the replication of HBV DNA with the inhibition rates of 46.58%,54.38%,72.43% ,69.92% and 27.09% ,28.77% ,34.71% ,32.68% respectively after 72 hours.There's no obvious tox-icity on cell.Conclusion Antisense LNA that targeting at HBV S gene has strong inhibition on HBV in vitro,and the optimal length of LNA sequence might be in the range of 15 base to 25 base.It has a therapeutic potential in the treatment of patients infected with HBV.
8.Establishment of a mouse model of cutaneous protothecosis

Objective To develop a mouse model of cutaneous protothecosis.Methods Totally,48 BALB/c mice were randomly and equally divided into four groups:high-and low-concentration immunosuppressive groups-immunosuppressed mice inoculated with P.zopfii var.portoricensis suspension of 1 × 109 and 1 × 106 colony forming units (CFU) conidia/ml respectively,high-concentration healthy group-healthy mice inoculated with P.zopfii var.portoricensis suspension of 1 × 109 CFU conidia/ml,and control group-healthy mice inoculatedwith sodium chloride physiological solution.The P.zopfii suspension or sodium chloride physiological solution was subcutaneously inoculated to the abdominal skin of mice,with the inoculation volume being 200 μl.Skin appearance at the inoculation site was observed,and four mice were sacrificed in each group on day 7,14 and 28 after the inoculation.Skin specimens were resected from the inoculation sites of mice and subjected to pathological and mycological examinations.Results Mice in the three experiment groups inoculated with the P.zopfi suspension were all infected,with the appearance of papules and abscess at the inoculation sites of all mice as well as ulcer and crusts in some mice.Meanwhile,no mice were infected in the control group.Significant differences were noted in the diameter of skin lesions between the three time points in these experiment groups (all P < 0.05),and the largest diameter of lesions was observed on day 7,which was (6.75 ± 1.09) mm in the high-concentration immunosuppressive group,(5.88 ± 1.17) mm in the low-concentration immunosuppressive group,and (5.96 ± 0.99) mm in the high-concentration healthy group.Comparisons of lesion diameter between the three experiment groups revealed a statistical difference on day 28 (F =8.91,P < 0.05),with the largest lesion diameter observed in the high-concentration immunosuppressive group (4.38 ± 0.86 mm),but no statistical difference was found on day 7 or 14 (both P > 0.05).Pathology of skin specimens consistently revealed necrosis,abscess and granuloma formation in the three experiment groups.Spores were found in the lesions of infected mice by haematoxylin-eosin staining,periodic acid-Schiff staining,and direct microscopy.Culture of tissue samples from the experiment groups grew Prototheca.Conclusion The mouse model of protothecosis can be established by subcutaneous inoculation of Prototheca suspension in the abdominal skin of healthy or immunosuppressed mice.

10.The protective effect of exercise on osteopenia in ovariectomized rats

Objective To investigate the protective effect of exercise on osteopenia female rats caused by ovariectomy and its mechanism. Methods Twenty-four female rats aged 6 months were randomly divided into 3 groups: control group(CON),ovariectomized model group(OVX) and exercise group(Ex).The ovariectomy operation was performed with rats in OVX and Ex groups. Rats in Ex group were trained on a treadmill at a velocity of 18m/min for 1h, once daily for 5 days a week. Each group of rats were sacrificed at 12 weeks postoperatively and Dual-energy X-ray absorptiometry was carried out to measure the bone mineral density(BMD) of the femur. IGF-1 proteins in trabecular bone of proximal femur were examined by using immunochemical stain and the intensity of IGF-1 was determined by use of imaging analysis techniques. The apoptotic osteocytes in the trabecular bone of proximal femur were detected by using terminal-deoxynucleotidyl mediated nick end labeling (TUNEL) technique. Results The BMD of CON, Ex and OVX group was (0.170?0.011)g/cm2,(0.154?0.013)g/cm2 and(0.167?0.013)g/cm2, respectively. The difference of the BMD level between Ex and CON groups was not significant, however, the BMD level in OVX group was significantly lower than that of the Ex and CON groups (P
